Effect of Jasmonic Acid and NaCl on the Growth of Spearmint(Mentha spicata L.) (Jasmonic Acid 및 NaCl 처리가 스피아민트의 생육에 미치는 영향)

  • This study investigated the effects of NaCl and jasmonic acid (JA) on the growth and physiological responses of spearmint (Mentha spicata L.). Spearmint was hydroponically grown for 3 weeks in modified Hoagland solution containing 0 (untreated control), JA ($20{\mu}M$ JA pretreatment), NaCl (50 mM NaCl treatment) and JA + NaCl ($20{\mu}M$ JA pretreatment + 50 mM NaCl treatment). Growth characteristics, chlorophyll, vitamin C, proline contents, DPPH scavenging activity and inorganic ion contents were evaluated. As a results, there were significant decreases in the plant height, leaf length, leaf width, and fresh weight of plants, treated with NaCl compared with control. On the other hand, the dry matters of shoot and root treated with JA + NaCl combination were better than control or NaCl treatment. Chlorophyll a and b contents in JA treatment was the highest. Vitamin C, antioxidant activity, and proline content in shoot were increased in NaCl treatment which showed low level of growth rate. The K/Na ratio, which is known to indirectly reflect the balance of ion uptake, was higher in a single treatment of JA than the control group, while lower in salt treatment (NaCl and JA + NaCl) because of high $Na^+$ absorption. In conclusion, these results showed that moderate stress treatment such as low level salt treatment and plant growth regulator jasmonic acid (JA) application would be potential strategies to improve the quality of spearmint by inducing the accumulation of secondary metabolites containing high antioxidant activity and essential oil.

Growth and Histological Characteristics of Barley (Hordium vulgare L.) Seedling to NaCl Stress (NaCl Stress에 따른 보리 유묘의 생육특성 및 세포학적 반응)

  • This study was conducted to determine the morphological responses of barley seedlings to NaCl stress and to investigate histological changes of cells with transmission electron microscope(TEM) after NaCl stress. Plant height and root length of 10-day old barley seedlings with NaCl stress were reduced and inhibition level was found to be more severe in the plant height than in the root length. The leaf length, leaf width and leaf area were shorter as well with NaCl stress than without NaCl stress. However, there was no difference in the number of roots between NaCl treatments. The weight of dry matter decreased at higher NaCl concentrations, especially at 100mM NaCl. The water content of shoots tend to decrease at higher NaCl concentrations, but there was no difference in the water content of roots, The reduced sugar content was greatly increase than starch. Cellulose content was higher in NaCl stressed-plant than control, and tended to decreased at higher NaCl concentrations. Lignin content also decreased NaCl stressed-plant but there was no tendency at NaCl stress concentrations. Electric conductivity of cell sap with seedlings was high with NaCl stressed-plant. Amount of cell sap gradually increased with time in the roots than in the shoots, The grana of chloroplasts was changed by 150mM NaCl concentration. The christe of mitochondria in root meristematic sells ruined in structure and cell wall of leaf and root was also ruined by NaCl stress.

The effect on photosynthesis and osmotic regulation in Beta vulgaris L. var. Flavescens DC. by salt stress

  • This study was to investigate the effect of salt stress on physiological characteristics such as plant growth, photosynthesis, solutes related to osmoregulation of Beta vulgaris. A significant increase of dry weight was observed in 50 mM and 100 mM NaCl. The contents of Chl a, b and carotenoid were lower in NaCl treatments than the control. On 14 day after NaCl treatment, photosynthetic rate (PN), the transpiration rate (E) and stomatal conductance of CO2 (gs) were reduced by NaCl treatment. On 28 day after NaCl treatment, the significant reduction in gs and E was shown in NaCl 200 mM. However, PN and water use efficiency (WUE) in all NaCl treatments showed higher value than that of control. Total ion contents (TIC) and osmolality were higher than the control. On 14 day after treatment, the contents of proline (Pro) increased significantly in 200 mM and 300 mM NaCl concentration compared with control, whereas on 28 day in all treatments it was lower than that of the control. The contents of glycine betaine (GB) increased with the increase of NaCl concentration. The contents of Na+, Cl-, GB, osmolality and TIC increased with the increase of NaCl concentrations. These results suggested that under severe NaCl stress conditions, NaCl treatment did not induce photochemical inhibition on fluorescence in the leaves of B. vulgaris, but the reduction of chlorophyll contents was related in a decrease in leaf production. Furthermore, increased GB as well as Na+ and Cl- contents resulted in a increase of osmolality, which can help to overcome NaCl stress.

