High Efficiency Process Development for Methane Production by Anaerobic Fermentation (혐기성 발효에 의한 고효율 메탄 제조 공정 개발)

  • This study is to investigate the kinetics of anaerobic process, the effect of mass transfer on process, and the characteristics of the conventional anaerobic bioreactor, and develop new high efficiency bioreactor. In the new bioreactor wastewater containing highly concentrated organic materials, was treated without diluting wastewater. In this experiment the high COD removal rate (about 88%) and gas production(about 200l/d) was showed with short residence time(1.5 day). This performance is about 10 times as large as the conventional reactor.

In Vitro Mass Propagation and Economic Effects of Bioreactor Culture in Ever-bearing Strawberry 'Goha' (Bioreactor를 이용한 사계성 딸기 기내대량증식과 경제성)

  • This study was conducted to compare various culture methods and evaluate economic feasibility of each method for mass propagation of new ever-bearing strawberry 'Goha'. Four different methods such as semi-solid culture, solid culture, liquid suspension culture and bioreactor culture were compared. The solid culture and bioreactor culture showed the shortest and longest root length, such as 3.6 cm and 8.3 cm, respectively. Fresh weights of plants cultured in bioreactor were 2,261 mg, which were heavier than those of cultures. Dry weights of plants cultured in bioreactor were the heavier compared to those in other cultures. The number of axillary bud developed in bioreactor was seven, but axillary bud was not developed in other cultures. Production cost through bioreactor culture was calculated to be 303 won per plant which was 542 won less than that of solid culture. As a result, we found that the bioreactor culture was the most cost effective culture method for in vitro mass propagation in new ever-bearing strawberry 'Goha'.

Improved Optimization of Indirubin Production from Bioreactor Culture of Polygonum tinctorium

  • Effect of the two-stage operation and cell concentration on indirubin production was investigated using bioreactor culture of Polygonum tinctorium. Two-stage culture was operated successfully for 110 days without any adverse effects on continuous indirubin production. Maximum indirubin concentration was found to be at 80 mg/bioreactor. Initial cell concentration significantly affected indirubin production. The indirubin production at 29.2% PCV was improved by 845%, compared to that at 5% PCV. For high-density bioreactor culture of P. tinctorium, a maximum production rate of 10.2 mg indirubin/L day was obtained. Indirubin recovery for bioreactor operation was also examined using XAD-2, XAD-4, XAD-7, and solid silicon. XAD-4 was 1.6-fold more effective than that for solid silicon in indirubin recovery.

  • Recently, developments of large scale and high density cell culture methods have been the objects of many researches, because the demand of various pharmaceutical products produced by animal cell culture has been rapidly increasing. The cell culture equipment should have the requirements such as sufficient oxygen transfer and mixing, low shear stress and surface tension, and small foaming. In order to develop a proper bioreactor meeting these requirements simultaneously, a perfluorocarbon having high solubility of oxygen was sprayed into the medium as an oxygen carrier instead of air. Also, a new impeller was developed and combined together with the perfluorocarbon spraying system so as to design a new bioreartor for cell cultivation. The new impeller had better characteristics of mixing and oxygen transfer than the paddle and cell-lift impellers based on the same, shear rate. But, it was observed that the volumetric oxygen transfer coefficient of the new bioreactor decreased with increasing cell density during E. coli fermentation.

Construction of an Oscillator Gene Circuit by Negative and Positive Feedbacks

  • Synthetic oscillators are gene circuits in which the protein expression will change over time. The delay of transcription, translation, and protein folding is used to form this kind of behavior. Here, we tried to design a synthetic oscillator by a negative feedback combined with a positive feedback. With the mutant promoter PLacC repressed by LacIq and PLux activated by AHL-bound LuxR, two gene circuits, Os-LAA and Os-ASV, were constructed and introduced into LacI-deleted E. coli DH5α cells. When glucose was used as the carbon source, a low level of fluorescence was detected in the culture, and the bacteria with Os-ASV showed no oscillation, whereas a small portion of those carrying Os-LAA demonstrated oscillation behavior with a period of about 68.3 ± 20 min. When glycerol was used as the carbon source, bacteria with Os-ASV demonstrated high fluorescence value and oscillation behavior with the period of about 121 ± 21 min.

