Purification and Structure Determination of Antifungal Phospholipids from a Marine Streptomyces

  • A series of antifungal compounds were obtained from the methanol extract of the mycelium from marine actinomycetes M428 which was identified as a Stereptomyces species by fatty acid composition and biochemical characteristics. These compounds were purified by combined chromatographic techniques and the structures were characterized with spectroscopic methods including 1D and 2D NMR, and mass spectrometry as sn-l lysophosphatidyl inositols. The side chains were established by chemical degradation followed by GC analysis to be 14-methyl pentadecanoic acid (iso-palmitic acid, i-C16:0, compound A) and 13-methyl tetradecanoic acid (iso-pentadecanoic acid, i-C15:0, compound B). These compounds displayed highly selective antifungal activity against C. albicans with MIC values of $5{\;}\mu\textrm{g}/ml$ (compound A) and $2.5{\;}\mu\textrm{g}/ml$ (compound B), while it had almost negligible antibiotic activity against E. coli and P aerogenosa with MIC value higher than $50{\;}\mu\textrm{g}/ml$ and no cytotoxic activities against human myeloma leukemia K562 ($IC_{50}>100{\;}\mu\textrm{g}/ml$).

HPLC 및 GC/MSD를 이용한 식물 중의 Higenamine 함량 분석 (Analysis of Higenamine Contents in Plants with HPLC and GC/MSD)

  • Higenamine is known as a cardiotonic principle of Aconiti tuber. The analytical procedures were established for the detection of higenamine in plants. The amounts of higenamine in several Aconiti tubers and the embryo of Nelumbo nucifera, another plant species known to contain higenamine, were determined. The $H_2O$ soluble fraction prepared from MeOH extract was first treated with AD-2 resin and then applied to either HPLC or GC/MSD systems. With HPLC, $6.4{\sim}19.2\;{\mu}g/g$ of higenamine were detected from various Aconiti tubers and $182.3\;{\mu}g/g$ of higenamine from the embryo of Nelumbo nucifera. The results obtained with GC/MSD also provided comparable data with those obtained with HPLC.

Identification of Meat Species Using PCR-RFLP Marker of Cytochrome b Gene

  • Food labeling regulations require that the meat species in various meat products are accurately declared to the consumer. Substitution or adulteration of costly meat with a cheaper one is one of the most common problems in the meat industry. In this study, PCR-restriction fragment length polymorphism(RFLP) method of the mitochondrial cytochrome b(mt cyt b) gene has been applied for identification of the origin of six mammalian meat species(beef, port horse, goat, mutton and deer) and three poultry meat species(chicken, turkey and duck) as raw materials for meat products. PCR was used to amplify a variable region of mt cyt b gene. Meat species differentiation was determined by digestion of the amplified products with a 359 bp fragment using HaeIII and HinfI restriction enzymes, which generated species-specific RFLP patterns. This PCR-RFLP DNA marker of mt cyt b gene could be very useful for the accurate and reliable identification and discrimination of animal meat species in routine analysis.

Tissue Factor Inhibitory Sesquiterpene Glycoside from Eriobotrya japonica

  • Tissue factor (TF, tissue thromboplastin) is a membrane bound glycoprotein, which acceler-ates the blood clotting, activating both the intrinsic and the extrinsic pathways to serve as a cofactor for activated factor VII (Vila). The TF-factor Vila complex (TF/VIIa) proteolytically activates factors IX and X, which leads to the generation of thrombin and fibrin clots. In order to isolate TF inhibitors, by means of a bioassay-directed chromatographic separation technique, from the leaves of Eriobotrya japonica Lindley (Rosaceae), a known sesquiterpene glycoside (2) and ferulic acid (3) were isolated as inhibitors that were evaluated using a single-clotting assay method for determining TF activity. Another sesquiterpene glycoside (1) was also isolated but was inactive in the assay system. Compound 3 was yielded by alkaline hydrolysis of compound 2. The structures of compounds 1, 2, and 3 were identified by means of spectral analysis as $3-O-{\alph}-L-rhamnopyranosyl-(1{\rightarrow}4)-a-L-rhamnopyranosyl-(1{\rightarrow}2)-[{\alph}-L-rhamnopyrano-syl-(1{\rightarrow}6)]-{\beta}-D-glucopyranosyl nerolidol$ (1), $3-O-{\alph}-L-rhamnopyranosyl-(1{\rightarrow}4)-{\alph}-L-rhamnopyr-anosyl-(1{\rightarrow}2)-[{\alph}-L-(4-trans-feruloyl)-rhamnopyranosyl-(1{\rightarrow}6)]-{\beta}-D-glucopyranosyl$ nerolidol (2) and ferulic acid (3), respectively. Compounds 2 and 3 inhibited 50% of the TF activity at con-centrations of 2 and $369{\;}\mu\textrm{m}/TF$ units, respectively.

