Screening for Chemosensitizers from Natural Plant Extracts through the Inhibition Mechanism of P-glycoprotein

  • P-gp plays a critical role in drug disposition and represents a mechanism for the development of multidrug resistance. Flavonoids, a major class of natural compounds widely present in foods and herbal products, have been shown to inhibit P-gp. Therefore, the aim of this study was to identify new candidate chemosensitizers by screening various plant extracts. The ability of natural plant extracts to inhibit P-gp activity was assessed by measuring cellular accumulation of calcein AM, daunorubicin and vincristine in P-gp overexpressing MDCKII-MDR1 cells. Among more than 800 plant extracts, eight were found to inhibit P-gp activity. Curcuma aromatica extract produced greatest inhibition, followed by Curcuma longa and Dalbergia odorifera extracts. Extracts of Aloe ferox, Curcuma zedoariae rhizome, Zanthoxylum planispinum, and Ageratum conyzoides showed moderate inhibitory effects. Curcumin and quercetin exhibited similar inhibition of P-gpmediated efflux of daunorubicin and vincristine, and flavones had a lesser effect. When chemosensitizing effect was evaluated by measuring daunorubicin sensitivity to MDCKII-MDR1 cells in the presence of natural plant extracts, Curcuma aromatica showed the most potent chemosensitizing effect based on daunorubicin cytotoxicity. In conclusion, natural plant extracts such as Curcuma aromatica can potently inhibit P-gp activity and may have potential as a novel chemosensitizers.

Antimicrobial activity of Mongolian medicinal plants

  • The antimicrobial activity of seventy five ethanol extracts obtained from 67 different kinds of plant species of the Mongolian flora were evaluated by means of the disc diffusion method against five species of microorganisms, Escherichia coli, Enterococcus faecalis, Staphylococcus aureus, Micrococcus luteus and Pseudomonas aeruginosa. Among the plant extracts examined, 34 kinds of extracts demonstrated significant antibacterial activity against one or more species of microorganisms, respectively. Especially, the root extract of Paeonia anomala, the whole herb extract of Myricaria alopecuroides, the whole herb extract of comarum zalesovianum, the whole herb extract of Agrimonia pilosa and some other plant extracts demonstrated a particularly potent antimicrobial activity. The ethylacetate fractions obtained from the whole herb extract of Myricaria alopecuroides and from those of Sedum aizoon, Paeonia anomala, Sedum hybridum and Dasiphora fruticosa exhibited a particularly potent antibacterial activity especially against Staphylococcus aureus and Micrococcus luteus.

구강 질환 유발 미생물에 대한 항생작용을 갖는 천연물 추출물 검색 (Screening of the Natural Plant Extracts for the Antimicrobial Activity on Dental Pathogens)

  • 구강청정제에 사용되는 많은 항미생물 성분들은 치주조직의 착색 및 작열감 등의 부작용을 가지고 있다. 이러한 취약점을 해결하고, 좀 더 생체친화적인 항미생물 물질을 찾기 위하여,한국식물추출물은행에서 소장하고 있는 2,125종의 식물추출물을 검색하였다. 항미생물 능력을 확인하기 위해, 잘 알려진 치과 병원균(치주질환 3종, 충치 1종)에 대한 성장억제 효과를 측정하였다. 대표적인 치주질환 유발세균인 Porphyromonas gingivalis에 대해서 19중의 식물 추출물이 항생효과를 나타내었으며, 17종이 Prevotella intermedia에 대해, 45종이 Haemophilus actinomycetemcomitans에 대해, 그리고 85종의 추출물이 충치 유발세균인 Streptococcus mutans 에 대해 효과를 나타내었다. 이중에서 총8종의 식물추출물이 실험에 사용한4종의 미생물 중 3종 이상에 대해 항생작용을 나타내었다. 또한 Cudrania tricuspidata, Morus bombycis와 Mallotus japonicas가 실험에 사용된 4종의 치과 병원균 모두에 대해 항생작용을 나타내어 치과용 재료 혹은 양치액에 항미생물 물질로 사용될 가능성을 나타내었다.