  • This paper was carried out to investigate the relation of pH, aciidity and sourness during Kimchi fermentation and preservatives on Kimchi fermentation. Na-acetate, Na-malate, K-sorbate and K-sorbate+acetic acid were added to Kimchi samples. These Kimchi samples were fomented for 7 days at $37^{\circ}C$ and $20^{\circ}C$. In the experiment about the sourness and buffer action by organic salts which showed that the intensity of sourness was differented by the difference of pH in the same acidity. Na-acetate (0.3%) and Na-malate (0.3%) acted as good buffer, whereas K-sorbate (0.1%) and K-sorbate (0.1%)+acetic acid (0.05%) acted as lactobacilli growth enhancer in the fermentation.

Effect of NaCl Concentration on Photosynthesis and Mineral Content of Barley Seedlings under Solution Culture

  • This study was conducted to elucidate the changes of photosynthetic ability and cation content in barley cultivar seedlings cultured for 10 and 30 days with different NaCl concentrations containing 1/4 Hoagland solutions. At the highest NaCl concentration, the weight of dry matter and the shoot/root ratio (S/R ratio) were decreased. Thus, shoots were affected more than roots by NaCl treatment. The S/R ratio decreased more in 'Neulssalbori' than in 'Bunong' by the NaCl treatment. The. internal $Na^+$ concentration increased greatly with the highest NaCl concentration, but $K^+$ concentration in plants decreased with the highest NaCl treatment. The $Ca^{2+}$ concentration had a small change with NaCl concentrations. Thus $Na^+$/$K^+$and $Na^+$/$Ca^{2+}$ratios increased with the highest concentration. The chlorophyll content (%/dry weight) of seedlings decreased at higher NaCl levels except for Bunong in 30 day old seedlings. The photosynthetic ability decreased only for Neulssalbori in the 10 days NaCl treatment. The stomatal conductance, and transpiration had decreased in the 10 day old seedlings, but not with 30 day old seedlings.

Antibacterial Synergic Effect and Cellular Responses of Nalidixic Acid-Resistant Salmonella typhimurium Exposed to Tea Polyphenols and Nalidixic Acid (Nalidixic Acid 내성인 Salmonella typhimurium의 녹차 폴리페놀과 Nalidixic Acid에 의한 살균상승 효과 및 세포반응)

  • The purpose of this work was to investigate the synergically bactericidal effects and cellular responses of green tea polyphenols (TPPs) and nalidixic acid (NA) on nalidixic acid-resistant (NAR) Salmonella typhimurium. The bactericidal activities of $>3,500{\mu}g/ml$ TPPs and $<256{\mu}g/ml$ NA were investigated for S. typhimurium of which initial cell number was approximately adjusted to 107 cell/ml. Complete elimination of NAS-S. typhimurium was achieved within 6 hr of incubation at the concentrations of $3,500{\mu}g/ml$ TPP or $256{\mu}g/ml$ NA, whereas only partial bactericidal effect was achieved under the same conditions. However, the combinations of $3,000{\mu}g/ml$ TPPs and $32{\mu}g/ml$ NA against NAS-S. typhimurium and $3,500{\mu}g/ml$ TPPs and $64{\mu}g/ml$ nalidixic acid against NAR-S. typhimurium showed complete removal within 5 hr of incubation. The stress shock proteins (SSPs) were induced at different concentrations of TPP o rNA used as stressors against cell culture of S. typhimurium. The proteins were identified as 70-kDa DnaK and 60-kDa GroEL by SDS-PAGE and Western blot. SSPs induced by the stressors were found to increase in proportion to the TPPs or NA. Scanning electron microscopy analyses revealed the presence of perforations and irregular rod shape with wrinkled surfaces for cells treated with TPPs or NA.

Effect of NaCl on Nitrogen Content of Barley Seedlings

  • This study was conducted to determine the effects of NaCl stress on nitrogen, ${NH_4}^+$, and ${NO_3}^-$ content of 4 barley cultivar seedlings that were cultured for 10 and 30 days with different NaCl levels (0, 50, 100, and 150 mM) contain-ing 1/4 Hoagland solutions. The sodium ion content in the shoot of barley seedlings sharply increased with an increase of NaCl concentration. After 30 days of NaCl treatment, the sodium content of the shoot at 150 mM NaCl was 27 times higher than in non-saline conditions. The sodium content in the root linearly increased with increasing NaCl concentration. Nitrogen content in the shoot linearly increased with increasing NaCl concentration, but nitrogen content in the root declined above the point where the $Na^+$ content was 3.0 mM/g $Na^+$ in the barley seedling. ${NO_3}^-$ content also decreased with increasing NaCl concentration. ${NH_4}^+$ content in the shoot decreased with NaCl condition, but its content in the root increased with NaCl condition. A positive correlation between ${NO_3}^-$ and ${NH_4}^+$ content was found in the shoot, but their relationship was negative in the root.

