양조간장에서 분리한 갈색물질의 항산화성 (Antioxidative Activity of Browning Products Fractionated from Fermented Soybean Sauce)

  • 동결건조한 양조간장의 분말로 부터 갈색물질을 Sephadex G-10으로 겔-여과 크로마토그라피법에 의하여 분획한 다음 이를 동결건조하여 분말로 만든 후, 동갈색물질의 항산화력을 다른 항산화제인 부틸히드록시아니졸(BHA) 및 ${\alpha}-토코패롤과$ 비교하여 보았다. 갈색물질 및 양조간장은 다 같이 지방산의 산화반응에 있어서 과산화물의 생성을 크게 저해시켰고, 갈색물질이 양조간장 자체보다 항산화 효과가 더 높았으며 농도에 비례하여 항산화성이 증대되었다. 그러나 갈색물질의 항산화력은 과산화물 생성억제 능력 그리고 conjugated dienoic acid 생성저해 능력 등에서 BHA 및 ${\alpha}-토코페롤$ 보다는 낮았다. 이와같은 양조간장의 갈색물질의 항산화성은 그 획분이 대량 섭취도고 있는 발효조미식품이라는 점에서 중요한 의의를 가질 것으로 판단된다.

Potential of Benthic Diatoms Achnanthes longipes, Amphora coffeaeformisand Navicula sp. (Bacillariophyceae) as Antioxidant Sources

  • Recently, interest in plant-derived food additives has developed natural antioxidants, in order to alternate syn-thetic antioxidants with several disadvantages. In the present study, different organic fractions from solvent parti-tions of 80% methanol extract from Jeju benthic diatoms, Achnanthes longipes, Navicula sp. and Amphora coffeaeformis was assessed for their potential antioxidant effects. Among the solvent fractions tested, n-hexane (80.4%) and 80% methanol extract (76.6%) from A. longipes, chloroform (63.2%) from Navicula sp. and n-hexane (67.4%) from A. cof-feaeformis were effective in DPPH free radical scavenging. Fractions of chloroform (53.4%) and n-hexane (53.1%) from A. longipes exhibited higher activities on $H_2O_2$ scavengin. Fraction of n-Hexane from A. longipes exhibited the highest hydroxyl radical scavenging activity and NO. scavenging activity (56.5% and 75.6%, respectively). Aqueous residue from A. coffeaeformis (75.6%) showed the highest metal chelating effect. chloroform and ethyl acetate frac-tion of all the diatoms exhibited significant antioxidant activities in lipid peroxidation inhibitory activity. In particu-lar, both chloroform and the ethyl ecetate fraction from A. longipes and A. coffeaeformis exhibited lipid peroxidation inhibitory activity significantly higher than that of $\alpha$- tocopherol. These data suggest that the Jeju benthic diatoms tested are rich in hydrophilic and hydrophobic antioxidative compounds with different antioxidative properties that can be applied in food industry.

수 종의 국화과 식물의 항산화 쵸과 (Antioxidative Effects of Several Compositae Plants)

  • 전남 순천지역 들판에서 채취한 수종의 국화과 식물의 항산화 능력을살펴보았다. Rancimat와 TBA 방법을 이용하여 국화과 식물 중 항산화 효과가 높게 나타난 서양민들레, 조뱅이, 쑥을 선발하였다. 유지 산화안정도에 대한 실험에서 BHT와 ascorbic acid가 각각 3.86과 2.43 AI(Antioxidative Index)를 나타냈고 한련초 잎이 1.51 AI를 나타냈다. 이것은 서양민들레 잎이 유지산화안정도에 높은 항산화 효과를 나타냄을 의미한다. 한편 건조된 분쇄물과 메탄올 추출물을 이용한 장기간의 산화 억제에 대한 TBA실험에서 저장 4주 째에 BHT, ascorbic acid가 각각 0.05와 0.09 TBARS치를 나타낸 반면, 서양민들레 잎은 0.12의 수치를 나타내 대조구 1.01 TBARS치 보다 월등히 낮은 수치를 보여 우수한 항산화 효과를 보였다. 이상의 결과로 서양민들레, 조뱅이, 쑥 등의 국화과 식물에서 비교적 높은 항산화 활성을 확인할 수 있었다. 특히 이 식물체들은 기존의 인공 항산화제에 대한 하나의 대체수단으로서 식물유래 천연 항산화제로의 개발 가능성 을 시사해 주었다.

