The effect of Baeckji, Seasin, Baeckgangjam and Myunkanbang on skin-whitening (백지, 세신, 백강잠 및 면간방이 피부미백에 미치는 영향)

  • To develope new natural substances for whitening agent, Baeckji(Angelica dahuria;BK), Seasin(Asarum sieboldii;SS). Baecjgangjam(Bombycis corpus;BGJ) and Mynkanbang(MKB) were selected and extracted by hot water and 70% EtOH, respectively. Hydrothermal and ethanolic extracts of BK, SS, BGJ and MKB were evaluated for anti-oxidative effect, tyrosinase activity, melanogenesis of B16 melanoma cells and changes in level of tyrosinase expression by using Western blotting. All hydrothermal and ethanolic extracts showed scavenging activities of free radicals against 1,1-diphenyl-2-picrylhydrazyl(DPPH) but no inhibitory effect on tyrosinase activity. Ethanolic extracts of BK, SS, and BGJ and especially highly, those of MKB inhibited production of melanin in B16 melanoma cells and were able to reduce the level of tyrosinase expression in B16 melanoma cells. These results suggest that ethanolic extracts of BK, SS, BGJ and MKB can be an effective whitening agent from natural plant.

In Vivo Antifungal Activities of the Methanol Extracts of Invasive Plant Species Against Plant Pathogenic Fungi

  • Plants are the promising reservoirs for natural products with their diverse secondary metabolites. Many invasive plants have been introduced in Korea, which adversely affect on the native ecosystem but holds difficulty removing them due to their proliferation. In this study, we evaluated disease control efficacy of methanol extracts from four invasive plant species against 7 representative crop pathogens. Methanol extract of Phytolacca americana effectively suppressed rice blast, tomato gray mold, and tomato late blight in a dose dependent manner. The methanol extract of Amorpha fruticosa also exhibited potent antifungal activity against pepper anthracnose in a concentration dependent way. These data suggest that the extracts of P. americana and A. fruticosa can be developed as plant disease protection agents against rice blast, tomato gray mold, tomato late blight, and pepper anthracnose. Furthermore, more extensive research will be required to identify and isolate active compounds from problematic invasive plant species to develop valuable agrochemicals.

Combination of Grapefruit and Rosemary Extracts Has Skin Protective Effect through MMPs, MAPKs, and the NF-κB Signaling Pathway In Vitro and In Vivo UVB-exposed Model

  • Long-term ultraviolet (UV) exposure accelerates the phenomenon of skin photo-aging by activating collagenase and elastase. In this study, we aimed to investigate the effects of a combination of grapefruit and rosemary extracts (cG&Re) on UVB-irradiated damage in HaCaT cells and dorsal mouse skin. In HaCaT cells, cG&Re recovered UVB-reduced cell viability and inhibited protein expression of mitogen-activated protein kinases (MAPKs), such as extracellular signal-regulated kinases (p-Erk), c-Jun N-terminal kinases (p-JNK), and a class of MAPKs (p-P38). Also, cG&Re suppressed UVB-induced collagen and elastin degradation by decreasing matrix metalloproteinases (MMPs) and nuclear factor kappa light chain enhancer of activated B cells (NF-κB) expression, which is a transcription factor. Similar results were observed in dorsal mouse skin. Taken together, our data indicate that cG&Re prevent UVB-induced skin photo-aging due to collagen/elastin degradation via activation of MAPKs, MMPs, and the NF-κB signaling pathway in vitro and in vivo.

