양배추 추출물의 간보호 및 간기능 개선 효과 (The Effects of Protecting Liver and Improving Liver Function on Cabbage Extract)

  • 본 연구는 인체에 유익한 효과가 있고 부작용 없이 장기간 안전하게 사용할 수 있으며 간보호 및 간기능 개선에 탁월한 효과가 있는지 양배추출물의 간보호 효과에 대하여 알아보았다. 약물(APAP)에 의해 유도된 간손상 동물모델에서 수행된 실험에서, 양배추 추출물은 간손상 마커인 AST 및 ALT를 효과적으로 감소시킴으로써 간 보호 및 간 기능 개선 효과를 나타냄을 알 수 있었다. 양배추 추출물은 간질환의 예방 또는 치료용 약학적 추출물로서 유효하다는 것이다. 특히, 양배추 추출물은 간손상에 수반되는 급성 염증 반응에 관여하는 염증 매개체 iNOS 및 COX-2 및 전염증성 사이토카인 $IL-1{\beta}$의 발현을 감소시킴으로써 간염 염증을 치료하는데 효과적이었다. 그 결과, $100{\sim}150^{\circ}C$에서 열처리 한 배추 추출물은 생배추 추출물에 비해 간기능 개선 효과 또는 항염증 효과가 우수 하였다.

  • 본 연구는 인체에 유익한 효능을 지니고 부작용없이 장기간 안전하게 사용할 수 있으며 간보호 및 간기능 개선에 탁월한 효능을 갖는 가열 브로콜리 추출물을 연구하기 위해 수행되었다. 열처리된 브로콜리 추출물은 세포 독성을 나타내지 않으므로 안전하게 사용할 수 있었다. 약제에 의한 간손상 동물모델 (APAP)에서 수행 한 실험에서, 열처리된 양배추 추출물은 간손상 마커인 AST 및 ALT를 효과적으로 감소시킴으로써 간보호 및 간기능 개선 효과를 나타냄을 알 수 있었고, 이는 열처리된 브로콜리 추출물이 간질환을 예방하거나 치료하기 위한 약학적 추출물로서 효과적이라는 것을 시사한다. 특히, 열처리된 브로콜리 추출물은 간손상을 동반한 급성염증 반응에 관여하는 염증 매개체 iNOS 및 COX-2 및 염증 유발성 사이토카인 $IL-1{\beta}$의 발현을 감소시킴으로써 간염 염증을 치료하는데 효과적이라고 할 수 있다.

DNCB로 유도한 아토피 유사 피부염에 지모 추출물이 미치는 영향 (Effects of Anemarrhena asphodeloides Extract on Atopic-Dermatitis like Skin Lesions in DNCB-induced Balb/c Mice)

  • Under constant environmental pollution, the incidence of Atopic Dermatitis (AD) caused by air pollutants and allergens has increased. AD is an allergy inflammatory skin disease characterized by pruritus, eczema, and skin dryness. In herbal medicine, Anemarrhena asphodeloides (Anemarrhenae Rhizoma; AR) has been utilized to treat Alzheimer's disease, osteoporosis, hypertension, and inflammation. The purpose of study evaluated the effect of AR in a mouse model of 2,4-dinitrochlorobenzene (DNCB)-induced AD-like skin lesions. After acclimatization for 5 days, the mice (6-week-old, male Balb/c) were divided into five groups (n=6/group): NC (normal control), DNCB (control), Dex (5 mg·kg-1, p.o.), AR100 (100 mg·kg-1, p.o.), and AR300 (300 mg·kg-1, p.o.). On days 1 and 3, 1% DNCB was applied to the skin and ears. After 4 days, 0.5% DNCB was applied once every 2 days for 2 weeks. Then, skin and ears eczema area and severity index (EASI); skin nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and prostaglandin E2 (PGE2) levels; and plasma immunoglobulin E (IgE) levels were examined. The AR groups showed lower EASI, skin and ear thickness, mast cell count, and IgE levels than the control groups. Moreover, AR reduced iNOS, COX-2, and PGE2 levels. Therefore, AR possesses anti-inflammatory properties and can improve skin damage, indicating its therapeutic potential against AD.

