  • BACKGROUND/OBJECTIVES: Supplementation with n-3 polyunsaturated fatty acids (PUFAs) has been shown to generally decrease levels of innate immune markers and inflammatory cytokines, but the specific associations between blood levels of PUFAs and those of innate immune markers have not been investigated. Thus, the present study was conducted to test the hypothesis that innate immune markers as well as cytokines are negatively associated with n-3 PUFAs but positively associated with n-6 PUFAs in healthy adults. MATERIALS/METHODS: One hundred sixty-five healthy Korean adults aged 25-70 years old were included in this cross-sectional study. RESULTS: Serum levels of n-3 PUFAs, such as 18:3n3, 20:5n3, 22:5n3, and 22:6n3 were negatively correlated with eosinophil and basophil counts and $TNF-{\alpha}$, $IFN-{\gamma}$, IL-4, and IL-10 levels. Multivariate analysis also showed that serum levels of n-3 PUFAs were negatively associated with monocyte, eosinophil, and basophil counts and $TNF-{\alpha}$, $IFN-{\gamma}$, IL-4, and IL-12 levels. Additionally, the ratio of 20:4n6 to 20:5n3 was positively correlated with eosinophil counts and associated with $TNF-{\alpha}$, $IFN-{\gamma}$, and IL-4 levels. However, NK cell activity was not associated with serum fatty acid composition. CONCLUSIONS: Innate immune markers such as eosinophil, monocyte, and basophil counts were inversely associated with serum levels of n-3 PUFAs, but were positively associated with the 20:4n6/20:5n3 ratio in this population.

  • Water polution status of urban streams in Daegu city were observed to provide the basic information for the effective purification of urban sewage and the conservation of Keumho river. Periodically, pH, DO, COD, nitrate and phosphate of water were investigated at Yee cheon, B$\ddot{o}$m$\ddot{o}$ cheon, Chilsung cheon, Dalseo cheon and Kongdan cheon. The results were as follows. 1) The ranges of average values of analyzed components for 12 months at six sampling sites were pH 7.3-8.2, DO trace-6.5ppm, COD 20.4-116.9ppm, T-N 23.2-31.7ppm, $NH_4$-N 18.3-27.7ppm, $NO_2$-N 0.08-1.89ppm, $NO_3$-N 0.19-1.51ppm, $PO_4$-P 2.50-17.28ppm. 2) At Kongdan cheon, the most heavily polluted site, average values of components were pH 8.2, DO trace, COD 116.9ppm, T-N 23.2ppm, $NH_4$-N 18.3ppm, $NO_2$-N 1.89ppm, $NO_3$-N 1.51ppm, $PO_4$-P 17.28ppm. 3) The values of pH, DO, COD, T-N and $NH_4$-N at winter urban streams were higher than those at summer urban streams. And the values of $NO_2$-N and $PO_4$-P were more or less higher at summer urban streams.

  • Pig slurry, urine and fermented liquid manure were evaluated measuring electrical conductivity (EC), dry matter (DM) contents and other components. Samples were collected during a period from February to April, 2001: slurry samples from 70 storage tanks, urine samples from 19 and 20 fermented liquid manure samples from 109 pig farms in Jeju. Samples were analyzed for pH, EC, DM, $NH_4$-N, K, P, Ca, Mg, Na, BOD, S-S, Cd, Cr, Cu, Fe, Mn, Ni, Pb, Zn and OM. Relationships between EC or DM and various mineral and $NH_4$-N contents were examined using linear regression. EC, DM, all mineral contents and $NH_4$-N were higher in slurry samples than in urine or fermented liquid manure. EC, $NH_4$-N, Ca, Mg and Na concentration was similar in urine and fermented liquid manure, but K and DM values in liquid manure were twice as much as in urine. P concentration was higher in urine than in liquid manure. $NH_4$-N and Na in slurry, $NH_4$-N in urine and $NH_4$-N, P and Ca in fermented liquid manure were highly correlated with EC, while $NH_4$-N, P, Ca and Mg in slurry, $NH_4$-N and Ca in urine, and K, P, Ca, Mg, and Na in fermented liquid manure with DM (P<0.05). BOO in slurry (22,520 mg/mL) was higher than that in urine (4,763) and fermented liquid manure (2,701). Results indicated that slurry is a better fertilizer source than urine or fermented liquid manure. However, slurry may pollute soil more than urine or fermented liquid manure when applied to land, although the levels are not above the permit.

  • The monomer N,N'-bis(3-pyrrol-1-yl-propyl)-4,4'-bipyridinium$(PF_6)_2$ was electrochemically polymerized on glassy carbon electrode surface. This polymer film electrode has electroactive sites on its bipyridinium ions distributed at the polymer strands. The formal potentials of the electrodes were -0.41V and -0.81V(vs. SSCE) for each step at phosphate buffer(pH=5.70). The diffusion coefficients of the dopants ions into the polymer matrix were $1.57{\times}10^{-4}$ and $4.35{\times}10^{-5}cm^2s^{-1}$ for first and second redox couple, respectively. The rate constants of electron transfer at $V^{2+/+}$ of the first step was a $57.53s^{-1}$, which was 22 times higher than $V^{+/0}$ one having $2.63s^{-1}$ in the solution. The charge transfer resistance of the polymer film was influenced by the dopant ion of the electrolyte. Thus the resistances were 22.63, 16.81, 12.44 and $11.36k{\Omega}$ for $LiClO_4,\;NaClO_4,\;KClO_4$, and phosphate buffer, respectively. The reaction order of the electropolymerization was first order and the rate constant of the polymerization was $1.31{\times}10^{-1}s^{-1}$ as determined by EQCM method. The G.C./p-BPB type electrode doped with phosphate ions showed a stability and reproducibility in CV procedure over 20 cycles.

  • The ion selective microelectrode (ISME) has been used for measuring the ion profile of DO, $NH_4{^+}-N$, $NO_2{^-}-N$ and $NO_3{^-}-N$ in biofilm. In this study we evaluated the detection limit and validity of ISME and applied ISME for the continuous measurement of $NH_4{^+}-N$ and $NO_3{^-}-N$ concentration in the modified Ludzack-Ettinger (MLE) process. Average detection limits of $NH_4{^+}-N$ and $NO_3{^-}-N$ ISME were $10^{-4.44}M$ and $10^{-4.62}M$, respectively. Since the ISME with $5{\sim}10{\mu}m$ of tip diameter showed a faster response time than that of $1{\sim}5{\mu}m$, the ISME with a tip diameter of $5{\sim}10{\mu}m$ was fabricated and used to make real-time ion detections. Direct monitoring of $NH_4{^+}-N$ and $NO_3{^-}-N$ concentrations in the aerobic (2) tank causes the instability of the electromotive force (EMF) for the initial 5~8 hours and also causes remarkable error values of $NH_4{^+}-N$ and $NO_3{^-}-N$ concentration. This phenomenon is caused by aeration and mixing in the reactor. Thus, the measuring chamber was newly designed for the aerobic (2) tank and then the EMF of the ISME were stabilized in less than 1 hour. Errors of $NH_4{^+}-N$ and $NO_3{^-}-N$ concentration were decreased after stabilization of the EMF. The ISME analysis were well corresponded to the results of auto analyzer and ion chromatography. Consequently, the concentration of $NH_4{^+}-N$ and $NO_3{^-}-N$ could be continuously measured for 178 hours by the ISME.
