Lifestyle intervention might easily improve blood pressure in hypertensive men with the C genotype of angiotensin II type 2 receptor gene

  • BACKGROUND/OBJECTIVES: Recent studies have reported an association of the angiotensin II type 2 receptor (AT2R) 3123Cytosine/Adenine (3123C/A) polymorphism with essential hypertension and cardiovascular diseases. The purpose of the study was to investigate whether the AT2R 3123C/A polymorphism affects blood pressure for free-living hypertensive men during a 5-month intervention period. SUBJECTS/METHODS: The subjects were free-living hypertensive Japanese men aged 40 to 75 years who agreed to intervention in the period from 2004 to 2011. Detection of the AT2R 3123C/A polymorphism was determined by polymerase chain reaction. The dietary intervention was designed to decrease salt level and to increase potassium level through cooking instructions and self-monitoring of the diet. The exercise session consisted of activities such as stretching, resistance training, and walking. Blood pressure, urinary sodium and potassium excretion, dietary and lifestyle data, and non-fasting venous blood sample were collected at baseline and after the intervention period. RESULTS: Thirty nine subjects were eligible for participation and the follow-up rate was 97.4%. The C allele proportion was 57.9%. AT2R 3123C/A polymorphism was X-chromosome-linked, therefore we analyzed the C and A genotypes. At baseline, no significant differences were observed between the genotype groups. After the intervention, there were no significant differences in lifestyle habit between the groups. Nevertheless, the estimated salt excretion (g/day) was significantly decreased only in the C genotype (13.0-10.3, P = 0.031). No significant change was observed in systolic blood pressure (SBP) (mmHg) in the A genotype, but a significant decrease was observed in the C genotype (150.0-141.5, P = 0.024). CONCLUSTIONS: In the C genotype, it might be easy to improve SBP through lifestyle intervention in free-living hypertensive Japanese men, however generalization could not be achieved by the small sample size.

Presence of Transcription Factor OCT4 Limits Interferon-tau Expression during the Pre-attachment Period in Sheep

  • Interferon-tau (IFNT) is thought to be the conceptus protein that signals maternal recognition of pregnancy in ruminants. We and others have observed that OCT4 expression persists in the trophectoderm of ruminants; thus, both CDX2 and OCT4 coexist during the early stages of conceptus development. The aim of this study was to examine the effect of CDX2 and OCT4 on IFNT gene transcription when evaluated with other transcription factors. Human choriocarcinoma JEG-3 cells were cotransfected with an ovine IFNT (-654-bp)-luciferase reporter (-654-IFNT-Luc) construct and several transcription factor expression plasmids. Cotransfection of the reporter construct with Cdx2, Ets2 and Jun increased transcription of -654-IFNT-Luc by about 12-fold compared with transfection of the construct alone. When cells were initially transfected with Oct4 (0 h) followed by transfection with Cdx2, Ets2 and/or Jun 24 h later, the expression of -654-IFNT-Luc was reduced to control levels. OCT4 also inhibited the stimulatory activity of CDX2 alone, but not when CDX2 was combined with JUN and/or ETS2. Thus, when combined with the other transcription factors, OCT4 exhibited little inhibitory activity towards CDX2. An inhibitor of the transcriptional coactivator CREB binding protein (CREBBP), 12S E1A, reduced CDX2/ETS2/JUN stimulated -654-IFNT-Luc expression by about 40%, indicating that the formation of an appropriate transcription factor complex is required for maximum expression. In conclusion, the presence of OCT4 may initially minimize IFNT expression; however, as elongation proceeds, the increasing expression of CDX2 and formation of the transcription complex leads to greatly increased IFNT expression, resulting in pregnancy establishment in ruminants.

Enhancement of Thermoelectric Properties in Cold Pressed Nickel Doped Bismuth Sulfide Compounds

  • Nanostructured Ni doped $Bi_2S_3$ ($Bi_{2-x}Ni_xS_3$, $0{\leq}x{\leq}0.07$) is explored as a candidate for telluride free thermoelectric material, through a combination process of mechanical alloying with subsequent consolidation by cold pressing followed with a sintering process. The cold pressing method was found to impact the thermoelectric properties in two ways: (1) introduction of the dopant atom in the interstitial sites of the crystal lattice which results in an increase in carrier concentration, and (2) introduction of a porous structure which reduces the thermal conductivity. The electrical resistivity of $Bi_2S_3$ was decreased by adding Ni atoms, which shows a minimum value of $2.35{\times}10^{-3}{\Omega}m$ at $300^{\circ}C$ for $Bi_{1.99}Ni_{0.01}S_3$ sample. The presence of porous structures gives a significant effect on reduction of thermal conductivity, by a reduction of ~ 59.6% compared to a high density $Bi_2S_3$. The thermal conductivity of $Bi_{2-x}Ni_xS_3$ ranges from 0.31 to 0.52 W/m K in the temperature range of $27^{\circ}C$ (RT) to $300^{\circ}C$ with the lowest ${\kappa}$ values of $Bi_2S_3$ compared to the previous works. A maximum ZT value of 0.13 at $300^{\circ}C$ was achieved for $Bi_{1.99}Ni_{0.01}S_3$ sample, which is about 2.6 times higher than (0.05) of $Bi_2S_3$ sample. This work show an optimization pathway to improve thermoelectric performance of $Bi_2S_3$ through Ni doping and introduction of porosity.

