토끼 두개골 결손부에서 전기 방사된 나노실크-수산화인회석 복합체를 이용한 골재생 효과에 대한 연구 (Electrospun Silk Nano-Fiber Combined with Nano-Hydoxyapatite Graft for the Rabbit Calvarial Model)

  • Purpose: The objective of the present study was to determine the capability of electrospun silk fibroin as a biomaterial template for bone formation when mixed with nano-hydoxyapatite in vivo. Materials and Methods: Ten New Zealand white rabbits were used for this study and bilateral round shaped defects were formed in the parietal bone (diameter: 8.0 mm). The electrospun silk fibroin was coated by nano-hydroxyapatite and grafted into the right parietal bone (experimental group). The left side (control group) did not receive a graft. The animals were sacrificed at 6 weeks and 12 weeks, humanly. The microcomputerized tomogram (${\mu}CT$) was taken for each specimen. Subsequently, they were undergone decalcification and stained for the histological analysis. Results: The average value of all measured variables was higher in the experimental group than in the control at 6 weeks after the operation. BMC in the experimental group at 6 weeks after operation was $48.94{\pm}19.25$ and that in the control was $26.17{\pm}16.40$ (P = 0.027). BMD in the experimental group at 6 weeks after operation was $324.59{\pm}165.24$ and that in the control was $173.03{\pm}120.30$ (P = 0.044). TMC in the experimental group at 6 weeks after operation was $19.50{\pm}6.00$ and that in the control was $10.52{\pm}6.20$ (P = 0.011). TMD in the experimental group at 6 weeks after operation was $508.88{\pm}297.57$ and that in the control was $273.54{\pm}175.91$ (P = 0.06). Gross image of both groups showed higher calcification area at 12 weeks than them in 6 weeks. The average value of ${\mu}CT$ analysis was higher at 12 weeks than that in 6 weeks in both groups. BMC in the experimental group at 12 weeks after operation was $51.21{\pm}8.81$ and that in the control was $33.47{\pm}11.13$ (P = 0.010). BMD in the experimental group at 12 weeks after operation was $323.39{\pm}21.54$ and that in the control was $197.75{\pm}76.23$ (P = 0.012). TMC in the experimental group at 12 weeks after operation was $21.44{\pm}5.30$ and that in the control was $13.31{\pm}4.17$ (P = 0.008). TMD in the experimental group at 12 weeks after operation was $524.47{\pm}19.37$ and that in the control was $299.60{\pm}136.20$ (P = 0.016). Conclusion: The rabbit calvarial defect could be successfully repaired by electrospun silk nano-fiber combined with nano-hydroxyapatite.

AN EXPERIMENTAL STUDY OF NEWLY DESIGNED IMPLANT WITH RBM SURFACE IN THE RABBIT TIBIA : RESONANCE FREQUENCY ANALYSIS AND REMOVAL TORQUE STUDY

  • Statement of problem. The importance of fixture design and surface treatment. Purpose. The clinical success of dental in plants is affected by many factors such like as degree of osseointegration, the effective load dispersion for the prostheses, and a lot of attempts have been made to overcome the difficulties. In this study, efforts were made to find the possibility of clinical acceptance of the dental implants of newly designed surface and resorbable blast media surcace. Materials and methods. In this study, two groups of custom-made, screw-shaped implants were prepared. The first with the consisting of Branemark clone design and the other with the new design. These implants were divided into four groups according to the kinds of surface treatment. Four implants($AVANA^{(R)}$, Osstem, Busan, Korea)of each group were installed in twenty rabbits. Group A was consisted of Branemark done implant left as machined, Group B with Branemark clone implants with RBM(Resorbable blast media) surface, Group C with newly designed implants left as machined and Group D with newly designed implants with RBM surface. One of the twenty rabbits died from inflammation and the observation was made for six weeks. Specimens from four groups were observed using scanning electron microscopy with 40, 100, 1000 magnification power and microsurface structures were measured by white-light scanning interferometry for three dimensional surface roughness measurements(Accura $2000^{(R)}$, Intek-Plus, Korea.). Removal torque was measured in 17 rabbits using digital torque gauge(MGT 12R, Mark-10 corp., NY, U.S.A.) immediately after the sacrifice and two rabbits were used for the histologic preparation(EXAKT $310^{(R)}$, Heraeus Kulzer, wehrheim, Germany) of specimens and observed under light microscope. Resonance frequency measurement($Osstell^{(R)}$) was taken with the 19 rabbits at the beginning of the implant fixation and immediately after the sacrifice. Results. Following results were taken from the experiment. 1. The surface of the RBM implants as seen with SEM had rough and irregular pattern with reticular formation compared to that of fumed specimens showing different surface topographies. 2. The newly designed implant with RBM surface had high removal torque value among four groups with no statistical significance. The average removal torque was $49.95{\pm}6.70Ncm$ in Group A, $51.15{\pm}4.40Ncm$ in Group B, $50.78{\pm}9.37Ncm$ in Group C, $51.09{\pm}4.69Ncm$ in Group D. 3. The RFA values were $70.8{\pm}4.3Hz$ in Group A, $71.8{\pm}3.1Hz$ in Group B, $70.9{\pm}2.5Hz$, $72.7{\pm}2.5Hz$ in Group D. Higher values were noted in the groups which had surface treatment compared to the untreated groups with no statistical significance. 4. The results from the histomorphometric evaluation showed a mean percentage of bone-to-implant contact of $45{\pm}0.5%$ in Group A, $55{\pm}3%$ in Group B, $49.5{\pm}0.5%$ in Group C, and $55{\pm}3%$ in Group D. Quite amount of newly formed bone were observed at the surface RBM-treated implants in bone marrow space.