골조직 재생 촉진을 위한 Novel Perfusion Bioreactor

  • In vitro 상에서 골조직을 원활하게 재생하기 위해서는 3차원 지지체를 이용한 세포 배양과 세포 배양 시 세포의 형태와 기능을 유지/향상시키기 위한 인체 내 미세 환경 재현은 필수적이다. 따라서 본 연구에서는 뼈 성분과 유사한 생체 활성 물질인 hydroxyapatite (HA)와 생분해성 고분자인 poly $\varepsilon$-caprolactone (PCL)를 복합재료로 이용하여 내부 연결성이 우수한 골조직 재생용 3차원 지지체를 제작하였으며, 골 재생 능력 향상을 위하여 인체내 골조직의 기계적 미세 환경을 체외에서 구현한 새로운 형태의 perfusion bioreactor system을 개발/적용하였다. 또한 본 연구에서 개발된 perfusion bioreactor system의 생물학적 평가를 위해 MG63 (osteoblast like cell, 한국 세포주 은행)과 New Zealand White Rabbit에서 분리한 중간엽 줄기세포를 골조직 재생용 3차원 지지체에 파종하였다. 48시간 동안 안정화 후 perfusion bioreactor system을 이용하여 기계적 자극을 파종된 세포에 인가하였으며, 배양 기간 동안 세포의 증식 확인 및 형태학적 관찰을 실시하였다. 본 연구 결과, perfusion bioreactor system을 이용하여 기계적 자극을 인가한 실험군에서 세포의 증식 및 활성도가 대조군에 비해 우수함을 확인 할 수 있었다. 따라서, perfusion bioreactor를 이용한 세포 배양은 세포의 활성 향상 및 골조직 재생에 도움이 될 것으로 사료된다. 차후 perfusion bioreactor를 이용한 다양한 패턴의 자극이 골재생 능력 및 중간엽 줄기세포의 골 분화능에 미치는 영향에 대한 연구가 필요할 것으로 사료된다.

Optimization and High-level Expression of a Functional GST-tagged rHLT-B in Escherichia coli and GM1 Binding Ability of Purified rHLT-B

  • The Escherichia coli heat-labile enterotoxin B subunit (HLT-B) is one of the most powerful mucosal immunogens and known mucosal adjuvants. However, the induction of high levels of HLT-B expression in E. coli has proven a difficult proposition. Therefore, in this study, the HLT-B gene was cloned from pathogenic E. coli and expressed as a fusion protein with GST (glutathion S-transferase) in E. coli BL2l (DE3), in an attempt to harvest a large quantity of soluble HLT-B. The culture conditions, including the culture media used, temperature, pH and the presence of lactose as an inducer, were all optimized in order to obtain an increase in the expression of soluble GST-rHLT-B. The biological activity of the purified rHLT-B was assayed in a series of GMI-ELISA experiments. The findings of these trials indicated that the yield of soluble recombinant GST-rHLT-B could be increased by up to 3-fold, as compared with that seen prior to the optimization, and that lactose was a more efficient alternative inducer than IPTG. The production of rHLT-B, at 92 % purity, reached an optimal level of 96 mg/l in a 3.7 L fermentor. The specific GM1 binding ability of the purified rHLT-B was determined to be almost identical to that of standard CTB.

Fouling analysis and biomass distribution on a membrane bioreactor under low ratio COD/N

  • This paper deals with the influence of chemical oxygen demand to nitrogen ratio ((COD/N) ratio) on the performance of an membrane bioreactor. We aim at establishing relations between COD/N ratio, organisms' distribution and sludge properties (specific resistance to filtration (SRF) and membrane fouling). It is also essential to define new criteria to characterize the autotrophic microorganisms, as the measurements of apparent removal rates of ammonium seem irrelevant to characterize their specific activity. Two experiments (A and B) have been carried on a 30 L lab scale membrane bioreactor with low COD/N ratio (2.3 and 1.5). The obtained results clearly indicate the role of the COD/N ratio on the biomass distribution and performance of the membrane bioreactor. New specific criteria for characterising the autotrophic microorganisms activity, is also defined as the ratio of maximum ammonium rate to the specific oxygen uptake rate in the endogenous state for autotrophic bacteria which seem to be constant whatever the operating conditions are. They are about 24.5 to 23.8 $gN-NH_4{^+}/gO_2$, for run A and B, respectively. Moreover, the filterability of the biological suspension appear significantly lower, specific resistance to filtration and membrane fouling rate are less than $10^{14}m^{-2}$ and $0.07\;10^{12}m^{-1}.d^{-1}$ respectively, than in conventional MBR confirming the adv < antage of the membrane bioreactor functioning under low COD/N ratio.

Characteristics of Immobilized Culture of Mentha piperita Cells for Oil Production

  • To investigate the characteristics of immobilized peppermint (Mentha piperita) cells, dry cell weight (DCW), change of cell viability, and oil productivity of the immobilized cells were determined. Peppermint cells were immobilized in polyurethane (PU) foams of $5{\times}5{\times}5$ mm and cultured in a shaking flask. The maximum DCW was 2.1 mg per foam piece after 20 days of cultivation and the cell density was approximately 420 mg per flask containing 200 foams in 200 ml medium. For the first five days of cultivation, the cell viability was about 80$%$ and decreased to 70$%$ during 5 to 20 days of cultivation. The maximum oil productivity, 148 mg/l was achieved after 40 days of cultivation. The immobilized cells were also cultivated in a bioreactor, equipped with a round spiral type impeller, containing 2, 400 PU foams. The cell viability after 30 days of cultivation with chitosan as an elicitor in the bioreactor was 67$%$ and DCW was 2.0 mg per foam piece. Though the cell viability was relatively high in the bioreactor system, the oil productivity was relatively lower than that of the flask system.
