본 연구는 재구성육제품내 첨가제를 줄이는데 있어 예비혼합물에 첨가제를 전량 첨가하여 그 기능적 특성을 최대한 부여시킬 수 있는 첨가제의 종류와 이온강도 효과의 조건을 설정하고자 실시하였다. 육제품의 10%를 예비혼합들로 설정하여 돼지전지부위에 NaCl(0, 4.5, 9.0%), STPP (0, 2.5, 5.0%), PP (0, 2.44, 4.88%)를 각각 세단계로 이온강도를 같게 설정한 후 조합첨가하여 27가지 예비혼합물을 제조하고 pH와 추출한 단백질의 기능적 특성을 조사하였다. pH는 STPP와 PP의 첨가에 의해 증가하는 경향을 나타냈으며(p<0.01), 동일 이온강도에서 PP가 STPP에 비해 높은 pH증진효과를 보였다. 예비혼합물에서 추출한 단백질(1 M NaCl, 동일한 염추출 용액 사용)의 특성 중 총 단백질추출성에 대하여 동일 이온강도 하에서 phosphate 이온이 chloride 이온보다 추출효과가 뛰어났다. 단백질의 용해성은 NaCl 첨가로 용해성이 낮아졌으며(p<0.05), 단백질추출성이 좋을수록 용해성이 감소하는 것으로 나타났다. 원료육 내의 SH기 함량은 단백질추출성이 좋은 경우 SH기 함량이 높은 경향을 보였다. 단백질의 가열처리를 가상한 $65^{\circ}C$, 7분간의 단백질 추출물의 열처리 효과로 원료육의 SH기 함량이 낮아지고 소수성은 증가하였다(p<0.1). 그러나 세 첨가제에 의한 27가지의 예비혼합물에서 추출한 단백질의 전기영동상 형태는 차이가 나타나지 않았다. 결론적으로 첨가제의 조합에 의해 이온강도가 높아짐에 따라 기능적 특성이 증가되지는 않았으나 인산염 중 동일이온강도를 감안할 때 첨가제를 줄이는 방안으로는 STPP보다 PP가 좋은 것으로 판단되었다.
Little is known concerning the mechanisms responsible for the transplacental transfer of thiamine. So, the aim of this work was to characterize the placental uptake of thiamine from the maternal circulation, by determining the characteristics of $^3H$-thiamine uptake by a human trophoblast cell line (BeWo). Uptake of $^3H$-thiamine (50-100 nM) by BeWo cells was: 1) temperature-dependent and energy-independent; 2) pH-dependent (uptake increased as the extracellular medium pH decreased); 3) $Na^+$-dependent and $Cl^-$-independent; 4) not inhibited by the thiamine structural analogs amprolium, oxythiamine and thiamine pyrophosphate; 5) inhibited by the unrelated organic cations guanidine, N-methylnicotinamide, tetraethylammonium, clonidine and cimetidine; 6) inhibited by the organic cation serotonin, and by two selective inhibitors of the serotonin plasmalemmal transporter (hSERT), fluoxetine and desipramine. We conclude that $^3H$-thiamine uptake by BeWo cells seems to occur through a process distinct from thiamine transporter-1 (hThTr-1) and thiamine transporter-2 (hThTr-2). Rather, it seems to involve hSERT. Moreover, chronic (48 h) exposure of cells to caffeine ($1\;{\mu}M$) stimulated and chronic exposure to xanthohumol and iso-xanthohumol (1 and $0.1\;{\mu}M$, respectively) inhibited $^3H$-thiamine uptake, these effects being not mediated through modulation of the expression levels of either hThTr-1 or hSERT mRNA.
Background: In Changbai Mountains, Panax ginseng (ginseng) was cultivated in a mixture of the humus and albic horizons of albic luvisol in a raised garden with plastic shade. This study aimed to evaluate the impact of ginseng planting on soil characteristics. Methods: The mixed-bed soils were seasonally collected at intervals of 0-5 cm, 5-10 cm, and 10-15 cm for different-aged ginsengs. Soil physico-chemical characteristics were studied using general methods. Aluminum was extracted from the soil solids with $NH_4Cl $(exchangeable Al) and Na-pyrophosphate (organic Al) and was measured with an atomic absorption spectrophotometer. Results: A remarkable decrease in the pH, concentrations of exchangeable calcium, $NH_4^+$, total organic carbon (TOC), and organic Al, as well as a pronounced increase in the bulk density were observed in the different-aged ginseng soils from one spring to the next. The decrease in pH in the ginseng soils was positively correlated with the $NH_4^+$ (r=0.463, p<0.01), exchangeable calcium (r=0.325, p<0.01) and TOC (r= 0.292, p < 0.05) concentrations. The $NO_3^-$ showed remarkable surface accumulation (0-5 cm) in the summer and even more in the autumn but declined considerably the next spring. The exchangeable Al fluctuated from $0.10mg\;g^{-1}$ to $0.50mg\;g^{-1}$ for dry soils, which was positively correlated with the $NO_3^-$ (r=0.401, p < 0.01) and negatively correlated with the TOC (r=-0.329, p < 0.05). The Al saturation varied from 10% to 41% and was higher in the summer and autumn, especially in the 0-5 cmand 5-10 cm layers. Conclusion: Taken together, our study revealed a seasonal shift in soil characteristics in ginseng beds with plastic shade.