Antioxidative and Fibrinolytic Activities of Several Medicinal Plant Extracts (수종(數種)의 한약재(韓藥材) 추출물(抽出物)의 항산화능(抗酸化能)과 혈전용해능(血栓溶解能))

  • Objectives : This study was conducted to evaluate the antioxidative and fibrinolytic activity of the water and ethanol extracts from medicinal plants. Methods : Five kinds of medicinal plants(Carthami Flos, Glycyrrhizae Radix, Schisandrae Fructus, Atractylodes Rhizoma, Shiitake mushiroom) were extracted with distilled water and 70% ethanol, and the extracts were tested for their antioxidative and fibrilytic activities. Results : The highest polyphenol contents of the water and ethanol extracts from medicinal plants were 812.52 mg and 685.44 mg per 100 g of Carthamus tinctorius and Schizandra chinensis, respectively. The electron donating abilities (EDA) of the water extracts from all medicinal plants except Lentinus edodes were about 90% at 1,000 ppm and ethanol extracts were higher than those of water extracts. The highest SOD-like activity and nitrite scavenging abilities (NSA) were both of water and ethanol extracts from Schizandra chinensis. Five kinds of medicinal plants had fibrinolytoc activity and the highest activities were water and ethanol extracts from Glycyrrhiza uralensis. Conclusion : These results suggest that the medicinal plants can be used as natural antioxidant to prevent oxidative damage in normal cells probably because of their antioxidative and fibrinolytic activities.

Aldose reductase inhibitory activity of the methanol extracts from Korean folk plants (민속식물의 알도즈 환원효소 억제작용)

  • To search for the aldose reductase (AR) inhibitors from Korean folk plants, the inhibition of rat lens AR in vitro using the methanol (MeOH) extracts from Korean folk plants was investigated. Among fifty four Korean folk plants tested, the MeOH extract of Cedrela sinensis showed highest inhibition of AR ($IC_{50}$ value, 2.52 ${\mu}g/ml$). The plant C. sinensis has a possibility of new natural resources for the development of AR inhibitor for the prevention of diabetic complications.

Anti-inflammatory Effect of Natural Plant Extracts on in vitro Rumen Fermentation and Methane Emission (천연 식물 추출물의 항염 효과가 in vitro 반추위 발효성상과 메탄 생성에 미치는 영향)

  • This study was conducted to investigate the effects of anti-inflammatory plant extracts on the in vitro rumen fermentation characteristics and methane emission. Anti-inflammatory plant extracts from Morus bombycis Koidz, Mallotus japonicus L., Morus alba L., Paulownia coreana Uyeki, Isodon japonicus Hara and Ginkgo biloba L. were used in the study. The ruminal fluid(5 mL), McDougall buffer(10 mL), timothy as a substrate(0.3 g) and each anti-inflammatory plant extract(5% of substrate) were dispensed anaerobically into 50mL serum bottle. The mixtures were incubated for 3, 9, 12, 24, 48 and 72h at $39^{\circ}C$ without shaking. Supplementation of the anti-inflammatory plant extracts did not effects characteristics(pH, digestibility of dry matter, glucose concentration, ammonia concentration, protein concentration, VFA) on rumen fermentation. Total gas was showed a different pattern depending on treatments. Carbon dioxide was significantly(p<0.05) higher in Morus alba and Isodon japonicus than in control at 48h. Methane was significantly(p<0.05) lower in treatment than in control at initial fermentation. However the more incubation time was increased, the more methane emission was higher in treatment than in control. The concentrations of polyphenol and flavonoid were higher in Ginkgo biloba. In conclusion, supplementation of the anti-inflammatory plant extracts did not effect on rumen fermentation and methane emission was decreased in initial fermentation.

Inhibitory Effects of Natural Plant Extracts on ICAM-1/LFA-1 Mediated Adhesion of HL-60 Cells (자생식물 추출물의 세포접착인자 저해활성 검색)