약용약침(藥茸藥鍼)이 LPS 유발(誘發) 관절염(關節炎)의 윤활관절막내(潤滑關節膜內) 과다염증반응(過多炎症反應) 조절(調節)에 미치는 영향(影響) (The Effect of Deer Antler Herbal Acupuncture Control to Hyper-inflammatory Responses on Synovial Membrane by LPS-induced Arthritis)

  • Objectives: To evaluate expression of MIF mRNA, MIF, $TNF-{\alpha}$, $IL-6R-{\alpha}$, STAT-3, $NF-{\kappa}B$ p65, COX-2 and iNOS, MMP-9mRNA after injecting deer antler herbal acupuncture solution in a LPS rat model. Methods: The experiment was divided in category of the control group, RA group, and NA group. RA was induced in the mice via injecting 300ug/kg LPS. The deer antler herbal acupuncture solution 50ug/kg was applied on $ST_{35}$(犢鼻) and EX-LE201(內膝眼) for 19days from $3^{rd}$ day of RA inducement. Results: 1. In the deer antler herbal acupuncture solution treated RAW 264.7cell, the mRNA expression of cytokines, RA related inflammation factors, such as the MIF, COX- 2, iNOS, and MMP-g reduced concentration dependently. 2. In the deer antler herbal acupuncture treated mice's synovial membrane, decrease in the cell replication of synovial joint cells, regeneration of blood vessel, fibrosis and fibroblastic cells expansion were observed. 3. Positive reaction of RA-related cytokines MIF, $TNF-{\alpha}$, $IL-6R-{\alpha}$, STAT3, COX-2, iNOS, $NF-\;{\kappa}B$ p65, MMP-9 was reduced. Conclusion : On the basis of the results, it was concluded that deer antler herbal acupuncture extract has significant protecting ability against acute progressive RA by inhibiting the production of MIF, as a top in cytokines related to inflammation.

NF-${\kappa}B$ 조절(調節)을 통한 충울자(茺蔚子)의 염증억제효과 (Leonuri Fructus Ameliorates Acute Inflammation via the Inhibition of NF-${\kappa}B$-mediated Nitric Oxide and Pro-inflammatory Cytokine Production)

  • Leonuri Fructus, a semen of Leonuri Herba, has been used for the treatment of menstrual disorders such as amenorrhea, dysmenorrhea and leukorrhea and for the remedy of hyperemia. The present study was conducted to evaluate the anti-inflammatory effects of the Leonuri Fructus extract (Leonurus japonicus Houtt. EtOH extract; LJE) in vivo and in vitro. In vitro study, the MTT assay for cell viability was conducted to determine the non-cytotoxic concentration of LJE treatment in media. The levels of NO were measured with Griess reagent. Pro-inflammatory cytokines were detected by ELISA method. The inflammation-related proteins of this study were detected by immunoblot anlaysis. The increases of NO production and iNOS expression were detected in LPS-treated cells compared with control, but LJE attenuated the increases of NO and iNOS by LPS. LJE reduced the production of TNF-${\alpha}$ and IL-$1{\beta}$ induced by LPS stimulation. LJE suppresses the signaling pathways of NF-${\kappa}B$ and MAPKs in LPS-induced macrophage cells. In vivo study, carrageenan-induced hind paw acute edematous inflammation rat model was used for evaluation of anti-inflammatory activity of LJE. LJE significantly inhibited the increases of hind paw swelling, skin thicknesses and inflammatory cell infiltrations, and decreased the numbers of mast cell induced by carrageenan injection. These results suggest that LJE has an anti-inflammatory therapeutic potential, which is mediated through modulating NF-${\kappa}B$ activation and MAPK phosphorylation. Inhibition of the rat paw edema induced by carrageenan is considered as direct evidence that LJE may be a useful source to treat inflammation.

