  • This study was conducted to further purify and characterize a polysaccharide fraction from whole parts of Duchesnea indica (Andr.) Focke, which was previously reported to show an antitumor activity by us. Mild acid-treatment of the fraction by 5% acetic acid yielded three subtractions (Fr. A,B and C) on a Sephadex G-25. Fr. A was composed of uronic acid, hexose, pentose and phenolic substance, Fr. C mainly phenolic substance, and Fr. B contained hexose. All three fractions exhibited colony stimulating factor activity.

  • Tissue factor (TF) is a cell surface receptor of coagulation factor Ⅶ and is the principal initiator of the vertebrate coagulation cascade. TF is found in high levels in some organs such as brain, lung and placenta, whereas blood monocytes, endothelial cells contain only trivial amount of TF when quiescent, and is stimulated to synthesize TF by infections or vascular lesions. TF is reported to be found in high levels in atherosclerotic plaques, cancer cells. TF activation in various cells in many infectious or immunologic diseases tells us the physiologic importance of TF. We screened many edible vegetables for TF inhibitor, by measuring the prothrombin time to detect the TF activity, and we picked Aster scaber to isolate the TF inhibitory substance. Aster scaber showed two kinds of anti thrombotic activity, one is TF inhibition and the other is elongation of plasma recalcification time. The anti thrombotic substances were found to be saponins which has echinocystic acid as aglycone.

  • 1. Amylase producing bacteria have been isolated from natural sources and was identified out. The above strain is identical to Bacillus subtillis in every details of physiological and morphorogical characteristics by this investigations. 2. Sikworm chrysalis nad chlorella extracts are prove to be further effective for the stimulation of amylase production than any other sources. The former silkworm extract is more stronger in its activity. 3. It is found that effective results had been accepted by a less soybean meal addition per that of wheat bran in the mixture ratio of them, also less addition of corn is needed in the mixture ratio of between corn and whear bran. In the mixing ratio of wheat bran per silkworm or chlorella extracts, the addition of wheat bran is necessary for stimulation of amylase production. 4. Fro the stimulation of amylase production, the effective substances from natural sources and its order of activity in cultral media cultral media is proved as follows, silkworms ext.>chlorella ext.>alcohol>skim-milk and peptone>fermented milk>radish leaf ext.

  • To investigate skin whitening natural substances, the effects on melanogenesis by measuring the tyrosinase activity and the melanin contents of three hydrolysable tannins, $1,2,6-tri-O-galloyl-{\beta}-D-glucose$ (1), 2,3-(S)-HHDP-D-glucose (2) and pedunculagin (3) in B16 melanoma cells were examined. $1,2,6-Tri-O-galloyl-{\beta}-D-glucose$ (1), 2,3-(S)-HHDP-D-glucose (2) and pedunculagin (3) inhibited tyrosinase activity in B16 melanoma cells in a dose-dependent manner.

  • Quantitative structure-activity relationship (QSAR) methodologies have been applied for many years, to correlate the relationship between physicochemical properties of chemical substances and their biological activities to generate a statistical model for prediction of the activities of new chemical entities. The basic principle behind the QSAR models is that, how structural variation is responsible for the difference in biological activities of the compounds. 3D-QSAR has emerged as a natural extension to the classical Hansch and Free-Wilson approaches, which develops the 3D properties of the ligands to predict their biological activities using various chemometric techniques (PLS, G/PLS, ANN etc). It has served as a valuable predictive tool in the design of pharmaceuticals and agrochemicals. This review seeks to provide different 3D-QSAR approaches involved in drug designing process to develop structure-activity relationships and also discussed the fundamental limitations, as well as those that might be overcome with the improved methodologies.

Synthesis of Quercetin and Luteolin Derivatives with Cell Proliferation Inhibitory Activity and Toxicity in B16 Melanoma Cells

  • Melanoma is a malignant skin tumor caused by damage to melanocytes that can spread to other organs. Hence, various studies have been conducted on preventing the spread of melanoma. Flavonoid-structured substances such as apigenin and galanzin are effective therapeutic agents for inhibiting the proliferation and metastasis of melanoma. In this study, luteolin, quercetin, and their respective derivatives were synthesized. These compounds inhibited cell proliferation of B16 melanoma cells. These results confirmed that the derivatives of quercetin and luteolin may be useful as therapeutic agents to prevent melanoma metastasis.