Constituents from the Fruiting Bodies of Ganoderma applanatum and Their Aldose Reductase Inhibitory Activity

  • Eight compounds were isolated from the fruiting bodies of Ganoderma applanatum, and were identified as 2-methoxyfatty acids (1), 5-dihydroergosterol (2), ergosterol peroxide (3) $3{\beta},7{\beta},20,23{\zeta}-tetrahydroxy-11,15-dioxolanosta-8-en-26-oic$ acid (4), $7{\beta},20,23{\zeta}-trihydroxy-3,11,15-trioxolanosta-8-en-26-oic$ acid (5), cerevisterol (6), $7{\beta},23{\zeta}-dihydroxy-3,11,15-trioxolanosta-8,20E(22)-dien-26-oic$ acid (7), and $7{\beta}-hydroxy-3,11,15,23-tetraoxolanosta-8,20E(22)-dien-26-oic$ acid methyl ester (8) by spectral analysis. All compounds were isolated for the first time from this fruiting bodies, and their effect on rat lens aldose reductase (RLAR) activity was tested. Among these eight compounds, ergosterol peroxide (3) was found to exhibit potent RLAR inhibition, its $IC_{50}$ value being $15.4\;{\mu}g/mL$.

Gas Chromatography Residue Analysis of Bifenthrin in Pears Treated with 2% Wettable Powder

  • This study was conducted to monitor the level of bifenthrin residues in pear sprayed with 2% bifenthrin wettable powder (WP) at the recommended rate at four different schedules prior to harvest. The target analyte was extracted with acetone, partitioned into dichloromethane, and then purified by florisil chromatographic column. The residue determination was performed on a DB-5 capillary column using GC with electron capture detector (ECD). Linearity of this method was quite good ($r^2$ = 0.9951) in the concentration ranged from 0.2 mg/kg to 10 mg/kg. Recovery test was carried out at two concentration levels, 0.2 mg/kg and 1.0 mg/kg, in three replicates, and their rates were from 82.9% to 107.2%. No quantitative bifenthrin was detected in pear of all kinds of treatments including the treatment sprayed 4 times until 7 days before harvest. This sensitive and selective method can be used to monitor the trace residual amounts of bifenthrin in pear in a quite low concentration level.

Extraction of seven major compounds from Agastache rugosa (Fisch. & C.A.Mey.) Kuntze: optimization study using response surface methodology

  • The purpose of this study is to demonstrate the potential enhancement of the flavonoid contents from Agastache rugosa, which can be obtained as raw materials for functional products in the food medicine industry by identifying important factors for efficient preparation to save costs and time in terms of economic factors. For this reason, response surface methodology using Box-Behnken design was used to optimize the extraction conditions for the maximum yield of seven major compounds from A. rugosa. The optimum conditions were obtained with an ethanol concentration of 60.0%, a temperature of 50 ℃, and an extraction time of 33.6 min, meaning that the regression analysis fits the experimental data well. Under these conditions, the seven major compounds 1-7 had observed values of 2.169, 2.135, 0.697, 2.485, 0.105, 1.247, and 0.551%, respectively. These results show that the observed values are in good agreement with the predicted values in the regression model. This process for optimization study exhibited a basic protocol for obtaining stable ingredients from A. rugosa that are appropriate for the development of effective functional products.