in Vitro와 ex vivo에서 황백 온수추출물의 신생혈관 억제효과 (The Anti-angiogenic Potential of a Phellodendron amurense Hot Water Extract in Vitro and ex Vivo)

  • 신생혈관형성을 억제하는 것은 최근들어 많은 고형암의 치료에 있어서 매우 유용한 접근 방법이다. 현재까지 가장 잘 알려진 접근 방법은 신생혈관형성의 핵심인자인 혈관내피세포성장인자(VEGF)를 표적으로 하는 방법이다. 다양한 화학예방 물질들을 포함하는 많은 자연의 생산물들이나 추출물들은 그들의 항신생혈관형성 성질을 통하여 악성종양의 성장을 억제하고 있다. 황백은 항종양, 항균, 항염증 및 그외 다른 생물학적 작용을 가지고 있는, 오래전부터 널리 사용되어온 한국 전통 약재이다. 우리는 종양의 성장, 침투, 전이에 있어서 매우 중요한 과정인 신생혈관형성에 미치는 황백 온수추출물의 효과를 연구하였다. 황백 온수추출물의 항신생혈관형성 효과를 확인하기 위해서 혈관 내피세포의 성장, 이동, 침투, 관형성 그리고, 자이모그램 분석을 수행하였으며, 흰쥐 대동맥 주변 미세혈관 발아실험을 진행하였다. 그 결과, 황백 온수추출물은 혈관내피세포성장인자(VEGF)에 의해 유도되는 혈관내피세포의 성장, 이동, 침투, 관형성 그리고, 대동맥의 혈관발아를 억제하는 효과를 in vitro와 ex vivo 실험을 통해서 확인하였다. 또한, 황백 추출물은 VEGF에 의해 유도되는 기질금속단백질분해효소 (MMP)-2와 -9의 활성화를 저해하였다. 본 연구 결과들은 황백 온수추출물의 신생혈관형성 억제작용이 암과 같은 혈관신생과 관련된 질병을 치료하는데 좋은 소재가 될 수 있음을 시사한다

인터넷을 통한 반려견 식품 구매자의 인식 조사 (Awareness Survey of Consumers buying Companion Dog Food through Internet)

  • 본 연구는 인터넷을 통하여 반려견 식품을 구매하는 소비자의 반려견을 위한 지출 품목 및 비용, 반려견 푸드 선택 시 고려사항 등을 조사하였다. 성별로는 남성 39명(6.8%) 여성 532명(93.2%)으로 반려견 식품을 구입할 때 대부분 여성들이 인터넷 구매를 한다는 것을 알 수 있었다. 견주 연령대로는 20대 이하 184명(32.2%), 30대 241명(42.2%), 40대 이상 146명(25.6%)로 나타났다. 반려견의 연령을 보면 3세 이하가 200(35%)이고, 8세 이상 202(35.4%), 4~7세 169(29.6%)으로 나타났다. 반려견을 위한 구매빈도 주기는 1달 이상 주기로 구매한다고 가장 많이 답하였으며, 구매 식재료 중에서는 닭고기를 가장 많이 선호하는 것으로 나타났다. 월지출비용에서도 10만원 미만이 가장 높게 나타났으며, 반려견 푸드 주요 구입처로는 반려견 전문 온라인 쇼핑몰, 그 다음으로는 동물병원에서 구입한다고 하였다. 건강기능 향상을 위해서는 피부·모질개선을 가장 중요하게 여기고 있으며 그 다음으로 치아 뼈 형성, 소화력 개선 순으로 나타났다. 반려견 푸드 선택 시 전제품의 영양성분을 가장 고려할 사항이라고 여겼으며 그 다음으로는 인지도가 있는 브랜드 제품, 색·냄새·형상, 기능개선제품, 제품의 가격 순으로 나타났다. 따라서 반려견 푸드 신제품을 개발할 때는 전제품의 영양성분 뿐만 아니라 브랜드 제품, 색·냄새·형상, 영양밸런스, 기능개선제품, 제품의 가격 등을 고려해야 할 것으로 보여진다.

