Effects of a botulinum toxin type A injection on the masseter muscle: An animal model study

  • Background: The aim of this study was to investigate the effect of a botulinum toxin type A (BTX-A) injection in the masseter muscle using electromyography (EMG) in an animal model. Methods: Ten male adult (>3 months of age) New Zealand white rabbits were used. Muscle activity was continuously recorded from 8 hours before to 8 hours after BTX-A injection. The rabbits received unilateral BTX-A injections of either 5 units (group 1, n = 5) or 20 units (group 2, n = 5). Results: The masseter muscle activity of the rabbits was significantly reduced immediately after BTX-A injection (P < 0.05 for both groups). When the results from group 1 were compared with those from group 2, only the peak voltage was significantly decreased in group 2 (P = 0.013). Conclusion: Masseter muscle activity measured by EMG was immediately decreased after a BTX-A injection.

  • LB00013, a newly developed recombinant human Interferon $\beta$, was tested for primary eye and skin irritation in male New Zealand White rabbits. In the primary eye irritation test, 0.1 ml of a solution of LB00013 was instilled into the eye. In the rinsing group, the eye was washed with water at 30 seconds after instillation. No reaction was produced at the cornea, iris and conjunctivae by LB00013. In the primary skin irritation test, LB00013 was applied to the back of rabbits for 24 hours. Primary irritation index was "0" in test and control sites of all animals. Thus LB00013 was evaluated as a non-irritant on the basis of the criteria of Draize et al. (1944).l. (1944).

The influence of leukocyte-platelet-rich plasma on accelerated orthodontic tooth movement in rabbits

  • Objective: To determine the effects of a local injection of leukocyte-platelet-rich plasma (L-PRP) on orthodontic tooth movement in rabbits. Methods: Twenty-three male New Zealand white rabbits were included in a split-mouth design. Tooth movement with a 100-g nickel-titanium closed-coil spring was performed on the maxillary first premolars. L-PRP was injected submucosally at the buccal and lingual areas of the first premolar in one random side of the maxilla and the other side served as the control and received normal saline. The amount of tooth movement was assessed on three-dimensional digital models on days 0, 3, 7, 14, 21, and 28. Histological findings and osteoclast numbers were examined on day 0 as the baseline and on days 7, 14, and 28. Results: The L-PRP group showed significantly greater cumulative tooth movement at all observed periods. However, a significantly higher rate of tooth movement was observed only on days 0-7 and 7-14. The osteoclast numbers were significantly increased in the L-PRP group on days 7 and 14. Conclusions: Local injection of L-PRP resulted in a transient increase in the rate of tooth movement and higher osteoclast numbers.

  • The acute dermal and eye irritation tests of Photorhabdus temperata, a symbiotic bacterium of Heterorhabditis megidis Gwangju strain, were carried out in New Zealand white rabbit (Oryctolagus cuniculus), following the guidelines of OECD and Rural Development Administration (RDA) of Korea. In both tests, neither dermal nor eye responses were found from all the P. temperata treated rabbits and the results were classified as non-irritating. That is, erythema, eschar, edema, and any other dermal critical signs were not observed from all the experimental rabbits in the dermal irritation test. In the acute eye irritation test, no clinical signs of cornea, iritis, conjunctiva (redness, edema, lacrima, and chemosis) were observed from all the experimental rabbits. Individual ocular irritation, mean ocular irritation, and acute ocular irritation were calculated as 0.0. The results of dermal and eye irritation studies on P. temperata indicated that this bacterium could be a safe and effective alternative bionematicide against the most serious plant-parasitic root-knot nematodes in the genus Meloidogyne.

