한약재복합 추출물의 인간피부섬유아세포 HS68에 대한 항노화 효과 (Antiaging Activity of Mixed Extracts from Korean Medicinal Herbs on HS68 Skin Fibroblast)

  • Objectives : The purpose of this study was to investigate antiaging and antioxidant effects on cultured human skin fibroblast with 80% ethanol extracts of plants including of stem of Dendropanax morbifera, Corni fructus and Lycii Fructus. Methods : An ethanol extract of three medicinal plants including stem of Dendropanax morbifera, Corni fructus and Lycii Fructus. Extracts were assessed to determine the mechanism of antioxidant and antiaging activities. Antioxidant activity of extract was evaluated by two different assays as 2,2-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and super oxide dismutase (SOD) like activities. These extracts were tested for cell viability on HS68 skin fibroblast by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay. We investigated the effects of Ultraviolet-B irradiation on cytotoxicity, type 1 collagen, elastin level and oxidative damage in cultured human skin fibroblast (HS68). Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. Results : The extracts obtained dose-dependently increased the scavenging activity on DPPH radical scavenging activity and SOD like activity. The extracts of complex herbal medicine showed low cytotoxicity as more than 100% cell viability in 100ppm/ml concentration. HS68 fibroblasts were survived 70% at 120 $mJ/cm^2$ UVB irradiation and treated tumor necrosis factor (TNF)-alpha. The levels of aging factors and cytotoxicity were decreased by ethanol extract of complex herbal medicine. Conclusions : These results suggest that ethanol extracts of complex medicinal plants of including of stem of Dendropanax morbifera, Corni fructus and Lycii Fructus may have value as the potential antioxidant and antiaging medicinal plant.

폴리에톡시레이티드레틴아마이드를 함유한 입방상 액정 젤 및 큐보좀의 제조 및 평가 (Preparation and Evaluation of Cubic Liquid Crystalline Phase Gel and Cubosome containing Polyethoxylated Retinamide)

  • The objective of this study is to prepare a stable delivery systems containing polyethoxylated retinamide(PERA) - derivatives of retinoic acid, effective anti-wrinkle and anti-acne agent. Cubic liquid crystalline phase gel (CLCPG) and cubosomes containing various concentrations of PERA were prepared to investigate the physicochemical properties. Furthermore, stability and transdermal absorption efficacy of the CLCPG containing PERA were investigated in comparison with oil-in-water (O/W) emulsions which are predominantly used as a topical formulation. CLCPG increase the stability of PERA in comparison with O/W emulsion. For tropical application, CLCPG containing PERA shows higher moisturizing effect than that of O/W emulsion. In skin permeation test, CLCPG shows higher PERA deposit on epidermis. With its specific physicochemical property caused by the glyceryl oleate, CLCPG itself could be used for stabilizer of various actives and applied as an effective delivery system for topical application. Cubosome, nano-sized dispersed CLCPG, is also expected to be applied in a various field of industry like food, cosmetics and pharmaceuticals.

Triterpenoid-Containing Liposome by Micelle-to-Vesicle Transition and Their Biological Activities

  • Ursolic acid (UA) and oleanolic acid (OA) are pentacyclic triterpenoids which are widely distributed in plants, and their derivatives are aglycones of many naturally occurring saponins. It is known that pentacyclic acids may possibly enhance the mechanical barrier functions of cell membranes in plants. Recently, it has been reported that OA and UA have interesting biological activities on skin, such as anti-inflammatory and anti-wrinkle activities. Since triterpenoids are extremely insoluble and their solubility problem limits skin-care application, OA and UA were encapsulated in liposomes via micelle-to-vesicle transition to overcome poorly soluble property and enhance biological efficacy. Optimal molar ratio of OA to lecithin was found to exist for producing liposomes of small hydrodynamic size and liposomal suspensions without recrystallized precipitation of OA. From electron micrograph and dynamic light scattering studies, reconstituted OA-containing liposomes without severe mechanical treatment showed small hydrodynamic size about 150 nm. Wide-angle X-ray diffraction coupled with dynamic light scattering revealed that optimal amount of OA in liposome was 25.4 mole %. In biological evaluation, OA-containing liposome significantly increased filaggrin and transglutaminase as markers of keratinocyte differentiation in epidermal layer of hairless mouse, whereas ursolic acid-containing liposome did not show noticeable increase of filaggrin and transglutaminase compared to empty liposome. It is concluded that nano-scaled liposomes containing triterpenoids were spontaneously prepared by vesicular transition from mixed micelle and liposomal triterpenoids can enhance skin absorption of triterpenoid and biological efficacy.

