성장하는 증거는 소포체 (ER) 스트레스의 매개 세포 사멸이 알츠하이머병을 포함한 신경 퇴행성 질환의 병리학적 발달에 중요한 역할을 한다. 로디올라 사크라(ERS)의 에탄올 추출물은 ER 스트레스 유도제인 호모시스테인(Hcy)세포 사멸과 ER 스트레스의 신경 neuro -2A 세포를 보호할 수 있는지를 조사한다. 뉴런 세포에서 Hcy는 MTT 분석에 의해 확인된 바와 같이 세포 생존 가능성은 현저히 감소시켰고, Annexin V 양성 세포의 사멸을 유도했다. ERS로 전처리한 Hcy세포 생존력 및 세포 사멸 손실은 약화되었으며, Hcy는 C/EBP 상 동성 단백질과 78-kDa 포도당 조절 단백질의 발현 및 X-box 결합 단백질 -1 (xbp1) mRNA의 접합에 스트레스를 유도했다. ESR은 Hcy에 의해 유도된 xbp-1 mRNA 접합, GRP78 및 CHOP 세포를 감소시켜 Hcy-induced ER 스트레스 및 세포 사멸에 대한 보호를 나타내며, Western blotting 분석에 heme oxygenase-1의 발현 및 HO-1 효소 활성 억제는 hemin에 의한 세포 사멸을 감소시키는 등 신경 퇴행성 질환에 치료적 가치를 보여준다.
This study examined the effect of the Mediterranean-DASH Intervention for Neurodegenerative Delay (MIND) diet of the Korean multi-domain dementia prevention program on the cognitive functions of the elderly with dementia risk factors. We developed the program including nutrition, exercise, cognitive training, vascular disease prevention, and motivation. One- hundred and fifty-three participants aged 60~79 years with at least 1 dementia risk factor were randomly assigned in a 1:1:1 ratio to the facility-based intervention (FMI), home-based intervention (HMI), and the control group. The nutrition education program consisted of 10 classes over 24 weeks: the FMI received 7 group sessions and three 1:1 sessions, the HMI received 4 group sessions and three 1:1 sessions with 3 homework sessions. The Nutrition Quotient for Elderly (NQ-E) and the Mini Nutritional Assessment (MNA) were used to evaluate nutritional status. The Repeatable Battery for the Assessment Neuropsychological Status (RBANS), Korean Mini-Mental State Examination (K-MMSE), and the Cognitive Complaint Interview (CCI) were used to evaluate cognitive functions. A total of 136 people completed the program with an 11.1% dropout rate. The NQ-E (P=0.009) and RBANS (P=0.001) scores significantly increased in the FMI (N=45) and HMI (N=49) groups compared to the control group (N=42) after the study. The changes in the score of MNA and CCI did not differ significantly between groups. In conclusion, the nutritional intervention which focused on the MIND diet as a part of a multi-domain intervention program had a positive effect on the improvement of healthy eating habits and cognitive function scores in the high-risk dementia group.
Objectives : Cyclooxygenase (COX) plays a central role in the inflammatory cascade by converting arachidonic acid into prostaglandin. COX-2 is typically induced by inflammatory stimuli in the majority of tissues, it is responsible for propagating the inflammatory response and thus, considered as the best target for anti-inflammatory drugs. The present study investigated the modulatory effect of ginsenoside Rg3, a principle active ingredient in Panax ginseng, on COX-2 expression in the brain tissue induced by systemic lipopolysaccharide (LPS) treatment in C57BL/6 mice. Methods : Because systemic LPS treatment induces COX-2 expression immediately in the brain, ginsenoside Rg3 was treated orally with doses of 10, 20, and 30 mg/kg at 1 hour before the LPS (3 mg/kg, i.p.) injection. At 4 hours after the LPS injection, COX-2 mRNA was measured by real-time polymerase chain reaction method, COX-2 protein levels were measured by Western blotting. In addition, COX-2 expressions in brain tissue were observed with immunohistochemistry and double immunofluoresence labeling. Results : Ginsenoside Rg3 (20 and 30 mg/kg) significantly attenuates up-regulation of COX-2 mRNA and protein expression in brain tissue at 4 hours after the LPS injection. Moreover, ginsenoside Rg3 (20 mg/kg) significantly reduced the number of COX-2 positive neurons in the cerebral cortex and amygdala. Conclusion : These results indicate that ginsenoside Rg3 plays a modulatory role in neuroinflammation through the inhibition of COX-2 expression in the brain and suggest that ginsenoside Rg3 and ginseng may be effective on neurodegenerative diseases caused by neuroinflammation.
