Study on diuretic activity and electrolytes excretion of methanol extract of Lippia nodiflora (Verbenaceae) in rats

  • In the Indian traditional medicine, Lippia nodiflora (Verbenaceae) whole plant is claimed to possess powerful diuretic activity. However, the diuretic potential of this plant is not yet investigated. The aim of this study was to evaluate the diuretic potential of methanol extract of Lippia nodiflora (MELN) in rats. Control (0.9% saline solution, 25 ml/kg, b.w) or urea (1 g/kg b.w) or frusemide (5 mg/kg b.w) and different concentrations of MELN (200 and 400 mg/kg b.w) were intraperitoneally administered (n = 6 per each treatment group) to hydrated rats and their urine output was monitored over a period of 5 h and 24 h after drug administration. The diuretic responses with its electrolyte excretion potency of the extract were highly remarkable in comparison with control animals. The extract at doses of 200 and 400 mg/kg shows a significant increase in volume of urine with increase in $Na^{+}$, $Ca^{2+}$ and $Cl^{-}$ excretion accompanied by the excretion of $K^{+}$ in dose dependent manner. This study suggests that the active component(s) in MELN had similar diuretic effect to that of frusemide. These results validate the traditional use of Lippia nodiflora as a diuretic agent.

  • Each parching Anemarrhenae Rhizoma with 25% ethanol and normal saline has been used for anti-inflammatory, expectorant, antipyretic, sedative and diuretic, and so on. In order to investigate the differences of referential efficacy about each fractionated part of the aqueous extracts, pharmacological studies were carried out. The results of studies were summerized as follows : 1. By the administration of 25% ethanol treated preparation (F-I-1), the increase in urinary volume and $Na^+$ extraction was significantly recognized. 2. The excretion of electrolyte $Na^+$ by saline solution treated preparation (F-II-1) was significantly recognised in normal rats. 3. The increase of urinary volume and urinary $Na^+$ and $Cl^-$ excretion by F-I-1 and n-butanol insoluble parts of F-I-1 (F-I-3) was significantly recognized in $HgCl_2-induced$ acute renal failure of rats. 4. The increase of plasma $Na^+$ by F-I-1 and urea nitrogen by F-I-1 and F-I-3 in acute renal failure of rats was significantly recognized. 5. The increase of urinary volume by F-I-1 and F-I-3 was recognised in mice.

고지혈증 모델 흰쥐에서 칼슘과 소디움 섭취수준이 체내 지질대사에 미치는 영향 (Effects of Dietary Calcium and Sodium Levels on Lipid Metabolism in Hyperlipidemic/Hypercholesterolemic Rats)

  • The effects of dietary Ca and Na levels on lipid metabolism in hyper lipidemic/hypercholesterolemic rats were examined. In Expt. 1, normal rats were divided into six groups and fed high fat(15%, w/w)/cholesterol(1%, w/w) diet containing two levels of Na, low (0.05) or high(1.5%) and three levels of Ca, low(0.1%), normal (0.5%), or high(1.5%) for 8 weeks. In Expt. 2, hyperlipidemia / hypercholesterolemia rats were induced by feeding high fat / cholesterol diet for 4 weeks. They were divided into four groups and fed the high fat / cholesterol diet, containing two levels of Na, low or high and two levels of Ca, low or high for 4 weeks. In Expt. 1, total lipid and total cholesterol contents in serum and liver were significantly lower in rats fed high Ca diet than in rats fed normal or low Ca diet regardless of dietary Na levels. Serum TG was the highest in rats fed low Ca and low Na diet. In Expt. 2, Serum total lipid, TG, and total cholesterol levels decreased by 24, 35, 26% respectively in rats fed high Ca diet regardless of dietary Na levels. Serum total lipid level tended to increased in rats fed low Na diet. The total lipid and TG contents in liver slightly decreased in rats fed high Ca diet. Another observation was that high Ca intake significantly faciliated the fecal lipid and cholesterol excretion regardless of dietary Na levels. There results suggest that the hypolipidemidc/hypocholesterolemic effects of high Ca diet could be partly due to increase in lipid and cholesterol excretion and these effects may be independent of dietary Na levels.