  • Atherosclerosis is a progressive disease characterized by the accumulation of lipids and fibrous elements in the arteries. Monocyter/macrophages are involved in many aspects of the development of atherosclerotic plaques. It is known that the intercellular adhesion molecule-1(ICAM-1) expressed preferentially on endothelial cells of atherosclerotic plaque, promotes local adhesion and transendothelial migration of monocytes, neutrophils, and lymphocytes. Using the human promyelocytic leukemia HL-60 cell line, we investigated the inhibitory effects of methanol extracts of 175 natural plants on ICAM-1/LFA-1 mediated cell adhesion. Eight kinds of methanol extracts of tested plants inhibited PMA-induces homotypic aggregationof HL-60 cells without cytotoxicity at the concentration of $6.25\;{\mu}g/ml$. They were divided two fractions of $CHCI_3$ and $H_2O$ to use solvent partition. Among them, $CHCI_3$ extract $(1.0\;{\mu}g/ml)$ of Saururus chinensis and Chloranthus japonicus singificantly inhibited aggregation of HL-60 cells without cytotoxicity, respectively.

Screening of Natural Antimicrobial Plant Extract on Food Spoilage Microorganisms (식품 부패미생물의 증식을 억제하는 천연 항균성물질의 검색)

  • Certain parts of 36 kinds of plant were extracted by 75% ethanol and water. The extracts were tested their microbial inhibition activities against several food spoilage microorganisms, Bacillus subtilis, Lactobacillus plantarum, Leuconostoc mesenteroides, Pseudomonas fluorescens, Bacillus cereus and Saccharomyces cerevisiae. The ethanol extract of amur cork was shown inhibitory effect on all 6 species of the microorganisms tested. Chinese pepper, sesame cake, gromwell and oak were on 5 species except S. cerevisiae or P. fluorescens and bamboo leaves, lycopi herba, paulownia and rigida were on 4 species. In general amur cork exhibited the strongest inhibition with a few exceptions on certain species. By disc diffusion method, the ethanol extract of leaf mustard showed the highest inhibition effect on B. subtilis, amur cork on L.plantarum, L. mesenteroides and B. cereus, and amur cork and gallnut on P. fluorescens. Mostly the ethanol extracts in comparison with water extracts showed higher inhibition in most of plants but a few exhibited higher in water extracts.

Overview of Studies on the Use of Natural Antioxidative Materials in Meat Products

  • Studies conducted in the past decade related to the use of natural antioxidants in meat products revealed the prevalent use of plant-based antioxidative materials added as powders, extracts, or dried or raw materials to meat products. The amount of antioxidative materials varied from 7.8 ppm to 19.8%. Extracts and powders were used in small amounts (ppm to grams) and large amounts (grams to >1%), respectively. Antioxidative materials used in meat products are mainly composed of phenolic compounds and flavonoids, which are able to inhibit lipid peroxidation of meat products, thereby preserving meat quality. However, the main ingredients used in processed meat products are the traditional additives, such as sodium erythorbate, sodium hydrosulfite, and synthetic antioxidants, rather than natural antioxidants. This difference could be attributed to changes in the sensory quality or characteristics of meat products using natural antioxidants. Therefore, novel research paradigms to develop meat products are needed, focusing on the multifunctional aspects of natural antioxidants.

Antithrombotic Effects of Some Traditional Plant Medicines (수종 생약의 항혈전활성)

  • Twentyfive plant crude drugs with traditional reputation were selected for the antithrombotic screening. MeOH (80%) extract of each plant was given (500 mg/Kg, p.o.) to mice 1 hr prior to the thrombotic challenge (1.8 mg collagen plus 180 ug epinephrine/Kg, i.v.). Seven extracts (1, 2, 8, 10, 18, 22 and 23) showed either comparable or better recovery than aspirin (50 mg/Kg, p.o.) from paralysis caused by the pulmonary thrombosis. 1, 2, 8, 10, 18, 22 and 23 were fractionated to $CH_2Cl_2$ soluble (fr I) and $H_2O$ soluble (fr II) fractions and each fraction (100 mg/Kg, p.o.) was also tested. Mice treated with fr I of 2 (Angelica koreana), 10 (Corydalis spp.) or 23 (Schizonepeta tenuifolia) showed higher recovery and lower mortality rates than aspirin (50 mg/Kg, p.o.) treated mice.
