The Theoretical Investigation of Phased Array Guided Waves (위상배열 유도초음파 검사의 이론적 고찰)

  • Guided waves inspection techniques that are different with inspection technique by bulk waves are widely used in pipe line evaluation due to advantages of long distance inspection. However, most of pipe lines at industrial fields are buried and/or coated. In this case, due to the attenuation effect from soil and/or coating material, there are a lot of difficulty on inspection by conventional ultrasonic technique. In this paper, guided waves propagating patterns are calculated with respect to excitation mode by Normal Mode Expansion(NME). Guided waves patterns based on excited by single transducer and guided wave focusing technique have employed to analyze focusing pattern on a pipe. A longitudinal mode and high order flexural modes are used with various number of transducers to determine sensitivity. Guided waves energy excited by multi transducer with focusing algorithm was successfully focused at a desired point.

The effect of μ-opioid receptor activation on GABAergic neurons in the spinal dorsal horn

  • The superficial dorsal horn of the spinal cord plays an important role in pain transmission and opioid activity. Several studies have demonstrated that opioids modulate pain transmission, and the activation of ${\mu}$-opioid receptors (MORs) by opioids contributes to analgesic effects in the spinal cord. However, the effect of the activation of MORs on GABAergic interneurons and the contribution to the analgesic effect are much less clear. In this study, using transgenic mice, which allow the identification of GABAergic interneurons, we investigated how the activation of MORs affects the excitability of GABAergic interneurons and synaptic transmission between primary nociceptive afferent and GABAergic interneurons. We found that a selective ${\mu}$-opioid agonist, [$D-Ala^2$, $NMe-Phe^4$, Gly-ol]-enkephanlin (DAMGO), induced an outward current mediated by $K^+$ channels in GABAergic interneurons. In addition, DAMGO reduced the amplitude of evoked excitatory postsynaptic currents (EPSCs) of GABAergic interneurons which receive monosynaptic inputs from primary nociceptive C fibers. Taken together, we found that DAMGO reduced the excitability of GABAergic interneurons and synaptic transmission between primary nociceptive C fibers and GABAergic interneurons. These results suggest one possibility that suppression of GABAergic interneurons by DMAGO may reduce the inhibition on secondary GABAergic interneurons, which increase the inhibition of the secondary GABAergic interneurons to excitatory neurons in the spinal dorsal horn. In this circumstance, the sum of excitation of the entire spinal network will control the pain transmission.

Ab Initio Conformational Study on Ac-Pro-$NMe_2$: a Model of Polyproline

  • We report here the results on N-acetyl-N'-dimethylamide of proline (Ac-Pro-NM $e_2$) calculated using the ab initio molecular orbital method with the self-consistent reaction field (SCRF) theory at the HF level with the 6-31+G(d) basis set to investigate the conformational preference of polyproline depending on the cis/trans peptide bonds and down/up puckerings along the backbone torsion angle $\square$ in the gas phase, chloroform, and water. In the gas phase, Ac-Pro-NM $e_2$ has seven local minima of tFd, tFu, cFd, cFu, cAu, tAu, and cAd conformations. In particular, polyproline conformations tFd, tFu, cFd, and cFu are found to be more stable than $\square$-helical conformations cAu, tAu, and cAd. In contrast, Ac-Pro-NHMe has seven local minima of tCd, tCu, cBd, cAu, tAu, cFd, and cFu conformations. Conformations tCd and tCu are found to be most stable, which is ascribed to the intramolecular hydrogen bond between C=O of acetyl group and $N^{~}$ H of N'-methyl amide group. The stability of the cFd conformation (i.e., the polyproline I structure) in chloroform is somewhat increased, relative to that in water, although tFd and tFu conformations (i.e., the polyproline II structure) are dominate both in chloroform and water. The population of backbone conformations feasible in chloroform and water is consistent with the experiments. This work is supported by a Korea Research Foundation Grant (KRF-2002-041-C00129).

Prediction of Arsenic Uptake by Rice in the Paddy Fields Vulnerable to Arsenic Contamination

  • There is an increasing concern over arsenic (As) contamination in rice. This study was conducted to develope a prediction model for As uptake by rice based on the physico-chemical properties of soil. Soil and brown rice samples were collected from 46 sites in paddy fields near three different areas of closed mines and industrial complexes. Total As concentration, soil pH, Al oxide, available phosphorus (avail-P), organic matter (OM) content, and clay content in the soil samples were determined. Also, 1.0 N HCl, 1.0 M $NH_4NO_3$, 0.01 M $Ca(NO_3)_2$, and Mehlich 3 extractable-As in the soils were measured as phytoavailable As concentration in soil. Total As concentration in brown rice samples was also determined. Relationships among As concentrations in brown rice, total As concentrations in soils, and selected soil properties were as follows: As concentration in brown rice was negatively correlated with soil pH value, where as it was positively correlated with Al oxide concentration, avail-P concentration, and OM content in soil. In addition, the concentration of As in brown rice was statistically correlated only with 1.0 N HCl-extractable As in soil. Also, using multiple stepwise regression analysis, a modelling equation was created to predict As concentration in brown rice as affected by selected soil properties including soil As concentration. Prediction of As uptake by rice was delineated by the model [As in brown rice = 0.352 + $0.00109^*$ HCl extractable As in soil + $0.00002^*$ Al oxide + $0.0097^*$ OM + $0.00061^*$ avail-P - $0.0332^*$ soil pH] ($R=0.714^{***}$). The concentrations of As in brown rice estimated by the modelling equation were statistically acceptable because normalized mean error (NME) and normalized root mean square error (NRMSE) values were -0.055 and 0.2229, respectively, when compared with measured As concentration in the plant.

Identifying Genes Related with Radiation Resistance in Oral Squamous Cell Carcinoma Cell Lines (두경부 편평상피세포암주에서 방사선 내성 관련 유전인자의 확인)

  • Background and Objective : Radiation resistance(RR) is one of main determinants of treatment outcome in oral squamous cell carcinoma(OSCC), but accurate prediction of RR is difficult. We aim to establish RR OSCC cell lines and identify genes related with RR by a measurement of altered gene expression after inducing RR. Material and Methods : OSCC cell lines, SCC15, SCC25 and QLL1, were treated with 2Gy radiation per session, and parts of them were alive in finally accumulated dosage of 60Gy through 30 times repetition of radiotherapy for inducing RR cell lines. We compared results of cDNA array and proteomics in non-radiated cell lines and RR cell lines to detect changes of gene expression. Western blot was used for the validation of results. Results : cDNA array revealed 265 commonly up-regulated genes and 268 commonly down-regulated genes in 3 RR cell lines comparing their non-radiated counterpart. Among them, 30 cancer related genes were obtained. Proteomics showed 51 commonly altered protein expressions in 3 RR cell lines and 18 cancer related proteins were obtained. Among the detected genes, we found NM23-H1 and PA2G4 were over-expressed in both cDNA array and proteomics. Western blot showed increased expression of NME1 in RR cell lines but not in PA2G4. Conclusion: We concluded that NM23-H1 may be a candidate of RR related gene and over-expression of NM23-H1 could be a biomarker to predict RR in OSCC.