고혈압 환자 가족과 정상인에 있어서 혈압과 Na, K 섭취간의 상관관계 (A Study on Correlation between Blood Pressure and Na, K Intakes Pattern in the Family Members of Normal and Hypertension Patients)

  • 고혈압은 한국인에 있어서 발병율이 높은 성인병이며 또한 식이 중 Na 섭취와 관련이 있는 질병으로 알려져 있는바, 본 연구는 건강한 구성원만을 가진 정상인군과 고혈압 환자가 있는 가족의 구성원을 대상으로(환자 자신은 제외 ) 이들이 섭취하는 음식물, 소변 중의 Na 및 K과 혈압을 측정함으로서 이들 항목이 고혈압과 어떤 상관관계가 있는지를 알아보고자 하였다. 고혈압 환자의 수축기 $혈압(126\pm18.0mmHg)과$ 이완기 $혈압(77.6\pm14.6mmHg)은$ 정상인 가족의 수축기 $혈압(119.3\pm17.2mmHg)과$ 이완기 $혈압(71.6\pm12.5mmHg)$ 보다 유의적으로 높은 수치를 보였다. 수축기 혈압과 조사 항목간의 상관관계는 다음과 같다. 정상인군에서는 나이, 체중, 된장내 Na, 된장내 Na는 양의 상관관계를 보였고, 고추장내 K, 된장내 K, 고기반찬류내 K 등과 부의 상관관계를 나타냈다. 고혈압 환자 가족군에서는 나이, 체중, 소금 섭취량, 된장내 Na 등과 양의 상관관계를, 소변내 K배설량과 부의 상관관계를 보였다.

  • The influence of prazosin (0.1 mg/kg i.v.) on the excretion and diuretic action of furosemide (2mg/kg i.v.) in rabbits was studied to investigate an interaction between ${\alpha}-adrenergic$ blocking agent, prazosin and furosemide. The results were as follows; 1) With the combined administration of prazosin and furosemide, the plasma concentration of furosemide was increased, the urinary excretion rate and renal clearance of furosemide were reduced, and tile biological half-life of furosemide was increased. 2) The diuretic action of furosemide was significantly reduced with the combined administration of prazosin: maximal decrease in urine volume, urinary electrolytes, clearance of $Na^+$ and $Cl^-$, and GFR and RPF, as well as maximal increase in $Na^+$ reabsorption rate were noted 10 minutes after administration of furosemide (2mg/kg i.v.)

Acetazolamide 의 Carbonic Anhydrase 활성 억제 작용에 대한 Testosterone 의 영향 (Effects of Testosterone on Carbonic Anhydrase Inhibiting Action of Acetazolamide)

  • This study was carried out to observe the effect of testosterone on carbonic anhydrase inhibiting action of acetazolamide. Carbonic anhydrase activities in the kidneys of mice were measured by Philpot and Philpot method(1936) at 30, 90 and 150 minutes after intravenous administration of saline(0.5 ml/10 g) or acetazolamide (0.25 mg/10 g) in mice pretreated with testosterone (0.1 mg/10 g). The changes in volume and pH of urine as well as those in urinary electrolytes, such as $Na^+,\;K^+\;and\;Cl^-$ were measured at 15 minutes interval for 150 minutes in the rabbit pretreated with double administrations of testosterone(10 mg/kg), 1 hour and 18 hours, prior to the administration of acetazolamide (10 mg/kg). The results were as follows: 1. Carbonic anhydrase activities in the kidneys of mice of testosterone-pretreated groups were significantly higher than those of acetazolamide-treated group at 30 minutes. No significant changes of carbonic anhydrase activities were observed in testosterone-pretreated groups compared with saline-treated groups. 2. Combined administrations of acetazolamide and testosterone exhibited higher carbonic anhydrase activity than those group of acetazolamide alone in the kidney of mice through observed period of 150 minutes. 3. There were no significant changes in the excretion rate of urine and urinary electrolytes in the group of rabbits with testosterone administerone alone. Urine volume as well as $Na^+\;and\;Cl^-$ excretion rates in the combined treated group of acetazolamide and testosterone were significantly lower than that of acetazolamide group throughout experimental period except 15 minutes after drug administration at the time transient increase was shown. 4. Generally lower $K^+$ excretion rate was observed in the combined treated group of acetazolamide and testosterone compared with the single acetazolamide-treated group and the testosterone-pretreated group shows lowest excretion rate of potassium.