Sandblasted large grit, acid etched 표면처리에 따른 교정용 미니 임플랜트의 제거회전력에 관한 연구 (Removal torque of sandblasted large grit, acid etched treated mini-implant)

  • Sandblasted large grit, acid etched(SLA)표면처리 된 교정용 미니 임플랜트와 평활면을 가진 교정용 미니 임플랜트 사이에 제거회전력과 조직학적 소견을 통해 표면처리된 교정용 임플랜트의 임상적 가능성에 대하여 알아보고자 하였다. 실험재료로는 길이가 9.5 mm, 외경이 1.8 mm인 custom made, screw shaped, titanium implants가 사용되었다. 미니 임플랜트는 두개의 군으로 분류되었는데 SLA군은 20개의 SLA 표면처리된 미니 임플랜트이었고, 평활면군은 크기와 형태가 같지만 SLA처리공정이 생략되어 제작된 20개의 미니 임플랜트로 구성되었다. 이들은 10마리 가토의 경골에 식립되었다. 각각의 가토의 우측 경골에는 SLA군의 미니 임플랜트 2개가 식립되었고, 좌측 경골에는 평활면군의 미니 임플랜트 2개가 식립되었다. 각 군에는 식립 직후 Ni-Ti coil spring에 의해 약 150 g의 지속적인 견인력이 주어졌다. 식립 6주 후에 10마리의 가토를 희생하였고, 안정된 상태에서 Ni-Ti coil spring을 제거하였으며 digital torque gauge를 이용하여 제거 회전력을 측정하였다. 식립 6주 후에 SLA군의 경우 (8.29 Ncm) 평활면군 (3.34 Ncm)에 비해 더 높은 평균 제거회전력을 나타내었고 조직학적 소견에서도 screw 하방에서의 신생골 형성이 관찰되었다. SLA 표면처리된 미니 임플랜트는 평활면 미니 임플랜트에 비해 좀 더 강한 교정력에 저항할 수 있을 것으로 예상되었다.

  • 목 적 : 내측 반월상 연골후각의 후방 경골 부착부위의 절단 파열은 흔하지 않고 기존의 수기로는 봉합이 어렵다. 이에 저자들은 내측 반월상 연골의 후방 경골 부착부위의 절단 파열의 임상 양상 및 pullout suture를 통한 새로운 봉합술을 소개하고자 한다. 대상 및 방법 : 년 9월부터 1999년 7월까지 9명의 환자에서 후방 경골 부착부위의 절단 파열을 경험 하였다. 평균 나이는 59.3세로 38세에서 70세까지의 분포였다. 전예에서 자기공명영상과 임상양상으로 진단하였고, 슬관절경을 통해서 확진과 치료를 하였다. 1예에서 2차적 관절경을 시행하여 봉합 된 반월상 연골의 치유여부를 확인하였다. 임상 양상 : 대개 중년이후의 나이에서 호발하며, 특별한 외상의 과거력이 없다. 슬관절 후면의 통증을 호소하였으나, 그 위치가 불명확한 특징이 있었다. 경도내지 중등도의 슬관절의 종창을 호소하였으며, 특징적으로 슬관절을 굴곡 시킬수록 심해지는 술관절 후면의 통증으로 완전한 굴곡의 장애가 있었다. 단순 방사선 사진에서 퇴행성 변화는 없거나, 나이에 비해서 미미한 양상이었다. 술 전 자기 공명영상이 진단에 있어서 매우 중요한 역할을 하며, 특히 관상면에서 반월상 연골의 후각이 후방 경골 부착부위에서 분리되어 있는 형태를 보인다. 수술 수기 : 4개의 표준 portal 로 도달하여 후각부의 절단파열을 확인하고, 변연절제술로 시야를 확보하고 후각의 후방 부착부위의 연골하골을 노출 시킨다. 전내측 또는 후내측 도달법으로 절단단율 PDS 로 봉합한다. PDS 봉합사를 전내측 도달법으로 밖으로 꺼내 놓는다. 경골 내측 근위부에 종절개를 가한 후에 ACL tibla guide를 이용하여 경골 내측 근위부에서 후각의 후방 경골부착부위까지 골 터널을 만든다. 강선 루프를 경골 터널을 통하여 삽입하여서 전내측 도달법으로 빼내고, PDS 봉합사를 사이에 끼워서 다시 경골 터널을 통해 아래쪽으로 당겨서 끌어낸다. PDS 봉합사를 경골 내측 근위부에 post-tie 방법으로 고정한다. 결 론 : 내측 반월상연골 후각의 후방 경골부착부위의 절단피열은 임상적으로 흔하지 않고 보고도 드물다. 임상 양상과 자기공명영상이 진단에 있어서 중요한 역할을 한다. 관절경적 pullout 봉합이 이런 형태의 파열에 대한 치료로서 유용할 수 있을 것으로 사료된다.

