Pharmacokinetics and Tissue Distribution of UTI in the Rat (랫드에서의 UTI의 약물동태학 및 조직 분포)

  • The purpose of this study was to determine pharmacokinetic parameters and tissue distribution patters of urinary trypsin inhibitor(UTI) in Sprague-Dawley rats. $Na^{125}$I was conjugated to UTI to make $^{125}I-UTI$ and the concentrations were determined by $\gamma$-counter. With the aid of nonlinear least-square regression analysis for i.v bolus injection of 1,000 unit UTI including $^{125}I-UTI$, the temporal concentration curves were best fitted by 2-compartment open model. The distribution phase half-life was 0.39$\pm$0.02 hours whereas the elimination half-life was 12.99$\pm$1.05 hours in male rats. The volume of distribution and total body clearance in male rats were 0.28$\pm$0.01 1/kg and 83.16$\pm$1.15 ml/kg/h, respectively. We could not find any difference of pharmacokinetic parameters of UTI between male and female rats. UTI were distributed widely in rat organs. In both male and female rats, the kidney was the highest distributed organ. Amount of UTI in 24 hour cumulative urine in male rats was 36.22$\pm$8.74% and that in 48 hours was 43.32$\pm$10.55%. Excretion via feces was very scanty, with the 24 hours cumulative amount being only 2.76$\pm$0.97%. This data suggest the main excretion route of UTI is urine.

  • The purpose of this study was administered to the body for examination and treatment, high-energy radioactive isotope(F-18, I-131) in vitro discharge experiments. Increasing exposed dose of radiation to health professionals is caused by the increase of PET/CT use and a radioactive isotope. Therefore, the high-energy isotope F-18 and I-131 after administration about using Metabolite excretion was studied. As a results of this study, patients had plenty of fluids for testing and treatment alone administered radiopharmaceuticals can be more than twice as fast excretion induced emissions. Also was able to get a better image space to reduce the dose rate.

Effect of a 6-month Low Sodium Diet on the Salt Taste Perception and Pleasantness, Blood Pressure and the Urinary Sodium Excretion in Female College Students (6개월간의 저염식이 여자 대학생의 짠맛에 대한 인지와 기호, 혈압 및 뇨 나트륨 배설량에 미친 영향)

  • The study aim was to examine the effect of sensory responses of subjects after 6-month dietary sodium reduction with the aid of nutritional education. Fourteen female college students voluntarily restricted their sodium intake for 6 months, during which time they received nutritional education on the low sodium diet. As a control group, 10 students, whose anthropometric measurement, sodium intake behavior, and blood pressure were not different from those of the experimental group, were maintained on a normal diet. For the sensory responses of subjects, the salt taste perception and pleasantness for graded (0.15-1.3%) NaCl solutions were measured by a 9-point hedonic scale. The optimum sodium concentration, urinary sodium excretion, and blood pressure were measured. All the measurements were done at the beginning and end of the experiment. The sensory evaluation revealed an absence of any difference between the two groups in salt taste perception and pleasantness responses at the beginning. After 6-month adaptation, the experimental group subjects showed higher responses to low NaCl solution (0.15, 0.3, 0.5%) in salt taste perception and pleasantness evaluation while the control group subjects exhibited the opposite response. The optimum sodium concentration was reduced from 105.6 mmol to 80.7 mmol (p = 0.015) and the urinary sodium excretion was also reduced from 1,398 mg to 906 mg (p = 0.041) only in the experimental group. Systolic blood pressure was significantly reduced in the experimental group, although there was no correlation between the urinary sodium excretion and blood pressure. The optimum sodium concentration was negatively correlated with the urinary sodium excretion (r = 0.418, p = 0.053), indicating that adaptation to low sodium diet can reduce sodium intake. Further study on the individual responses of subjects on a low sodium diet by periodical evaluation may provide useful data for setting the duration needed to stabilize a lowered appetite for sodium.

