Abamectin offers great protection against Bursaphelenchus xylophilus, a well-known devastating pathogen of pine tree stands. Trunk injection of nematicides is currently the most preferred method of control. This study aimed to evaluate the potency of the commonly used formulations of abamectin against B. xylophilus. Twenty-one formulations of abamectin were evaluated by comparing their sublethal toxicities and reproduction inhibition potentials against B. xylophilus. Nematodes were treated with diluted formulation concentrations in multi-well culture plates. And, populations preexposed to pre-determined concentrations of the formulations were inoculated onto Botrytis cinerea culture, and in pine twig cuttings. Potency was contrastingly different among formulations, with LC95 of 0.00285 and 0.39462 mg/ml for the most, and the least potent formulation, respectively. Paralysis generally occurred at an application dose of 0.06 ㎍/ml or higher, and formulations with high sublethal toxicities caused significant paralysis levels at the tested doses, albeit the variations. Nematode reproduction was evident at lower doses of 0.00053-0.0006 ㎍/ml both on Botrytis cinerea and pine twigs, with significant variations among formulations. Thus, the study highlighted the inconsistencies in the potency of similar product formulations with the same active ingredient concentration against the target organism, and the need to analyze the potential antagonistic effects of the additives used in formulations.
The pine wood nematode (PWN), Bursaphelenchus xylophilus is a well-known devastating pathogen of economic importance in the Republic of Korea and other countries. In the Republic of Korea, trunk injection of nematicides is the preferred method of control. In this study, the efficacy of 16 locally produced formulations of emamectin benzoate against the PWN are compared through determining their sublethal toxicities and reproduction inhibition potentials. Nematodes were treated with varying concentrations of the tested chemicals in multi-well culture plates, and rates of paralysis and mortality were determined after 24 h. Reproduction inhibition potential was tested by inoculating pre-treated nematodes onto Botrytis cinerea, and in pine twig cuttings. Despite the uniformity in the concentration of the active ingredient, efficacy was contrastingly different among formulations. The formulations evidently conformed to three distinct groups based on similarities in sublethal activity (group 1: LC95 of 0.00768-0.01443 mg/ ml; group 2: LC95 of 0.03202-0.07236 mg/ml, and group 3: LC95 of as high as 0.30643-0.40811 mg/ml). Nematode paralysis generally occurred at the application dose of 0.0134-0.1075 ㎍/ml, and there were significant differences in nematode paralysis rates among the products. Nematode reproduction was only evident at lower doses both on B. cinerea and pine twigs, albeit the variations among formulations. Group 1 formulations significantly reduced nematode reproduction even at a lower dose of 0.001075 ㎍/ml. The variations in efficacy might be attributed to differences in inert ingredients. Therefore, there is need to analyze the potential antagonistic effects of the large number of additives used in formulations.
Effects of organic matter content in soil on activity. vertical migration. and persistence of two nernaticides. carbofuran and ethoprophos. to root-knot nematode, Mrloitlog!~iei ~lcognita.w ere investigated. As the organic matter content increased. activity of the nernaticides tended to be reduced. Both nematicides exhibited control values of more than 80'k' to M. iix.o,yilit~iln 0-2 cm depth soil layer from the surface. regi~rdless of organic matter content in soil. In 2-4 cm depth soil layer. however. the control value of the neniaticides varied with the organic niatter content in soil. The control value of carbofuran in the soil layer was ranged from I0 to 30'2,. depending on the soil organic niatter content. In contrast. ethoprophos had no control value against M. i/ic.o,ytlitcr in the \oil layer, except that the nematicide had a control value of 30% when the organic matter content was 0.4%. Furthermore. ethoprophos had no effect on controlling M. i/ic.o,gtiitrr in soil layer of below 4cm. whereas control values of carbofuran were approximately from 5 to 20% in all test soils having different organic matter contents. These results indicate that carbofuran has more vertical migration effect than ethoprophos. Persistence of the two neniaticides was also decreased with increasing soil organic matter content. Half life of carbofuran was 2-3 weeks in soil containing 0.4% organic matter, whereas it was found to be I week in soils containning 0.8 ant1 1.6% of organic matter. On the other hand. activity of ethoprophos was reduced to half in 3-3 weeks and in 2-3 weeks in soil containing 0.4 and 0.8%, and 1.6%) of organic matter, respectively. However, no activity of the both nernaticides was found in soil containing 3.2% of organic matter.

