Manufacture of Cutting-Gouda Cheese using Lactobacillus rhamnosus_p1 and the Physicochemical Properties of Gouda Cheese during Ripening Periods (Lactobacillus rhamnosus_p1을 이용한 절단형 고다치즈 제조방법 및 숙성 중 품질특성)

  • The aim of this study was to manufacture Cutting-Gouda cheese and to investigate the change in physicochemical properties of Cutting-Gouda cheese made with Lactobacillus rhamnosus_p1. Lactic acid bacteria were isolated from Gouda cheese ripened for more than 1 year. They were identified as 2 strains of L. rhamnosus, Lactobacillus casei, Lactobacillus curvatus, and Staphylococcus saprophyticus by 16S rDNA sequencing and named L. rhamnosus_p1, L. casei_p2, L. curvatus_p3, L. rhamnosus_p4 and S. saprophyticus_p5. The proteolytic activities of isolated strains against casein were measured using prepared skim milk agar plates. L. rhamnosus_p1 showed the highest proteolytic activity. Cutting-Gouda cheese was made with L. rhamnosus_p1, and its physicochemical properties (moisture, protein, fat, ash and free amino acid content) were measured during ripening periods. Because of the modified atmosphere packaging ($N_2{^-}$), there was no change in moisture, protein, fat, and ash in the experimental group. The total amount of free amino acids in the control and experimental group gradually increased during ripening periods. The sensory evaluation showed that the experimental group was preferable to the control group. This result suggests that L. rhamnosus_p1 has potential to be developed as a new starter for Gouda cheese.

Community Structure of Macrobenthic Fauna in the Hallyeohaesang National Park from Korea Strait, Korea (한려해상국립공원 조하대 해역 대형저서동물의 군집구조)

  • The study was performed to investigate the community structure of macrobenthic fauna during June 2005, in Hallyeohaesang National Park from Korea Strait. A total of 284 macrobenthic fauna were collected. The overall average macrobenthic density and biomass were 2,002 individuals m$^{-2}$ and 154.92 gWWt m$^{-2}$, respectively. Based on the Lebris (1988) index, there were 20 species accounting for approximately 47.64% of total individuals. And the highest densities were found in the polychaetes Scoletoma longifolia, Isolda pulchella, Mediomastus californiensis, Minuspio multibranchia, Tharyx sp. 1 and the bivalve Theora fragilis. On the other hand, the top twenty species made up 70.47% of the total biomass while the three most abundant, the echinoderms Schizaster lacunosus, Amphiura vadicola and the bivalve Fulvia mutica. The conventional multivariate statistics (cluster analysis and non-metric Multi-Dimensional Scaling) applied to assess spatial variation in macrobenthic assemblages. As a result of cluster analysis and nMDS ordination, this study area was divided into two different groups of stations and species. In case of station, there are two groups: one is a mud dominated station group and the other is dead shell and sand dominated group. And the 2 faunistic groups were established as follows: 1) a group of relatively higher abundances in mud dominated sediment. There were numerically dominated by the polychaetes Magelona japonica, Sternaspis scutata, Paraprionospio pinnata, Tharyx sp. 1 and the amphipods Monocorophium sinense and Eriopisella sechellensis. 2) a group of relatively higher densities dead shell and sand dominated sediment. These groups were characterized by the polychaetes I. pulchella, M. multibranchiata, the amphipods M. acherusicum, Gammaropsis japonicus and the echinoderm A. vadicola.

Isolation and Evaluation of an Antiviral Producing Serratia spp. Strain Gsm01 against Cucumber mosaic virus in Korea (한국에서 CMV에 항바이러스 효과를 나타내는 Serratia spp. Gsm01 균주의 분리 동정 및 효과 검정)