생약조성물(MH-30)의 면역조혈계 및 재생조직 방사선 손상에 대한 방호 효과 (Protective Effects of New Herbal Composition (MH-30) against Radiation Injuries in Hematopoietic and Self-Renewal Tissues)

  • 본 연구팀에서는 방사선에 의한 면역조혈계 및 위장관계 손상에 대한 방호를 위하여 당귀, 천궁, 작약 혼합물의 열수 추출물로부터 그 다당체 함량을 강화한 생약조성물 Hemo HIM을 개발한 바 있다. 본 연구에서는 열수 추출물에 기반을 두어 제조된 HemoHIM에 비하여 지용성 폴리페놀 성분을 강화함으로써 더 뛰어난 생리활성을 갖는 생약조성물을 개발하고자 30% 에탄올 추출과 열수 추출을 함께 시행하여 얻은 추출물을 기반으로 생약조성물 MH-30을 제조하였다. MH-30과 HemoHIM의 에탄올 분획 내의 성분을 HPLC로 비교 분석한 결과 수용성 성분에는 큰 차이가 없었으나 여러가지 지용성 성분이 MH-30에서 크게 증가하는 것을 확인하였으며, 특히 decursin의 함량은 8.7배로 크게 증가하였다. 다음으로 MH-30의 시험관 내 항산화 및 면역세포 활성화 효과와 방사선 조사 마우스에서 면역조혈계 및 재생조직의 방호 효과를 HemoHIM과 비교하여 관찰하였다. 항산화 활성을 비교하기 위한 시험관 내 hydroxyl radical 및 superoxide anion 소거 활성 평가에서 MH-30이 HemoHIM보다 높은 항산화 활성을 보였다. 림프구 증식능을 이용한 시험관 내 면역세포 활성화 시험에서는 MH-30과 Hemo HIM은 거의 비슷한 활성을 나타내었다. 방사선 조사 마우스에서 MH-30은 내재성 비장 조혈세포 집락수를 증가시키고 골수조직 내 세포사멸을 줄였으며, 위장관 재생조직인 소장움의 생존율을 증가시키는 등 방사선에 의한 면역조혈계와 재생조직 손상을 방호하는 효과를 보여주었다. 또한, 종합적인 방사선 방호 효과를 평가하는 지표인 방사선 조사 마우스의 30일 생존율도 MH-30 투여로 유의하게 증가함을 확인하였다. 특히 상기한 모든 마우스 실험에서 MH-30은 Hemo HIM보다 더 뛰어난 방호 효과가 있음을 관찰할 수 있었다. 이상의 결과들은 새롭게 개발한 생약조성물 MH-30이 면역 조혈계와 재생조직의 방사선 손상을 줄여주는 효과가 있으며 기존에 개발된 HemoHIM에 비해 더 뛰어난 활성을 갖고 있음을 보여주었다. 따라서 MH-30은 방사선 사고 또는 암환자의 방사선치료 시 발생할 수 있는 면역조혈계 및 재생조직의 손상을 경감시킬 수 있는 방사선 방호 물질로서 유용하게 활용될 수 있을 것으로 생각한다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. On this base, we composed of the radial pulse diagnosis system in korean traditional medicine. The system composed of a radial pulse wave detection system and a radial pulse diagnosis system. With a detection system, we detected Inyoung and Cheongu radial pulse wave and processed it.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. The important parameters of this process are the conduction loss to laser power absorption (defined as the conduction-to-laser power parameter, Nk), the convection heat transfer at the surface to conduction loss (defined as Bi), dimensionless extinction coefficient (defined as Br.), and dimensionless irradiation time (defined as Fo). Dependence of Fo on Nk and Bi has been observed by experiment, and the results have been compared with the numerical results obtained by solving a 2-dimensional conduction equation. Fo and explosion depth (from the surface to the point of maximun temperature) are increased when Nk and Bi are increased.To find out the minimum laser power for explosive evaporative removal process, steady state analysis has been also made. Optimal stimulus parameters, which can minimize muscle fatigue and can achieve effective muscle contraction were determined in slow and fast muscles in Sprague-Dawley rats. Stimulus patterns of our stimulator were designed to simulate electromyographic activity monitored during locomotion of normal subjects. Muscle types of the lower extremity were classified according to their mechanical property of contraction, which are slow muscle (msoleus m.) and fast muscle (medial gastrocneminus m., rectus femoris m., vastus lateralis m.). Optimal parameters of electrical stimulation for slow muscles were 20 Hz, 0.2 ms square pulse. For fast muscle, 40 Hz, 0.3 ms square pulse was optimal to produce repeated contraction. Higher stimulus intensity was required when synergistic muscles were stimulated simultaneously than when they were stimulated individually. Electrical stimulation for each muscle was designed to generate bipedal locomotion, so that individual muscles alternate contraction and relaxation to simulate stance and swing phases. 방사선에 대한 생약복합조성물(HemoHIM)의 재생조직 및 면역계 방호 · 회복촉진 효과 (Protective Effect of a Herbal Preparation (HemoHIM) on the Self-Renewal Tissues and Immune System against γ-Irradiation)

