Antioxidant Activity of Peanut Flours with Germination and Roasting (볶음 및 발아처리한 땅콩분말의 항산화 활성)

  • The purpose of this study was to evaluate the antioxidant activity of roasted and germinated peanut flours. This study also aims to utilize it as a functional material to be applied to processed foods. The moisture, crude protein, crude fat and ash carbohydrate contents of the common peanut powder used in this study were 1.27, 25.63, 42.19, 2.38, 28.20 g / 100 g, respectively. The moisture content, crude protein, crude fat and ash carbohydrate in germinated peanut powder were 1.47, 25.86, 42.86, 2.25 and 26.66 g / 100 g, respectively. 26.52, 45.02, 2.33, 24.70, g / 100 g, and the dietary fiber content of peanut, roasted peanut and germinated peanut powder was 12.27, 13.05 and 14.22 g / 100g, respectively. The antioxidants and radical scavenging ability of polyphenols and flavonoids in peanut powder treated with germination and germination compared to ordinary peanuts. Resverasterol content was high in the germinated peanut powder. Especially, germinated peanut powder can act as a natural antioxidant.

Enhancing the Effect of Aronia Extract on Hyaluronic Acid Synthesis through Liposome Formation

  • Background and objective: Aronia melanocarpa, called black chokeberry, is a natural product belonging to the family rosaceae, and is known to contain polyphenolic antioxidants including cyanidin-3-galactoside, cyanidin-3-arabinoside, cyanidin-3-xyloside, and cyanidin-3-glucoside Because of the abundance of anthocyanins, Aronia has been studied to be used in various industries. Methods: Aronia melanocarpa extract was treated 24 hours a day to RAW 264.7 cells with inflammations induced by LPS. After extracting total RNA, the amount of inflammatory cytokine expression was measured using RT-PCR. After processing the Aronia liposome using Aronia extract and the layer-by-layer electrostatic deposition method in keratinocyte cells at the same time, we checked the synthesis of Hyaluronic acid enhanced through the formation of Aronia liposome using ELISA. Results: The treatment of Aronia extract in inflammation-induced RAW 264.7 cells conducted to check the anti-inflammatory efficacy of Aronia extract inhibited inflammatory cytokines including TLR4, TNF-α, IL-1β, COX-2, and iNOS and increased the mRNA expression of HAS2 genes related to moisturizing. Based on the anti-inflammatory and moisturizing effect of Aronia extract, the Aronia liposome technology was introduced to Aronia extract to produce Aronia liposome. Conclusion: The liposome formation of Aronia extract is expected to be used as a functional material in treating various inflammatory skin diseases by controlling the moisture content of the corneocytes by increasing the expression rate of genes associated with the synthesis of hyaluronic acid, while retaining the efficacy of its components.

Anti-inflammatory Activity of the Methanol Extract from the Stem of Coriandrum Sativum in RAW 264.7 Cells

  • Objectives : Coriandrum sativum is a medicinal herb that is used to enhance organoleptic quality and food flavor and as source of natural antioxidants. This research investigated the anti-inflammatory activity of Coriandrum sativum stem methanol extract (CSSE) using RAW 264.7 cells. Methods : Production of tumor necrosis factor-${\alpha}$(TNF-${\alpha}$), interleukin (IL)-$1{\beta}$, IL-6, and nitric oxide (NO) in the culture supernatant, protein expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and nuclear factor-kappa B (NF-${\kappa}B$) in the extract were assayed. Results : Treatment with CSSE ($100{\mu}g/m{\ell}$) resulted in inhibited levels of protein expression of lipopolysaccharide- (LPS-) induced iNOS, COX-2, and NF-${\kappa}B$ as well as production of TNF-${\alpha}$, IL-$1{\beta}$, IL-6, and NO induced by LPS. Conclusions : These results demonstrate that CSSE exhibits anti-inflammatory activities via decreasing production of pro-inflammatory mediators through suppression of the pathways of NF-${\kappa}B$ in LPS-induced RAW 264.7 cells. Thus, CSSE may have therapeutic potential for a variety of inflammation-mediated diseases.