  • An Antiviral producing bacterial strain was isolated from ginseng root environment in Hongcheon, Kangwon province of Republic of Korea. Identification of this bacterial strain was performed by physiological and biochemical tests along with 16S rRNA analyses. The results revealed that the bacterium was closer to genus Serratia, which was named as Gsm01. The strain was grown in Mannitol-Glutamate-Yeast (MGY) broth for 48 h. The culture was centrifuged and the filtrate obtained was tested for its ability to control Cucumber mosaic virus strain Y (CMV-Y) in greenhouse and field experiments. In the green house experiments, CF was evaluated for its ability to protect local host, Chenopodium amaranticolor and systemic host of CMV, Nicotiana tabacum cv. Xanthi-nc. It was found that, CF treatment reduced viral infection by 98% in local host; C. amaranticolor. The N. tabacum cv. Xanthi-nc plants treated with CF did not show visible viral symptoms 15 days post inoculation (dpi) and remained symptomless throughout the periods of the study. To evaluate effectiveness of CF under field conditions, experiment was carried out in a polyvinyl house. It was observed that, 52% plants were protected from viral diseases compared to non-treated plants, increasing the crop yield. This is the first report showing antiviral activity of a Serratia spp. against CMV.

Effects of Supplemental Alkali Feldspar-Ilite on Growth Performance and Meat Quality in Broiler Ducks (알칼리장석-일라이트가 육용오리의 생산성 및 육질에 미치는 영향)

  • This experiment was conducted to investigate the effect of the supplemental alkali feldspar-ilite(feldspar) on growth performance and meat quality in broiler ducks for 43 days. One hundred eighty broiler ducks were divided into 5 groups of 12ducks. Dietary levels of feldspar 0, 0+antibiotics, 0.5, 1.0 and $1.5\%$ were added to experimental diets of each of the groups. Daily weight gain was slightly increased in 1.0 and $1.5\%$ feldspar treatments. Feed intake was slightly increased at all feldspar treatments. Glucose concentration of serum profile was decreased whereas BUN concentration was significantly increased (p<0.05) at $0.5\%$ feldspar. Cholesterol concentration was decreased at all feldspar treatments, this difference was especially observed in supplemental levels of $0.5\%$ feldspar(p<0.05). Carcass weight was increased at all feldspar treatments. Moisture and crude fat contents of proximate chemical composition in duck meat were decreased at all feldspar treatment, this difference especially was observed in supplemental levels of $1.5\%$ feldspar(p<0.05) on crude fat content. Lightness and yellowness was increased at all feldspar treatment. Cholesterol contents and TBA in meat were decreased, but this parameters were not difference by feldspar treatment. The composition of saturated fatty acids(SFA) was decreased, whereas unsaturated fatty acids(USFA) was slightly increased by feldspar treatment. The Pb content of heavy metal concentrations was increased with compared control, but not difference. The appearance of sensory evaluation was improved by supplemental feldspar, especially in supplemental feldspar, 1.0 and $1.5\%$(p<0.05). The results of this study indicate that the supplemental alkali feldspar may improve the production and meat quality of broiler ducks.

Characteristics and Clinical Course of Ovarian Hernias in Infants (1세 미만 여아 난소 탈장의 특성과 임상 경과)

  • Purpose : Inguinal hernias are common in children and sometimes are associated with dangerous complications, such as incarceration. There are no established management guidelines for ovarian hernias. We have reviewed the clinical course of ovarian hernias in infants. Methods : We reviewed the medical records of female infants diagnosed with ovarian hernias by ultrasonogram at Kwandong University College of Medicine, Cheil General Hospital, and the Women's Healthcare Center between March 2001 and August 2007. We analyzed the patients gestational age, birth weight, age at the time of detection of the inguinal mass, the patients chief complaints, operative time, post-operative complications, and ultrasonographic findings. Results : Eight female infants had ovarian hernias, four of whom were born prematurely. Seven infants had left-sided ovarian hernias, and one infant had a right-sided ovarian hernia. Five infants underwent surgery and there were no postoperative complications or recurrences. Three girls did not have surgery, and the ovarian hernias regressed spontaneously, with no recurrences or complications. The regression time of inguinal masses ranged from 70-161 days after birth. Conclusion : Physical examination to detect movable masses within the labium major in premature female infants is important because the incidence of premature inguinal hernias is much higher than in term infants. No rational medical treatment plans for female ovarian hernias have been published to date. We cared for three girls with spontaneous regression of ovarian hernias. Pediatricians should be aware whether emergent surgery for ovarian hernias is indicated.