HPLC-DAD-ion trap-MS를 이용한 인동 생리활성 물질의 동시분석 (Simultaneous Analysis of Bioactive Metabolites from Caulis Lonicera japonica by HPLC-DAD-ion trap-MS)

  • A high-performance liquid chromatography (HPLC) with DAD detector and electrospray ionization mass spectrometry (ESI-MS) was established for the simultaneous determination of coniferin (1), loganic acid (2), demethylsecologanol (3), sweroside (4) and loganin (5) from caulis Lonicera joponica. The optimal chromatographic conditions were obtained on an ODS column ($5{\mu}m$, $4.6{\times}150mm$) with the column temperature $35^{\circ}C$. The mobile phase was composed of (A) water with 0.1% formic acid and (B) methanol with 0.1% formic acid using a gradient elution, the flow rate was 0.3 ml/min. Detection wavelength was set at 254 nm. All calibration curves showed good linear regression ($r^2$>0.998) within test ranges. The developed method provided satisfactory precision and accuracy with overall intra-day and interday variations of 0.16~3.28% and 0.14~1.99%, respectively, and the overall recoveries of 99.39~105.89% for the five compounds analyzed. The verified method was successfully applied to quantitative determination of the two types (phenolic compounds and iridoids) of bioactive compounds in 24 commercial caulis L. japonica samples from different markets in Korea and China. The analytical results demonstrated that the contents of the five analytes vary significantly with sources.

Development of Molecular Biological Methods to Analyze Bacterial Species Diversity in Freshwater and Soil Ecosystems

  • A new method was developed for the rapid analysis of diverse bacterial species in the natural environment. Our method is based on PCR-single-strands-conformation polymorphism (PCR-SSCP) and selective isolation technique of single-stranded DNA. Variable V3 fragments of 16S rDNA were amplified by PCR with bacterial 16S rDNA primers, where one of the primers was biotinylated at the 5'-end. The biotinylated strands of the PCR products were selectively isolated by using streptavidin paramagnetic particles and a magnetic stand, to prevent SSCP analysis producing heteroduplexes from heterogeneous DNA samples. The selected strands were separated by electrophoresis on a polyacrylamide gel, and detected by silver staining. Analysis of PCR products from 8 bacterial strains demonstrated their characteristic DNA band patterns. In addition, changes in the structure of the bacterial community and species diversity in the microcosm treated with phenol could be monitored. After 3 weeks of incubation, phenol and its intermediate, 2-hydroxy-muconic-semialdehyde, were degraded by indigenous bacteria. These dominating bacterial populations were identified as strong bands on an SSCP gel. Therefore, this study provides useful tools for microbial community analysis of natural habitats.

천연 탄산음료 제품의 소비자 이용 실태 및 인식도에 관한 연구 (A Study of Consuming Status and Consumer Awareness for Natural Carbonated Drinks)

  • Purpose: This research was to understand the consumer awareness and consumption of natural carbonated drinks in Korea. Methods: A survey was conducted enrolling adults over the age of 20, residing in Seoul and its metropolitan area, and who consumed natural carbonated drinks. Data analysis of 544 valid samples was performed using SPSS 17.0. Results: The study showed that consumers mostly purchased natural carbonated water the most, primarily as a water substitute. However, many other usages that the consumer selected showed a possibility for a more versatile usages of natural carbonated drinks. As for the reason of purchase, 'interest in a new product' showed the highest mean value. The factor 'interest in a new product', 'interest in its effectiveness on skin care and diet', 'lower-calorie products', 'health benefits from the usage of natural flavoring and coloring' and 'recommendation from acquaintances' showed statistically significant higher values for women than men. The most frequent and preferred location for purchase were large discount stores, and the most preferred, major informants were in the order 'tasting events at marts and department stores', 'TV/radio advertisement' and 'recommendations from family/friends'. Analysis of the purchase intent showed high interest for non-alcoholic natural carbonated drinks. Conclusion: The study results can be used as baseline data to establish marketing strategies in the emerging natural carbonated beverage market.