Verification of Utility of Simple Mensuration of Cl- from Urine to Estimate the Amount of Sodium Intake (나트륨 섭취량 추정을 위한 소변 중 Cl-의 간이적 측정방법의 유용성 검증)

  • The purpose of this study was to measure concentrations of $K^+$, $Na^+$, $Cl^-$ by ionometer with check salt strip, simple salimeter and Ion-selective electrode (ISE) and compare the results of each mensuration; furthermore, the possibility of inferring the $Na^+$ concentration from $Cl^-$ concentration of urine and the impact of $K^+$ on the concentration of each ion was examined. The results showed that ISE determined $Na^+$ and $Cl^-$ concentrations in the urine are highly interrelated (R=0.9039); in addition, concentrations of $Cl^-$, measured with strip and ISE from urine are highly interrelated (R=0.9338). The concentration of $Na^+$ in urine, inferred by measuring $Cl^-$ concentration with strip, has a high relationship (R=0.8580) with the concentration of $Na^+$ in urine, measured by ISE. The results of our study will increase awareness of $Na^+$ intake and the utility of check salt strip, as well as the possibility of inferred $Na^+$ concentration from measures of $Cl^-$ concentration as a screening test for reducing sodium intake.

A Study on Salt Intake and Urinary Sodium Excretion by Groups Educated Regarding Low Salt Diet (일부 저염식 피교육자의 식염섭취 및 뇨중 Sodium 배설양상 -장류 및 김치류 등 고식염 함유 식품을 중심으로-)

  • To estimate the amount of daily salt intake by source of food and urinary sodium excretion, a dietary survey was conducted from August 27th to September 3, 1980. The salt concentration of six kinds of food was measured. The families of 25 womens' club leaders and 25 families of hypertensive patients were studied after they had been given a short course of education on lowsalt diet. The results were as follows: 1. The average amount of high-salt foods eaten daily by the study group were as follows: 4.5 Gm. in the form of table salt 16.8 ml of home-made soy sauce 6.20 ml of commercial soy sauce 11.9 Gm. of home-made red-pepper bean paste 12.0 Gm. of home-made soy-bean paste 120.7 Gm. of Kimchi 2. The average NaCl concentration of each food was as follows: 99.5% in table salt, 22.5% in home made soy sauce, 11.9% in Commercial soy sauce, 6.8 Gm% in home made red pepper bean paste, 9.2 Gm% in home-made soy-bean paste and the average concentration of various kinds of Kimchi was 2.7 Gm%. 3. The total amount of daily salt intake per adult was calculated as 14.3 Gm. By sources of food 4.5 Gm from table salt, 3.9 Gm from home-made soy sauce and 3. 3 Gm from Kimchi were taken daily Three other kinds of food were also minor sources of salt intake.4. The average amounts of individual daily salt intake were not significantly different between the hypertensive group and the normotensive group: the median of the normotensive group (11.7Gm.) was significantly lower than that of the hypertensive group (14.9Gm.). Therefore the womens' club leaders appeared to respond more quickly than the hypertensive group after low-salt diet education. 5. The average amount of sodium excretion in the 24 hour urine specimen was 234.7 mEq. From this finding the daily NaCl intake was estimated to be 15.8Gm.

A Study on the Intkae-Balance of Sodium and Potassium of College Men of Korea (한국 남자 대학생의 Sodium 과 Potassium 평형에 관한 연구)

  • This study measured the daily intake and excretion of sodium and potassium of eight 20-26years old college men during four weeks by means of analyzing their food intake, urine and feces, keeping their normal living pattern and body weight. This study also compared the actual measurement value of sodium and potassium intake by atomic absorption spectrophotometer with the conversion value of them by food table. The results are as follows ; Daily mean sodium intakes conversed ($2.36{\pm}0.03g/day$) was about 63% lower than those intakes meansured($6.36{\pm}0.13g/day$). Daily mean potassium intakes conversed($1.71{\pm}0.03g/day$) was not different of sodium and potassium were $5.49{\pm}0.19g/day\;and\;1.33{\pm}0.08g/day$, respectively. Daily mean fecal excretions of sodium and potassium were $0.24{\pm}0.02g/day\;and\;0.45{\pm}0.03g/day$, respectively. Mean prooportion of Na/K in urine was $4.3{\pm}0.1$.

Effect of Monensin or Salinomycin Supplementation in a 50% Concentrate Diet on Mineral Utilization of Growing Goats

  • An experiment was conducted to clarify the effects of dietary supplementation of monensin or salinomycin on mineral utilization of growing goats. Six goats weighing 10.54 kg initially were randomly assigned to treatments in a replicated $3{\times}3$ Latin square design. Treatments were a basal diet, basal plus 30 ppm monensin and basal plus 20 ppm salinomycin on a DM basis. The basal diet was a mixture of Italian ryegrass wafer, soybean meal, ground maize and $CaCO_3$ with DM proportions of 50, 13.76, 36 and 0.24%, respectively. Each period lasted for 21 days, and the apparent absorption and retention of minerals were measured during the last 7 days of each period. Salinomycin supplementation improved NDF digestibility and plasma glucose. The apparent absorption and retention of Ca, P, Mg, Na and K were not influenced by the treatments. The concentrations of plasma Ca, P, Mg, Na, K and Cl were similar in all treatments. The ionophore supplementation had no significant effect on acid excretion. The results suggest that 30 ppm monensin or 20 ppm salinomycin supplementation is not effective in improving the utilization of Ca, P, Mg, Na and K in growing goats fed a diet composed of the 50% concentrate.