Streptomyces sampsonii KK1024 having strong chitinolytic activity was isolated from crab-shell rich soil at Muan, Jeolanamdo. The KK1024 produced chitinase, protease, gelatinase and lipase. When 50% of KK1024 culture broth was treated to juveniles and eggs of root-knot nematode, juvenile mortality at 3 days was 81.67% and egg hatch rate at 5 days was 2.00%. When $183.7{\mu}g\;mL^{-1}$ of crude enzyme produced by KK1024 was treated, juvenile mortality at 3 days was 96.00% and egg hatch rate at 5 days was 5.33%. At 1% of butanol extract from KK1024, juvenile mortality was highest with 90.00% and egg hatch rate was lowest with 0%. The comparison of the effect of KK1024 culture broth with only medium, synthetic fertilizer, and commercial nematicide on tomato growth and nematode infection was examined in pot trials. KK1024 culture broth showed lower number of egg mass and gall in plant, and population of juveniles in soil compared with only medium and synthetic fertilizer treatment, but not in commercial nematicide. However, the highest shoot weight and length was discovered in KK1024 culture broth. These results suggest that Streptomyces sampsonii KK1024 producing lytic enzymes and nematicidal compounds can be one of candidates for biocontrol agents against root-knot nematodes.

Efficacy of novel nematicide, imicyafos GR was evaluated against two species of the root-knot nematodes in pot and greenhouse conditions. When tested in pots, the population of Meloidogyne incognita and M. hapla was reduced sixty days after treatment, with mortality rate of 91.5% and 90.6%, respectively. Suppression effect of imicyafos GR on root galling in tomato was tested. The number of root galls caused by M. incognita and M. hapla was reduced 60 days after nematode inoculation, with the efficacy of 94.2% and 95.1%, respectively. Under greenhouse conditions planted with watermelon, melon, cucumber, and tomato, the efficacy of imicyafos GR on M. incognita persisted up to 60 days after treatment, showing 90% of control efficacy. Moreover, the number of root galling was more reduced than fosthiazate treatment, with the potential as a control agent.

Pine wood nematode (PWN), Bursaphelenchus xylophilus is one of the most serious pests of pine tree. Trunk injection of some nematicides in tree is well known as an effective control method. However there are some limitating factors which hindering the efficacy of trunk injection in field or potted tree. In this study we suggested easy and useful alternative screening methods of nematicides against PWN. Reproduction of PWN was influenced by tree twig moisture (high reproduction in high moisture trig) and paraffin coating was 78.6% reduced moisture loss in tested twig. There were no reproduction different in up and down site from infection site of twig at 1 month after inoculation of PWN and also distance (5 and 10 cm) from inoculation site of PWN did not influence the reproduction of PWN. Numbers of reproduced PWN were higher with decreasing diameter of twig. Numbers of reproduced PWN were similar to P. thunbergii and P. densiflora. However reproduction was increased depending on high inoculation density and longer propagation period. When inoculation of PWN on cut twig injected with emamectin benzoate 2.15% EC and morantel tartrate 8% SL in trunk of Pinus thunbergii in the field, PWN number were significantly reduced than untreated control. We suggest this screening method for PWN control agents.
This study examined the control effects of amino acid biochemical agent extracted from chicken feathers on Meloidogyne incognita for the purpose of developing an environmentally friendly nematicidal agent that can replace chemical control of root-knot nematodes (RKN). We investigated the lethal effects of J2 juveniles for 19 types of commercial amino acids. As a result, five kinds of amino acids (L-asparagine, L-aspartic acid, L-methionine, L-tyrosine, L-cysteine) showed mortality rate of more than 50% at a concentration of 50 mM. L-asparagine showed the highest mortality rate at 94%. We also investigated the lethal effect of J2 juveniles and suppressive effects of egg hatching by feather amino acids (FAA) biochemical agent. It showed that the mortality rate of J2 juveniles was more than 80% and suppression rate of egg hatching was 74% at 1/50 concentrations of FAA. As a result of conducting a tomato pot culture experiment for 60 days after treating 1/50 concentrations of FAA biochemical agent in rhizosphere soil, it showed that the control effects were 63% of juveniles density in the soil, 59% of egg mass and 61% of root gall index, respectively. Based on the above results, it is considered that the FAA biochemical agent extracted from chicken feathers can be used as an environmentally friendly nematicidal agent of RKN.