Purpose: Oxidative stress was thought to play a critical role in neurodegenerative disease. Many in vivo and in vitro reports explained the possible pathway of human aging. But in therapeutic aspects, there was no clear answers to prevent aging associated with neural diseases. In this study, we investigated the antioxidant and neuroprotective effects of the Insamyangyung-tang (IYT). Methods: To estimate the antioxidant effects, we carried out 1.1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay, 2,2'-azinobis-(3- ethylbenzothiazoline-6- sulfonic acid (ABTS) radical cation decolorization assay, and measurement of total polyphenolic content. To evaluate neuroprotective effect of IYT in vitro. We performed thiazolyl blue tetrazolium bromide (MTT) assay, reactive oxygen species (ROS) creation in SH-SY5Y. Tyrosine hydroxylase (TH) immunocytochemistry, nitric oxide (NO) assay, and TNF-${\alpha}$ assay in primary rat mesencephalic dopaminergic neurons. Results: The $IC_{50}$ values were $571.6{\mu}g/m{\ell}$ and $202.3{\mu}g/m{\ell}$ in DPPH and ABTS assay respectively. Total polyphenolic content was 1.05%. In SH-SY5Y culture, IYT significantly increased the decreased cell viability by 6-OHDA at the concentrations of $10{\mu}g/m{\ell}$ in pre-treatment group, $10-100{\mu}g/m{\ell}$ in post-treatment group, and $100{\mu}g/m{\ell}$ in co-treatment group. The production of ROS induced by 6-OHDA was significantly inhibited in IYT treated group. In mesencephalic dopaminergic cell culture, the IYT group reduced the dopaminergic cell loss against 6-OHDA toxicity and the production of No and TNF-${\alpha}$ at the concentration of $0.2{\mu}g/m{\ell}$. Conclusion: These results showed that IYT has antioxidant and neuroprotectctive effects in the dopaminergic cells through decreasing the production of ROS, NO and TNF-${\alpha}$ which can cause many neurodegenerative changes in brain cell.
파킨슨병은 두번째로 많이 발병하는 퇴행성 신경질환이며 약 5-10%는 유전된다. Leucine-rich repeat kinase 2(LRRK2)는 그 돌연변이의 일부가 파킨슨병을 일으키는 유전자이다. LRRK2에는 인산화효소와 GTPase 기능이 있는 도메인과 함께 단백질 상호작용에 관여하는 Leucine-rich repeat (LRR), WD40 도메인이 존재하여, LRRK2와 상호작용하는 단백질이 파킨슨병 발병에 중요한 역할을 함을 암시한다. 우리는 이러한 LRRK2와 상호작용하는 단백질을 규명하여 그 단백질의 세포내 기능을 통해 역으로 LRRK2의 기능을 밝히고자 하였다. NIH3T3 세포 용해물을 LRRK2 항체와 IgG로 각각 면역침강하여 LRRK2 항체 침강반응에서만 특이적으로 나타나는 단백질 밴드를 질량 분석한 결과, methionyl-tRNA synthetase (MRS)로 나타났다. LRRK2와 MRS의 상호작용은 면역침강반응과 GST-pull down assay를 통해 확인됐다. 병을 유발하는, LRRK2의 돌연변이인 G2019S가 인산화효소 활성을 증가시키므로 LRRK2가 MRS를 인산화하는 지를 조사한 결과, LRRK2재조합단백질은 MRS 단백질을 인산화 하지 않았다. 또한 이들 두 단백질의 각각의 양 증가가 상대 단백질의 양 증가, 즉 안정성에 영향을 미치는 지를 조사하였으나 안정성의 변화를 관찰하지 못하였다. 결론적으로, MRS는 LRRK2와 상호작용을 하지만 LRRK2 인산화효소의 기질은 아니다.