Screening and Identification of Natural Herbicidal Active Substance in Rye and Oat Extracts (호밀, 귀리 추출물로부터 제초활성물질 탐색 및 동정)

  • This study was conducted to identify allelopathic compounds in rye and oat straw extracts by HPLC analysis. These extracts were analyzed with 12 standard chemicals including salicylic acid. 11 chemicals in rye extract except for naringin and in oat extract except for catechin were identified. Salicylic acid(8.34mg/g) in rye straw extracts and naringin(7.50mg/g) in oat straw extracts among these standard chemicals were identified as the largest amount substance. The germination of Chenopodium ablum seeds was significantly inhibited by these chemicals at $10^{-3}$ and $10^{-4}M$ concentrations as compared to control. Salicylic acid in rye and naringin in oat were considered as the major allelopathic substances although allelopathy may be caused by an interaction of many substances. Yet many unidentified chemical compounds are present in both extracts.

Formulation Of Some Mathematical Models For The Estimations Of The Most Probable Salts Derived From The Major Mineral Constituents In Natural Water

  • By extensive studies with the quantitative status of the mineral constituents of thousands of water samples, it was found that almost in all natural waters irrespective of the surface or sub-surface sources, minimum 99.5% of the total amount of the cationic constituents are generally the contributions of 3 commonly present parameters viz. Ca, Mg and Na and that of the anionic species are same and contributed by $HCO_3$, Cl and $SO_4$ only. In the field of water works, all these major mineral substances are conventionally measured as their individual ions. But till now, no reliable and generalised methods or rules have been developed for the determination of the exact kinds of the individual salt components and their amounts from these ionic constituents normally present in water. As salt content, only the TDS (Total Dissolved Solids) parameter is frequently measured by evaporation of the water sample. But TDS can tell nothing about the kinds and amounts of the individual salts present in it. Considering the analytical importance of the estimation of the mineral substances as their individual salts, some generalised mathematical models have been developed by this research which are based on the 'hypothetical order of chemical combinations' as may occur among the ionic constituents. With the help of these models, one can easily assume the most probable salts with approximate quantities derived from the ionic constituents. In addition, approximate amount of Na content can also be estimated mathematically with simultaneous verification of the correctness of the water analysis results. The models are stated in this paper with practical illustrations and descriptions of the method of applications.

  • Antimicrobial activities of ethanol extracts of marta rosemary (Rosemarinus officinalis L.) against Bacillus subtilis and Salmonella typhimurium were investigated under conditions similar to processing of general food materials to develop natural food preservatives. The strongest antimicrobial activities were shown against the tested organisms at pH 7. In B. subtilis, the herbal extract extended the lag phase at low concentrations, and reduced the specific growth rate during logarithmic phase at high concentrations. In S. typhimurium the herbal extract reduced the specific growth rate even at lower concentrations. These trends were obtained at low pHs of culture solution. Antimicrobial substances of herbs were efficiently extracted with low polar solvents. The thermal stability of antimicrobial substances decreased with heating time at $100^{\circ}C$. S. typhimurium was more sensitive to herbal extracts at $100\;and\;121^{\circ}C$ than B. subtilis.

  • Chinese herbs have been used for a long period of time and less side effects than synthesized chemical drugs. Therefore, using Chinese herbs as natural additives in cosmetics becomes popular in recent years. The methanol extracts of Scutellariae Radix, Lithospermi Radix, Lonicerae Flos, Andrographitis. Herba, Angelicae Dahuricae Radix, Ligustici Rhizoma et Radix, Hedyotis Diffusae Herba, Isatidis Folium, Magnoliae Liliflorae Flos, Forsythiae Fructus, Anmarrhenae Rhizoma, Spirodelae Herba, Gardeniae Fructus, Sophorae Flavescentis Radix, Coptidis Rhizoma, Prunellae Spica, Equiseti Hiemalis Herba, Gentianae Radix, Moutan Radicis Cortex, Fraxini Cortex, Lycii Radicis Cortex, Violae Herba, Lophatheri Herba, Matricariae chamomillae Flos, Taraxaci Herba and Scutellariae Barbatae Herba are used to test the efficiency of inhibiting acne pathogens. Twenty-six Chinese herbs are extracted by methanol, and then condensed to dried powder. These extracts are divided into water-soluble part and DMSO soluble part. These two type solutions are tested for the effect on acne pathogens by paper disc diffusion method. The results show that the substances of water soluble part which are Coptidis Rhizoma, Moutan Radicis Cortex, Scutellariae Barbatae Herba have medium to high activity of inhibiting acne pathogents, and the substances of DMSO soluble part which are Coptidis Rhizoma, Ligustici Rhizoma et Radix, Sophorae Flavescentis Radix, Moutan Radicis Cortex, Scutellariae Radix, Scutellariae Barbatae Herba also have medium to high activity of inhibiting acne pathogens. Using Chinese herbs as natural additives in cosmetics is convenience and valuable application in cosmetceutical research and development. Therefore, it is worth that re-investigation and find out the potential of Chinese herbs being use in cosmetics.
