Treatment of acute bovine theileriosis in grazing Korean native cattle (방목중인 한우에서 발생한 급성 타일레리아증 치료)

  • Bovine theileriosis caused by Theileria sergenti is a tick-borne hematoprotozoan disease that is characterized by chronic anemia and fever in cattle. In this study, results of microscopic examination and PCR detection confirmed 17 Korean native cattle with emaciation and fever as acute bovine theileriosis caused by T. sergenti. Buparvaquone was injected as treatment, but was proved to be an inappropriate measure according to our study. After 6 months of injection, clinical signs and hematological values were recovered, but T. sergenti was still identified in blood sample as a result of microscopic exam and PCR. These results suggest that continuous management is necessary to control bovine theileriosis. Therefore, findings of this study may provide significant guideline on the control of bovine theileriosis.

Korean native wild herbal-based functional ingredient for skin health: Agatri® (Agastache rugosa extract) (피부 건강을 위한 국내 자생 천연 식물 원료: Agatri® (배초향 추출물))

  • "Eating cosmetic" market comes to prominence worldwide. Inner Beauty has established itself as the top trend nowadays and is leading the health functional food market. Another noticeable trend other than beauty-from-within is that products using "plant-based" natural botanical raw material are increasing. Based on these trends above, we have developed a plant extract named Agatri® for skin health using Agastache rugosa. By ingesting Agatri®, it is expected that the formation of skin wrinkles is suppressed, and skin elasticity is increased by suppressing the decrease in collagen content caused by ultraviolet rays. At the same time, it is possible to prevent skin aging by improving skin hydration and trans-epidermal water loss (TEWL). The studies show that it significantly increased production of hyaluronic acid and collagen, and decreased production of matrix metalloproteinases (MMPs). And Agatri® has been approved as an "Individually recognized functional ingredient for skin health by the Ministry of Food and Drug Safety, Korea in 2020. As a result, Agatri® is a good solution for those who are in need of skin protection from exposure to UV and aging.

Ecological Restoration Planting Design Awareness Survey (생태복원 사업에서의 식재 설계에 관한 인식 조사)

  • Much controversy has been generated over whether the ecological restoration project in Korea is faithful to the basic approach of ecological restoration compared to projects aimed at creating parks or green spaces. This study was aimed at understanding the level of awareness of practitioners in the planting design method, which is recognized as relatively important in landscape architecture and ecological restoration. The proficiency of practitioners in planting design techniques for ecological restoration projects was investigated and analyzed in seven areas: overall planting design awareness, plant species selection method, planting tree sizes, planting density, use of native species, vegetation climatic zone, soil environment, and weed control. There was no statistically significant difference between the group who thought they were experts in the field of ecological restoration and the group who thought they were experts in the field of parks and green spaces. Moreover, there was no statistically significant difference between the degree of work performance, that is, the group whose main industry was ecological restoration, the group that performed relatively more ecological restoration work, and the group that rarely performed it. Therefore, to apply desirable ecological restoration techniques in the future and to clearly differentiate between the approach in the field of parks and green spaces, the appropriate tree size or planting density selected in the ecological restoration planting design stage must be chosen. To this end, the awareness of expert groups must be improved and related laws, systems, and guidelines must be revised.

  • Species are constantly using for clothing, food, shelter and health are distributed in various places. Convention on Biological Diversity made for conserve resources and enhance the value in many countries around the world. Each country is in the process of building a database for to protect the native biological resources and establish sovereignty. This studies analyzed the distribution of such traditional knowledge of native biological resources and builded a database through using standardized form of collected data made by our research. The result, almost native biological resources used for food and medical treatment.

