Inhibitory Effect of Rosa laevigata on Nitric Oxide Synthesis and $NF-{\kappa}B$ Activity in lipopolysaccharide-stimulated Macrophages (lipopolysaccharide로 자극된 대식세포에서 금앵자의 Nitric Oxide 생성 및 $NF-{\kappa}B$ 활성 억제 효과)

  • Nitric oxide (NO) has been suggested to play an important role in endotoxin-mediated shock and inflammation. In this study, we investigated the effect of Rosa laevigata Michx. (Rosaceae) on the production of NO and the molecular mechanism of its action. Rosa laevigata inhibited NO generation and iNOS expression in LPS-stimulated murine macrophages. Activity of nuclear $factor{-\kappa}B\;(NF{-\kappa}B)$ and the degradation of $I{\kappa}B-{\alpha}$ were suppressed by Rosa laevigata. Furthermore, extracellular signal-stimulated kinase (ERK), which is known to be involved in $NF{-\kappa}B$ activation, is inhibited by Rosa laevigata. These results suggest that Rosa laevigata could exert its anti-inflammatory actions by suppressing the synthesis of NO through inhibition of $NF{-\kappa}B$ activity.

Inhibitory Effect of Esculetin on the Inducuble Nitric Oxide Synthase Expression in TNF-stimulated 3T3-L1 Adipocytes

  • While nitric oxide (NO) produced by inducible nitric oxide synthase (iNOS) is beneficial for host survival, it is also detrimental to the host. Thus, regulation of iNOS gene expression may be an effective therapeutic strategy for the prevention of unwanted reactions at various pathologic conditions. During the screening process for the possible iNOS regulators, we observed that esculetin is a potent inhibitor of cytokine-induced iNOS expression. The treatment of 3T3-L1 adipocytes with the tumor necrosis factor-${\alpha}$ (TNF) induced iNOS expression, leading to enhanced NO production. TNF-induced NO production was inhibited by esculetin in a dose-dependent manner. Esculetin inhibited the TNF-induced NO production at the transcriptional level through suppression of iNOS mRNA and subsequent iNOS protein expression. These results suggest esculetin, a component of natural products, as a naturally occurring, nontoxic means to attenuate iNOS expression and NO-mediated cytotoxicity.

Yomogin, an Inhibitor of Nitric Oxide Production in LPS-Activated Macrophages

  • In activated macrophages the inducible form of nitric oxide synthase (i-NOS) generates high amounts of toxic mediator, nitric oxide (NO) which contributes to the circulatory failure associated with septic shock. A sesquiterpene lactone compound (yomogin) isolated from medicinal plant Artemisia princeps Pampan inhibited the production of NO in LPS-activated RAW 264.7 cells by suppressing i-NOS enzyme expression. Thus, yomogin may be a useful candidate for the development of new drugs to treat endotoxemia and inflammation accompanied by the overproduction of NO.

  • Poly(dimethyl siloxane) (PDMS) has been employed as a microchip material for DNA separation in microfluidic condition. Different sieving molecules such as cellulose derivatives having glucose building block (methyl cellulose (MC), hydroxyethyl cellulose (HEC), and hydroxypropyl methyl cellulose (HPMC)) and polyethylene oxide (PEO) having linear (ring-opened ethylene oxide) unit were used and their performance was compared in terms of separation efficiency and resolution. In general, PEO showed better separation performance than cellulose derivatives probably due to the nature of linear shape polymer conformation. It was possible to perform at least 15 consecutive running with 1.2% PEO at the electric field strength around 200 V/cm. Fast analysis of the standard $\Phi$X 174 RF DNA/Hae III (less than 130s) was obtained with the number of the theoretical plate around 250,000/m. Our PMDS microchip was applied to the measurement of CAG repeat number, which is related to male infertile disease.

Heavy Metal Removal from Drinking Water using Bipolar Surface Modified Natural Mineral Adsorbents (천연광물의 양극성 표면개질을 이용한 상수원수 중 중금속제거 특성)

  • Objectives: The most commonly detected heavy metals in rocks and soils, including Pb, Cd, Cu, Fe, Mn and As, are representative pollutants discharged from abandoned mines and have been listed as potential sources of contamination in drinking water. This study focused on increasing the removal efficiency of heavy metals from drinking water resources by surface modification of natural adsorbents to reduce potential health risks. Methods: Iron oxide coating and graft polymerization with zeolites and talc was conducted for bipolar surface modification to increase the combining capacity of heavy metals for their removal from water. The removal efficiency of heavy metals was measured before and after the surface modification. Results: The removal efficiency of Pb, Cu, and Cd by surface modified zeolite showed 100, 92, and 61.5%, respectively, increases compared to 64, 64, and 38% for non-modified zeolite. This implies that bipolar surface modified natural adsorbents have a good potential use in heavy metal removal. The more interesting finding is the removal increase for As, which has both cation and anion characteristics showing 27% removal efficiency where as non-modified zeolite showed only 2% removal. Conclusions: Zeolite is one of the most widely used adsorptive materials in water treatment processes and bipolar surface modification of zeolite increases its applicability in the removal of heavy metals, especially As.

Pseudoguaianolides Isolated from Iunla britannica vats. chinenis as Inhibitory Constituents against Inducible Nitric Oxide Synthase

  • Three pseudoguaianolide type sesquiterpenes, bigelovin(1), 2,3-dihydroaromaticin (2), and ergolide (3) were isolated as inhibitory constituents against inducible nitric oxide synthase (iNOS) from the flowers of Inula britannica var. chinensis. Bigelovin (1) exhibited a highly potent inhibitory activity on lipopolysaccharide (LPS)-induced iNOS in murine macrophage RAW 264.7 cells with an $IC_{50}$ value of 0.46 mM, which is about 8 times more potent than the known selective inhibitor of iNOS, $L-N^6-(1-iminoethyl)Iysine{\;}(IC_{50}{\;}3.49{\;}{\mu}M)$. 2,3-Dihydroaromaticin (2) and ergolide (3) also exhibited potent inhibitory activities on LPS-induced iNOS with $IC_{50}$ values of 1.05 and $0.69{\;}{\mu}M$, respectively.

  • The objective of this study is screening the anti-oxidative effects of several plant MeOH extracts against oxidative stress in Neuroblastoma cell. Oxidative stress has been implicated in the pathogenesis of many neurotoxicity, neurodegenerative disorders and cell death. This oxidative stress is generated by ROS (Reactive Oxygen Species) such as nitric oxide, nitrogen dioxide, peroxyl, superoxide ($O_2^-$), hydroxyl, alkoxyl. So, in the present study, we induced oxidative stress by treatment of sodium nitroprusside (2.5 mM) in human neuroblastoma SH-SY5Y cell which was treated samples before 24hr, and cell viability was measured by MTT reduction assay. Of those tested, the extracts of Paeonia japonica (roots), Eucommia ulmoides (炒)(barks), Paeonia japonica (曝乾)(roots), Phyllostachys bambusoides (stems), Polygala tenuifolia (去心, 炒)(roots), Paeonia japonica (roots), Polygala tenuifolia (roots), Machilus thunbergii (barks), Mallotus japonicus (leaves), Poria cocos (whole), Sophora flavescens (roots), Angelica tenuissima (roots), Angelica gigas (當歸尾)(roots) showed anti-oxidative effects[$EC_{50}$<15.20 ${\mu}g$/ml(Carnosine:Positive control)]in dose dependent manner.