Development of Natural Antioxidants and Whitening Agents for Cosmeceuticals

  • Oxidative stress have known to be a risk factor for the degenerative processes and closely related to a lot of diseases. It is well established that antioxidants are good in protection and therapeutic means against oxidative damage. There is increasing interest in natural antioxidants and many natural antioxidants have been found and utilized as the possible protection for various diseases and skin aging. We have screened natural antioxidant agents for cosmeceuticals, nutraceuticals, and drugs as therapeutic and preventive means against oxidative stress, and have developed a number of novel antioxidants from various natural sources. A novel melanin synthesis inhibitor, Melanocin A, isolated from the metabolite of a fungal strain Eupenicillium shearii F80695 inhibited mushroom tyrosinase and melanin biosynthesis of B16 melanoma cells with $IC_{50}$ value of 9.0 nM and MIC value of $0.9\;{\mu}M$, respectively. Melanocin A also exhibited potent antioxidant activity by scavenging of DPPH and superoxide anion radicals. UV was found to increase the level of hydrogen peroxides and other reactive oxygen species (ROS) in skin tissues. This increase in ROS may not only alter the structure and function of many genes and proteins directly but may also modulate their expressions through signal transduction pathways and, ultimately, lead to skin damage. We investigated the effect of Melanocin A on UV-induced premature skin aging. Firstly, the effect of Melanocin A on UV-induced matrix metalloproteinase (MMP)-9 expression in an immortalized human keratinocyte cell line, HaCaT in vitro was investigated. Acute UV irradiation induced MMP-9 expression at both the mRNA and protein levels and Melanocin A suppressed this expression in a dose-dependent manner. We then investigated UV-induced skin changes in hairless mice in vivo by Melanocin A. Chronic exposure of hairless mouse dorsal skin to UV increased skin thickness and induced wrinkle formation and the gelatinase activities of MMP-2 and MMP-9. Moreover, Melanocin A significantly suppressed UV-induced morphologic skin changes and MMP-2 and MMP-9 expression. These results show that Melanocin A can prevent the harmful effects of UV that lead to skin aging. Therefore, we suggest that Melanocin A should be viewed as a potential therapeutic agent for preventing and/or treating premature skin aging. Terrein is a bioactive fungal metabolite isolated from Penicillium species. Terrein has a relatively simple structure and can be easily synthesized. However, the biologic effects of terrein are comparatively unknown. We found for the first time that terrein potently inhibit melanin production in melanocytes and has a strong hypopigmentary effect in a spontaneously immortalized mouse melanocyte cell line, Mel-Ab. Treatment of Mel-Ab cells with terrein (10-100 mM) for 4 days significantly reduced melanin levels in a dose-dependent manner. In addition, terrein at the same concentration also reduced tyrosinase activity. We then investigated whether terrein influences the extracellular signal-regulated protein kinase (ERK) pathway and the expression of microphthalmia-associated transcription factor (MITF), which is required for tyrosinase expression. Terrein was found to induce sustained ERK activation and MITF down-regulation, and luciferase assays showed that terrein inhibits MITF promoter activity in a dose-dependent manner. To elucidate the correlation between ERK pathway activation and a decreased MITF transcriptional level, PD98059, a specific inhibitor of the ERK pathway, was applied before terrain treatment and found to abrogate the terrein-induced MITF attenuation. Terrein also reduced the tyrosinase protein level for at least 72 h. These results suggest that terrain reduces melanin synthesis by reducing tyrosinase production via ERK activation, and that this is followed by MITF down-regulation.