A Study on the Growth Characteristics under Seedling Types of Native Woody Plants After Planting on the Disturbed Slope (훼손비탈면에 식재된 자생목본류의 묘유형별 생육특성에 관한 연구)

  • This research was initiated to investigate the revegetation characteristics under seedling types of native woody plants on a disturbed slope. 4 different seedling types of woody plants (control, unit seedling plot, nature seedling plot, pot seedling plot) with the same woody plants (Rhus chinensis Mill, Albizzia julibrissin Duraz., Quercus acutissima Carr., Pinus densiflora S. et Z.) were treated with 3 replications on the experimental disturbed slope. Data such as the died number of native woody plants, growth height and visual quality were analyzed. The died number of plants were high in unit seedling plot when compared to the rest plots of seedling types. From the standpoint of died plant species, the died number were high in Rhus chinensis and Albizzia julibrissin whereas the died number were low in Quercus acutissima and Pinus densiflora. That's because we used 1-year old seedlings of Rhus chinensis and Albizzia julibrissin and 3-year old seedling Quercus acutissima and Pinus densiflora. This result indicated that the died reason of native woody seedling was not species but seedling age. Both growth height and visual quality were ranked first in the pot seedling plot, second in the nature seedling plot, third in the unit seedling plot, and last in the control plot, respectively. However, the difference of both growth height and visual quality in each seedling plot was not observed on the 11 months after planting. Especially, the main reason of low rooting and growth value in the unit seedling plot was due to wooven fabric outer crust of pot that obstruct seedling rooting into the soil. We concluded that pot seedling of native woody plants was effective in the viewpoint of the ecological disturbed slope revegetation.

Effects of Sasa borealis silage on proximate composition, amino acid and fatty acid contents, and antioxidant activity in fresh meat of Korean native goat fed with total mixed ration

  • Jeju Sasa borealis (S. borealis) is indigenous to the Halla Mountain area of Jeju Island, Republic of Korea. However, its dominance has retarded the development of other plant species and lowered biodiversity in this region. The aim of this study was to determine whether S. borealis silage (SS) supplementation affects the chemical composition and antioxidant activity in the fresh meat of Korean native goats (Capra hircus coreanae). The experiment was conducted on 12 Korean native goats at the finisher stage. The feeding groups were the Control (total mixed ration, TMR) and the Treatment (80% TMR + 20% SS). The animals were adapted for two weeks and then subjected to a six-month breeding experiment. Meat samples were excised from the neck, loin, rib, front leg, and hind leg of the slaughtered animals. The meat derived from the treatment group contained more taurine and anserine than that derived from the control group. Both groups did not significantly differ in terms of ω-6/ω-3 fatty acid ratio. The loin and front leg of the treatment group contained significantly higher vitamin E levels than those of the control group. DPPH, ABTS, and FRAP analyses disclosed that the loin and front leg had significantly higher antioxidant activity (p<0.05) than the other parts. Moreover, the loin and front leg cuts of the treatment group had higher antioxidant activity than those of the control group. The present study demonstrated that S. borealis supplementation could effectively improve Korean native goat meat quality.

Native and Foreign Proteins Secreted by the Cupriavidus metallidurans Type II System and an Alternative Mechanism

  • The type II secretion system (T2SS), which transports selected periplasmic proteins across the outer membrane, has rarely been studied in nonpathogens or in organisms classified as Betaproteobacteria. Therefore, we studied Cupriavidus metallidurans (Cme), a facultative chemilithoautotroph. Gel analysis of extracellular proteins revealed no remarkable differences between the wild type and the T2SS mutants. However, enzyme assays revealed that native extracellular alkaline phosphatase is a T2SS substrate, because activity was 10-fold greater for the wild type than a T2SS mutant. In Cme engineered to produce three Ralstonia solanacearum (Rso) exoenzymes, at least 95% of their total activities were extracellular, but unexpectedly high percentages of these exoenzymes remained extracellular in T2SS mutants cultured in rich broth. These conditions appear to permit an alternative secretion process, because neither cell lysis nor periplasmic leakage was observed when Cme produced a Pectobacterium carotovorum exoenzyme, and wild-type Cme cultured in minimal medium secreted 98% of Rso polygalacturonase, but 92% of this exoenzyme remained intracellular in T2SS mutants. We concluded that Cme has a functional T2SS despite lacking any abundant native T2SS substrates. The efficient secretion of three foreign exoenzymes by Cme is remarkable, but so too is the indication of an alternative secretion process in rich culture conditions. When not transiting the T2SS, we suggest that Rso exoenzymes are probably selectively packaged into outer membrane vesicles. Phylogenetic analysis of T2SS proteins supports the existence of at least three T2SS subfamilies, and we propose that Cme, as a representative of the Betaproteobacteria, could become a new useful model system for studying T2SS substrate specificity.

