Effects of Yeast Culture Supplementation on Rice Straw Digestibility and Cellulolytic Bacterial Community in the Rumen (볏짚 조사료에 대한 효모 배양물 첨가가 반추위 소화율 및 섬유소 분해균의 군락 변화에 미치는 영향)

  • In vitro and in situ incubation studies were conducted to determine effects of yeast culture supplements (Saccharomyces cerevisiae) on cellulolytic bacterial function and fiber digestion in rice straw. In vitro dry matter digestibility of rice straw gradually increased according to supplemental levels of yeast culture (0.0, 0.2, 0.4, 0.6, 0.8 and 1.0%). Digestibility of rice straw started to increase apparently when yeast culture was added more than 0.6% level (p<0.05). Also, we reconfirmed that in vitro dry matter digestibility was significantly increased by 0.6% of yeast culture addition in 4% NaOH treated and non-treated rice straws (p<0.05). When in situ dry matter digestibility was tested in Korean native goats fed basal diet or experimental diet which contained 1.0% of yeast culture, the yeast culture feeding improved in situ dry matter digestibility in both 4% NaOH treated and non-treated rice straws (p<0.05). In case of real-time PCR monitoring cellulolytic bacterial function, the bacterial population attached on rice straw showed the increasing trends with higher level of yeast culture spraying on rice straw. F. succinogenes and R. flavefaciens were significantly increased in accordance to spraying levels of yeast culture (0.0, 0.1 and 0.3%) at both 12 and 24 hrs of in situ incubation (p<0.05). R. albus was significantly higher population in yeast culture spraying than non-soraying at 12 hrs of in situ incubation (p<0.05). These bacterial populations were showed the increasing trends with digestibility enhancement of rice straw according to the higher levels of yeast culture supplement. Overall, these results clearly suggest that the presence of yeast culture result in noticeable increase of rice straw digestion, which is modulated via good effect on cellulolytic bacterial attachment to fiber substrates.

Study for Reduction Effect of Pool Top Radiation in Research Reactor by Using Ion Exchanger of Hot Water Layer (고온층계통의 이온교환기에 의한 연구로 수조 상부 방사선의 저감효과에 대한 연구)

  • A hot water layer (HWL hereinafter) was installed at the depth of 1.2 m from the pool surface to reduce the radiation level at the pool top. After the HWL system was improved by the replacement of the filter with the Ion Exchanger to capture the Na-24, to purify the pool water of HWL and finally to reduce the radiation at the pool top. It was confirmed by the performance test of the pump and the measurement of the pressure difference through the Ion Exchanger and the strainer, that the flow characteristics of HWL system was not adversely affected after the system modification. Also the flow analysis using the pressure loss coefficients of the Ion Exchanger and strainer, calculated by the Darcy formula, could predict the flow variations by pressure changes within $10\%$ error in comparison with the field test results. It was also confirmed that HWL was maintained with the depth of 1.2 m from the pool surface because each electric water heater was electrically and thermodynamically maintained at 30 kW and the temperature of HWL was maintained with $5^{\circ}C$ higher temperature than that of pool water. Finally, it was confirmed that the pool top radiation was saturated and stabilized below 10000 nG/hr within 24 hours as the ion exchanger captured the main nucleus, Na-24 and purified the pool water of HWL.

  • To improve the strength of glass is being studied in order to contribute to weight saving of flat panel displays. Generally, the strength achieved of glass-ceramics is higher as is the fracture toughness by the formation of a heterogeneous phase inside glass. In this study, Ag-doped $45SiO_2-24CaO-24Na_2O-4P_2O_5\;and\;70SiO_2-10CaO-24Na_2O-10TiO_2$ glasses were irradiated to strengthen by crystallization using femto-second laser pulse. XRD, Nano-indenter and SEM etc., irradiation of laser pulse without heat-treated samples was analyzed. Samples irradiated by laser had higher value($4.4{\sim}4.56^*10-3Pa$) of elastic modulus which related with strength of glass than values heat-treated samples and these are $1.2{\sim}1.5$ times higher values than them of mother glass. This process can be applicable to the strengthening of thinner glass plate, and it has an advantage over traditional heat-treatment and ion-exchange method.

