Cancer Cell Recognition by Fuzzy Logic

  • This paper proposes the new method based on fuzzy logic which recognizes between normal and abnormal. The object image was the Thyroid Gland cell image that was diagnosed as normal and abnormal(two types of abnormal : follicular neoplastic cell, and papillary neoplastic cell), respectively. The nuclei were successfully diagnosed as normal and abnormal. The multiple feature parameters (pre-obtained 16 feature parameters of image data) were used to extract the features of each nucleus. As a consequence of using fuzzy logic algorithm, proposed in this paper, average recognition rate of 98.25% was obtained.

  • 18세령 중성화 암컷 요크셔테리어종 개가 우측 4번 및 5번 유선 부위에 위치한 대형 복부 종괴로 내원하였다. 외과적으로 적출한 유선 종괴가 병리학적 진단을 위하여 의뢰되었다. 병리조직학적 소견으로 종괴는 표피에서 피하조직에 걸쳐 다발성 및 융합된 종양성 집락들로 구성되어 있었다. 종양성 집락들은 면포상, 고형상, 사상 및 미세유두상 등의 매우 다양한 형태를 나타내었으며, 큰 종양성 유선 도관은 주변조직으로의 강한 침습성을 나타내고 있었다. 진피표층에 위치한 다수의 림프관 얼기들과 진피의 림프관에는 무수히 많은 종양세포의 색전을 가지고 있었다. 종양을 구성하고 있는 세포들의 약 50%이상이 면포상의 형태를 띠고 있기 때문에, 본 증례는 최근의 개 유선 종양의 분류에 근거하여 개의 유선에서 발생한 유선 면포 암종으로 진단하였다.

  • A new method of digital image analysis technique for discrimination of cancer cell was presented in this paper. The object image was the Thyroid eland cells image that was diagnosed as normal and abnormal (two types of abnormal: follicular neoplastic cell, and papillary neoplastic cell), respectively. By using the proposed region segmentation algorithm, the cells were segmented into nucleus. The 16 feature parameters were used to calculate the features of each nucleus. A9 a consequence of using dominant feature parameters method proposed in this paper, discrimination rate of 91.11% was obtained for Thyroid Gland cells.

Metastatic thyroid C-cell carcinoma in a beagle dog

  • An adult beagle dog was presented with a cervical mass detected by palpation and computed tomography. Fine needle aspirates revealed numerous epithelial cells with plasmacytoid appearance and frequent naked nuclei. Histologically, the mass consists of multiple packets of neoplastic cells and extensive areas of necrosis and fibrosis. Neoplastic cells were also found in submandibular lymph nodes. Immunohistochemistry showed that neoplastic cells were positive for calcitonin and negative for thyroglobulin. Based on these findings, the cervical mass was diagnosed as thyroid C-cell carcinoma. Almost one year after the surgical excision, the dog remains healthy without any symptom of recurrence or metastasis.

  • This paper proposes the new method based on contour following method with directional angle to segment the cell into the nuclei. The object image was the Thyroid Gland cell image that was diagnosed as normal and abnormal(two types of abnormal : follicular neoplastic cell, and papillary neoplastic cell), respectively. The nuclei were successfully diagnosed as normal and abnormal. this paper, improved method of digital image analysis required in basic medical science for diagnosis of cells was proposed. The object image was the Thyroid Gland cell image with difference of chromatin patterns. To segment the cell nucleus from background, the region segmentation algorithm by edge tracing was proposed. And feature parameter was obtained from discrete Fourier transformation of image. After construct a feature sample group of each cells, experiment of discrimination was executed with any verification cells. As a result of experiment using features proposed in this paper, get a better segmentation rate(70-90%) than previously reported papers, and this method give shape to get objectivity and fixed quantity in diagnosis of cells. The methods described in this paper be used immediately for discrimination of neoplastic cells.

Acrylamide가 인체상피세포의 발암화에 미치는 영향 (ACRYLAMIDE-INDUCED NEOPLASTIC TRANSFORMATION OF HUMAN EPITHELIAL CELLS IN CULTURE)