본 연구에서는 심야전력 및 폐열의 회수이용을 위한 잠열축열재를 개발하기 위하여, 상변화 온도가 $50{\sim}80^{\circ}C$의 범위에 있는 상변화 물질중 잠열량이 크고, 물성안정이 비교적 용이한 Octacosane($C_{28}H_{58}$)과 Sodium Acetate Trihydrate($CH_3COONa{\cdot}3H_2O$)를 선택하여 상변화 온도의 변화특성과 물성안정방법 그리고 잠열특성을 분석하여 다음과 같은 결과를 얻었다. 1. 공업용 수준 Octacosane과 Sodium Acetate Trihydrate의 상변화 온도는 각각 $60.7^{\circ}C,\;57.4^{\circ}C$로서 시약용 수준에 비하여 Octacosane은 $1.1^{\circ}C$, Sodium Acetate Trihydrate는 $0.6^{\circ}C$ 낮았으며, 잠열량에 있어서는 Octacosane은 60.6kcal/kg으로 시약용 수준과 비슷한 값을 보였으나, Sodium Acetate Trihydrate는 51.1kcal/kg으로 시약용 수준에 비하여 5.4% 작은 값을 나타내었다. 2. Octacosane의 상변화 사이를 증가에 따른 잠열량을 분석한 결과, 초기에는 60.6kcal/kg이었으나, 시약용의 증가와 더불어 점차로 감소하여 200cycle에서 47.2kcal/kg이 되었고, 그 이후에는 거의 일정한 값을 유지하고 있는 것으로 보아 더 이상의 잠열량 감소는 없을 것으로 판단된다. 3. Sodium Acetate Trihydrate의 과냉현상을 제어하기 위한 조핵제로서 SPD(Sodium Pyrophosphate Decahydrate)를 3wt 이상 첨가하여 $25.7^{\circ}C$의 과냉도를 $1^{\circ}C$ 미만으로 줄였고, 증점제로서 CMC-Na를 3wt% 이상 첨가하여 상분리 현상을 해결하였으며, 안전성을 고려한 조핵제 및 중점제의 적정 첨가량은 4wt%였다. 4. UREA($NH_2CONH_2$)를 첨가하여 Sodium Acetate Trihydrate($CH_3COONa{\cdot}3H_2O$)의 상변화 온도를 $57.4^{\circ}C$에서 $46.2^{\circ}C$까지 다양하게 변화시켜 이용온도 수준에 적합한 잠열축열재를 선택할 수 있도록 하였으며, UREA 함량에 따른 잠열량을 분석한 결과 UREA의 함량이 증가함에 따라 잠열량이 점차로 감소하여 10wt%의 UREA를 첨가하였을 경우에는 38.3kcal/kg였으며, UREA의 함량이 20wt%까지 증가하는 동안 더 이상의 잠열량 감소는 없었다. 5. $30^{\circ}C$에서 $90^{\circ}C$까지의 가열과정에서 현열재인 물, 그리고 돌(화강암)과 잠열축열재의 축열량을 비교 분석한 결과, Octacosane은 상변화 온도 $60{\sim}70^{\circ}C$에서의 축열량이 물의 2.45배, 돌의 12.5배였으며, S.A.T.(Sodium Acetate Trihydrate)는 상변화 온도 $57.4^{\circ}C$에서의 축열량이 물보다 2.53배, 돌보다는 12.91배 큰 것으로 나타났다.
