Antioxidant Activity of Extract from Walnut Uuglans sinensis Dode) and Its Protective Effect on Cell Injury and Lipid Peroxidation in Renal Cortical Slices (호두 추출물의 항산화 활성과 신피질에서 세포 손상과 지질과산화 방지효과)

  • To investigate the antioxidant activity of extract from the raw walnut, Juglans sinensis Dode, we prepared five fractions (methanol (MeOH), dichloromethane $(CH_2Cl_2)$, ethyl acetate (EtOAc), n-buthanol (n-BuOH) and dehydrogen monooxide $(H_2O)$ fractions) and examined. The effect of walnut extract on the oxidative stress was investigated in vitro. The DPPH (2,2-Di (4-tert-octylphenyl)-1-picrylhydrazyl) free radical scavenging activity of extract from raw walnut was shown in the following order: $EtOAc\;fraction layer. The result showed that the highest activity $(0.56{\mu}g/ml,\;IC_{50}.)$ was observed in EtOAc fraction, whereas n-BuOH fraction, MeOH fraction, $CH_2O_2$ fraction and $H_2O$ layer of $IC_{50}$ were $2.34{\mu}g//ml,\;3.88{\mu}g/ml,\;8.06{\mu}g/ml,\;and\;8.19{\mu}g/ml$, respectively. The radical scavenging activity assay of each fraction showed that the antioxidative activity was observed in the following order: EtOAc fraction $(74.27\pm1.56\%)>MeOH\;fraction\;(60.76\pm3.4\%)>n-BuOH\;fraction\;(59.32\pm0.88\%)>H_2O\;layer\;(41.69\pm2.06\%)$. These results revealed that all fractions, except for $CH_2Cl_2$ fraction, showed high antioxidative activity. Furthermore, the peroxynitrite $(ONOO^-)$ scavenging activity was assayed in each fraction. The result showed that the $ONOO^-$ scavenging activity of EtOAc fraction, MeOH fraction and n-BuOH fraction from raw walnut was $95.14\pm0.36\%,\; 90.02\pm1.19\%\;and\;89.41\pm0.81\%$, respectively. The tert-butylhydroperoxide (t-BHP) treatment in vitro increased lactate dehydrogenase release and lipid peroxidation in renal cortical slices. Such changes were completely prevented by addition of MeOH fraction, EtOAc fraction and n-BuOH fraction of walnut. These results indicate that the walnut extract exerts the benedicial effect against t-BHP-induced cell injury and its effect may be due to antioxidant action. In addition, it is suggested that walnut extract might be developed as the effective scavenger for the prevention of oxidative stress.

Anti-inflammatory Effects of Inhalation of Injured Starfish Extracts on Formaldehyde Exposure (손상된 불가사리 추출물 흡입이 포름알데히드 노출에 의한 항염증 작용에 미치는 효과)

  • Formaldehyde (FA) is widely used in industries, and it is an indoor and outdoor pollutant. Exposure to FA may cause inflammation and respiratory oxidative stress. Studies have demonstrated that FA can cause cancer in animal models. During the regeneration process of injured starfish (Asterina pectinifera), several changes have been observed in the expression of cytokines. In particular, higher TGF-${\beta}1$ expression has been detected in arm cut starfish extract after eight days. The current study was designed to elucidate the in-vitro and the in-vivo pharmacological effects of starfish extract on FA exposure. We investigated the protective effects of intact starfish extract and arm cut starfish extract on an IMR-90 cell line and on mouse lung injury in response to FA exposure. In the presence of FA, inhalation of the arm cut starfish extract was associated with more promising cell proliferation, TNF-${\alpha}$, NF-${\kappa}B$ decrement, and $I{\kappa}-B{\alpha}$ increment. In the experimental group, the pulmonary structure of the arm cut starfish extract-treated group in the presence of FA exposure was similar to the control group, whereas the FA exposure group showed damage to the pulmonary structure. Moreover, the arm cut starfish extracts was more effective than the intact starfish extracts in terms of the expression of TNF-${\alpha}$, NF-${\kappa}B$, $I{\kappa}-B{\alpha}$, and surfactant protein A. The results obtained in this study demonstrate that arm cut starfish extracts are more effective in protecting pulmonary structure and function against FA exposure than intact starfish extracts.

