  • Arachidonic acid(AA), which is stored in membrane glycerophospholipids, is liberated by phospholipase $A_2(PLA_2)$ enzymes and is sequentially converted to cyclooxygenase (COX) and lipoxygenase (LOX) then to various bioactive prostaglandins (PGs,) and leukotrienes (LTs). In order to find the specific inhibitors of AA metabolism enzymes such as $PLA_2$, COX-2, 5-LO and lyso PAF acetyltransferase. 195 Korean indigenous plant extracts were evaluated for their inhibitory activity on $PGD_2,\;LTC_4$ production from cytokine-induced mouse bone marrow-derived mast cells (BMMC) and arachidonic acid released from phospholipid and PAF production from lyso PAF. From this screening procedure, methanol extract of eight plants such as Saururus chinensis, Aster tataricus, Chrysanthemum cinerariaefolium, Reynoutria japonica, Disocorea nipponica, Epimedium koreanum, impatiens textori, Veronica rotunda var. subintegra were found to inhibit production of inflammatory mediators in vitro assay system.

Development of the Turnip Aphid, Lipaphis erysimi Kaltenbach (Homoptera: Aphididae), and Test of Insecticidal Efficacy of Some Commercial Natural Products

  • The turnip aphid is a worldwide pest, damaging mainly to crucifers. In order to understand the life parameters of Lipaphis erysimi for the eventual goal of control, the developmental periods, survival rates, lifespan, and fecundity of the species were investigated under five temperature regimes ($15^{\circ}C-35^{\circ}C$). Furthermore, the efficacy of several environment-friendly agricultural materials (EFAMs) that are on the market was subjected to test in order to obtain further accurate information. The developmental period of the turnip aphid nymph was longest at $15^{\circ}C$ as 16.9 days, shortened as temperature goes up to $25^{\circ}C$ (5.4 days), and then somewhat increased at $30^{\circ}C$ (5.9 days), suggesting that the most efficient temperature for nymphal development could be around $25^{\circ}C$. Mortality of the nymphal turnip aphid was obvious at $35^{\circ}C$, whereas it was minimal at other temperature schemes. The longevity of adults shortened as temperature goes up to $30^{\circ}C$. In particular, the maximum lifespan for adults continued for 55 days at $15^{\circ}C$, but shortened to 21 days at $30^{\circ}C$. The total fecundity was 35.7 at $15^{\circ}C$, 81 at $20^{\circ}C$, 64.2 at $25^{\circ}C$, and 6.6 individuals at $30^{\circ}C$, showing the highest fecundity at $20^{\circ}C$. After the turnip aphids were successfully stabilized in indoor environment the insecticidal activity was tested and mortality was determined 12, 24, 36, and 48 hrs after EFAMs are treated. Several on-the-market EFAMs showed more than 90% of insecticidal activity within 24 hrs or 48 hrs, but a few showed less than 90% activity, signifying importance of selection of proper EFAMs.

전남 서부 해안가에서 생산되는 세발나물의 유통실태, 이화학적 성분 및 생리활성 (Distribution Status, Physicochemical Composition, and Physiological Activity of Spergularia marina Cultivated in the Western Region in Jeon-Ra-Nam-Do)

  • This study was conducted to examine the distribution status, the physicochemical composition, and the physiological activity of Spergularia marina grown in the western region in Jeon-Ra-Nam-Do, looking for its potential as a local specialty goods. The amount sold of Spergularia marina to different areas from a rural market place was examined during April, 2008. The proportions of Spergularia marina are as follows; Yeongam(30.5%), Muan(24.3%), Naju(20.3%), Hampyeong(18.0%), and Haenam(7.0%), which were shown higher than those of Pimpinella brachycarpa. Total vitamin C and total chlorophyll contents in Spergularia marina were 68.61mg/100g DW and 58.11mg/g DW, respectively. Total amino acid contents were 331.94mg/100g DW and 20 kinds of amino acids were separated and classified. The contents of inorganic matters in Spergularia marina were as follows; Na(64.23mg/L), Mg(15.88mg/L) K(9.99mg/L), Ca(3.83mg/L). The total phenol and the total flavonoid contents of methanol extracts from Spergularia marina were 41.7mg/L and 18.9mg/L, respectively, with an extracting concentration of 2,000mg/L. The nitrite scavenging activity was highest (83.4%) in hot water extract. These results may be useful as basic data for using Spergularia marina as a local specialty goods.