Aldose Reductase Inhibitory Activity of Methanol Extracts from the Korean Plants

  • We examined methanol extracts prepared from the Korean plants for their inhibitory effects on rat lens aldose reductase (RLAR) activity in vitro. Among 41 plant extracts, the extracts of Lagerstroemia indica, Punica granatum, Eurya japonica, Liquidambar styraciflua, and Vitis coignetiae exhibited highest inhibitory potency, even more potent than tetramethylene glutaric acid (TMG), known as one of typical aldose reductase inhibitors $(IC_{50}\;value,\;0.51\;{\mu}g/ml)$. Especially, the extract of Lagerstroemia indica (Lythraceae), exhibited the most inhibitory potency $(IC_{50}\;value,\;0.069\;{\mu}/ml)$ on RLAR.

Evaluation of the Antioxidant Potential of Natural Products Mediated by Inhibition of Xanthine Oxidase Activity

  • Since reactive oxygen species (ROS) play an important role in carcinogenesis and other several human diseases, antioxidants present in consumable fruits, vegetables, and beverages have received considerable attention as cancer chemopreventive agents. Thus, in order to identify antioxidants in plant extracts, potential activity was assessed by determining with inhibition of a xanthine/xanthine oxidase assay system. Approximately 170 plant extracts of Korean herbal medicines were primarily evaluated for the potential of antioxidant activity. As a result, 13 plant extracts were found to be active $(IC_{50}$<$100\;{\mu}g/ml)$. Especially, Juncus effusus, Selaginella tamariscina, Pueraria thunbergiana and Sedum albroseum showed strong inhibitory activity in this process. Further studies for the identification of active principles from these active lead plant extracts might be warranted.

식물성 천연 추출액을 첨가한 고추장의 특성 (Studies on the Properties of kochujang by Addition of Natural Plant Extracts)

  • 식물성 천연 추출액을 첨가하여 60일간 숙성시킨 고추장과 일반 고추장의 유리 아미노산 분석 결과, cysteine, isoleucine, tyrosine, lysine을 제외한 나머지 유리 아미노산들의 함량이 일반 고추장에 비해 식물성 천연 추출액을 첨가한 고추장에서 높게 나타났고, 식물성 천연 추출액을 첨가한 고추장에서 감칠맛의 주요 성분인 gutamic acid 함량이 높게 증가함을 나타내었다. 한편 glutamic acid 외에도 aspartic acid, proline, valine, histidine등의 함량이 식물성 추출액을 첨가한 고추장에서 높았다. 식물성 천연 추출액을 첨가한 고추장의 관능적 특성에 미치는 색, 향, 구수한 맛, 매운맛, 단맛, 짠맛, 전체적인 기호도를 조사한 결과, 일반 고추장에 비해 향, 구수한 맛, 단맛에서 높은 값을 보였고, 색이나 매운 맛, 짠맛은 거의 비슷한 값을 나타내었다. 전체적인 기호도에서 일반 고추장에 비해 기호도가 높은 것을 볼 수 있었다. 따라서 고추장에 식물성 천연 추출액을 첨가하여 숙성시키는 것이 고추장의 구수한 맛, 향, 단맛을 향상시킬 뿐만 아니라 천연물의 한방적인 효능이 있을 것으로 생각된다.

Methanol Extract of Cinnamomum cassia Represses Cellular Proliferation and Gli-mediated Transcription in PANC-1 Human Pancreatic Cancer Cells