본 연구의 목적은 알츠하이머질환(Alzheimer's disease: AD) 동물 모델을 대상으로 트레드밀 운동(Treadmill exercise: TE)과 환경강화(environmental enrichment: EE) 처치가 인지기능, 근 기능, 및 밀착연접 단백질 발현에 미치는 영향을 확인하는데 있다. AD 동물 모델을 제작하기 위해 aluminum chloride(AlCl3)를 90일간(40mg/kg/하루) 투여 하였으며 동시에 TE(10-12m/min, 40-60min/day) 혹은 EE에 노출시켰다. 그 결과 AlCl3 투여에 의한 인지기능 저하와 근 기능 감소가 TE와 EE에 의해 완화된 것으로 나타났다. 또한, TE와 EE는 AD 질환에서 나타나는 β-amyloid(Aβ), alpha-synuclein 및 tumor necrosis factor-α(TNF-α) 단백질의 발현 증가를 감소시킨 것으로 나타났다. 게다가 TE와 EE는 AlCl3 투여에 의해 감소된 밀착연접 단백질(Occludin, Claudin-5 및 ZO-1)의 발현을 통계적으로 유의하게 증가시킨 것으로 나타났다. 마지막으로 Aβ 단백질과 밀착연접 단백질과의 상관분석을 실시한 결과 부적 상관관계(Occludin: r=-0.853, p=0.001; Claudin-5 : r=-0.352, p=0.915; ZO-1 : r=-0.424, p=0.0390)로 나타났다. 따라서 이를 종합해 보면 TE 혹은 EE 처치는 AD에 나타나는 병리학적 특징들을 일부 완화시켜 인지기능과 근 기능을 일부 개선 시킬 수 있는 효과적인 운동 방법이라고 생각된다.
최근 중추신경계에서 면역기능을 담당하는 미세아교세포(microglia)의 염증반응을 효율적으로 조절하는 것은 알츠하이머 병, 파킨슨 병 및 헌팅턴 병과 같은 퇴행성 뇌질환의 치료를 위한 중요한 타겟으로 인식되고 있다. 왕귀뚜라미(Teleogryllus emma)는 다양한 치료효능으로 인해 세계적으로 널리 이용되고 있으며, 본 연구팀에서는 최근 왕귀뚜라미의 전사체 분석을 통하여 항균활성을 가지는 다양한 종류의 새로운 항균 펩타이드(antimicrobial peptide; AMP) 후보들을 선별한 바 있다. 항균 펩타이드는 미생물에서부터 포유류까지 매우 다양한 종으로부터 발견되었으며, 현재는 항균활성뿐만 아니라 염증반응과 같은 다양한 질병의 치료제 개발을 위한 후보 물질로 관심을 받고 있다. 본 연구에서는 선행연구를 통하여 선별된 왕귀뚜라미 유래 항균 펩타이드들 중에서 Teleogryllusine(VKWKR-LNNNKVLQKIYFVKI-NH2)으로 명명된 항균 펩타이드의 신경염증 억제 효능을 관찰하였다. Teleogryllusine의 신경염증 억제 효능을 관찰 하기 위하여 immortalized mouse microglia 세포주인 BV-2 세포에 Teleogryllusine을 1시간 전처리 한 후 LPS를 이용하여 BV-2 세포의 염증 반응을 유도하였다. 그 결과 Teleogryllusin은 최대 처리 농도인 80 ㎍/ml까지 세포독성 없이 nitric oxide (NO) 생성을 현저히 감소시킴을 확인할 수 있었다. 또한 염증반응 매개인자인 iNOS와 COX-2 및 cytokine (Il-6, TNF-α)의 발현을 유전자 수준과 단백질 수준에서 확인한 결과 Teleogryllusine 처리농도에 의존적으로 감소됨을 확인할 수 있었다. 또한 Teleogryllusine의 신경염증 억제작용 기작을 확인한 결과 mitogen activated protein kinases (MAPKs)와 IκB의 인산화 및 proteosome에 의한 IκB의 분해를 억제함으로서 BV-2 세포의 신경염증반응이 조절됨을 확인할 수 있었다. 이러한 결과로 보아 왕귀뚜라미 유래 Teleogryllusine 펩타이드는 신경염증반응에 의해 유도되는 퇴행성 뇌질환 치료 및 예방을 위한 의료용 소재로 사용될 수 있을 것으로 기대된다.