자외선 흡수제 처리 시 은나노/수지 첨가가 면직물의 기능성에 미치는 영향 (Effect on Functional Properties of the Cotton Fabrics Treated by UV-absorbers with Nanosilver/DMDHEU Treatment)

  • Silver ions or silver nanoparticles have multi-functional properties. The cotton fabrics for providing multi-functional properties were treated with a nanosilver powder, UV-absorbers, and dimethyloldi-hydroxyethyleneurea (DMDHEU) alone and mixed solution. The physical properties, UV protection, antibacterial, and deodorizing properties of treated cotton fabrics were evaluated. The results were as a follows. The UV protection of cotton fabrics were increased by the application of a nanosilver and Uv-absorbers mixture. The UV protection of treated fabrics were improved by nanosilver/DMDHEU/UV-absorbers mixed solution. The wrinkle recovery properties of fabrics treated with DMDHEU and nanosilver improved. The stiffness of fabrics are decreased by a nanosilver/DMDHEU/UV-absorbers mixed solution. The antibacterial properties of the fabrics treated with nanosilver/DMDHEU/UV-absorbers mixed solution is 99.99%. The functional properties of cotton fabrics are shown to be better with aanosilver/DMDHEU/UV-absorbers mixed than treated with nanosilver alone.

올레아노익산의 폴리글리세릴계 나노에멀젼에서의 안정화 및 인체적용 유효성평가에 대한 연구 (Study of stabilizing and efficacy evaluation in human of Oleanoic acid with poly-glyceryl nano emulsion system)

  • 올레아놀릭산은 항암, 신행혈관 생성방지, 항염증, 항산화 및 주름 개선효과가 알려져 있다. 본 연구자들은 천연에서 분리한 올레아놀릭산의 항산화효과에 주목하여 연구하였고 미백효과가 있음을 확인하였다. 본 연구에서는 천연유래의 폴리글리세릴계 계면활성제를 사용하여 간단한 교반만으로 올레아놀릭산을 안정화하였고 고가의 장비인 마이크로풀루다이저를이용하여 제조한 레시틴리포좀과 경피흡수투과율을 비교하였다. 0.4% 올레아놀릭산을 안정화한 폴리글리세릴 나노에멀젼의 12시간 후 경피흡수투과율은 95%였다. 레시틴 리포좀은 92%로 유사하였으나 폴리글리세릴 나노에멀젼은 3시간 경피 흡수투과율이 65%로 레시틴리포좀의 45%에 비해 속방성의 특징을 보였다. 인체대상 임상시험결과 올레아놀릭산을 무배합한 대조군에 비해 올레아놀릭산을 배합한 폴리글리세릴 나노에멀젼은 MEXAMETER의한 멜라닌 색소감소효과가 2주차 25%, 4주차 58%, 8주차 58%이상 높았다.

