Characteristic Features of Cytotoxic Activity of Flavonoids on Human Cervical Cancer Cells

  • Cervical cancer is the most common gynecologic malignancy worldwide and development of new therapeutic strategies and anticancer agents is an urgent priority. Plants have remained an important source in the search for novel cytotoxic compounds and several polyphenolic flavonoids possess antitumor properties. In this review article, data about potential anticarcinogenic activity of common natural flavonoids on various human cervical cancer cell lines are compiled and analyzed showing perspectives for the use of these secondary metabolites in the treatment of cervical carcinoma as well as in the development of novel chemotherapeutic drugs. Such anticancer effects of flavonoids seem to differentially depend on the cellular type and origin of cervical carcinoma creating possibilities for specific targeting in the future. Besides the cytotoxic activity per se, several flavonoids can also contribute to the increase in efficacy of conventional therapies rendering tumor cells more sensitive to standard chemotherapeutics and irradiation. Although the current knowledge is still rather scarce and further studies are certainly needed, it is clear that natural flavonoids may have a great potential to benefit cervical cancer patients.

Validations of Analysis Methods for Decursin and Decursinol Angelate of Angelicae gigantis Radix by Reversed-Phase Liquid Chromatography

  • A reversed-phase liquid chromatographic method for decursin and decursinol angelate of Angelicae gigantis Radix, an important crude drug in Korean traditional medicine, was developed and validated. Decursin and decursinol angelate, the structure isomer (pyranocoumarin) each other, are the main organic constituents in Angelicae gigantis Radix. This method was developed using a RP-18 column, UV detector at 280 nm and 50% acetonitrile solution containing 0.01 M sodium dodecyl sulfate and 25 mM sodium dihydrogen phosphate (pH 5.0) as the mobile phase. Various validation parameters were included and evaluated satisfactorily. Linearity was established in range 2-75 mg/ml of decursin and decursinol angelate (correlation coefficient = 0.9997 and 0.9995, respectively). This analytical method showed good accuracy (98.1% and 99.5%, respectively). Precision (repeatability) revealed a relative standard deviation value of 1.71% (decursin) and 3.19% (decursinol angelate). For intermediate precision measure the considered variables were equipment and days. A robustness test showing the influence of deferent counter-ion concentration in mobile phase was also performed.

A Study on the Installation Method of Down Conductors of the Lightning Protection System, using Rebar in Reinforced Concrete Structures (철근콘크리트 구조물 내의 철근을 활용한 피뢰설비 인하도선의 설치방법에 관한 연구)

  • IEC Standards on lightning protection system specify the use of natural components as down conductors. This paper provides an analysis of problems revealed from our field investigation and survey conducted for the relevant experts in the construction site where natural components are used as down conductors. It also considers a suitable condition for installing natural components as down conductors in accordance with the latest version of the standard. As a result, when rebars of reinforced concrete are used as down conductors, vertical bars consisting of rebars of which thickness is not less than D13($127mm^2$) should be connected by welding, clamps or bound joints, using appropriate connection components conforming to IEC Standards. The lashed joints, however, shall not be applied for down conductors.

  • A new method for the analysis of coumarin constituents in the roots of Peucedanum japonicum by high performance liquid chromatography was established. Among two coumarin constituents identified, peucedanol was confirmed to be applicable to a standard compound. A reversed phase system with a ${\mu}Bondapak$ C_{18}$, column using $H_2O-MeOH=5\;:\;4$ as a moble phase was developed. Peucedanol and a minor constituent, umbelliferone were detected at 333nm and the analysis was successfully carried out within 20 min.

An Antioxidant Capacity Assay Using a Polyvinyl Alcohol-Based DPPH Pellet

  • To assay for antioxidant capacity of natural products considered important in producing human health benefits, a practical and economical method using pellet techniques was developed. A standard visualizing reagent, 1,1diphenyl-2-picryl-hydrazyl (DPPH), was mixed with a water-miscible polyvinyl alcohol (PVA), serving as a solid phase support for the DPPH reagent. A DPPH pellet was prepared by dropping a small volume of the DPPH solution onto PET film, and drying in an oven. The PVA-based DPPH pellet was dissolved into water, in which the water-miscible PVA plays as a non-ionic surfactant to help the DPPH reagent to be dissolved into the solvent. Using the DPPH assay, the antioxidant capacity of water-soluble extracts of black soybean, barley, green tea, and green gram was examined. Among the natural products tested, green tea showed the highest antioxidant capacity. This PVA-based DPPH antioxidant assay can be further applied in the natural food, raw plant material, and health product inspection field.

  • Natural gas is converted in to LNG by chilling and liquefying the gas to the temperature of $-162^{\circ}C$, when liquefied, the volume of natural gas is reduced to 1/600th of its standard volume. This gives LNG the advantage in transportation. The pressure dorp of the cascade liquefaction cycle was investigated and simulated using HYSYS software. The simulation results showed that the pressure drop in the LNG heat exchanger is set to 50 kPa considering the increase in the compressor work of cryogenic cascade liquefaction cycle.

  • The new assay method was developed by using young chicks instead of frogs for screening of anti-emetic compounds from natural sources. Comparing with our previous method using leopard and ranid frogs, the advantages of the new method were not only completely parallel results but also decreasing standard errors. Hinesol and eudesmol were isolated from MeOH extract of Atractylodes lancea rhizome as the anti-emetic principles.

  • In this study, by using the natural language processing of the field of artificial intelligence, automated index was performed. And then, the Natural Language Processing Interface for knowledge representation(NALPI) has been developed. Furthermore, the DEsign KnOwledge DataBase(DEKODB) has been also developed, which is designed to interlock the knowledge base. The DEKODB processes both the documented design-data, like a concrete standard specification, and the design knowledge from an expert. The DEKODB is also simulates the design space of structures accordance with the production rule, and thus it is determined that DEKODB can be used as a engine to retrieve new knowledge and to implement knowledge base that is necessary to the development of automatic design system. The application field of the system, which has been developed in this study, can be expanded by supplement of the design knowledge at DEKODB and developing dictionaries for foreign languages. Furthermore, the perfect automation at the data accumulation and development of the automatic rule generator should benefit the unified design automation.