Quantitative Analysis of Antioxidants in Sesame Seed (참깨 종실의 항산화 성분 정량분석 연구)

  • This study was conducted to obtain basic informations on the lignan components from sesame seed. Two major lignans, sesamin and sesamolin, were isolated and identified by means of spectral methods, and quantitative analysis was by HPLC from sesame variety Danbaeggae. Separation was achieved by isocratic elution and reversed phase chromatography Develosil ODS column. The content of the major lignan components were about 0.42% and 0.30% for sesamin and sesamolin, respectively.

Enzymatic preparation and antioxidant activities of protein hydrolysates from hemp (Cannabis sativa L.) seeds

  • Hemp (Cannabis sativa L.) seeds have recently been attracting attention as a new high-value-added food material owing to their excellent nutritional properties, and research on the development of functional food materials using hemp seeds is actively progressing. This study aimed to evaluate the antioxidant properties of hemp seed protein hydrolysates. Protein hydrolysates were prepared from defatted hemp seed powder (HS) by enzymatic hydrolysis using five different proteases (alcalase, bromelain, flavourzyme, neutrase, and papain). 2,4,6-trinitrobenzene sulfonic acid (TNBS) assay and SDS-PAGE analysis revealed that HS showed a high degree of hydrolysis after treatment with each enzyme except papain. The total polyphenol content of the protein hydrolysates (<3 kDa) and the RC50 values obtained from two different antioxidant tests showed that alcalase hydrolysate (HSA) had a relatively high level of antioxidant capacity. In addition, treatment with HSA (25-100 ㎍/mL) significantly inhibited linoleic acid peroxidation. These results suggest that hemp seed protein hydrolysates are potential sources of natural antioxidants. Future studies will focus on the identification of active peptides from HSA.

Comparison of in vitro antioxidant capacities of Phragmites communis Trin. and Phragmites japonica Steud.

  • The rhizome of Phragmites communis Trin. is used for vomiting and belching by clearing stomach and the sprout is used as tea. Phragmites japonica is similar with P. communis except the color of sheath is purple. This study is aimed to compare the in vitro antioxidant activity, total polyphenol and flavonoids contents of P. communis and P. japonica. The antioxidant activities of fractions from the two Phragmites plants were evaluated by 1,1-diphenyl-2- picrylhydrazyl (DPPH) and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging assay. The antioxidant activity varied with plant parts and extract solvents. The fractions of leaf extract from the two Phragmites plants (4.06±1.32-16.47±1.28%) showed higher antioxidant activity by DPPH assay compared with rhizome fractions of two Phragmites plants (0.00±0.00-14.15±0.07%), these are lower compared with ascorbic acid and butylhydroxyanisole (BHA). The highest ABTS radical scavenging activity was found for rhizome ether fraction, namely 74.95±0.56% and 73.04±1.85% for P. communis and P. japonica, these are higher than BHA. The total polyphenol and flavonoids contents were different with plant parts and extract solvents, likewise antioxidant activity. A significant correlation was shown between DPPH and ABTS radical scavenging activity. Considering the results of this study, the leaves and stems of P. communis and P. japonica are expected to be used as natural antioxidants.

Physicochemical characteristics and volatile profile of novel lemon varieties, Minimon and Jeramon

  • Although most lemons are imported into Korea, their consumption is increasing. Development of domestic varieties of Jeramon and Minimon is currently underway in an effort to meet the increasing demand for fresh lemons. In this study, an analysis of the physicochemical characteristics of Jeramon and Minimon, including their flavor components and antioxidant properties, was conducted. The results revealed that these new varieties of Korean lemon can be regarded as good sources of antioxidants and phytochemicals. Compared to Sunkist, the most consumed lemon variety in Korea, Minimon contained more than twice as much β-cryptoxanthin, and the content of L-ascorbic acid was more than three-four-fold higher in both Minimon and Jeramon. In addition, results from measurement of DPPH and ABTS radical scavenging activities indicated that Jeramon extract exhibited the highest antioxidant activity. In the volatile profile analysis, the profiles of volatile components showed high similarity among the three lemon samples, and the composition ratio was largely dominated by terpenoids. A markedly higher ratio of d-limonene and thymol was detected in the Minimon variety compared with that in the other two varieties. Collectively, the findings from this study on Korean lemon varieties provide a basis as well as valuable guidance for breeding domestic lemon varieties.

