MicroRNAs (miRNAs) are abundant in bovine milk and milk derived from other livestock, and they have functional roles in infants and in the secretion process of mammary glands. However, few studies have evaluated miRNAs in dairy processes, such as during cheese making and ripening. Thus, we investigated the characteristics of milk-derived miRNAs during the manufacturing and ripening of Camembert cheese as well as the microbiota present using the quantitative reverse transcription polymer chain reaction (RT-qPCR) and 16S rRNA pyrosequencing, respectively. Pyrosequencing showed that the cheese microbiota changed dramatically during cheese processing, including during the pasteurization, starter culture, and ripening stages. Our results indicated that the RNA contents per $200mg/200{\mu}l$ of the sample increased significantly during cheese-making and ripening. The inner cheese fractions had higher RNA contents than the surfaces after 12 and 22 days of ripening in a time-dependent manner (21.9 and 13.2 times higher in the inner and surface fractions than raw milk, respectively). We performed a comparative analysis of the miRNAs in each fraction by RT-qPCR. Large amounts of miRNAs (miR-93, miR-106a, miR-130, miR-155, miR-181a, and miR-223) correlated with immune responses and mammary glands were present in aged cheese, with the exception of miR-223, which was not present on the surface. Considerable amounts of miRNAs were also detected in whey, which is usually disposed of during the cheese-making process. Unexpectedly, there were no significant correlations between immune-related miRNAs and the microbial populations during cheese processing. Taken together, these results show that various functional miRNAs are present in cheese during its manufacture and that they are dramatically increased in amount in ripened Camembert cheese, with differences according to depth.
The purpose of this study was to investigate the feasibility of As(V) reduction by aqueous Fe(II), and subsequent As(III) immobilization by the precipitation of As(III) incorporated magnetite-like material [i.e., co-precipitation of As(III) with Fe(II) and Fe(III)]. Experimental results showed that homogeneous As(V) reduction did not occur by dissolved Fe(II) at various pH values although the thermodynamic calculation was in favor of the redox reaction between As(V) and Fe(II) under the given chemical conditions. Similarly, no heterogeneous reduction of sorbed As(V) by sorbed Fe(II) was observed using synthetic iron (oxy)hydroxide (Goethite, ${\alpha}$-FeOOH) at pH 7. Experimental results for the effect of As(V) on the oxidation of Fe(II) by dissolved oxygen showed that As(V) inhibited the oxidation of Fe(II). These results indicate that As(V) could be stable in the presence of Fe(II) under the anoxic or subsurface environments.

This study was performed to investigate the reaction between alkylketene dimer(AKD) and cellulose molecules in AKD-sized paper sheet. AKD was added to highly beaten($80{\pm}3^{\circ}SR$) SwBKP(ca. 0.8% on pulp) in order to have much AKD retention in the paper sheet. This AKD-sized paper sheet was aged at different temperatures, $60^{\circ}C,\;80^{\circ}C,\;105^{\circ}C\;and\;125^{\circ}C$. Changes in FT-IR spectra of AKD in paper sheet during the ageing were measured. In addition, sizing degrees of the AKD-sized paper sheet pretreated for 30 sec. at $105^{\circ}C$ were measured by HST size tester during the storage at different temperature. IR spectra of AKD-sized paper sheet preheated at $105^{\circ}C$ for 30 sec. showed unchanged spectra two absorption bands at $1849cm^{-1}\;and\;1722cm^{-1}$ which refer to the typical AKD IR bands. However, these typical AKD bands were gradually reduced with increasing ageing, completely disappeared after 6 hrs. and formed new absorption band at $1706cm^{-1}$, which refers to carbonyl stretching vibration of dialkylktone. Eventually the AKD molecule was hydrolyzed to diakylketone without formation of ${\beta}$-ketoester with cellulose in paper sheet. After 6 days ageing, a little amount of ${\beta}$-ketoester bands was identified in 6 or 7 days ageing, because of the absence of water due to long-term heating. The same tendency was observed at different ageing conditions. At the practical papermaking process, AKD reacts prevailing with water, and mostly seems to be hydrolyzed to dialkylketene. Concerned to the sizing development, AKD-sized paper sheet was shown no sizing development at the initial stage of ageing at $60^{\circ}C$ after heating treatment at $105^{\circ}C$ for 30 sec., and gradually increased the sizing degree with increasing ageing, such as Hercules Sizing Tester (HST) 130 see for 12 hr, HST 300 sec. for 3 days and HST 400 sec. for 5 days. It was concluded that hydrolyzed AKD could contributed to the sizing development of the paper sheet.