개의 고관절 이형성 치료를 위한 변형 3중 골반 절골술의 적용 (Application of a Modified Triple Pelvic Osteotomy for Treatment of Hip Dysplasia in Dogs)

  • 개의 양측성 고관절 이형성을 치료하기 위해 실시하는 슬로컴의 편측 3중 골반 절골술을 개선하기 위해 고안된 변형 3중 골반 절골술의 효과에 대해 알아보고자 본 연구를 실시하였다. 변형 3중 골반 절골술은 양측 장골 절골술과 치골결합 절골술을 동시에 실시하는 방법이다. 변형 3중 골반 절골술을 실시한 36마리와 편측 3중 골반 절골술을 실시한 7마리의 개에 대한 품고, 체중, 수술 시간, 골 유합기간, 복합증 등의 자료를 조사, 기록하였다 임상 증상과 고관절 이형성의 등급, Norberg angle, percentage of femoral head coverage, 골반 직경에 대해 수술 전, 직후, 수술 후 2, 4, 8, 12, 24주에 각각 측정, 기록하였다. 편측 3중 골반 절골술을 실시한 개체의 평균 연령과 체중은 각각 $9.0\pm4.9$개월, $24.8\pm7.5kg$ (n=7)이었다. 수술 후 평균 $3.0\pm1.0$일에 스스로 일어서고 $8.3\pm0.6$일부터는 걸을 수 있었다(n=3). 수술 전과 24주 후의 평균 임상 증상 등급은 각각 $2.2\pm0.42$ (n=6), $3.5\pm0.7$ (n=2)이었다. 평균 수술 시간은 $107.3\pm38.9$분 (n=4)이었다. 변형 3중 골반 절골술을 실시한 개체의 평균 연령과 체중은 각각 $8.7\pm3.0$개월, $26.9\pm7.6kg$ (n=36)이었다. 이중 25마리는 수술 후 평균 $4.9\pm3.7$일부터 스스로 일어날 수 있었고 평균 $7.3\pm4.8$일 후부터 보행이 가능하였다 변형 3중 골반 절골술을 실시한 개체의 수술 전과 24주 후의 평균 임상 증상 등급은 각각 $2.3\pm1.5$ (n=27), $3.2\pm0.7$ (n=9)로 수술 전보다 유의적으로 개선되었다.(P<0.01). 평균 수술 시간은 $143\pm42.8$분(n=24)으로 두 번의 편측 3중 골반 절골술보다 단축되었다 변형 3중 골반 절골술을 실시한 개체의 수술 전 방사선학적 평균 고관절 등급은 양측 모두 $3.2\pm0.9$이었고 수술 직후의 좌 우측 평균 고관절 등급은 각각 $2.7\pml.1,\;2.7\pm0.9$ (n=36) 이었다. 수술 직후와 2, 4, 8, 12, 24주 후의 고관절 등급이 수술 전에 비하여 유의적으로 개선된 것을 확인하였다.(P<0.01). 수술 후 정기적인 검사 시에 측정한 Norberg angle, percentage of femoral head coverage도 수술 전과 비교해 유의성 있게 증가하였다(P<0.01). 변형 3중 골반 절골술 직후의 평균 골반직경은 수술 전의 골반직경보다 유의적으로 증가하였으며(P<0.01)(n=36) 수술 후 평균 9.3L2.7주에 절골선 유합이 종료되었다(n=21). 반면 편측 3중 골반 절골술을 실시한 경우에는 수술 후 골반경이 수술 전과 비교해 증가하지 않았다. 변형 3중 골반 절골술후에 장액종 형성(1마리), 스크류 변위(4마리), 스크류 부러짐(1마리), 편측성 신경마비(1마리) 등의 부작용이 발생하였다. 이상의 결과를 토대로, 변형 3중 골반 절골술은 개의 고관절 이형성의 효과적인 치료 방법이라 사료된다.