  • Twenty five methanolic plant extracts were investigated to determine the anticancer activity against sonic hedgehog (shh)/Gli signaling pathway dependent cancer, PANC-1 human pancreatic cancer cells, through three screening programs. All extracts were inspected their inhibitory properties on sonic hedgehog-conditioned medium (shh-CM) induced alkaline phosphatase (ALP) activity in C3H10T1/2 mouse mesenchymal stem cells to examine whether the plant extracts affect the shh/Gli signaling pathway. Next, plant extracts were screened the ability to suppress the cell proliferation of PANC-1 human pancreatic cancer cells. Finally, active plant extracts from the two screening systems were evaluated for the suppressive effect on Gli-mediated transcriptional activity in PANC-1 cells. Among active plants, Cinnamomum cassia suppressed Gli-mediated transcriptional activity leading to the down-regulated expression of Gli-target genes such as Gli-1 and Patched-1 (Ptch-1). This study provides the consideration for the important role of natural products in drug discovery process as well as the basis for the further analysis of active plant and potential identification of novel bioactive compounds as inhibitors of Gli and therapeutic candidates against shh/Gli signaling pathway dependent cancers.

치주염 원인균에 대한 천연 식물 추출물의 항균효과 (Antimicrobial Effect of Natural Plant Extracts against Periodontopathic Bacteria)

  • 이 연구는 34종의 천연 식물 추출물을 이용하여 치주염 원인균 Actinobacillus actinomycetemcomitans와 Prevotella intermedia에 대한 항균효과를 조사하였다. 이에 본 연구는 치주염 원인균에 대한 천연 식물 추출물의 항균효과를 연구하고자 유기용매 추출물을 이용하여 생육저지활성을 측정하고, 시료 추출물의 최소생육저해농도(Minimum Inhibition Concentration, MIC)를 측정한다. 1차적으로 34종의 각 천연 식물 추출물을 에탄올로 추출하여 디스크 확산법(disk diffusion method)을 이용하여 각 추출물의 생육저지환(clear zone, mm)의 크기를 측정한 결과 나복자, 목통, 백작약, 사간, 선복화, 어성초, 연교, 진교, 천련자, 황금, (천)황련, 황백, 해동피 등 13종에서 천연 식물 추출물이 치주염의 원인균인 A. actinomycetemcomitans에 대해서 생육저지 활성을 나타내었다. P. intermedia의 경우, 나복자, (일)당귀, 목통, 사간, 선복화, 어성초, 육계, 자완, 천련자, 황금, (천)황련, 황백, 해동피 등 13종에서 생육저지 활성을 나타내었다. A. actinomycetemcomitans에 대해서는 사간, 육계, 해동피, 황백, (천)황련에서 항균효과가 나타났다. (천)황련은 A. actinomycetemcomitans의 모든 유기용매 분획에서 가장 우수한 생육저지 활성을 나타내었다. P. intermedia는 사간, 육계, 천련자, 황백, 황련에서 생육저지 활성이 나타났다.

Inhibition of Aldose Reductase from Rat Lenses by Methanol Extracts from Korean Folk Plants

  • The inhibition of rat lens aldose reductase (AR) in vitro using methanol extracts from Korean folk plants was investigated. Among them, the extracts of Saussurea grandifolia and Rumex crispus showed highest inhibition of AR. $IC_{50}$ values of the extracts from S. grandifolia and R. crispus were demonstrated 0.07 and 0.05 mg/ml, respectively. Korean folk plants such as S. grandifolia and R. crispus has a possibility of new natural resources for the inhibition of AR.

천연 식물추출물과 감마선이 막걸리에서 분리한 유산균에 미치는 영향 연구 (Effects of Natural Plant Extracts and Gamma Rays on Lactobacillus Isolated from Korean Traditional Raw Rice Wine)

  • Recently, Korean traditional raw rice wines (RRW) have received attention because they are a nutritious food with health properties. But the rapid deterioration of fermented RRW is one of the serious problems for brewing and marketing in the world beyond Korea. The goal of this study was to develop a way to enhance the quality and to lengthen the period of circulation of the RRW. A lactic acid bacterium was isolated from RRW. It was identified as Lactobacillus fermentum (98%) based on its biochemical properties, and 16S rRNA sequence. Treatments of RRW with gamma radiation and green tea extracts reduced the bacterial population except for yeasts and Lactobacillus in the RRW. This result suggested that the natural plant extracts and catechin products can be used as an effective natural storage agent.
