A New Record of Neosartorya aureola Isolated from Field Soil in Korea

  • A new species of Neosartorya was recovered during investigation of the fungal community in soil samples collected from different locations in Korea; Neosartorya aureola KNU14-7 was isolated for the first time from field soil in Korea and identified based on the internal transcribed spacer region of rDNA and morphological characteristics. The species has not been officially reported from Korea and we report it here with description and figures.

  • Collections of Gymnopus sect. Levipedes from the Republic of Korea have been studied. Two new species, Gymnopus dryophiloides and G. brunneodiscus, are described based on their macro- and micromorphological and phylogenetic characteristics. Three other species, referred to as Gymnopus spp. 1, 2, and 3, are distinguished as separate taxa without formal descriptions. Taxonomic and phylogenetic positions of all taxa have been inferred and confirmed by analyses of ITS and LSU sequence data. Their detailed descriptions, illustrations and an identification key are provided.

  • The herb Pedicularis cheilanthifolia var. albida (Pennell) P. C. Tsoong is reported and collected for the first time from the Rudraprayag District of Uttarakhand. The species was seen growing along a glacial stream in an alpine region between 4,100 and 4,400 m asl. Morphological characteristics of the species were examined and processed for herbarium deposition. To simplify taxon identification, a brief taxonomic description and illustration of the taxon are provided.

북미산 주요 전나무속 수종 목재에 있어서의 결정 (Crystals in North American Commercial Woods of Abies Species)

  • 침엽수재에 있어서는 결정을 지니는 세포가 드물게 존재하기 때문에 전나무속, 가문비나무속, 은행나무속 및 소나무속과 같은 경우 이들의 존재 여부는 수종 식별 인자로 귀중한 가치를 지니게 된다. 따라서 본 연구는 광학현미경과 주사전자현미경을 이용하여 북미산 전나무속 수종의 목재에 존재하는 결정의 분포와 형태를 조사하여 목재 해부 및 식별에 필요한 정보를 추가적으로 제공하기 위해 실시하였다. 출현 빈도 측면에서 볼 때 다각형 결정, 신장형 결정 및 주정이 Abies concolor, A. grandis 및 A. magnifica, A. nobilis(=A. procera), A. lasiocarpa 그리고 A. amabilis의 순으로 적게 관찰되었지만 A. balsamea 및 A. fraseri에서는 그 존재가 전연 확인되지 않았다. 다른 수종과는 달리 A. lasiocarpa는 다각형 결정보다는 신장형 결정 및 주정이 더 많이 지니고 있었다. A. concolor에서는 사정, 다각형 결정, 신장형 결정 및 주정이 축방향유세포 및 방사유세포 모두에서 관찰된 반면 나머지 수종 모두에서는 다각형 결정, 신장형 결정 및 주정이 방사유세포에만 존재하였다. 사정을 지니는 방사유세포 그리고 사정, 다각형 결정, 신장형 결정과 주정을 지니는 축방향유세포는 아마 본 연구에서 최초로 밝혀진 것으로 여겨진다. 결론적으로 결정의 존재 여부는 A. nobilis (=A. procera), A. lasiocarpa, A. amabilis, A. balsamea 및 A. fraseri로부터 A. concolor, A. grandis 및 A. magnifica를 식별해 내는데 있어 중요한 가치를 지니는 것으로 밝혀졌다.

DNA 바코드를 이용한 가정간편식 제품의 원재료 모니터링 연구 (Monitoring of Raw Materials for Commercial Home Meal Replacement Products Using DNA Barcode Information)

  • 본 연구는 최근 소비가 크게 증가하고 있는 가정간편식의 원료에 대한 모니터링을 수행하였다. 다양한 유형의 가정간편식 제품을 구입하여 112개 원료의 DNA 바코드를 분석하였다. 원재료의 종을 동정하기 위하여 DNA 바코드 증폭에 주로 이용되는 미토콘드리아의 16S ribosomal RNA 유전자 부위를 증폭하는 프라이머 세트를 이용하였다. PCR 산물은 정제하여 염기서열을 분석한 후, 이를 이용하여 미국국립보건원에서 제공하는 BLAST search를 수행하였다. GenBank에 등록되어 있는 종의 염기서열과 유사도(Identity)와 매치 점수(Match score)를 비교하여 원료의 종을 판별하였다. 112개의 원료에서 24개의 종(Species)과 3개의 속(Genus)를 동정하였다. 3개의 속은 Identity의 기준이 되는 98% 이내에 해당하는 종이 다수 존재하여 속 수준에서 판별하였다. 판별 결과를 「식품의 기준 및 규격(제2019-57호)」 중 '(별표 1) 사용할 수 있는 원료 목록'에서 제시하는 사용 가능한 원료와 비교하여 국명 및 섭취 가능 여부를 판단하였으며, 등재되어 있지 않은 6개 종은 국제적으로 공인된 기구에서 어획량에 대한 정보를 확인하고, 식용 근거, 학명·이명 등을 확인하여 식용 가능 여부를 판단하였다.