Anti-Cancer Effects of Imperata cylindrica Leaf Extract on Human Oral Squamous Carcinoma cell line SCC-9 in Vitro

  • Imperata cylindrica, a tall tufted grass which has multiple pharmacological applications is one of the key ingredients in various traditional medicinal formula used in India. Previous reports have shown that I. cylindrica plant extract inhibited cell proliferation and induced apoptosis in various cancer cell lines. To our knowledge, no studies have been published on the effect of I. cylindrica leaf extract on human oral cancers. The present study was undertaken in order to evaluate the anticancer properties of the leaf extract of I. cylindrica using an oral squamous cell carcinoma cell line SCC-9 as an in vitro model system. A methanol extract from dried leaves of I. cylindrica (ICL) was prepared by standard procedures. Effects of the ICL extract on the morphology of SCC-9 cells was visualized by microscopy. Cytotoxicity was determined by MTT assay. Effects of the ICL extract on colony forming ability of SCC-9 cells was evaluated using clonogenic assay. Cell cycle analysis was performed by flow cytometry and induction of apoptosis was determined by DNA fragmentation assay. The ICL extract treatment caused cytotoxicity and induced cell death in vitro in SCC-9 cells in a dose-dependent manner. This treatment also significantly reduced the clonogenic potential and inhibited cell proliferation by arresting the cell cycle in the G2/M phase. Furthermore, DNA fragmentation assays showed that the observed cell death was caused by apoptosis. This is the first report showing the anticancer activity of the methanol extracts from the leaves of I. cylindrica in human oral cancer cell line. Our data indicates that ICL extract could be considered as one of the lead compounds for the formulation of anticancer therapeutic agents to treat/manage human oral cancers. The natural abundance of I. cylindrica and its wide geographic distribution could render it one of the primary resource materials for preparation of anticancer therapeutic agents.

Dendropanax morbifera Extract Inhibits Intimal Hyperplasia in Balloon-Injured Rat Carotid Arteries by Modulating Phenotypic Changes in Vascular Smooth Muscle Cells

  • The plant Dendropanax morbifera Léveille is effective folk medicines for the treatment of several conditions, such as infectious diseases, skin diseases, and other illnesses. Although the inhibitory effects of D. morbifera on the proliferation and migration of vascular smooth muscle cells (VSMCs) have been shown in our previous study, its effects in vivo remain to be elucidated. In this study, we aimed to investigate the protective effects of the extracts from D. morbifera (EDM) on neointimal hyperplasia of rat carotid artery and explore the underlying mechanisms. We observed that the ratio of intima to media thickness (I/M) was significantly decreased in the EDM-treated groups by ~80% compared to that of the control. The expression of Ki-67 and proliferating cell nuclear antigen was decreased by ~70% in the EDM-treated groups compared to that of the control. In addition, matrix metalloproteinase (MMP)2 and MMP9 significantly reduced in the neointimal layer of the EDM-treated groups. Moreover, the decreased levels of contractile phenotypic markers of VSMCs, such as α-smooth muscle actin, myocardin, and smooth muscle-myosin heavy chain, were successfully restored by EDM treatment. Furthermore, the levels of synthetic phenotypic markers, cellular retinal binding protein 1 and connexin 43 were also restored to normal levels. These results suggest that EDM inhibits vascular neointimal hyperplasia induced by balloon injury in rats via phenotypic modulation of VSMCs. Therefore, EDM may be a potential drug candidate for the prevention of restenosis.

카르노스산에 의한 화이트소시지의 항산화 및 항균 효과 (Antioxidant and Antibacterial Effects of Carnosic Acid on White Sausage)

  • 로즈마리의 주요성분으로는 carnosic acid, carnosol, rosmarinic acid, ursolic acid 등의 폴리페놀(polyphenolic)로 구성되어 있다. 식물기원의 페놀혼합물은 항산화 작용과 항균작용을 포함한 유용한 기능과 영양적인 효과에 주목을 끌고 있다. 본 연구는 로즈마리 추출물의 하나인 카르노스산을 화이트소시지 원료에 첨가한 후 실온 저장기간 동안 pH, TBARS, VBN, 전자공여능 및 총균수의 변화를 조사하였다. 카르노스산의 첨가는 무첨가구인 대조구에 비해 낮은 pH, TBARS값, 그리고 VBN값을 보여 주었으며, 높은 항산화 작용과 항균작용을 나타내었다. 이들의 활성은 카르노스산의 농도가 높을수록 증가하였다. 또한 카르노스산의 병원성균에 대한 항균활성을 측정한 결과, 두 병원성균(E. coli KCCM 11234와 S. enteritidis KCCM 12021)에 대하여 강한 항균활성을 보여주었다. 따라서 화이트소시지에 있어서 카르노스산은 천연보존제로서 충분히 이용 가능한 결과를 나타내었다.