Anti-stress and Sleep-enhancing Effects of Ptecticus tenebrifer Water Extract Through the Regulation of Corticosterone and Melatonin Levels (코르티코스테론 및 멜라토닌 수치 조절을 통한 동애등에 물 추출물의 항스트레스 및 수면 개선 효과)

  • P. tenebrifer (PT) belongs to the Diptera order and Stratiomyidae family. Recently, insect industry have been focused as food, animal feed and environmental advantages. γ-aminobutyric acid (GABA) and melatonin have been associated with regulating sleep and depression. GABA is the primary inhibitory neurotransmitter and is synthesized via biotransformation of monosodium glutamate (MSG) to GABA by lactic acid bacteria. In this study, we first used a GABA-enhanced PT extract, wherein GABA was enhanced by feeding MSG to PT. The underlying mechanisms preventing stress and insomnia were investigated in a corticosterone (CORT)-induced endoplasmic reticulum (ER) stress and chronic restraint stress (CRS)-exposed mouse model, as well as in pentobarbital (45 mg/kg)-induced sleep behaviors in mice. In the present study, the GABA peak was detected in high-performance liquid chromatography-evaporative light scattering detector (HPLC-ELSD) analysis and showed in Ptecticus tenebrifer water extract (PTW) but not in non-PTW extract. The results showed that PTW and Ptecticus tenebrifer with 70% ethanol extract (PTE) exerted neuroprotective effects by protecting against CORT-induced downregulation of phosphorylated extracellular signal-regulated kinase 1/2 (ERK1/2) and cAMP-response element binding protein (CREB) expression. In addition, PTW (300 mg/kg) significantly reduced CORT levels in CRS-exposed mice. Furthermore, PTW (100 and 300 mg/kg) significantly reduced sleep latency and increased total sleep duration in pentobarbital (45 mg/kg)-induced sleeping behaviors, which was related to serum melatonin levels. In conclusion, our results suggest that PTW exerts anti-stress and sleep-enhancing effects by regulating serum CORT and melatonin levels.

Inhibitors of Adipogenesis in 3T3-L1 Cells Isolated from Wheat Bran (밀겨 유래의 3T3-L1 세포 지방생성 억제물질)

  • As a part of our ongoing program on finding biologically active components from natural source we found three known constituents from the EtOH extract of the wheat bran. The known compounds were identified as tachioside (1), pinellic acid (2) and tryptophan (3). The structure and relative stereochemistry were determined from MS, 1D and extensive 2D NMR techniques as well as by comparison of their data with the published values. All isolates were tested their inhibitory effects on the adipogenesis in 3T3-L1 cells. The effect of compounds from wheat bran on 3T3-L1 adipocyte differentiation were measured by Oil Red O staining. These results demonstrate that tachioside (1) and pinellic acid (2) decreased lipid content in 3T3-L1 adipocytes by inhibiting lipogenesis. These compounds had shown antiobesity activities.

A Study on Colorfastness of the Natural Dye (천연식물성 염료의 염색견뢰도에 관한 연구)

  • The study was about on the colorfastness of the natural vegetable dye. Impatient Balsamina was chosen as a speciesof the natural vegetable dye. It has a long history for Korean Women‘s grooming on the nail to be reddened, Actually those were very easy to gather and easy to extract the colors from the leaves and the flowers, Also Impatient Balsamina were abundant near our environment. So this was a cue as a natural dye for the wool fabric, This study was designed to testify the possibility of the Impatient Balsamina as a natural vegetable. First, 2 kinds of extracts from the Impatient Balsamina were prepared. Second, 2 kinds of mordants and 3 kinds of dyeing process were used. And the dyed wool fabric were evaluated the color index using Chroma Meter, the result was recorded as L$^*$, a$^*$, b$^*$ value(65.86, 16.40, 36.80). The effect of the mordants was more effective in Cr mordant than Al mordant in the color fastness and the color affinity. The very interesting results were in the colorfastness to the drycleaning, that was graded 4 to 5, but the colorfastness to light was low, graded 2. Impatient Balsamina was veryfied, it could be a natural vegetable dye for the wool fabric.

A new method to detect cracks in plate-like structures with though-thickness cracks

  • In this paper, a simple two-step method for structural vibration-based health monitoring for beam-like structures have been extended to plate-like structures with though-thickness cracks. Crack locations and severities of plate-like structures are detected using a hybrid approach. The interval wavelet transform is employed to extract crack singularity locations from mode shape and support vector regression (SVR) is applied to predict crack serviettes form crack severity detection database (the relationship of natural frequencies and crack serviettes) using several natural frequencies as inputs. Of particular interest is the natural frequencies estimation for cracked plate-like structures using Rayleigh quotient. Only the natural frequencies and mode shapes of intact structures are needed to calculate the natural frequencies of cracked plate-like structures using a simple formula. The crack severity detection database can be easily obtained with this formula. The hybrid method is investigated using numerical simulation and its validity of the usage of interval wavelet transform and SVR are addressed.

