Non-viral siRNA Delivery Systems

  • The emergence of new biological drugs based on RNA interference (RNAi) technology has been one of the most attractive issues in the field of gene therapy for years. However, the use of siRNA therapeutics in clinical settings is still limited due to lack of appropriate delivery systems for the highly charged macromolecular drug. In this review, recent development of major non-viral siRNA delivery systems, including lipid, liposome, polymer, and peptide-based carriers, is to be summarized.

Toward Proper 3D-QSAR Datasets for Parameter Evaluation

  • 3D-QSAR techniques including CoMFA have been used a lot for more than two decades now. For now, the perspective of 3D-QSAR has been changed. The realization of gorge activity cliffs and higher chance correlation with many independent variables (IVs) has changed the requirements. Some suggested the benchmarking datasets for 3D-QSAR. However, were they still useful for right reasons? Here, we propose the requirement of any general purpose 3D-QSAR benchmarking datasets for lead optimization, especially for feasibility test of any IVs. Specifically, we summarize the conceptual requirements for an ideal settings for 3D-QSAR especially CoMFA.

Overview of RCT for Non-Alcoholic Fatty Liver Disease and Non-Alcoholic Steatohepatitis

  • Objective: This study aimed to get information on the current status of therapies to date for non-alcoholic fatty liver disease (NAFLD), including non-alcoholic steatohepatitis (NASH). Methods: All randomized clinical controlled trial (RCT)-derived papers for NAFLD or NASH were reviewed via PubMed Database. Results: 39 RCTs met the review criteria, of which 15 and 24 papers were for NAFLD and NASH, respectively. 83% of the papers were released since 2006, and 30 studies were conducted for western medicines, antioxidants and lifestyle intervention whereas nine trials were done using herbal medicine or acupuncture which showed positive outcome. Conclusions: NAFLD and NASH are new epidemic disorders which can be a target of traditional Oriental medicine. This study will be helpful for the Oriental medicine-based strategies or therapeutic development for them.

  • Arthritis may be broadly classified as degenerative - related to defects in cartilage and other joint constituents, often age-associated - or inflammatory disease. Inflammatory arthritis called as rheumatoid arthritis (RA) is a chronic inflammatory arthropathy and characterized by a destructive arthritis. RA encompasses infectious arthritis, arthritis caused by intra-articular deposits of crystalline material (gout), syndromes associated with genetic defects (familial Mediterranean fever), and the immune-mediated inflammatory arthropathy. Degenerative arthritis called as osteoarthritis (OA), which is most frequently occurring, causes degenerative figures of knee, waist and knuckle, and accompanies severe pain around the cartilage. Also, it may cause morning stiffness, gelling effect, tenderness, bone swelling, crepitus, and motion disorders.

활성화한 RAW 264.7 세 포주에서 인삼 Polyacetylene류의 Nitric Oxide 생성저해 (Inhibitors of Nitric Oxide Synthesis from Ginseng in Activated Macrophages)

  • Nitric Oxide (NO), derived from L-arginine, is produced by two types (constitutive and inducible) of nitric oxide synthase (NOS). The NO produced in large amounts by the inducible NOS is known to be responsible for the vasodilation and hypotension observed in septic shock. We have found three polyacetylene compounds which inhibited the production of NO in LPS-activated RAW 264.7 cells. Their structures were identified as panauynol, ginsenoyne A and PQ-6 by the spec- troscopic analysis (IC50 values were 32.3 $\mu$M, 2.3 $\mu$M, 1.5 $\mu$M, respectively). These polyacetylenes may be useful candidates for the development of new drug to treat endotoxemia and inflammation accompanied by the overproduction of NO.

Terpenoids from Citrus unshiu Peels and Their Effects on NO Production

  • Two new compounds, 3-methyl-but-2-en-1-yl-1-O-β-xylopyranosyl-(1"→2')-O-β-glucopyranoside (1) and 1-O-β-glucopyranosyl-6-hydroxy-2-methyl-hep-2-enoic acid (2), along with sixteen known terpenoids were isolated from the peels of Citrus unshiu Markov. Their structures were elucidated based on extensive NMR analyses (1H NMR, 13C NMR, DEPT, COSY, HMQC, and HMBC) and high-resolution mass spectrometry. In addition, all isolates (1 - 18) were tested their effects on nitric oxide (NO) production in RAW264.7 cells. Limonin (15) showed to inhibit LPS-induced NO production in a concentration-dependent manner without cytotoxicity.

Phosphagen Kinases of Parasites: Unexplored Chemotherapeutic Targets

  • Due to the possible emergence of resistance and safety concerns on certain treatments, development of new drugs against parasites is essential for the effective control and subsequent eradication of parasitic infections. Several drug targets have been identified which are either genes or proteins essential for the parasite survival and distinct from the hosts. These include the phosphagen kinases (PKs) which are enzymes that playa key role in maintenance of homeostasis in cells exhibiting high or variable rates of energy turnover by catalizing the reversible transfer of a phosphate between ATP and naturally occurring guanidine compounds. PKs have been identified in a number of important human and animal parasites and were also shown to be significant in survival and adaptation to stress conditions. The potential of parasite PKs as novel chemotherapeutic targets remains to be explored.

  • 나노바이오 테크놀로지는 최근 10년 동안 그 관심이 고조되어 왔다. 특히 의약, 약물전달 및 약리학 부분에서 분석 및 치료의 새로운 장을 여는 계기가 되었다. 나노바이오 테크놀로지는 전형적인 융합연구로서 생물학, 화학, 물리학, 공학 및 의학 연구자들의 협력을 그 기본으로 하고 있다. 본 총설에서는 나노바이오 생체분석, 나노 바이오칩/센서와 나노바이오소재를 내포하고 있는 최근 연구와 나노바이오 테크놀로지의 개발에 대한 설명 및 제시를 하고 있다.

Toxicity and Biomedical Imaging of Fluorescence-Conjugated Nanoparticles in Hematopoietic Progenitor Cells

  • Cellular uptake of nanoparticles for stem cell labeling and tracking is a critical technique for biomedical therapeutic applications. However, current techniques suffer from low intracellular labeling efficiency and cytotoxic effects, which has led to great interest in the development of a new labeling strategy. Using silica-coated nanoparticles conjugated with rhodamine B isothiocyanate (RITC) (SR), we tested the cellular uptake efficiency, biocompatibility, proliferation or differentiation ability with murine bone marrow derived hematopoietic stem/progenitor cells. The bone marrow hematopoietic cells showed efficient uptake with SR with dose or time dependent manner and also provided a higher uptake on hematopoietic stem/progenitor cells. Biocompatibility tests revealed that the SR had no deleterious effects on cell cytotoxicity, proliferation, or multi-differentiation capacities in vitro and in vivo. SR nanoparticles are advantageous over traditional labeling techniques as they possess a high level of cellular internalization without limiting the biofunctionality of the cells. Therefore, SR provides a useful alternative for gene or drug delivery into hematopoietic stem/progenitor cells for basic research and clinical applications.

Synthetic bio-actuators and their applications in biomedicine

  • The promise of biomimetic smart structures that can function as sensors and actuators in biomedicine is enormous. Technological development in the field of stimuli-responsive shape memory polymers have opened up a new avenue of applications for polymer-based synthetic actuators. Such synthetic actuators mimic various attributes of living organisms including responsiveness to stimuli, shape memory, selectivity, motility, and organization. This article briefly reviews various stimuli-responsive shape memory polymers and their application as bioactuators. Although the technological advancements have prototyped the potential applications of these smart materials, their widespread commercialization depends on many factors such as sensitivity, versatility, moldability, robustness, and cost.