미백 및 주름개선 성분을 함유하는 Poly(methylmethacrylate-co-trimethoxysilylpropylmethacrylate) 구의 제조와 평가 (Preparation and Evaluation of Poly(methylmethacrylate-co-trimethoxysilylpropylmethacrylate) Spheres Containing Whitening and Anti-wrinkle Agents)

  • 최근에 고분자, 지질 또는 그 밖의 여러 소재를 이용한 캡슐화 연구는 다양한 기능성 성분의 안정화 및 방출 제어를 목적으로 진행된 바 있다. 본 연구는 주요한 기능성 성분들을 캡슐화할 수 있는 또 하나의 새로운 방법으로써, methylmethacrylate (MMA)와 trimethoxysilylpropylmethacrylate (TMPMA)로 구성된 MMA-TMPMA 공중합체를 제조하였다. Poly(MMA-co-TMPMA)로 제조된 구는 속이 빈 형태의 미립구이며 레티놀, 레티닐팔미테이트, 토코페릴아세테이트, 아스코빌테트라이소팔미테이트와 같은 비타민유도체나 유용성감초추출물과 같은 기능성 성분들을 core 성분으로 캡슐화하는데 매우 유용하였다. 제조된 poly(MMA-co-TMPMA) core-shell 형태의 미립구 평균 입도는 $0.1{\sim}10{\mu}m$, 기능성 성분의 로딩 함량은 $15{\sim}25%$이며 로딩 수율은 90% 이상이었다. Poly(MMA-co-TMPMA) core-shell 형태의 미립구에 함유된 기능성 성분의 안정화는 캡슐 자체에 의한 안정화 외에 미립구 표면에 자외선차단 silane 전구체를 도입함으로써 향상시킬 수 있었다. 기능성 성분에 대한 광안정성은 자외선차단 전구체가 함유된 poly(MMA-co-TMPMA) 미립구의 경우, 자외선차단 전구체가 도입되지 않은 미립구에 비해 25%정도 더 항상됨을 확인할 수 있었다.

호두의 화장품 기능성 (Functional Properties of Walnut in Cosmetics)

  • 호두과육으로부터 용매별로 추출한 추출액중의 phenolic compound 함량은 열수, 에탄올, 메탄올, 아세톤 추출물 순서로 24.3, 34.4, 32.5, 15.1 mg/g의 함량을 나타내어 에탄올 추출물의 phenolic compound 함량이 가장 높았으며, 인체에 무해한 추출 용매인 열수 및 에탄올 농도별 추출물의 phenolic compound 비교한 결과 60% 에탄올 추출물에서 34.7 mg/g과 열수 추출물에서 24.6 mg/g 함량의 phenolic compound를 나타내었다. 호두추출물의 항산화 활성 중 전자공여능의 경우 열수추출물에서 78.1%, 60% 에탄올추출물에서 80.6%의 활성을 나타내었으며, ABTS radical cation decolorization 측정에서는 열수추출물과 60% 에탄올추출물 각각 98.1%와 98.3%의 높은 항산화 활성을 나타내었다. Antioxidant protection factor (PF)의 경우는 열수추출물과 60% 에탄올추출물에서 각각 $1.1{\pm}0.2$ PF와 $1.1{\pm}0.4$ PF로 높지는 않았으며, TBARs inhibitory activity 측정에서는 열수추출물과 60% 에탄올추출물에서 각각 60%와 75%로 비교적 높은 항산화력을 가졌음이 확인되었다. 호두추출물의 항염증효과를 살펴본 결과 100 ${\mu}g/ml$의 농도에서 50%의 NO발현이 억제되었고, western blot에 의한 iNOS 및 COX-2의 억제효과는 대조군에 비해 100 ${\mu}g/ml$의 농도에서 10% 정도의 낮은 저해 효과를 나타내었다. Tyrosinase의 저해 활성의 경우 60% 에탄올추출물에서 43%의 저해율을 보였으며, 수렴효과의 경우 60% 에탄올추출물에서 55%의 억제효과를 나타내어, 호두추출물은 미백 및 주름개선용 화장품소재로서 개발이 가능하다고 판단되었다. 본 연구는 고령친화산업 기업지원센터의 연구비 지원에 의하여 수행되었으며, 이에 감사드립니다.