  • 본 연구팀에서는 방사선으로부터 위장관과 면역 조혈계를 보호하기 위하여 당귀, 천궁, 백작약으로부터 새로운 생약복합물 HIM-I을 개발한 바 있다. 본 연구에서는 방사선 방호뿐 아니라 다양한 생체질환의 예방에 있어 면역조혈기능이 중추적인 역할을 한다는 관점에서, HIM-I로부터 면역조혈 기능을 더욱 증진시킨 생약복합조성물 HemoHIM을 제조하여 그 효능을 검증하였다. HIM-I를 에탄을 침지하여 에탄을 분획(HIM-I-E)과 조다당 분획 (HIM-I-P)을 얻은 후, HIM-I 에 조다당 분획을 첨가하여 HemoHIM을 제조하였다. 이렇게 얻은 HemoHIM, HIM-I 및 각 분획에 대하여 재생조직 및 면역계 방호와 회복촉진 효과를 비교 검증하였다. HemoHIM 과 HIM-I는 시험 관내에서 방사선에 의한 DNA 손상을 유의 적으로 억제하고 수산화 라디칼을 소거하는 효과가 있음이 관찰되었으며, HemoHIM과 HIM-I는 거의 비슷한 활성을 나타내었다. 시험 관내 면역 세포 활성화와 골수세포 성장촉 진 실험에서는 HemoHIM이 HIM-I 비하여 높은 활성을 보였으며, 이는 HIM-I에 비해 높은 조다당 함량에 기인한 것으로 보인다. 감마선 조사 마우스를 이용한 생체 보호효과를 살펴본 결과, HemoHIM은 HIM-I와 비슷한 정도의 소장 움 생존율 증가효과를 보였으나, 내재성 비장 조혈세포집락 형성 시험에서는 HemoHIM은 HIM-I보다 높은 효과를 나타내었다. 또한 방사선조사 마우스에서 HemoHTM의 투여는 방사선 조사후 급격히 감소된 말초 혈액내 백혈구 및 림프구수의 회복을 촉진시키고, 생존율을 증대시키는 효과가 관찰되었다. 이상의 결과들은 생약복합물 HIM-I 조다당 분획을 첨가하여 개발한 새로운 생약복합조성물 HemoHIM이 방사선에 의해 유발된 위 장관 및 면역계 조직의 손상을 감소 시켜 생존을 증가시키는 효과가 있음을 제시하였다. 특히 HemoHIM은 HIM-I와 비교하여 재생조직의 산화적 손상억제 효과는 비슷하게 유지되면서도 면역 조혈세포 방호 및 회복촉진 효과가 높은 것으로 관찰되어 방사선 방호제로서 뿐만 아니라 면역조혈기능 증진제로서 유용하게 활용될 수 있을 것으로 사료된다.