Cenozoic Geological Structures and Tectonic Evolution of the Southern Ulleung Basin, East Sea(Sea of Japan) (동해 울릉분지 남부해역의 신생대 지질구조 및 지구조 진화)

  • The Cenozoic geological structures and the tectonic evolution of the southern Ulleung Basin were studied with seismic profiles and exploration well data. Basement structure of the Korea Strait is distinctly characterized by normal faults trending northeast to southwest. The normal faults of the basement are most likely related to the initial liking and extensional tectonics of Ulleung Basin. Tsushima fault along the west coast of Tsushima islands runs northeastward to the central Ulleung Basin. The Middle Miocene and older sequences in the Tsushima Strait show folds and faults mostly trending northeast to southwest. These folds and faults may be interpreted as a result of compressional tectonics. The Late Miocene to Qauternary sequences are not much deformed, but numerous faults mostly N-S trending are dominated in the Tsushima Strait. The Ulleung Basin was in intial rifting during Oligocene, and then active extension and subsidence from Early to early Middle Miocene. Therefore SW Japan separated from Korea Peninsula and drifted toward southeast, and Ulleung Basin was formed as a pull-apart basin under dextral transtensional tectonic regime. During rifting and extensional stage, Tsushima fault as a main tectonic line separating SW Japan block from the Korean Peninsula acted as a normal faulting with right-lateral strike-slip motion as SW Japan drifted southeastward. During middle Middle Miocene to early Late Miocene, the opening of Ulleung basin stopped and uplifted due to compressional tectonics. The southwest Japan block converging on the Korean Peninsula caused compressional stress to the southern margin of Ulleung Basin, resulting in strong deformation under sinistral transpressional tectonic regime. Tsushima fault acted as thrust fault with left-lateral strike-slip motion. From middle Late Miocene to Quaternary, the southern margin of Ulleung Basin has been controlled by compressional motion. Thus the Tsushima fault still appears to be an active thrust fault by compressional tectonic regime.

A Stochastic Study for the Emergency Treatment of Carbon Monoxide Poisoning in Korea (일산화탄소중독(一酸化炭素中毒)의 진료대책(診療對策) 수립(樹立)을 위한 추계학적(推計學的) 연구(硏究))

  • Emergency medical service is an important part of the health care delivery system, and the optimal allocation of resources and their efficient utilization are essentially demanded. Since these conditions are the prerequisite to prompt treatment which, in turn, will be crucial for life saving and in reducing the undesirable sequelae of the event. This study, taking the hyperbaric chamber for carbon monoxide poisoning as an example, is to develop a stochastic approach for solving the problems of optimal allocation of such emergency medical facility in Korea. The hyperbaric chamber, in Korea, is used almost exclusively for the treatment of acute carbon monoxide poisoning, most of which occur at home, since the coal briquette is used as domestic fuel by 69.6 per cent of the Korean population. The annual incidence rate of the comatous and fatal carbon monoxide poisoning is estimated at 45.5 per 10,000 of coal briquette-using population. It offers a serious public health problem and occupies a large portion of the emergency outpatients, especially in the winter season. The requirement of hyperbaric chambers can be calculated by setting the level of the annual queueing rate, which is here defined as the proportion of the annual number of the queued patients among the annual number of the total patients. The rate is determined by the size of the coal briquette-using population which generate a certain number of carbon monoxide poisoning patients in terms of the annual incidence rate, and the number of hyperbaric chambers per hospital to which the patients are sent, assuming that there is no referral of the patients among hospitals. The queueing occurs due to the conflicting events of the 'arrival' of the patients and the 'service' of the hyperbaric chambers. Here, we can assume that the length of the service time of hyperbaric chambers is fixed at sixty minutes, and the service discipline is based on 'first come, first served'. The arrival pattern of the carbon monoxide poisoning is relatively unique, because it usually occurs while the people are in bed. Diurnal variation of the carbon monoxide poisoning can hardly be formulated mathematically, so empirical cumulative distribution of the probability of the hourly arrival of the patients was used for Monte Carlo simulation to calculate the probability of queueing by the number of the patients per day, for the cases of one, two or three hyperbaric chambers assumed to be available per hospital. Incidence of the carbon monoxide poisoning also has strong seasonal variation, because of the four distinctive seasons in Korea. So the number of the patients per day could not be assumed to be distributed according to the Poisson distribution. Testing the fitness of various distributions of rare event, it turned out to be that the daily distribution of the carbon monoxide poisoning fits well to the Polya-Eggenberger distribution. With this model, we could forecast the number of the poisonings per day by the size of the coal-briquette using population. By combining the probability of queueing by the number of patients per day, and the probability of the number of patients per day in a year, we can estimate the number of the queued patients and the number of the patients in a year by the number of hyperbaric chamber per hospital and by the size of coal briquette-using population. Setting 5 per cent as the annual queueing rate, the required number of hyperbaric chambers was calculated for each province and for the whole country, in the cases of 25, 50, 75 and 100 per cent of the treatment rate which stand for the rate of the patients treated by hyperbaric chamber among the patients who are to be treated. Findings of the study were as follows. 1. Probability of the number of patients per day follows Polya-Eggenberger distribution. $$P(X=\gamma)=\frac{\Pi\limits_{k=1}^\gamma[m+(K-1)\times10.86]}{\gamma!}\times11.86^{-{(\frac{m}{10.86}+\gamma)}}$$ when$${\gamma}=1,2,...,n$$$$P(X=0)=11.86^{-(m/10.86)}$$ when $${\gamma}=0$$ Hourly arrival pattern of the patients turned out to be bimodal, the large peak was observed in $7 : 00{\sim}8 : 00$ a.m., and the small peak in $11 : 00{\sim}12 : 00$ p.m. 2. In the cases of only one or two hyperbaric chambers installed per hospital, the annual queueing rate will be at the level of more than 5 per cent. Only in case of three chambers, however, the rate will reach 5 per cent when the average number of the patients per day is 0.481. 3. According to the results above, a hospital equipped with three hyperbaric chambers will be able to serve 166,485, 83,242, 55,495 and 41,620 of population, when the treatmet rate are 25, 50, 75 and 100 per cent. 4. The required number of hyperbaric chambers are estimated at 483, 963, 1,441 and 1,923 when the treatment rate are taken as 25, 50, 75 and 100 per cent. Therefore, the shortage are respectively turned out to be 312, 791. 1,270 and 1,752. The author believes that the methodology developed in this study will also be applicable to the problems of resource allocation for the other kinds of the emergency medical facilities.