가토(家兎)의 간장(肝臟) 및 신장(腎臟)에서의 색소분필(色素分泌) 과정(過程)에 관(關)하여 (Studies on the Dye-Secretory Processes of the Liver and the Kidney in the Rabbit)

  • That different mechanisms are involved in the secretory processes by the liver and the kidney of various dyes has been indicated by Sporter (1959), Kim and Hong (1963). Andrews (1958). suggested that a striking difference in the dye-secretory mechanism existed even in the same organ from species to species. Hence, the attempt has been made to study in the rabbit the secretory processes by the live. and the kidney of either phenol red (PSP), bromsulfalein (BSP) or green in the presence of Na-acetate, Na-taurocholate, P-Aminohippurate (PAH) or Benemid. In 37 rabbits, weighing about 2kg., anesthetized with ether, a dye was administered in such 8 manner that the plasma concentration was kept at a relatively constant level throughout the whole experimental period. Hepatic bile sad urine samples were quantitatively collected through the canulae which were previously inserted into the common bile duct (with the cystic duct ligated) and the urinary bladder, respectively, while arterial samples were taken from a femoral artery. After 50 min from the onset of dye administration, these samples were obtained every 10 mit for a period of 40 min. This was followed by the administration of either Na-acetate, Na-tauro-cholate, PAH or Benemid with a repetition of the same sample collecting procedures just stated. The results may be summarized as follows: 1) Na·acetate augmented urinary clearance of PSP by nearly 300 per cent, but lowered urinary BSP clearance by about 50 per cent. It enhanced biliary BSP clearance by 40% and had no effect on biliary psp clearance. 2) Na-taurocholate lowered biliary and urinary clearance of PSP by 10 per cent and 30 per cent respectively, and had no effect on both biliary and urinary clearance of BSP. 3) PAH lowered both biliary and urinary excretion of BSP and PSP, while it lowered the biliary excretion of indocyanine green which was excreted only in the bile. 4) Benemid suppressed BSP excretion by the liver and the kidney. 5) raper chromatographic analysis of PSP and of BSP in the bile and urine samples gave the following results: a) PSP Ivas excreted in the urine and bile only in free forms, and no modification in the excretory pattern was brought about by Na-taurocholate. b) BSP was excreted in the urine in 4 different conjugated froms and in the bile in both 3 different conjugated forms and in a free form. Na-taurocholate modified the excretory pattern of the urinary BSP.

방사성(放射性) 스트론듐의 체내오염(體內汚染)에 대한 응급처치제(應急處置劑)의 효과(效果) (Effect of First-Aid Drugs on Internal Contamination of Radiostrontium)

  • 방사성 물질 체내 오염에 대한 진단에 필요한 기초 자료를 얻기 위하여 $^{85}SrCl_2\;1{\mu}Ci$를 NIH(GP) 마우스의 복강내 투여로 체내 오염시키고, $CaNa_3DTPA$ 8.4 mg, sodium alginate 5 mg 및 saline 5 ml등을 병행 투여로 응급 처치한 후 경과 시간에 따라 도살 부검하여 중요 장기 및 배설물의 방사능을 측정하였다. 그 결과 방사성 스트론튬에 대한 단기간의 유효 반감기는 33 시간으로 나타났으며 각 처치제의 배설효과는 $CaNa_3DTPA$가 4.7배, sodium alginate가 1.7배, 그리고 saline이 2.4배 이었다. 스트론튬 단독 투여군의 체내 축적은 척추(脊椎), 대퇴골(大腿骨), 흉골(胸骨), 간장(肝臟)의 순이었으며, 배설 경로는 요(尿) 35.4%, 변(便) 64.4% 및 기타 0.2%이었고, $CaNa_3DTPA$ 및 saline 투여군은 주로 뇨, 그리고 sodium alginate 투여군은 변(便)을 통하여 배설하였다. 이로부터 방사성 스트론튬 체내 오염에 대한 응급 처치는 $CaNa_3DTPA$, sodium alginate 및 saline등을 병행 투여함으로써 방사선 피폭을 경감시킬 것으로 사료 되었다.