Pine wood nematode, Bursaphlenchus xylophilus is a fatal damaging pest, infecting pinewood trees and is one of the serious forest pest in Korea. Trunk injection is one of the most effective method to protect the live pine trees from nematode infection. The aim of this research was to reveal the efficacy of the nematicidal materials from 46 Kyrgyzstan, 88 Vietnam plant materials and 21 active materials (${\alpha}$-terpinene, ${\alpha}$-pinene, ${\beta}$-thujaplicin, cinnamaldehyde, eugenol, emodin, geraniol, limonone, methyl palmitate, matrine, myrcene, methyl gallate, nicotine, quassin, resveratrol, rotenone, thymol, thujaplicin, terthiophene, tuberstemonine, ${\gamma}$-terpinene) isolated from plants for using as trunk injection agents. Eclipta prostrata extract was highly effective against pine wood nematode in plant extracts and terthiophene has the best nematicidal activity from active materials isolated from plants. Foliar spray of terthiophene on Japanese black pine (Pinus thunbergii) tree reduced proliferation of inoculated pine wood nematode at 22 days after treatment, however no efficacy was found over 22 days. Eclipta prostrata extract and terthiophene can be used as nematicide for further nematicidal efficacy test against any other plant parasitic nematodes and trunk injection efficacy against pine wood nematode.
To determine the economic threshold level of Meloidogyne incognita on cucumber in greenhouse conditions, cucumber seedlings(Cucumis sativa L. cv. 'Super Manchon') grafted on 'Jangsushintozoa'(Cucurbita maxima x Cu. moschata) were planted in wooden boxes($30{\times}40{\times}15cm,\;L{\times}W{\times}D$) under a plastic house in August 01 and harvested from Sep. 01-Oct. 30, 2006. The initial nematode population densities(Pi) in the wooden boxes were adjusted to 0, 10, 30, 100, 300, and 600 second-stage juveniles(J2)/100 $cm^3$ soil. The relationship of total fruit yield to Pi level could be adequately described by a linear regression equation, $Y=0.82-0.04{\cdot}Log_{10}$(Pi+1). Initial nematode densities(Pi) before planting in excess of 5 J2/1,000 $cm^3$ soil caused in total yields loss that is equivalent to the costs of granular nematicide application; Pi level in excess of 25 J2/1,000 $cm^3$ soil caused in total yields loss that exceed the costs for application of fumigants at current control costs. We propose two different management strategies depending on nematode densities, (i) application of non-fumigant granular nematicides for M. incognita Pi level of 5 J2/1,000 $cm^3$ soil and (ii) fumigant treatment with Pi level over 25 J2/1,000 $cm^3$ soil. Soil samples to determine population density of Meloidogyne spp. for advisory purposes should be taken several months before planting time, which allows a period of time enough for implementing management procedures.
Surveys were conducted on the occurrence of nematodes in the root systems of 1-3-year old Cymbidium hybida Swartz cultivated for export in Korea. The most frequently detected plant-parasitic nematode was Ditylenchus sp. with 4.7, 43.7, and 49.7/200 cc growth medium in 1, 2, and 3 year-old cymbidiums, respectively. And the densities of non-parasitic nematodes, dorylaimids, were 35.3, 70.5, and 155.8/200 cc growth medium in 1, 2, and 3-year-old cymbidiums, respectively. Three-year-old cymbidiums collected from Siheung, Ansan, and Gimpo had low densities of Aphelenchus sp. and Aphelenchoides sp. with under 40 individuals/pot, and the dorylaimid densities were 56-824/pot. To evaluate the effects of nematicides on Ditylenchus sp. in cymbidium, Emamectin benzoate EC, Fosthiazate SL, and Cadusafos CS were tested at two farms in Ansan and Gimpo. Emamectin benzoate EC showed control effects of 75.7 and 89.5%, whereas Fosthiazate SL and Cadusafos CS showed 27.2 and 65.3% and 30.1 and 90.5% control effects in the tests.