Studies on Mechanism and Damage Occurrence for Foliar Spray of NaCl Solutions in Fruit Trees (NaCl 엽면살포에 따른 과수의 피해양상과 기작에 관한 연구)

  • We investigated the mechanism on occurring of briny injury in four fruit species (apple, pear, grape, peach). Briny injury of apple, pear, peach, and grape was induced by the foliar splay of saline solutions. Browning symptom was initiated one hour after the spray of 3% NaCl in apple, pearl and peach trees. while two hour were needed for grape, but the latter developed the symptom faster than the formers. Foliar spray of 3% NaCl resulted in 4-fold increase in ethylene production 24 h after the spray, and the production was returned to early stage rate after 72 h. Ethylene production of control was nearly constant with slight decrease after 4 h. Browning and defoliation response to the spray of 3% NaCl on lower side were not different from the spray on both sides regardless of fruit species, while no injury symptoms occurred by the spiny on upper side. In saline damage for treated date, lateral bud was only germinated in apple at 30 July. A shoot apex was 100% grew in all fruit tree. In result for hormonal content of leaf and shoot apex, control was high t-zeatin content in leaf. However, IAA and ABA content was increasing in high saline concentration.

Effects of NaCl/H3PO4 Flame Retardant Treatment on Lyocell Fiber for Thermal Stability and Anti-oxidation Properties (NaCl/H3PO4 내염화 처리가 라이오셀 섬유의 열 안정 및 내산화 특성에 미치는 영향)

  • The improved thermal stability and anti-oxidation properties of Lyocell fiber were studied based on flame retardant treatment by using NaCl/$H_3PO_4$ solution. The optimized conditions of flame retardant treatment were studied on various maxing ratio of NaCl and $H_3PO_4$ and the mechanism was proposed through experimental results of thermal stability anti-oxidation. The IPDT (integral procedural decomposition temperature), LOI (limited oxygen index) and $E_a$ (activation energy) increased 23, 30 and 24% respectively via flame retardant treatment. It is noted that thermal stability and anti-oxidation improved based on char and carbon layer formation by dehydrogenation and dissociation of C-C bond resulting the hindrance of oxygen and heat energy into polymer resin. The optimized conditions for efficient flame retardant property of Lyocell fiber were provided using NaCl/$H_3PO_4$ solution and the mechanism was also studied based on experimental results such as IDT (initial decomposition temperature), IPDT, LOI and $E_a$.

A Study on Mineral Contents in Domestic Mozzarella Cheese (Mozzarella Cheese의 무기질 함량에 관한 연구)

  • This experiment was carried out to find general composition and mineral contents such as Ca, P, Mg, Na, K and Fe of domestic commercials Mozzarella cheese. Twenty-five products of 5 company were analyzed. The results obtained were as fellow : The average contents of moisture, fat, fat in dry matter, protein and ash in Mozzarella cheese were 49.01, 22.54, 44.16, 24.36 and 2.55%, respectively and average pH was 5.29. The average contents of Ca, P, Mg, Na, K and Fe were 696.44, 466.31, 22.08, 310.8, 82.84 and 0.36mg, respectively. Among products, product C was highest in levels of Ca, P and Mg, which were 797.4, 536.6 and 25.5mg, respectively, Na was highest for product A which contained 450.8mg. K and Fe were highest for product B which contained 98.9 and 0.51mg, respectively. Generally, the ratio of Ca to P was 1.5 : 1 and within the proper range. It was found that 645g of Mozzarella cheese was able to fully meet the daily recommendation of minerals including Ca, P and Na.

Technological Characteristics and Safety of Enterococcus faecium Isolates from Meju, a Traditional Korean Fermented Soybean Food (메주 유래 Enterococcus faecium 균주의 기능적 특성 및 안전성)

  • In this study, we assessed the technological characteristics and safety of 88 Enterococcus faecium strains isolated from meju; the strains possess the glutamate decarboxylase gene gadA/B involved in γ-aminobutyric acid production. The study was conducted to evaluate the possibility of introducing E. faecium meju isolates as food fermentation starters. We observed that a NaCl concentration of 6% (w/v) facilitated the growth and acid production of all strains. At a NaCl concentration of 7%, 21 strains (24%) exhibited a low growth rate, 72 strains (82%) a weak acid production, and 16 strains (18%) showed no acid production. All strains exhibited protease activity at a NaCl concentration of 4%. At a NaCl concentration of 5%, 86 strains exhibited weak activity, and one strain showed no protease activity. We could not detect any lipase activity in the investigated strains. None of the strains exhibited an acquired antibiotic resistance to the seven antibiotics tested in the present study, namely ampicillin, chloramphenicol, ciprofloxacin, gentamicin, penicillin G, tetracycline, and vancomycin. We could identify the Enterococcus endocarditis antigen gene efaA and the tyrosine decarboxylase gene tdc contributing to tyramine production, in 88 meju isolates. We could not detect the Enterococcus surface protein gene esp, which is specifically possessed by human-originated E. faecium strains, in any of the 88 strains tested in the study.