원발성 월경통에 대한 계지복령환(桂枝茯苓丸) 열수 추출물의 진통 효과 (Analgesic Effects of Gyejibokryeong-hwan Aqueous Extracts on the Rat Model of Primary Dysmenorrhea)

  • 목 적: 이 생체 내 연구의 목적은 인도메타신과 비교했을 때 쥐의 원발성 월경통에 대한 계지 복령환 열수 추출물의 진통 및 개선 효과를 관찰하는 것이다. 방 법: 랫트 PD 모델을 만들기 위해, Estradiol benzoate를 10일간 투여한 다음 마지막 10회 estradiol benzoate 투여 1시간 후 1 U/kg의 oxytocin을 투여하였다. 계지복령환 열수 추출물을 500, 250 및 125 mg/kg 용량으로 매일 1회씩 10일간 투여하였다. 이후 체중 및 실험 기간 동안의 체중 증가량, 자궁 중량 및 육안부검 소견, 진통 활성을 나타내는 abdominal writhing test, 자궁 조직 내 지질 과산화(Malondialdehyde, MDA 함량) 및 항산화 방어 시스템 - glutathione (GSH) 함량, superoxide dismutase (SOD) 및 catalase (CAT) 활성, Nuclear factor-κB (NF-κB) 및 cyclooxygenase (COX)-2 mRNA의 발현, 자궁의 조직병리학적 변화, tumor necrosis factor (TNF)-α 및 inducible nitric oxide synthase (iNOS)로 나타나는 면역조직 화학적 변화를 관찰하였다. 계지복령환 열수 추출물의 결과는 인도메타신 투여 쥐의 결과와 비교하였다. 결 과: Estradiol benzoate 및 oxytocin 투여 결과, 현저한 체중 및 증체량, 자궁 GSH 함량, SOD와 CAT 활성의 감소와 abdominal writhing 반응, 자궁 지질 과산화(MDA 함량), 자궁 중량, NF-κB 및 COX-2 mRNA 발현의 증가가 TNF-α 및 iNOS 면역반응세포와 염증세포 침윤 증가, 자궁의 충혈 및 확대와 함께 관찰되었다. 이는 전형적인 염증 및 산화 스트레스성 원발성 월경통이 잘 유도되었음을 의미한다. 한편 이러한 소견은 계지복령환 열수 추출물의 투여에 의해 용량의존적으로 현저히 억제되었으며, 인도메타신의 억제 효과보다는 낮았다. 결 론: 이 연구에서 얻은 결과는 계지복령환 열수 추출물이 estradiol benzoate 및 oxytocin으로 유도된 원발성 월경통에 용량-의존적으로 유리한 진통 및 개선 활성을 가짐을 시사한다.

진교${\cdot}$위령선${\cdot}$하고초 복합방이 MIA 유발 골관절염 모델에서 중추신경내 통증관련물질에 미치는 영향 (GCP Treatment on the Expression of NOS, C-fos, Serotonin and Substance-P in Central Nerve System of Monosodium Iodoacetate-Induced Osteoarthritic Pain Model)

  • This study was carried out to investigate the effects of GCP treatment on the expression of NOS, c-fos, serotonin and substance P in central nerve system of monosodium iodoacetate(MIA)-induced osteoarthritic pain model. Arthritis was induced by injection of MIA(0.5 mg) into knee joint cavities of rats. Arthritic rats were divided into control(n=8) and treated(n=8) group. Control group was taken distilled water for 20 days. Treated group was taken extracts of GCP by oraly for same duration. Normal group(n=8) was infected with normal saline and was taken distilled water for 20 days. The numbers of NADPH-d positive cells in superficial dorsal horn of spinal cord of treated group($21{\pm}5$) was significantly (p<0.01) decreased compared with control($33{\pm}5$). The numbers of NADPH-d positive cell in dorsolateral periaqueductal gray matter of treated group($111{\pm}16$) was significantly(p<0.01) decreased compared with control($143{\pm}14$). The numbers of c-fos positive cells in dorsal periaqueductal gray matter of treated group($57{\pm}16$) was significantly(p<0.01) decreased compared with control($78{\pm}13$). The numbers of c-fos positive cells in paraventricular thalamic nucleus of treated group($60{\pm}15$) was significantly decreased compared with control($88{\pm}27$). The numbers of serotonin positive cells in median raphe nucleus of treated group($171{\pm}31$) was significantly(p<0.05) decreased compared with control($217{\pm}48$). On the basis of these results, we concluded that GCP treatment has inhibiting effects on the pain transmission in monosodium iodoacetate-induced osteoarthritic pain model in rat.