Characteristics of preschoolers' giftedness by parents' perception (부모의 지각에 의한 유아 영재의 발달 특성의 변화)

  • The purpose of this study was to investigate the characteristics of preschoolers' giftedness by their parents' perception. Total 3 groups of 148 subjects from age 30 months to 6 years 10 months old young gifted children's parents participated. The major findings were as follows : (1) There were critical characteristics of preschoolers' giftedness by parents' perception, which were 'good memory', 'high curiosity', 'read and understand of math', 'enjoy of learning and high motivation', 'high concentration', reading books', 'verbal ability', 'creativity', 'questions', and 'independency', (2) These characteristics of preschoolers' giftedness showed more strong and intense as they got older, and (3) Some characteristics revealed more, but the other characteristics revealed less as they got older. These findings suggested the consideration of child's age as the reliable identification process of young gifted children.

Studies on the Determinations of Dissolved Oxygen in Beverages (음료중 산소분석법에 관한 연구)

  • This study was performed to establish a simple and accurate method for the determination of oxygen that is a processing aid in various beverage. The quantitative determination of dissolved oxygen (DO) contents in 30 cases of samples were performed by traditional titration method and polarography. As a result of the study, the analysis of DO contents in fruit-extract beverages containing oxygen by titration method was time consuming and large sample volumes were needed. Besides, serious interferences with compounds such as hydroxylamine and nitric oxide were observed, leading to false response. Although the polarography is easily affected by $H_2S$, proteins, and various organic compounds, it is a simple and practical method that provides inexpensive and relatively rapid analysis. The polarography is best suited to the routine determination of DO in a large number of samples and it is expected that the polarography can directly be applied to the quality control of the beverages containing added oxygen. The analysis results of DO contents in various fruit-extract beverages with oxygen and without oxygen were as follows: 23.10 ppm to 32.60 ppm for various frutis extract beverages with oxygen, 0.70 pp to 2.54 ppm for mixed beverages without oxygen, 7.63 ppm to 8.28 ppm for drinking water.