In Vitro Antioxidant Activity Profiles of ${\beta}$-Glucans Isolated from Yeast Saccharomyces cerevisiae and Mutant Saccharomyces cerevisiae IS2

  • To explore the possible usefulness of ${\beta}$-glucans as natural antioxidants, the antioxidant profiles of ${\beta}$-glucan, extracted from Saccharomyces cerevisiae KCTC 7911, and water soluble and insoluble mutant ${\beta}$-glucan, isolated from yeast mutant S. cerevisiae IS2, were examined by five different in vitro evaluation methods: lipid peroxidation value (POV), nitric oxide (NO), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, reducing power, and ${\beta}$-carotene diffusion assay. The antioxidant activities of all ${\beta}$-glucans evaluated in POV test were comparable to or better than that of the known antioxidant, vitamin C. Remarkably, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan possessed 2.5-fold more potent activity than vitamin C at a dosage of 2 mg. Although vitamin C showed 100-fold greater activity than all ${\beta}$-glucans in NO and DPPH tests for measuring the radical scavenging capacity, all ${\beta}$-glucans revealed higher radical scavenging activity than the known radical scavenger, N-acetyl-L-cysteine (NAC), in DPPH test. The water insoluble mutant ${\beta}$-glucan had 2.6- and 5-fold greater antioxidative activity than water soluble ${\beta}$-glucan in NO and DPPH tests, respectively, showing that all ${\beta}$-glucans were able to scavenge radicals such as NO or DPPH. While all ${\beta}$-glucans revealed lower antioxidant profiles than vitamin C in both reducing power activity and ${\beta}$-carotene agar diffusion assay, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan did show a marginal reducing power activity as well as a considerable ${\beta}$-carotene agar diffusion activity. These results confirmed the potential usefulness of these ${\beta}$-glucans as natural antioxidants.

마치현 열수 및 에탄올 추출물의 항산화 활성 (Antioxidant Activities of Hot Water and Ethanol Extracts from Portulacae Herba)

  • Objectives : This study aims to provide basic data about Portulacae Herba (PH) extracts as natural antioxidants by considering diverse antioxidant activities of PH depending on solvents. Methods : The samples of PH were pulverized, and A hot water and a 70% EtOH were stir-extracted for two hours three times repeatedly in a water bath with a temperature of 95 degrees and at room temperature respectively to measure 7 kinds of antioxidant activities. Results : There were significant differences in total phenol content, because the total phenol content of the 70% EtOH extract was higher than the hot water extract's, and the total flavonoid content of the 70% EtOH extract($4.40{\mu}g/mg$) was nearly 3.8 times higher than the hot water extract's($1.16{\mu}g/mg$). DPPH and ABTS radical scavenging activities were 70% EtOH extract showed a little higher activity than the hot water extract, and at a concentration of $500{\mu}g/mg$, the highest scavenging activity was found in the 70% EtOH extract, not in the control group. Hydroxyl radical and $Fe^{2+}$ chelatingactivities were slightly higher in hot water extract than in 70% EtOH extract, and increased in a dose-dependent manner. Nitrite scavenging activities increased dose-dependently in the hot water and the 70% EtOH extract, regardless of the pH level, and scavenging activity of the 70% EtOH extract was higher at pH 1.2 than at pH 3.0. Conclusions : In conclusion, it is thought that PH hot water and 70% EtOH extract have antioxidant activities, and can be used as natural antioxidants in future.