Antioxidant Effects of Ascorbic Acid on Renal-Ischemia Reperfusion Injury in Rabbit Model

  • Renal ischemia-reperfusion (I/R) injury is great clinical important because viability of the organ depends on the tolerance to ischemia-reperfusion injury, an inevitable processing during surgery. The purpose of this study was to investigate the effects of premedicated ascorbic acid alone in I/R injury model induced by cross-clamping of renal vessels. In the rabbit models, 2-4 kg New Zealand white rabbits were subjected to 30 minutes of warm unilateral renal ischemia followed by removal of contralateral kidney and then divided into five groups, control (2) arid treatment groups (3). In control group 1, the rabbits only received right nephrectomy. In control group 2, the rabbits received I/R on left kidney after the right nephrectomy. In treatment group 1, the rabbits received ascorbic acid 50 mg/kg IV before the operation. In treatment group 2, the rabbits received ascorbic acid 100 mg/kg IV before the operation. In treatment group 3, the rabbits received ascorbic acid 200 mg/kg IV before the operation. Blood samples were collected from these rabbits for measurement of kidney function tests at the 0, 1 st, 3rd and 7th day and antioxidant enzyme( SOD, GSHPx, CAT) at 24 hours. Kidney function tests (serum creatinine and BUN) showed a significant difference between group 2 and group 4, 5. Activity of antioxidant enzymes in plasma were significant decrease in group 4, 5 compare to group 2. The result of this study suggested that the exogenous ascorbic acid had a role of attenuation of renal I/R injury in rabbit model.

Effects of grape pomace on the antioxidant defense system in diet-induced hypercholesterolemic rabbits

  • The effects of grape seeds extract and grape peels extract prepared from grape pomace on the activity of antioxidant enzymes, degree of lipid peroxidation in serum and liver tissue were investigated in rabbits fed on high cholesterol diet. New Zealand white rabbits were divided as follows ; 1) NOR (normal group); 2) CHOL (cholesterol group); 3) GSH (cholesterol + grape seed extract group); 4) GPE (cholesterol + grape peel extract); 5) GSP (cholesterol + grape seed powder); 6) GPP (cholesterol + grape peel powder); 7) GE (cholesterol + grape seed and peel extract); 8) GP (cholesterol + grape seed and peel powder). Eight groups of rabbits were studied for 8 weeks. At the end of the experimental period, rabbits were sacrificed and the liver tissue were removed. Then, GSH, GPx, GST, CAT and MDA in the liver were measured. In liver tissues, total glutathione contents (GSH), glutathione peroxidase (GPx) and catalase (CAT) activity, which was significantly higher by grape seed extract supplementation. The level of malondialdehyde (MDA) was lower in the serum of rabbits fed grape seed extract or grape peel powder plus cholesterol than in the serum of rabbits fed cholesterol alone. It is therefore likely that grape seed extract prepared from grape pomace functioned as antioxidants in vivo, negating the effects of the oxidative stress induced by 1% cholesterol diet. The grape seed extract was found effective in converting the oxidized glutathione into reduced glutathione, and in removing $H_2O_2$ that is created by oxidative stress. The grape peel powder was found to have small influence on reduced glutathione content, CAT and GPX activity, but it increased GST activity in liver tissues, resulting in promoting the combination of lipid peroxide and glutathione (GSH), and further, lowering the formation of lipid peroxide in the serum. Therefore, grape pomace (grape seed extract and grape peel powder) supplementation is considered to activate the antioxidant enzyme system and prevent damage with hypercholesterolemia.

Orally administered Lactobacillus casei exhibited several probiotic properties in artificially suckling rabbits

  • Objective: Lactobacilli in rabbit intestine is rare and its function in rabbit gut health is not fully understood. The present study aimed to evaluate in vivo the probiotic potential of Lactobacillus casei for suckling rabbits. Methods: Two healthy 5-day-old suckling rabbits with similar weights from each of 12 New Zealand White litters were selected and disturbed to control group and treatment group. All rabbits were artificially fed. The treatment group had been supplemented with live Lactobacillus casei in the milk from the beginning of the trial to 13 days of age. At 15 days of age, healthy paired rabbits were slaughtered to collect intestinal samples. Results: i) Oral administration of Lactobacillus casei significantly increased the proportion of Lactobacilli in the total intestinal bacteria (p<0.01) and obviously reduced that of Escherichia-Shigella (p<0.01); ii) treatment increased the length of vermiform appendix (p<0.05); iii) a higher percentage of degranulated paneth cells was observed in the duodenum and jejunum when rabbits administered with Lactobacillus casei (p<0.01); and iv) the expression of toll-like receptor 9, lysozyme (LYZ), and defensin-7-like (DEFEN) in the duodenum and jejunum was stimulated by supplemented Lactobacillus casei (p<0.05). Conclusion: Orally administered Lactobacillus casei could increase the abundance of intestinal Lactobacilli, decrease the relative abundance of intestinal Escherichia-Shigella, promote the growth of appendix vermiform, stimulate the degranulation of paneth cells and induce the expression of DEFEN and LYS. The results of the present study implied that Lactobacillus casei exhibited probiotic potential for suckling rabbits.