  • Purpose Acrylamide is present in significant quantities in a wide range of commonly consumed human foods. Carcinogenic risk of acrylamide through the consumption of food is a great public concern and in controversy, but it is not properly addressed due to the lack of evidence in humans. While a plenty of data is available on the carcinogenicity in animal models, the studies in humans are limited. Thus, the present study attempted to examine the carcinogenic potentials of acrylamide on the human epithelial cell, which is the target cell origin of the most cancers. Material and method & Result 1. Acrylamide was not cytotoxic up to $100{\mu}M$ as measured by MTT and LDH assays, indicating a relatively low toxicity of this substance in human epithelial cells. 2. The parameters of neoplastic cellular transformation such as cell saturation density, soft-agar colony formation and cell aggregation were analyzed to examine the carcinogenic potential of acrylamide. 3. The neoplastic transformation was further increased with the co-treatment of TPA 4. Antioxidants blocked the generation of Reactive Oxygen Species(ROS) and the GSH depleting agent dramatically increased the ROS production. 5. mRNA levels of fibronectin following acrylamide exposure was increased in a dose-dependent manner, indicating a possible biomarker of acrylamide-induced cellular transformation. Conclusion The present study will provide a valuable basis to compare the interspecies differences in response to carcinogenic potentials of acrylamide. The data on the interspecies differences are essential element in human risk assessment. Thus, our results obtained from the human epithelial cells will contribute to improving the risk assessment of human neoplasm including oral cancer.

Multicentric T cell lymphoma in a Maltese dog

  • A case of multicentric high grade T cell lymphoma is reported in a 5-year-old male Maltese dog with generalized lymphadenopathy. The dog showed depression, anorexia, blindness, jaundice, arrhythmia, and hematuria for 8 months. Complete blood count and chemistry profile revealed anemia and increased alanine transferase, alkaline phosphatase, total bilirubin, and total cholesterol. Grossly, most of lymph nodes, spleen, and liver were enlarged and neoplastic masses were occupied in these tissues. Histologically, massive accumulation of small noncleaved neoplastic lymphocytes with high mitotic figures was observed in all lymph nodes and spleen. Infiltration of neoplastic lymphocytes was also noted in the lung, liver, kidney, eye, skin, muscle, and bone marrow of femur. Immunohistochemistry revealed that tumor cells were CD3-positive and but CD79a-negative, consistent with T-cell lineage. In our best knowledge, this is the first report of multicentric lymphoma clarified the origin of tumor cells in Korea.

Differential Expression of Cytokeratin 13 in Non-Neoplastic, Dysplastic and Neoplastic Oral Mucosa in a High Risk Pakistani Population

  • Background: Gradual loss of cytokeratin 13 (CK13) may be linked with the severity of dysplastic changes and transformation to malignancy. In this study we assessed the differential expression of CK13 in normal, hyperplastic, dysplastic and cancerous oral mucosa. Materials and Methods: A total of 93 oral biopsies were collected during the 2011-2014 period. The biopsies were characterized as normal (19), hyperplastic (21), severely dysplastic/carcinoma in situ (16) and invasive oral squamous cell carcinoma (OSCC) (37) after morphological assessment. Formalin fixed paraffin embedded sections were stained with a monoclonal antibody against CK13 using the Envision technique. Immunohistochemically stained slides were then analyzed for CK13 expression. Results: CK13 was consistently and diffusely expressed in all normal and hyperplastic tissue biopsies from oral mucosa. Severely dysplastic/carcinoma in situ biopsies showed complete loss in 50% of cases, while in the remaining 50% expression was very focal and weak. OSCC cases showed complete or near complete loss of CK13 in all cases. Few cases showed weak expression in keratin pearls only. Conclusions: This study validates the utility of CK13 IHC as a useful immunohistochemical marker in routine diagnostic practice to make distinction between non-neoplastic from dysplastic and neoplastic (malignant) oral lesions.

  • Gliomas, much like other cancers, are composed of a heterogeneous mix of neoplastic and non-neoplastic cells that include both native and recruited cells. There is extensive diversity among the tumor cells, with differing capacity for In vitro and in vivo growth, a property intimately linked to the cell's differentiation status. Those cells that are undifferentiated, self-renewing, with the capacity for developing tumors (tumorigenic) cells are designated by some as cancer stem cells, because of the stem-like properties. These cells may be a critical therapeutic target. However the exact identity and cell(s) of origin of the socalled glioma cancer stem cell remain elusive. Here we review the current understanding of glioma cancer stem cell biology.

Atypical Fibroma and Fibrosarcoma Derived from Cutaneous Ganglion Cell-Like Cells in Ten Djungarian Hamsters (Phodopus sungorus)

  • Ten cutaneous masses from Djungarina hamsters (Phodopus sungorus) were diagnosed as nine atypical fibromas and one fibrosarcoma derived from cutaneous ganglion cell-like cells. Among these patients, nine were male and one was female. Histologically, these abnormal masses were composed of spindle-shaped or polygonal neoplastic 'ganglion cell-like' cells with abundant amphophilic vacuolated cytoplasm. Immunohistochemically, these neoplastic cells were stained for vimentin, S100, and neuron-specific enolase. Out of 9 males, 3 showed positive reactions to the androgen receptor. This report provides a detailed histologic and immunohistochemical characterization of atypical fibroma, fibrosarcoma, and the tumorigenesis of ganglion cell-like cells in Djungarian hamsters.