Decreased Expression of $Na^+/K^+$-ATPase, NHE3, NBC1, AQP1 and OAT in Gentamicin-induced Nephropathy

  • The present study was aimed to determine whether there is an altered regulation of tubular transporters in gentamicin-induced nephropathy. Sprague-Dawley male rats ($200{\sim}250\;g$) were subcutaneously injected with gentamicin (100 mg/kg per day) for 7 days, and the expression of tubular transporters was determined by immunoblotting and immunohistochemistry. The mRNA and protein expression of OAT was also determined. Gentamicin-treated rats exhibited significantly decreased creatinine clearance along with increased plasma creatinine levels. Accordingly, the fractional excretion of sodium increased. Urine volume was increased, while urine osmolality and free water reabsorption were decreased. Immunoblotting and immunohistochemistry revealed decreased expression of $Na^+/K^+$-ATPase, NHE3, NBC1, and AQP1 in the kidney of gentamicin-treated rats. The expression of OAT1 and OAT3 was also decreased. Gentamicin-induced nephropathy may at least in part be causally related with a decreased expression of $Na^+/K^+$-ATPase, NHE3, NBC1, AQP1 and OAT.

Inhibition of $N^{+}-K^{+}$ Adenosine Triphosphatase Activity in Fisher Rats by Uranyl Nitrate

  • An attempt was made to test the possibility of a major role for the $Na^{+}-K^{+}$ adenosine triphosphatase (ATPase)system in the diuresis induced by uranyl nitrate(UN). Fisher 344 rats were intravenously injected with UN(5 mg/kg, 15 mg/kg and 30 mg/kg). Urinary excretion of $Na^{+}\;and\;K^{+}$ significantly increased in 24 h exposure on the UN and then decreased below the normal level 3 days after the treatment. $Na^{+}-K^{+}$ ATPase activity of kidney was significantly inhibited in high dosages of UN 15mg/kg and UN 30 mg/kg 3-5 days after injection. And then the recovery of the enzyme activity was observed within 5-10 days after injection, at which the regeneration of the tubular cells occurred.

The antihypertensive effect of ethyl acetate extract of radish leaves in spontaneously hypertensive rats

  • Radish (Raphanus sativus L.) is a cruciferous vegetable, and its leaves have antioxidant and anticancer properties. This study was conducted to evaluate the effects of ethyl acetate extracts from radish leaves on hypertension in 11-week-old spontaneously hypertensive rats (SHRs). The SHRs were randomly divided into 3 groups of 6 rats each on the basis of initial systolic blood pressure (SBP) and were treated with oral administration of radish leaf extract (0, 30, or 90 mg/kg body weight [bw], respectively) for 5 weeks. Six Wistar rats were used as normotensive controls. The amount of the radish leaf extract had no effect on body weight. The SBP of the SHRs showed a decreasing trend with the consumption of the radish leaf extract. In the third week, the SBP of the group fed 90 mg extract/kg bw reduced from 214 mmHg to 166 mmHg and was significantly lower than that of the normotensive and hypertensive controls. The extract did not show a significant effect on the angiotensin-converting enzyme (ACE) activity in the serum, kidney, and lung. The extract increased the concentration of NO in serum and the activities of antioxidant enzymes such as glutathione peroxidase and catalase in red blood cells (RBCs). The serum concentrations of $Na^+$ and $K^+$ were not significantly different between all groups. However, the fecal concentrations of $Na^+$ and $K^+$ increased; the fecal concentrations of $Na^+$ and$K^+$for the normotensive and hypertensive controls were not different. Urinary excretion of $Na^+$ was higher in the normotensive Wistar rats than in the SHRs, while that of $K^+$ was not significantly different. These findings indicate that consumption of radish leaves might have had antihypertensive effects in SHRs by increasing the serum concentration of NO and fecal concentration of $Na^+$ and enhancing antioxidant activities.