In vivo Angiogenic Activity of Dichloromethane Extracts of Aloe vera Gel

  • The angiogenic activity of Aloe vera (Aloe baradensis), known as a good healing plant, was investigated. We have extracted and fractionated dichloromethane extract (G1M1D1) and methanol soluble fraction of dichloromethane extract (G1M1D1M1) which contain low-molecular weight substances of Aloe vera gel. G1M1D1 and G1D1M1 fractions induced a radially arranged, spoke-sheel-like vasculature in chick embryo chorioallantoic membrane (CAM) assay. The angiogenic activity was dose-dependent and the angiogenic pattern in the CAM assay. The angiogenic activity was dose-dependent and the angiogenic pattern in the CAM assay was very similar to that of phorbol 12-myristate-13-acetate (PMA) used as a positive control. The modified CAM assay, e simple and accurate quantitating method, was used to quntitate the angiogenic activity of G1D1M1 fraction. Application of G1M1D1M1 fraction ($100\mug/egg$) resulted in much more intense angiogenesis than in control while slightly less intense angiogenesis than in PMA (100 ng/egg).

초고압 저온처리에 의한 매자나무의 면역 활성 (Effect of Immune Activity on Berberis koreana Palibin by Ultra High Pressure Low Temperature Process)

  • This study was performed to investigate a methodology of improving immune activities of Berberis koreana Palibin by ultra high pressure on low temperature extraction process. Extraction yield was enhanced up to more than 35% by above process, compare to the control and extraction average. The cytotoxicity on human kidney cell (HEK293) was showed below 20.4%, 21.6% in adding 1.0 mg/ml of the highest concentration. Generally, the extracts by ultra high pressure extraction process showed less toxicity about 5% than the other processes. It could tell that toxic materials that existing in the plant could be reduced or broken by ultra high pressure process due to can be broken bond such the hydrogen bond, the electrostatic bond, the van der Waals bond, and the hydrophobic bond, can be broken by high pressure. The result could be employed to develop a new type of functional food from B. koreana Palibin by low temperature high pressure process.

Patent and Anti-wrinkle Cosmetics

  • In the 21st century, the development of cosmetics is led to pursue the high functionality of cosmetics with excellent effectiveness and safety. As Cosmetic Act took effect in 2000, functional cosmetics is provided in the law. As a result, the research and development of functional cosmetics has continually increased, and the number of patent applications in functional cosmetics has also rapidly increased as a plan for preoccupying in the functional cosmetics age. Now, the cosmetic industry has a great interest in developing anti-wrinkle cosmetics among functional cosmetics, because women's desire for having young resilient skin has increased since Korea entered an aging society thanks to the advanced medical technology. The patent application trends of anti-wrinkle cosmetics at home since 2000 particularly show the rapid increase in the applications in natural plant extracts. It may be because Korean consumers preference of vegetable cosmetics has resulted in the development of raw materials based on the traditional medicine. As for the existing preparation such as Retinoid or Ascorbic acid, the patent application itself will be an essential technical element in the future because patent applications are filed in the field of a preparation of cosmetics for stabilizing ingredients, reducing skin irritability or promoting absorbance, and in the field of cosmetic formulation technology. As there are many studies on the causes of skin wrinkles, it is expected that new raw materials of cosmetics can be developed due to new mechanisms, and that the number of patent applications in new technologies will increase due to a change in the thought of cosmetics accompanied by the integration of cosmetics with biotechnology using Genetic Engineering, including the practical application of the medicine previously used far treating skin diseases to an anti-wrinkle agent and the mass production of active ingredients of cosmetics.

Life Cycle of the Perilla Leaf Pyralid Moth, Pyrausta panopealis (Lepidoptera: Pyralidae) and Test of Larvicidal Effect of Some Commercial Natural Products

  • The perilla leaf pyralid moth, Pyrausta panopealis Walker (Lepidoptera: Pyralidae), is a serious pest damaging to leaf perilla. In order to establish the life parameters of P. panopealis for eventual purpose of control, the developmental span of each stage was investigated under five temperature regimes ($20^{\circ}C{\sim}30^{\circ}C$). The larval period of P. panopealis was longest as 26.8 days at $20^{\circ}C$ and shortened as temperature goes up to $30^{\circ}C$ as 11.3 days. Survivorship of the larval P. panopealis was the highest at $27.5^{\circ}C$ as 82.5%, whereas that of other temperatures ranged from 40% ($20^{\circ}C$) to 60.0% ($30^{\circ}C$), indicating that the P. panopealis appears to favor somewhat higher temperature. In addition to larval period, the duration of egg, prepupal, and pupal period also were shortened sharply as temperature goes up, whereas the duration of adult stage shortened a maximum of only two days as temperature goes up. After the perilla leaf pyralid moths were successfully stabilized in indoor environment the larvicidal efficacy of ten on-the-market environmentfriendly agricultural materials (EFAMs) that were previously selected from the result of other moth species was tested. Seven of the ten tested showed more than 90% of mortality within 12 hrs and reached nearly up to 100% within 24 hrs, but the remaining three showed less than ~70%.