A Study on the Seeding Mixture Improvements for the Restoration and Revegetation of the Slopes by the Thin-Layer-Soil-Media Hydroseeding Revegetation Measures (생육보조재 취부 공법에 의한 비탈면 녹화 식생배합의 적정성에 관한 연구)

  • This study aimed to address problems and suggest solutions in applying seed mixture design criterior of slope revegetation works according to the "Slope revegetation design and guidelines(proposed)" proposed by the Ministry of Construction and Transportation. To do this, a proper thickness of attaching soil media was identified in April experiment where the thickness and slopes varied. The results were as follows. In the test, the coverage rate of plot where exotic grass was not sowed increased with time and was 79.3% after three months of seeding, which indicated less risk of soil erosion. When applying the provisional standards of the plant mix proposed by the Ministry of Construction and Transportation, tree seeds and native plants were found ill in the early stage in all test lands due to exotic grass. This was because exotic grass which grew well in the early stage, could grow densely, so it might hamper the growth of other plants. When slope revegetation were planted and goals were set for herb, shrub and tree dominant mixture types, it was required to shift focus toward nursing up native plants first rather than early plantation of exotic grass.

Development of Green Retaining-wall System with Native Evergreen Plants Corresponding to the Southern Region - A Case Study of Tongyeong City in Gyeongsangnam-do - (남부지역의 특성을 고려한 상록벽면녹화 공법 개발 -경남 통영시를 사례로-)

  • This study was carried out to investigate and propose a green-wall system with evergreen plants for urban greening of Tongyeong City. To achieve these goals, the requirements and possibilities for wall greening were investigated and evaluated considering the location, topography, and climate of Tongyeong City. Existing walls were analyzed and then a suitable green wall system is proposed. Tongyeong City and its 151 islands covers the central and the southern parts of the Goseong peninsula. Most of the land is covered with hills and mountains; $43.9\%$ of the land area has a slope greater than $15\%$ and most hills and mountains near the urban area have a slope of more than $30\%$. As a result of the topographical properties, concrete retaining walls can often be seen along the streets in urbanized areas. These retaining walls are not only unattractive, but they also create environmental problems, and thus should be replaced with native evergreen plants. Options for replacing the retaining walls include evergreen vine-plants such as Hedaa spp. and Euonymus radicans, but native evergreen shrubs such as Pittosporum tobira, Nandina domestica, Raphiolepis umbellata, Ilex cornuta, flex crenata, Fatsia japonic, and Aucuba japonica may be a more attractive option. Current wall conditions are unsuitable for planting vines, therefore, a reservoir-drainage-type plant box filled with a light artificial substrate is required for greening these concrete retaining walls. These might be irrigated in the dry season and fertilized annually by an appropriate system. These plant boxes could be attached along the entire walls. An experiment investigating effects of substrates and bark-chip mulching on the growth of Hedera spp. showed that the mixture of cerasoil and field soil(v/v, 4:6) was superior to field soil alone and to the mixture of perlite small grain, large grain, and field soil(v/v/v, 2:2:6). Bark-chip mulching tended to increase the growth of Hedera spp..

Effect of Medium Component on Asymbiotic Germination of 3 Korean Native Orchid Species (자생란 3종의 기내 종자발아에 미치는 배지구성물질의 영향)

  • This study was carried out to investigate the factors affecting seed germination of Korean native orchids, such as Habenaria radiata, Bletilla striate and Cypripedium macranthum. In general, seeds of H. radiata was effectively germinated on solid medium and the highest germination (29.6%) was achieved on Hyponex solid medium. But shoot growth was better in MS liquid medium containing $100mL\;L^{-1}$ coconut water (MSC medium). Seed germination of B. striata was effectively induced on MS solid medium. However, no seed germination was observed in all liguid medium tested. Seed germination experiments of C. macranthum were performed using 6 kinds of media and seeds harvested 90 days after cross- and self-pollination. Cross-pollinated seeds were germinated on all the media with the best germination ratio of 4.45% on MSC medium. But no germination was observed from self-pollinated seeds.