비스페놀 A에 대한 기수산 물벼룩의 항산화 시스템의 변화 (Modulation of antioxidant defense system in the brackish water flea Diaphanosoma celebensis exposed to bisphenol A)

  • Bisphenol A (BPA)는 대표적인 내분비계장애물질로 수서생물의 성장, 발생, 그리고 생식에 유해한 영향을 주는 것으로 알려져 있다. 본 연구는 기수산 물벼룩(Diaphanosoma celebensis)에서 48시간 BPA 노출 후 산화적 스트레스 반응을 조사하기 위하여 qRT-PCR을 이용한 항산화 유전자 발현 변화, 항산화 효소 활성, 총 단백질 함량 분석을 수행하였다. $3mg\;L^{-1}$의 BPA에 48시간 노출된 D. celebensis에서 모든 항산화 유전자(Cu/Zn-SOD, Mn-SOD, CAT)의 발현량이 유의하게 증가하였다. 특히 세 종류의 GST isoforms (GST-kappa, GST-mu, GST-theta)는 가장 낮은 농도인 $0.12mg\;L^{-1}$ BPA에 48시간 노출된 실험군에서도 유의하게 증가하였으며, GST-mu의 발현양이 상대적으로 가장 높게 나타났다. SOD 활성은 BPA 농도에 의존적으로 유의하게 증가하였으며, 총 단백질 함량은 BPA 노출에 대해 감소되는 양상을 보였다. 이러한 결과는 BPA가 D. celebensis에서 산화적 스트레스를 유발하였고, 이 과정에서 이들 항산화 유전자가 생물방어기전으로 참여한다는 것을 의미한다. 본 연구는 BPA가 해양생물에 미치는 영향에 대한 분자적 기전을 이해하는 데 도움이 될 것이다.

Influence of Roasting Treatment on the Antioxidant Activities and Color of Burdock Root Tea

  • The major trend in the antioxidant market is the growing consumer demand for natural antioxidants. Tea, one of the most widely consumed beverages in the world, is an easy way to obtain antioxidant components from a natural source. Our objective was to develop burdock root tea (BRT) with potent antioxidant activity and good color quality. In order to obtain maximum antioxidant activity and quality, the effect of roasting was determined. The antioxidant capacities and total phenolic contents of BRT increased as roasting increased. The color of BRT became darker with increased roasting, extraction time, and amount of burdock roots. Color of BRT was also positively correlated with total antioxidant capacity. Roasting significantly enhanced the total antioxidant activities and color quality of BRT. These results suggest that roasting BRT increases beneficial antioxidant components from burdock roots.

Ethanol Extracts from Mistletoe (Viscum album L.) Act as Natural Antioxidants and Antimicrobial Agents in Uncooked Pork Patties during Refrigerated Storage

  • The antioxidant potential of mistletoe (Viscum album L. var. coloratum Ohwi; VAL) extract in uncooked pork patties was evaluated. Three concentrations of VAL extract (0.1 [T1], 0.5% [T2] and 1.0% [T3]) along with 0.02% ascorbic acid as a positive control (V) were added to ground pork and pork patties were prepared. Incorporation of VAL extract decreased (p<0.05) the pH of the pork patties throughout the storage time and reduced (p<0.01) the thiobarbituric acid reactive substance values after day 14 of storage. Total plate counts of the VAL extract-treated samples and V-treated samples were also significantly lower (p<0.01) than that of the control (C) throughout the storage period. In addition, odor scores of the VAL extract-treated patties were lower than those of the C- or V-treated samples on 3rd day of the storage period. These results demonstrated that the VAL extract acts as a natural antioxidant in uncooked pork products.

  • The environmental pollutions were a serious problem in Korea recently. So many researcher have studied the effect of environmental pollution on plants and agro-ecosystem, but the basic mechanisms of environmental stresses were various. One of the important mechanisms was oxidative stress caused by active toxic oxygen. The toxic oxygen was generated by several stresses, abnormal temperature, many xenobiotics, air pollutants, water stress, fugal toxin, etc. In the species of toxic oxygen which is primary inducer of oxidative stresses, superoxide, hydrogen peroxide, hydroxyl radical and singlet oxygen were representative species. The scavenging systems were divided into two groups. One was nonenzymatic system and the other enzymatic system. Antioxidants such as glutathione, ascorbic acid, and carotenoid, have the primary function in defense mechanisms. Enzymatic system divided into two groups; First, direct interaction with toxic oxygen(eg. superoxide dismutase). Second, participation in redox reaction to maintain the active antioxidant levels(eg. glutathione reductase, ascorbate peroxidase, etc.).