  • The selective transport characteristics of $K^+$ and $Na^+$ of cell membrane model which irradiated by 60Co ${\gamma}$-ray was investigated. The cell membrane model used in this experiment was a Na+ type sulfonated copolymerized membrane of styrene and divinylbenezene. The initial flux of the ion was increased with increase of both $H^+$ ion concentration. In this experiment range(pH 0.5-3, temperature $15-65^{\circ}C$), first, the selectivity of $K^+$ and the ratio K+/Na+ of membrane which was not irradiated was about 1.06 - 1.13 and second, that of K+ and the ratio $K^+/Na^+$ of membrane which was irradiated was near about 0. And the driving force of pH of irradiated membrane was significantly increased about 4-5 times than membrane which was not irradiated. As selective transport of K+ and Na+ of cell membrane model were abnormal, cell damages were appeared at cell.

Estrogen Effect on the Na,K-ATPase Activity Repressed by IgE-Dependent Histamine-Releasing factor in HeLa Cells (HeLa 세포에서 Estrogen이 Na,K-ATPase활성에 미치는 영향)

  • IgE-dependent histamine-releasing factor (HRF) is found extracellularly to regulate the degranulation process of histamine in mast cells and basophils and known to play a predominant role in the pathogenesis of chronic allergic disease. HRF has been also identified in the intracellular region of the cell. Previously, we reported that HRF interacts with the 3rd cytoplasmic domain of the alpha subunit of Na,K-ATPase and inhibits Na,K-ATPase activity. Since it is known that estroaen activates the sarcolemmal Na,K-ATPase, we tested whether estrogen recovers the Na,K-ATPase activity repressed by HRF. In this study, we showed that estrogen activates Na,K-ATPase repressed by HRF. RT-PCR and western blot analysis showed that estrogen doesn't reduce the expression level of HRF in HeLa cell, suggesting that this recovery effects of estrogen probably occur via indirect mechanism on HRF and Na,K-ATPase.

The Effects of Builders on Calcium Deposition on the Fabric (세척시 조제의 종류가 직물에의 Calcium 침착에 미치는 영향)

  • The influence of builders on calcium deposition on the fabric was studied by laundering the cotton fabric with sodium carbonate, sodium metasilicate, sodium tripolyphosphate and built detergents in hard water. The laundry variables were: 1) Washing cycles: 5, 10, 20, 30 and 40 cycles. 2) Water hardness: 100 ppm, 150 ppm, 200 ppm and 300 ppm. 3) Builders: $Na_2\;CO_3,\;Na_2\;SiO_3$ and STPP. 4) Detergents: Na-DBS, $Na-DBS+Na_2CO_3,\;Na-DBS+Na_2\;SiO_3,\;Na-DBS+STPP,\;Na-DBS+Na_2\;CO_3+STPP$, and $Na-DBS+Na_2\;SiO_3+STPP$. The fabric was washed for 15 minutes at 23+$1^{\circ}C$ in a washing machine(Gold Star WP 3007) under the similar condition with those of home laundering, and rinsed 3 times in the same water hardness for 5 minutes. The calcium deposits on the fabric was determined by EDTA-BACK titration methods. The results of this study were as follows: 1) The amount of calcium deposits on the fabric was increased with increasing wash cycles. This deposit was due to the build up of insoluble calcium carbonate. 2) As the water hardness increased, the amount of calcium deposits on the fabric was increased. 3) Alkaline builders, such as, $Na_2CO_3$ and $Na_2SiO_3$, promoted calcium deposition on the fabric, however STPP prevented calcium deposition on the fabric. 4) Fabric laundered with $Na-DBS+Na_2CO_3$ showed the highest calcium deposits on the fabric, and decreased with the order of $Na_2CO_3$, $Na-DBS+Na_2SiO_3$, and Na-DBS. And fabrics washed with phosphate-built detergents showed a small amount of calcium deposition.