Development of the NDIF Method Using a Sub-domain Approach for Extracting Highly Accurate Natural Frequencies of Arbitrarily Shaped Plates (임의 형상 평판의 고정밀도 고유진동수 추출을 위한 분할영역법 기반 NDIF법 개발)

  • The NDIF method based on a sub-domain technique is introduced to extract highly accurate natural frequencies of arbitrarily shaped plates with the simply-supported boundary condition. The NDIF method, which was developed by the authors for the eigen-mode analysis of arbitrarily shaped plates with various boundary conditions, has the feature that it yields highly accurate natural frequencies thanks to its effective theoretical formulation, compared with other analytical methods or numerical methods(FEM and BEM). However, the NDIF method has the weak point that it can be applicable for only convex plates. It was revealed that the NDIF method offers very inaccurate natural frequencies or no solution for concave cavities. To overcome the weak point, the paper proposes the sub-domain method of dividing a concave plate into several convex domains. Finally, the validity of the proposed method is verified in various case studies, which indicate that natural frequencies obtained by the proposed method are very accurate compared to the exact method and FEM(ANSYS).

Screening for Chemosensitizers from Natural Plant Extracts through the Inhibition Mechanism of P-glycoprotein

  • P-gp plays a critical role in drug disposition and represents a mechanism for the development of multidrug resistance. Flavonoids, a major class of natural compounds widely present in foods and herbal products, have been shown to inhibit P-gp. Therefore, the aim of this study was to identify new candidate chemosensitizers by screening various plant extracts. The ability of natural plant extracts to inhibit P-gp activity was assessed by measuring cellular accumulation of calcein AM, daunorubicin and vincristine in P-gp overexpressing MDCKII-MDR1 cells. Among more than 800 plant extracts, eight were found to inhibit P-gp activity. Curcuma aromatica extract produced greatest inhibition, followed by Curcuma longa and Dalbergia odorifera extracts. Extracts of Aloe ferox, Curcuma zedoariae rhizome, Zanthoxylum planispinum, and Ageratum conyzoides showed moderate inhibitory effects. Curcumin and quercetin exhibited similar inhibition of P-gpmediated efflux of daunorubicin and vincristine, and flavones had a lesser effect. When chemosensitizing effect was evaluated by measuring daunorubicin sensitivity to MDCKII-MDR1 cells in the presence of natural plant extracts, Curcuma aromatica showed the most potent chemosensitizing effect based on daunorubicin cytotoxicity. In conclusion, natural plant extracts such as Curcuma aromatica can potently inhibit P-gp activity and may have potential as a novel chemosensitizers.

  • Quercus mongolica (QM) is a species of Quercus native to Eastern Mongolia, Siberia, China, Japan, and Korea. QM has been used as Korean traditional medicine for the treatment of inflammation of the skin. Present work, the validation and content determination of the phenolic compounds which were isolated from the leaves of QM, including pedunculagin (1) and kaempferol-3-O-(6"-galloyl)-${\beta}$-D-glucopyranoside (2) were conducted through High-Performance Liquid Chromatography (HPLC). As a result, the contents of these compounds were 4.14% and 0.76% in QM extract, respectively.

Anti-oxidant Effect of Agastache rugosa on Oxidative Damage Induced by $H_2O_2$ in NIH 3T3 Cell

  • The plant Agastache rugosa Kuntze has various physiological and pharmacological activities. Especially, it has been regarded as a valuable source for the treatment of anti-inflammatory and oxidative stress-induced disorders. However, little has been known about the functional role of it on oxidative damage in mammalian cells by ROS. In this study, we investigated the DPPH radical, hydroxyl radical, hydrogen peroxide and intracellular ROS scavenging capacity, and $Fe^{2+}$ chelating activity of the extracts from Agastache rugosa. In addition, we evaluated whether the extract can be capable of reducing $H_2O_2$-induced DNA and cell damage in NIH 3T3 cells. These extracts showed a dose-dependent free radical scavenging capacity and a protective effect on DNA damage and the lipid peroxidation causing the cell damage by $H_2O_2$. Therefore, these results suggest that Agastache rugosa is useful as a herbal medicine for the chemoprevention against oxidative carcinogenesis.