혼합식염 (NaCl-KCl) 이용 조리시 나트륨${\cdot}$칼륨 섭취와 소변 중 나트륨${\cdot}$칼륨 배설에 관한 연구 (The Influence of Mixed NaCl-KCl Salt on Sodium Intake and Urinary Excretion of Sodium and Potassium)

  • 고혈압의 유발 원인과 관련이 있는 나트륨의 섭취를 줄일 수 있는 방법을 모색하기 위해 건강한 20대 성인여성 9명을 대상으로 NaCl-KCl 혼합식염으로 조리한 식이를 섭취하도록 한 후 24시간 소변으로 배설되는 나트륨 양을 조사하여 나트륨 섭취량이 평상시보다 어느 정도 감소되는가를 보고자 하였다. 1) 3일간의 일상식이 섭취상태를 조사한 결과 연구대상자들이 섭취한 1일 평균 열량은 1,872.0 kcal였으며 나트륨과 칼륨의 1일 평균 섭취량은 각각 178.2 ${\pm}$ 46.0 mEq, 56.4 ${\pm}$ 21.9 mEq로 나타났다. 2) 실험기간동안 제공된 실험 식이에서 대상자들이 섭취한 열량은 평균 2,031.2 kcal였으며 나트륨의 섭취량은 평균 130.2 mEq이었으며, 칼륨의 섭취량은 평균 120.4 mEq이었다. 3) 조사대상자들의 혈압은 실험 시작 전 수축기 혈압과 이완기 혈압의 평균이 각각 104.6 ${\pm}$ 4.9 mmHg, 69.2 ${\pm}$ 6.3 mmHg였다. 실험식이 섭취 기간 동안 수축기 혈압과 이완기 혈압은 각각 108.1 ${\pm}$ 8.0 mmHg, 70.4 ${\pm}$ 5.7 mmHg로 나타났다. 4) 일상식이 섭취기간동안 소변의 평균 나트륨과 칼륨의 배설량은 각각 139.6 ${\pm}$ 37.3 mEq, 27.7 ${\pm}$ 14.7 mEq이었다. 소변의 나트륨/칼륨 비율은 6.6 ${\pm}$ 4.1이었다. 실험식이 섭취기간동안 소변의 평균 나트륨과 칼륨의 배설량은 각각 100.2 ${\pm}$ 14.5 mEq, 37.1 ${\pm}$ 7.4 mEq이었다. 소변의 나트륨/칼륨 비율은 2.8 ${\pm}$ 0.6이었다. 소변 중 나트륨 배설량과 나트륨/칼륨 비율은 실험식이 섭취기간에 유의적으로 감소하였다 (p < 0.05). 본 연구 결과 NaCl-KCl 혼합식염 식이는 나트륨의 섭취를 감소시키고 칼륨의 섭취를 증가시켜 나트륨/칼륨의 섭취비를 이상적인 수준인 1에 가깝게 변화시켜 주는 것으로 나타나 고혈압의 예방에 효과가 있을 것으로 사료된다. 특히 짠맛에 대한 기호도가 높은 우리나라 사람들에게 짠맛에 대한 기호도를 변화시키지 않고도 실제로 식생활에서 나트륨 섭취를 감소시킬 수 있는 구체적 방안을 제시할 수 있을 것으로 기대된다. 그러나 본 연구는 정상혈압을 가진 성인 여성을 대상으로 하였으므로 고혈압 환자 또는 나트륨과 관련이 있는 환자를 대상으로 한 실험이 이루어져야 할 것이다. 또한 소금 섭취가 높은 대상자들이나 나트륨의 섭취를 제한해야 하는 사람들에게 KCl을 이용한 혼합식염을 공급하여 나트륨 섭취를 감소시킴으로써 얻을 수 있는 건강증진 효과에 대한 후속 연구가 필요하다고 생각된다. 그러나 한편으로는 염화칼륨을 보충제로 과도하게 섭취할 경우 위장장애 등을 일으킬 수 있다고 알려져 있으므로 적절한 섭취량을 찾는 연구가 필요할 것이다.