인지기능 저하는 장 단기 기억 및 주의력 소실과 우울증, 불안증의 증가를 특징으로 한다. 또한, 인지기능 저하는 알츠하이머, 파킨슨병과 같은 다양한 퇴행성 뇌질환과 연관되어 있다. 경제적 부담, 안전 위협을 포함하는 인지기능 저하와 관련된 사회적 문제는 고령화가 진행됨에 따라 증가하고 있다. 이러한 문제를 해결하기 위해 전 세계적으로 많은 연구가 수행되고 있다. 일반적으로 인지기능 저하를 유발할 가능성이 있는 원인으로는 노화에 따른 대사 및 호르몬 불균형, 감염, 약물 오남용, 신경세포 손상 등이 알려져 있지만 다양한 요인이 관련되어 있으므로 원인 규명이 어려운 한계점 때문에 뚜렷한 치료전략 수립이 어려운 실정이다. 최근의 연구에 따르면 퇴행성 뇌질환 발생의 원인과 이에 대한 치료전략 수립에 있어서 장내미생물의 역할이 중요하게 제시되고 있다. 특히, 알츠하이머병과 파킨슨병에서 장내미생물 조성의 변화 및 이들에 의한 대사산물에 따른 분자생물학적, 신경행동학적 증상의 변화가 밝혀졌다. 알츠하이머병 동물모델에서 장내미생물의 변화는 NMDA 수용체와 글루탐산의 변화를 통해 기억능력 소실을 야기하였다. 반면, 알츠하이머병 동물모델에 프로바이오틱스를 투여하였을 때, 비정상적인 신경학적 행동이 유의적으로 감소하였다. 파킨슨병은 장내미생물 군집의 변화와 직접적인 연관성을 보였으며 이는 이차적 증상인 변비 발생에도 영향을 미치는 것으로 나타났다. 파킨슨병 동물모델에 투여한 프로바이오틱스는 단쇄지방산 중 하나인 뷰티르산 증가를 통한 신경세포 보호효과를 나타내었다. 또한, 알츠하이머병과 파킨슨병에서 뇌-혈관장벽의 기능이상이 밝혀졌으며, 뇌-혈관장벽 변화는 장내미생물 불균형에 의한 전신성 염증에 따른 미세소관의 파괴 및 투과성 증가와 연관된 것으로 나타났다. 더불어 장내미생물 대사과정에서 생성된 대사산물은 퇴행성 뇌질환의 발생과 치료에 영향을 미친다. 본 논문에서는 인지기능 저하의 진행을 지연시킴으로써 심화를 방지할 수 있는 효과적인 접근법을 제시하기 위하여 인지기능 저하와 장내미생물의 연관성을 심층적으로 고찰하여 치료적 대안으로 제시하고자 한다.