상지추출물이 Dextran Sulfate Sodium으로 유도된 대장염 마우스 모델에 미치는 항염증 효능 (Anti-inflammatory effects of mulberry twig extracts on dextran sulfate sodium-induced colitis mouse model)

  • 본 연구는 DSS 유도 대장염 마우스 모델에서 MT열수 추출물의 항대장염 효능을 확인하고자 ICR 마우스에 5일 동안 3% DSS와 함께 MT추출물을 경구투여한 후 대장염의 임상적 증상 및 염증 지표들을 분석하였다. 그 결과 MT추출물의 투여는 DSS에 의해 유도된 마우스 대장염 증상의 지표들, 즉 체중 감소와 혈성 설사를 포함한 질병 활성도, 대장점막증식, 그리고 $TNF-{\alpha}$와 IL-6를 포함한 염증성 사이토카인량 증가를 개선시키는 것으로 나타났다. 또한 DSS에 의해 증가된 Cox-2, iNOS, p-Erk 등 염증 지표 단백질의 발현도 MT추출물 투여군에서 감소되었다. 이러한 결과들은 MT추출물이 항대장염 효능이 있음을 제시하며 향후 MT추출물에 포함되어 있는 생리활성물질의 효능과 관련기작이 규명된다면 상지를 물로 우려내어 섭취하는 상지차 형태의 대장염 예방 혹은 개선용 건강식품으로의 개발을 기대해 볼 수 있을 것으로 사료된다.

The Effects of Anti-Inflammatory and Liver Function using Heat-Treated Cabbage

  • The cabbage extract of the research does not show cytotoxicity, and thus can be used safely. In an experiment performed on an animal model with liver injury induced by a drug (APAP), it could be seen that the cabbage extract exhibited the effects of protecting liver and improving liver function by effectively reducing AST and ALT which are liver injury markers, indicating that the cabbage extract is effective as a pharmaceutical composition for preventing or treating liver disease. In particular, the cabbage extract was effective in treating inflammation of the liver by reducing the expression of the inflammatory mediators iNOS and COX-2 and the proinflammatory cytokine IL-1β, which are involved in acute inflammatory reactions accompanying liver injury. In the research, an extract of cabbage heat-treated at a temperature of 100 to 150℃ had a better liver function-improving effect or anti-inflammatory effect than an extract of raw cabbage.

히어리의 항염증 효능 (Anti-inflammatory Effects of MeOH Extract of Corylopsis gotoana Uyeki)

  • Corylopsis gotoana (Hamamelidaceae) has been used as a traditional medicine for the treatment various diseases including cold, edema and vomiting. However, previous studies regarding component analysis and pharmacological actions of C. gotoana are extremely limited until now. In this study, we investigated anti-inflammatory activities of the methanolic extract of the twigs of Corylopsis gotoana (MCG) both in vitro and in vivo. MCG effectively inhibited excessive NO production in IFN-${\gamma}$ and LPS-stimulated mouse peritoneal macrophages without notable cytotoxicity. In addition, we also found that MCG could attenuate the expression of inflammatory mediators such as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). We further tested in vivo anti-inflammatory activity of MCG using paw edema mouse model. Herein, MCG demonstrated significant suppression on the paw edema induced by both of trypsin and carrageenan. These results indicated that MCG has potent anti-inflammatory potential and may be useful for prevention and treatments of inflammatory diseases.