토끼에서 레몬그라스(Cymbopogon citratus) 정유의 일차피부자극시험 (Primary Dermal Irritation Study of Lemon Grass (Cymbopogon citratus) Essential Oil in Rabbits)

  • The present study was carried out to investigate the skin irritation potential of lemon grass essential oil in rabbits. A volume of 0.5 ml of test article was applied to intact and abraded skins, respectively, for 24 h in 6 healthy male New Zealand White rabbits. Parameters measured during 72 h observation period were mortality, clinical signs, body weight changes, and local irritation. All rabbits showed severe edema at both 24 h and 72 h after the application of test article. These animals also showed very slight to severe erythema and/or slight eschar formation at both 24 h and 72 h. The edema and erythema induced by lemon grass essential oil were recovered on day 7 of additional 11-day recovery period, but the eschar formation was not recovered at the end of recovery period. On the other hands, there were no treatment-related adverse effects on clinical sign, body weight and gross finding in rabbits. Based on these results, it was concluded that a single dermal application of the undiluted lemon grass essential oil caused very slight to severe edema, erythema and eschar formation in rabbits and showed a primary irritation index score of 5.63 indicating severe irritation. The results of this study strongly suggest that lemon grass essential oil should be used with care and in highly diluted forms especially when directly applied to the skin.

토끼의 비골 골절에서 산화구리(CuO, copper oxide)의 골재성 효과 (Bone Regeneration Effects of Copper Oxide on Fibular Fracture in Rabbits)

  • The bone regeneration effects of copper oxide on experimentally induced fibular fracture were examined in 36 New Zealand white male rabbits. They were divided into two groups: non-treated group (control group) and copper oxide treated group (treatment group). A fibular fracture was created by an osteotomy in the middle of the fibula and 62.5 mg/kg of copper oxide was orally administrated during 7 days after operation in the treatment group. Radiological findings, histopathological examinations and hematoserological findings were observed to evaluate the bone regeneration effects of copper oxide on fibula fracture during 9 weeks. In radiological findings, the area of bone regeneration at the fracture site of the treatment group was significantly wider from 3 weeks to 6 weeks after administration of copper oxide than those of the control group (p < 0.05). In histopathological examinations, fracture healing in treatment group was faster than in control group. Also, histopathological responses of thick bony trabeculae and new bone marrow formation were shown in the treatment group, whereas many fibrous tissues and cartilages were mainly observed in the control group. No specific effects of copper oxide on the body was found in hematological and serological test during experimental period. These results showed that the copper oxide had a potential therapeutic application in the treatment of fracture and bone trauma.

랫드 및 토끼에 대한 치자 황색색소에 관한 단회 투여 경구 독성 시험 (Single-Dose Oral Toxicity of the Gardenia Yellow Pigment in Rate and Rabbits)

  • 치자에서 분리한 황색색소에 대한 랫드 및 토끼에 있어서의 단회투여 경구 독성시험을 수행하였다. 황색색소는 랫드의 경우 5,000, 2,500, 1,250, 625, 312,5 및 1mg/kg의 용량으로 경구로, 토끼에는 5,000, 2,500 및 1,250mg/kg의 용량으로 경구로 암.수 각각에 1회 투여한 후 14일 동안 관찰하였다. 시험결과에 있어 랫드 및 토끼의 모든 황색색소 투여군에서 사망예, 임상증상, 체중변화, 육안적 소견 및 병리조직학적 소견에서 특기할만한 이상은 관찰되지 않았으며 랫드와 토끼에서 황색색소의 LD50치를 산출할 수 없었다. 따라서 LD50치는 랫드 암.수 모두에서 체중 kg당 5,000mg 이상인 것으로 판단되었다. 이상의 결과로부터 치자로부터 분리한 황색색소는 랫드 및 토끼에서 단회 투여 경구 독성시험에서 어떠한 독성 및 부작용을 유발하지 않는 안전한 제제로 사료된다.