Morphology and Ecological Notes on the Larvae and Pupae of Simulium (Simulium) from Korea

  • Morphological features of larvae and pupae of twelve species of Simulium (Simulium) from Korea are investigated and redescribed. Detailed descriptions of immature stages of each species are provided with photographs. For the Korean members of subgenus Simulium, combinations of the following characteristics can be used to separate them from other Korean subgenera: larval abdomen without a pair of conical-shaped ventral tubercles; postgenal cleft variously shaped, viz., miter shaped, spearhead, bullet shaped or triangular; rectal papillae 3 simple lobe or compound lobes; pupal gill of 6-16 filaments; cocoon shoe, boot or slipper shaped; and head trichomes 3 pairs (1 facial+2 frontal). Habitat information and other ecological aspects of each species are provided. Updated keys to larvae and pupae of the Korean species of the subgenus Simulium are also provided to facilitate species identification. Taxonomic treatment based on external morphologies of larvae and pupae of Korean species of the subgenus Simulium are focused and included in this study. To understand their interspecific relationships, phylogenetic analyses using multiple data from molecule, morphology, and ecology are warranted in future research. Also continued exploration for new character systems is needed to establish more stable boundaries for subgenus and species delimitations.

Identification of Aecial Host Ranges of Four Korean Gymnosporangium Species Based on the Artificial Inoculation with Teliospores Obtained from Various Forms of Telia

  • The objectives of this study were to identify the aecial host ranges of four Korean Gymnosporangium species, G. asiaticum, G. cornutum, Gjaponicum and Gyamadae, and to verify the morphological characteristics of telia as diagnostic keys to the species. Thirteen Korean Rosaceous woody species were artificially inoculated with teliospores obtained from Juniperus species. There was high specificity between telial and aecial hosts and the fungal species, providing the first experimental proof on host alternation of these rust fungi in Korea. Telia on the witches' broom and on the small galls were identified as new telial characteristics in G asiaticum and in G. yamadae, respectively. Aecial hosts of G. asiaticum and G. yamadae showed varying responses in their susceptibility and in the days required for formation and duration of spermogonia and aecia after inoculation. Four telial host species in Juniperus were confirmed for the first time in Korea, which include J. chinensis var. kaizuka, J. chinensis var. horizontalis and J. chinensis var. globosa for G. asiaticum; and J. chinensis var. kaizuka for G. yamadae.

Morphological and Molecular Characterization of Lymnaeid Snails and Their Potential Role in Transmission of Fasciola spp. in Vietnam

  • Freshwater snails of the family Lymnaeidae play an important role in the transmission of fascioliasis worldwide. In Vietnam, 2 common lymnaeid species, Lymnaea swinhoei and Lymnaea viridis, can be recognized on the basis of morphology, and a third species, Lymnaea sp., is known to exist. Recent studies have raised controversy about their role in transmission of Fasciola spp. because of confusion in identification of the snail hosts. The aim of this study is, therefore, to clarify the identities of lymnaeid snails in Vietnam by a combination of morphological and molecular approaches. The molecular analyses using the second internal transcribed spacer (ITS2) of the nuclear ribosomal DNA clearly showed that lymnaeids in Vietnam include 3 species, Austropeplea viridis (morphologically identified as L. viridis), Radix auricularia (morphologically identified as L. swinhoei) and Radix rubiginosa (morphologically identified as Lymnaea sp.). R. rubiginosa is a new record for Vietnam. Among them, only A. viridis was found to be infected with Fasciola spp. These results provide a new insight into lymnaeid snails in Vietnam. Identification of lymnaeid snails in Vietnam and their role in the liver fluke transmission should be further investigated.

Molecular Identification of Haemadipsa rjukjuana (Hirudiniformes: Haemadipsidae) in Gageo Island, Korea

  • There are 60 species of blood-feeding land leeches, 50 species belonging to the family Haemadipsidae and 10 species belonging to the family Xerobdellidae. Despite recent papers on the land leeches, their taxonomic identification is not fully understood, especially at a species level. In Korea, there have been no historical records of the terrestrial leeches, but recently an unrecorded blood-feeding land leech was discovered at Gageo-do (Island), Korea. Molecular analysis was used to identify the species of 29 leeches collected from Mt. Dock-Sil in Gageo-do. Conventional PCR was conducted using nuclear 18S rRNA and mitochondrial cytochrome c oxidase subunit 1 (CO1) genetic marker. The 18S rRNA sequences revealed that the leeches share 99.9% identity with Haemadipsa rjukjuana (inhabiting Taiwan), and the CO1 sequences revealed that the leeches are very close to H. rjukjuana (inhabiting Taiwan). The CO1 sequences were separated into 2 categories, 1 with 94.6% and the other with 94.3% similarity to the H. rjukjuana L00115A (inhabiting Taiwan). This new finding of the land leech is the first record in Korea. In addition, the north range of the distribution of the blood-feeding leech (Hirudiniformes: Haemadipisidae) should be reconsidered including Korea.