As port development in soft ground is actively promoted for international logistics and transportation, the Deep Mixing Method (DMM) is continuously applied to form an improved column body directly in the ground by mixing cement with soil to secure the stability of the structure. However, in the case of cement, there is a problem of emitting a lot of greenhouse gases during the production process, so the development and use of new alternative materials are socially required to achieve the national goal of carbon neutrality. Accordingly, in this study, CMD-SOIL, developed to induce a hardening reaction similar to cement by recycling recycled resources, was used as a ground improvement material for the DMM. In addition, it was attempted to determine the possibility of replacing cement by conducting on-site test construction and evaluating applicability. As a result of the study, the compressive strength of CMD-SOIL compared to the design reference strength was 1.46 to 2.64 times higher in the field mixing test and 1.2 to 5.03 times higher than in the confirmed boring. In addition, the ratio (λ) of the compressive strength in the field to the design reference strength was 0.63 to 1.14, which was similar to the previous research results. Therefore, in the case of CMD-SOIL, it is possible to express the compressive strength necessary to secure stability, and there is no difference in applicability compared to existing materials such as ordinary portland cement and blast furnace slag cement, so it was analyzed that it could be used as a ground improvement material for the DMM.
This study aimed to investigate the effect of cognitive-motor interaction-based instrument playing on the early social skills of children with autism spectrum disorder (ASD). Nine children with ASD, averaging 5.6 years of age, participated in twelve individual sessions lasting 30 minutes each. The intervention comprised five stages: self-regulation, motivation to engage in social interaction, acceptance of a partner in co-playing activities, interpersonal coordination with a partner, and engagement in joint music playing. To evaluate changes in early social skills, joint attention and social interaction behaviors were observed, and the Social Responsiveness Scale (SRS-2) was administered pre- and post-intervention. Social synchronization was measured through a dyadic drum task, measuring synchronization accuracy, duration, and timing. Following the intervention, all nine children demonstrated increased early social behaviors, although there were no significant differences in SRS scores. Moreover, synchronized movement improved significantly in accuracy and maintained duration but not in reaction time. This study highlights the significance of recognizing the cognitive-motor interplay as crucial element in facilitating early social skills development in children with ASD.

The molecular understanding of resistance and susceptibility of host plants to scab, a most threatful disease to pome fruit production worldwide, is very limited. Comparing resistant line '93-3-98' to susceptible one 'Sweet Skin' at seven time points of 0, 0.5, 1, 2, 3, 4, 8 days post inoculation, RNA-sequencing data derived from infected and mock-inoculated young leaves were analyzed to evaluate the tolerant response and to mine candidate genes of pear to the scab pathogen Venturia nashicola. Analysis of the mapped reads showed that the infection of V. nashicola led to significant differential expression of 17,827 transcripts with more than 3-fold change in the seven pairs of libraries, of which 9,672 (54%) are up- and 8,155(46%) are down-regulated. These included mainly receptor (NB-ARC domains-containing, CC-NBS-LRR, TIR-NBS-LRR, seven transmembrane MLO family protein) and transcription factor (ethylene responsive element binding, WRKY DNA-binding protein) related gene. An arsenal of defense response of highly resistant pear accessions derived from European pear was probably supposed no sooner had V. nashicola infected its host than host genes related to disease suppression like Polyketide cyclase/dehydrase and lipid transport protein, WRKY family transcription factor, lectin protein kinase, cystein-rich RLK, calcium-dependent phospholipid-binding copine protein were greatly boosted and eradicated cascade reaction induced by pathogen within 24 hours. To identify transcripts specifically expressed in response to V. nashicola, RT-PCRs were conducted and compare to the expression patterns of seven cultivars with a range of highly resistant to highly susceptible symptom. A DEG belonging to the PR protein family genes that were higher expressed in response to V. nashicola suggesting extraordinary role in the resistance response were led to the identification. This study provides the first transcriptional profile by RNA-seq of the host plant during scab disease and insights into the response of tolerant pear plants to V. nashicola.
[18F]Fluorocholine is a radiopharmaceutical used non-invasively in positron emission tomography to diagnose parathyroid adenoma, prostate cancer, and hepatocellular carcinoma by evaluating the choline metabolism. In this study, a radiolabeling method for [18F]fluorocholine was optimized using a solid phase extraction (SPE) cartridge. [18F]Fluorocholine was labeled in two steps using an automated synthesizer. In the first step, dibromomethane was reacted with [18F]KF/K2.2.2/K2CO3 to obtain the intermediate [18F]fluorobromomethane. In the second step, [18F]fluorobromomethane was passed through a Sep-PakⓇ Silica SPE cartridge to remove the impurities and then reacted with N,N-dimethylaminoethanol (DMAE) in a Sep-PakⓇ C18 SPE cartridge to label [18F]fluorocholine. The reaction conditions of [18F]fluorocholine were optimized. The synthesis yield was confirmed according to the number of silica cartridges and DMAE concentration. No statistically significant difference in the synthesis yield of [18F]fluorocholine was observed when using four or three silica cartridges (P>0.05). The labeling yield was 11.5±0.5% (N=4) when DMAE was used as its original solution. On the other hand, when diluted to 10% with dimethyl sulfoxide, the radiochemical yield increased significantly to 30.1±5.2% (N=20). In conclusion, [18F]Fluorocholine for clinical use can be synthesized stably in high yield by applying an optimized synthesis method.