영아형 바텐병은 PPT1 결핍 및 기능장애로 인해 발병하며, 12,500명 당 1명의 발병률을 가진 신경 퇴행 질환이다. 전 세계적으로 수많은 연구가 진행 중 이지만, 아직 명확하게 밝혀진 발병원인 및 치료방법에 대해서는 알려지고 있지 않다. 본 연구에서는 뇌에서 풍부하게 발현되는 neurogranin의 발현수준이 WT과 EBD KO 쥐에서 어떤 변화를 보이는지 확인하기 위해 mRNA, 단백질, 배양된 neurospheres를 이용하여 실험을 수행하였다. real-time PCR을 통한 neurogranin의 발현수준 비교 결과 WT에서는 노화와 무관하게neurogranin mRNA 수준에 차이가 없었으나, EBD KO 쥐에서는 노화가 진행됨에 따라 neurogranin mRNA 발현수준이 감소하였으며, 뇌에서 추출된 단백질을 이용한 western blot 분석에서도 real-time PCR과 동일한 결과를 확인할 수 있었다. 또한 WT, EBD KO 쥐의 태아로 부터 neural stem cell 인 neurospheres를 배양하여 western blot 분석을 수행한 결과 PPT1 결핍에 의해 neurogranin의 정상적인 인산화에 문제가 발생함을 확인하였다. 이러한 결과들을 바탕으로 neurospheres에 산화스트레스 유발물질인 $H_2O_2$를 처리하였고, 24시간 경과 후 항산화제인 NAC을 처리하자 $H_2O_2$를 처리한 시료에서는 mock control인 인산화된 neurogranin에 비해 그 수준이 증가하였으며, $H_2O_2$ 처리 후 NAC을 투여한 시료의 인산화 수준은 mock control 보다는 높았지만 $H_2O_2$만을 처리한 시료 수준보다 neurogranin의 인산화 정도가 감소하는 결과를 확인하였다. 이러한 결과들을 통해 PPT1 결핍으로 인해 신경세포 내에 과다하게 인산화된 neurogranin이 존재하며, neurogranin 인산화 정도는 세포가 지닌 산화스트레스 정도에 의해 변화함을 알 수 있었다. 또한 항산화제를 사용하여 세포의 산화스트레스 수준을 감소시킬 경우 neurogranin의 기능을 정상적으로 회복시킬 수 있는 가능성을 확인하였다.

Alzheimer's disease (AD) is a neurodegenerative disorder associated with progressive memory loss and neuronal cell death. Although numerous previous studies have been focused on disease progression or reverse pathological symptoms, therapeutic strategies for AD are limited. Alternatively, the identification of traditional herbal medicines or their active compounds has received much attention. The aims of the present study were to characterize the ameliorating effects of spinosin, a C-glucosylflavone isolated from Zizyphus jujuba var. spinosa, on memory impairment or the pathological changes induced through amyloid-${\beta}_{1-42}$ oligomer ($A{\beta}O$) in mice. Memory impairment was induced by intracerebroventricular injection of $A{\beta}O$ ($50{\mu}M$) and spinosin (5, 10, and 20 mg/kg) was administered for 7 days. In the behavioral tasks, the subchronic administration of spinosin (20 mg/kg, p.o.) significantly ameliorated $A{\beta}O$-induced cognitive impairment in the passive avoidance task or the Y-maze task. To identify the effects of spinosin on the pathological changes induced through $A{\beta}O$, immunohistochemistry and Western blot analyses were performed. Spinosin treatment also reduced the number of activated microglia and astrocytes observed after $A{\beta}O$ injection. In addition, spinosin rescued the $A{\beta}O$-induced decrease in choline acetyltransferase expression levels. These results suggest that spinosin ameliorated memory impairment induced through $A{\beta}O$, and these effects were regulated, in part, through neuroprotective activity via the anti-inflammatory effects of spinosin. Therefore, spinosin might be a useful agent against the amyloid ${\beta}$ protein-induced cognitive dysfunction observed in AD patients.

제 12 조 (서비스 이용 시간)
① 서비스 이용은 당 사이트의 업무상 또는 기술상 특별한 지장이 없는 한 연중무휴, 1일 24시간 운영을
원칙으로 합니다. 단, 당 사이트는 시스템 정기점검, 증설 및 교체를 위해 당 사이트가 정한 날이나 시간에
서비스를 일시 중단할 수 있으며, 예정되어 있는 작업으로 인한 서비스 일시중단은 당 사이트 홈페이지를
통해 사전에 공지합니다.
② 당 사이트는 서비스를 특정범위로 분할하여 각 범위별로 이용가능시간을 별도로 지정할 수 있습니다. 다만
이 경우 그 내용을 공지합니다.
제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