명태연육 냉동변성을 억제할 수 있는 냉동변성방지제의 적정량을 알아보기 위하여 crystalline sorbitol을 비롯한 세가지의 변성방지제 및 sorbitol/sucrose 혼합제제를 첨가하 여 $-25^{\circ}C$에서 16주간 저장했을 때의 단백질품질 변화를 실험하였다. $8\%$ 수준의 sucrose-sorbitol 혼합제제(1:1, w/w)를 $0.2\%$ Na-pyrophosphate/ Na-triphosphate(1:1)와 병용하였을 때 drip loss가 가장 적었으며, 염용성단백질량은 가장 많았다. 또한 혼합제제를 사용했을 때는 trypsin inhibitor량, 단백질소화율 및 단백효율비의 변화가 거의 없어 단백질품질 보전에 효과적이었으나, 냉동변성방지제를 처리하지 않았을 경우에는 단백효율비와 소화율은 급격히 떨어졌었다. Polyphosphate나 maltodextrin의 경우에는 처리 농도가 증가하여도 소화율저하 방지에는 별효과가 없었으나, $4{\sim}6\%$의 sorbitol, 또는 $10\%$수준의 sucrose는 polyphosphate를 병용하지 않아도 소화율저하는 효과적으로 막을 수 있었다. $8\%$ 수준의 sorbitol/sucrose 혼합제제(5:3, w/w)처리가 소화율 보전효과로 볼 때 냉동연육의 단백질품질저하 방지에 가장 효과적이었다.
In the process of bone remodeling, mineral phase of bone is dissolved by osteoclasts, resulting in elevation of calcium concentration in micro-environment. This study was performed to explore the effect of high extracellular calcium ($Ca{^{2+}}_e$) on mineralized nodule formation and on the expression of progressive ankylosis (Ank), plasma cell membrane glycoprotein-1 (PC-1) and osteopontin by primary cultured mouse calvarial cells. Osteoblastic differentiation and mineralized nodule formation was induced by culture of mouse calvarial cells in osteoblast differentiation medium containing ascorbic acid and ${\beta}$-glycerophosphate. Although Ank, PC-1 and osteopontin are well known inhibitors of mineralization, expression of these genes were induced at the later stage of osteoblast differentiation during when expression of osteocalcin, a late marker gene of osteoblast differentiation, was induced and mineralization was actively progressing. High $Ca{^{2+}}_e$(10 mM) treatment highly enhanced mRNA expression of Ank, PC-1 and osteopontin in the late stage of osteoblast differentiation but not in the early stage. Inhibition of p44/42 MAPK activation but not that of protein kinase C suppressed high $Ca{^{2+}}_{e^-}$induced expression of Ank, PC-1 and osteopontin. When high $Ca{^{2+}}_e$(5 mM or 10 mM) was present in culture medium during when mineral deposition was actively progressing, matrix calcifiation was significantly increased by high $Ca{^{2+}}_e$. This stimulatory effect was abolished by pyrophosphate (5 mM) or levamisole (0.1-0.5 mM), an alkaline phosphatase inhibitor. In addition, probenecid (2mM), an inhibitor of Ank, suppressed matrix calcification in both control and high $Ca{^{2+}}_{e^-}$treated group, suggesting the possible role of Ank in matrix calcification by osteoblasts. Taken together, these results showed that high $Ca{^{2+}}_e$ stimulates expression of Ank, PC-1 and osteopontin as well as matrix calcification in late differentiation stage of osteoblasts and that p44/42 MAPK activation is involved in high $Ca{^{2+}}_{e^-}$induced expression of Ank, PC-1 and osteopontin.

Forest tree species usually takes for long periods to be harvested and cultivated but spearmints are a good model system for woody plant because of reducing and shortening cultivation time. Spearmints are good model plants (Mentha species) for research about terpenoids production and industrial essential oil manufacture. Isopentenyl pyrophosphate isomerase (Iso) and limonene synthase (Limo) are the key enzymes of terpenoid biosynthesis pathway. Transgenic and wild spearmints (Mentha spicata, MS) were cultured in vitro and assessed for the essential oil contents. The content of essential oil of transgenic spearmint also was enhanced slightly depending on the target terpenoid genes. In an attempt to increase productivity of terpenoids further, salt and fungal elicitation strategy was adopted on transgenic Mentha spicata. The salt (800 mM NaCl) as abiotic and two fungi (Botrytis cinerea and Glomerella cingulata) as biotic were used for elicitors. In the absence of salt stress four terpenoids were detected from the spearmint extracts, all of them being monoterpenes. On the other hand, the transgenic (MSIso) extracts contained eleven terpenoids (10 monoterpenes and 1 phenylpropene) while transgenic (MSLimo) extracts contained seven monoterpenes. After 3 days of fungal infection, the resistance indices further increased to 4.38, 3.89 and 2.04 for wild type, MSIso and MSLimo, respectively. The salt and fungal elicitators proved beneficial towards modifying both the terpenoids profile and improvement in the composition of essential oil. These results have important applications for the large-scale production of essential oils and forest biotechnology with respect to spearmint.