Nanoscale zero-valent iron (NZVI) particles were tested as remediation media for groundwater contaminated by organic pollutants (e.g., TCE, trichloroethylene). The contaminated groundwater contained anions ($NO_3^-$, $Cl^-$, $SO_4^{2-}$, and $HCO_3^-$) and natural organic matter (NOM). Treatability of commercial NZVI particles (NANOFER 25, Nanoiron, Czech) was tested by using a synthetic groundwater and the field groundwater samples. More than 95% of 1.8 mM TCE was removed within 20 hours with a NZVI dosage of 25 g/L ($k=0.15hr^{-1}$). Repetitive degradation experiments revealed that the removal capacity of NANOFER 25 was 0.19 mmole TCE/g NZVI. TCE degradation reactions were not substantially affected by the presence of each anion with concentrations as high as 100 times the average field concentrations. However, when the four anions ($NO_3^-$, $Cl^-$, $SO_4^{2-}$, $HCO_3^-$) were present simultaneously. the degradation reactivity and removal capacity were decreased by 60% ($k=0.069hr^{-1}$) and 10%, respectively. The k value of TCE degradation in the presence of NZVI (25 g/L) with dissovled organic carbon of 2.5 mg/L was also decreased by 84% ($k=0.025hr^{-1}$). In the experiments with the field groundwater, more than 90% of $1.8{\mu}M$ TCE, which is the concentration of TCE at the source zone, was removed within 10 hours with a NANOFER 25 dosage of 25 g/L. The results imply that the contaminated groundwater can effectively be treated by NANOFER 25 with more information on the hydrogeology of the site.
Persistent organic pollutants (POPs) can affect epigenetic mechanisms and obesity development. Polybrominated diphenyl ethers (PBDEs)-widely used to make flames-are one of the important POPs. Prenatal exposure to endocrine disrupting chemicals (EDCs), such as POPs, may affect global DNA methylation in long interspersed nuclear elements (LINE-1), increasing the risk of obesity later in life. Therefore, pregnant Sprague-Dawley (SD) rats were used to elucidate whether BDE-47 and BDE-209 transferred through placenta and breast milk cause epigenetic changes in LINE-1 and increase genetic susceptibility to obesity as obesogen during the developmental periods. Global DNA methylation in LINE-1 and gene expression related to obesity were measured in dams and offspring, using a methylation-sensitive high resolution melting analysis (MS-HRM) and direct bisulfite sequencing and quantitative real time polymerase chain reaction (qPCR), respectively. The results of MS-HRM showed global DNA hypomethylation patterns in LINE-1 of exposed offspring (2 of total 4) at PND 4, but bisulfite sequencing showed no difference in both the exposed and non-exposed groups. Gene expression in dams related to ${\beta}$-oxidation pathway and those related to adipokines showed different patterns between the two groups. On the contrary, gene expressions of offspring showed a similar pattern. Gene expressions related to ${\beta}$-oxidation pathway and obesity were significantly increased when compared with 'at birth', but not $PPAR-{\alpha}$. In conclusion, this study demonstrated the possibility that co-exposure to BDE-47 and BDE-209-via the placenta and breast milk-may affect epigenetic changes and modulate gene expression levels related to obesity.
Purpose : The aim of this study was to identify mutations associated with macrolide resistance in Mycoplasma pneumoniae (MP) and to establish a cultural method to determine antimicrobial susceptibility. Methods : Nasopharyngeal aspirates (NPAs) were collected from 62 children diagnosed with MP pneumonia by a serologic method or polymerase chain reaction. The 23S rRNA and L4 ribosomal protein genes of MP were amplified and sequenced. To identify mutations in these 2 genes, their nucleotide sequences were compared to those of the reference strain M129. MP cultivation was carried out for 32 (28 frozen and 5 refrigerated) NPAs and M129 strain using Chanock's glucose broth and agar plate in a 5% $CO_2$ incubator at $37^{\circ}C$ and examined at 2-3 day intervals for 6 weeks. Results : Among the 62 specimens, 17 had M144V mutations in ribosomal protein L4. The A2064G mutation was observed in 1 specimen; its 23S rRNA gene was successfully sequenced. Culture for MP was successful from the M129 strain and 2 of the 5 NPAs that were refrigerated for no longer than 3 days. However, MP did not grow from the 28 NPAs that were kept frozen at $-80^{\circ}C$ since 2003. Conclusion : We found the M144V mutation of L4 protein to be common and that of domain V of 23S rRNA gene was relatively rare among MP. Studies on the prevalence of macrolide-resistant MP and the relationship between the mutations of 23S rRNA gene and ribosomal protein L4 will aid in understanding the mechanism of macrolide resistance in MP.