Antioxidant Activity, Total Phenolic Content, Total Flavonoid Content, and Total Anthocyanin Content of Vaccinium oldhamii Miq. Collected from 11 Regions of South Korea

  • We studied antioxidant activities of Vaccinium oldhamii Miq. from 11 regions in South Korea and blueberries, domestically produced and imported. Correlation analysis between V. oldhamii habitats, environmental factors, and antioxidant properties was conducted. DPPH RC50 values ranged from 220.44 to 902.38㎍/mL, ABTS from 524.29 to 1230.97 ㎍/mL, and FRAP from 1783.71 to 2235.78 ㎛ Fe (II)/g. V. oldhamii from Gumi showed highest DPPH activity, Taean and Haenam for ABTS, and Gimcheon for FRAP. V. oldhamii exhibited superior antioxidant activities compared to blueberries. Meteorological conditions correlated positively with ABTS and DPPH activities, negatively with wind speed and humidity affecting DPPH and phenolic, flavonoid, and anthocyanin contents. Based on these findings, it is suggested that V. oldhamii fruits collected from Gimcheon and Gumi regions can be effectively utilized as natural antioxidants derived from plant materials.

Antioxidant Activities of Achyranthes japonica Nakai Extract and Its Application to the Pork Sausages

  • Influence of Achyranthes japonica Nakai Extract (AJNE) on properties of pork sausages were studied in the present investigation. AJNE was added to sausages alone or in combination with ascorbic acid to obtain a comparative analysis on properties of control and ascorbic acid added-sausages. Results showed that addition of 0.05% AJNE led to a decrease in color $L^*$ and whiteness (W), and an increase in color $b^*$ of pork sausage samples (p<0.05). Although color $a^*$ of pork sausages containing AJNE was not significantly different, ascorbic acid added-sausages were highest amongst other treatments (p<0.05). Sausages containing AJNE had lower non-heme iron values and peroxide value (POV) than control sausages (p<0.05); however, high nitrosomyoglobin content was observed in AJNE added-sausages (p<0.05). Ascorbic acid led to a decrease in residual nitrite concentration of sausages (p<0.05), but no difference was found in AJNE added-sausages. Free radical scavenging analysis showed that AJNE did not affect 1,1-diphenyl -2-picrylhydrazyl (DPPH) activity of sausages, whereas ascorbic acid added-sausages showed relatively higher activity among the samples (p<0.05). Addition of AJNE had no influence on texture properties of sausages. In sensory evaluation, AJNE treatment had significant effects on color (p<0.05), but no significant effects on aroma, flavor, springiness, juiciness, and overall acceptability. In conclusion, the addition of AJNE, as a natural supplement may offer natural antioxidants for pork sausages, and appears to be particularly effective in inducing changes in non-heme iron concentration, POV value and nitrosomyglobin content.

Microbiological Characteristics and Cytoprotective Effects of Samjung-Hwan Fermented by Lactic Acid Bacteria (유산균을 이용한 발효삼정환의 미생물 특성 및 세포 보호 효과)

  • Objectives: To confirm microbiological change and cytoprotective effect of Samjung-hwan (SJH) which fermented by Lactic acid bacteria from natural fermented SJH. Methods: SJH was fermented by Lactobacillus brevis and Lactococcus lactis subsp. lactis from natural fermented SJH. After 1 week of fermentation, we analysed pH and microbial profiling. We also performed measuring total polyphenol total flavonoid contents and 1,1-Diphenyl-2-picryhydrazyl (DPPH) free radical scavenging activity to investigate antioxidant ability. Cell viability was performed by using HepG2 cell. Results: pH of lactic acid bacteria inoculated group and non-inoculated group was decreased and total counts of lactic acid bateria for both group was increased after fermentation of SJH. Total polyphenol and flavonoid contents and DPPH free radical scavenging activity was increased in both group. Total polyphenol contents of lactic acid bacteria Inoculated group is more increased than non-inoculated group. HepG2 cell viability was increased in both group. Conclusions: SJH fermentd by Lactobacillus brevis and Lactococcus lactis subsp. lactis shows change in microbiological character and has cytoprotective effect. Further studies are required for investigating function of lactic acid bacteria during fermentation of SJH.
