The improved Goldschmidt floating point reciprocal algorithm (개선한 Goldschmidt 부동소수점 역수 알고리즘)

  • Goldschmidt 알고리즘에 의한 부동소수점 1.f2의 역수는 q=NK1K2....Kn (Ki=1+Aj, j=2i)이다. 본 논문에서는 N과 A 값을 1.f2의 값에 따라서 선정하고 Aj의 값이 유효자리수의 반이하 값을 가지면 연산을 종료하는 개선된 Goldschmidt 부동소수점 역수 알고리즘을 제안한다. 1.f2가 1.01012보다 작으면 N=2-1.f2, A=1.f2-1로 하며, 1.01012보다 크거나 같으면 N=2-0.lf2, A=1-0.lf2로 한다. 한편 Goldschmidt 알고리즘은 곱셈을 반복해서 수행하므로 계산 오류가 누적이 된다. 이러한 누적 오류를 감안하면 배정도실수 역수에서는 2-57, 단정도실수 역수에서는 2-28의 유효자리수까지 연산해야 한다. 따라서 Aj가 배정도실수 역수에서는 2-29, 단정도실수 역수에서는 2-14 보다 작아지면 연산을 종료한다. 본 논문에서 제안한 개선한 Goldschmidt 역수 알고리즘은 N=2-0.1f2, A=1-0.lf2로 계산하는 종래 알고리즘과 비교하여 곱셈 연산 회수가 배정도실수 역수는 22%, 단정도실수 역수는 29% 감소하였다. 본 논문의 연구 결과는 테이블을 사용하는 Goldschmidt 역수 알고리즘에 적용해서 연산 시간을 줄일 수 있다.

Immuno-regulatory Activities of Various Fractions from Ehpedrae Sinica STAPF, Rubus Coreanus Miq. and Angelica gigas Nakai Extracts with Ultrasonification (초음파 병행 추출을 이용한 마황과 복분자, 당귀 분획물의 면역활성 조절 효과)

  • This study was performed to examine immuno-regulatory activities of Ehpedrae Sinica STAPF, Rubus Coreanus Miq. and Angelica gigas Nakai extracts in conbination with ultrasonification. The extract yields of plants were the highest in the extraction system of $60^{\circ}C$ and 40 kHz of ultrasonification. The immune cell growth ratio of human immune B and T cells was increased compared to other fractions by the water fraction of the plants at $60^{\circ}C$ and 40 kHz. The water fractions of the plants at $60^{\circ}C$ and 40 kHz increased the specific secretion of IL-6 and $TNF-{\alpha}$ of human immune B and T cells compared to other fractions of the plants. The water fraction of Ehpedrae Sinica STAPF among the plants was observed to show the highest specific secretion of IL-6 and $TNF-{\alpha}$. Also, NK-92 MI cells growth was increased in adding the water fractions of the plants at $60^{\circ}C$ and 40 kHz. The water fraction of Ehpedrae Sinica STAPF among the plants showed the highest in NK-92 MI cell growth ratio. The differentiation activity of the HL-60 cells significantly increased in adding the water fraction of Ehpedrae Sinica STAPF compared to other fractions of the plants. These results suggest that the water fractions of the plants in extraction system of temperature $60^{\circ}C$ and ultrasonification 40 kHz have marked useful immuno-stimulatory activities.

Inhibitory Effects of Soamsan1 on Lung Metastasis of B16 Melanoma Cells (B16 흑색종 세포의 폐전이에 대한 소암산1의 억제효과)

  • We have examined whether Soamsan 1 (SA 1) augment the inhibitory effect of oral administration of Soamsan (SA) on lung metastasis of mouse 816 melanoma cells. The inhibitory effect was slightly enhanced by increase in administration dosage of SA 1. SA 1 as well as SA inhibited effectively the lung metastasis regardless of the pretreatment with anti-mouseNK monoclonal antibody. However, in the case of 2-chloroadenosine-pretreated mice, the inhibitory effects of SA and SA 1 were decreased by 18 and 23%, respectively. In vitro stimulation of the mouse splenocytes with mitogens showed that SA or SA 1 significantly augmented the proliferation of mouse splenocytes. Especially, the activity was more prominent in the presence of a B cell mitogen. LPS than a T cell mitogen, Con A. These results suggest that oral administration of SA 1 or SA inhibited lung metastasis of B16 melanoma cells, possibly through a mechanism mediated by the activation of macrophages and B lymphocytes in the host immune system. However, SA 1 did not showed more significant augment of the activation of immune system than SA.

Safety Test of Brown Rice Expressing Arabidopsis Calcium Transporter by Feeding Trial in Mice (애기장대 칼슘수송체를 발현하는 형질전환 현미의 생쥐 식이를 통한 안전성 평가)

  • Previously, we found that the transgenic rice plants over-expressing the Arabidopsis $H^+/Ca^{2+}$ antiporter CAX 1 (accession no. U57411) gene accumulated 2.7 to 7.5-fold more calcium in the T3 rice grains as compared to those of control. To examine physiological safety of the $T_3$ rice grains, the effect of the $T_3$ brown rice on change in levels of body weight and white blood cells was compared with that of the control Ilpum brown rice by feeding trial in mice. During the feeding trial for one month, there was no significant difference between two mice groups, which were fed by the $T_3$ brown rice or Ilpum brown rice. There were no detectable differences in their effects on immune functions including plaque-forming unit, peritoneal macrophage number, and NK-cell activity. In addition, biochemical analysis of the blood failed to exhibit any difference between two mice groups. Together, these results suggested that the $T_3$ brown rice, which was produced from a genetically modified organism (GMO), might be safe and possess a potential to be applicable as calcium-fortified feed or food. Long-term safety of the $T_3$ brown rice, however, remains to be elucidated.

Immuno-stimulating Activities of Polysaccharide Fractions Isolated from Persimmon Leaves (감잎에서 분리한 다당의 면역자극 활성)

  • In order to develop new physiologically active polysaccharides from persimmon leaves, two different crude polysaccharides were prepared using hot water (PLW-0) and pectinase digestion (PLE-0) and their immuno-stimulating activities were estimated. PLW-0 and PLE-0 showed similar sugar compositions with 15 different sugars, including rarely observed sugars in general polysaccharides such as 2-O-methyl-fucose, 2-O-methyl-xylose, apiose, aceric acid, 3-deoxy-D-manno-2-octulosonic acid, and 3-deoxy-D-lyxo-2-heptulosaric acid, but the uronic acid content of PLE-0 was lower than that of PLW-0 caused by pectinase treatment. Both PLW-0 and PLE-0 showed potent anti-complementary activity in a dose-dependent manner which was similar to a known immuno-stimulating polysaccharide, PSK, from Coriolus versicolor. The activity of PLE-0 at a low concentration ($100{\mu}g/m{\ell}$) was higher than that of PLW-0. In an in vitro cytotoxicity analysis, PLW-0 and PLE-0 (up to $1,000{\mu}g/m{\ell}$) did not affect the growth of peritoneal macrophages and Colon 26-M3.1 carcinoma cells. In contrast, they enhanced lymphocyte proliferation activity. Peritoneal macrophages stimulated with PLW-0 and PLE-0 produced various cytokines, such as IL-6 and IL-12. However, PLE-0 was more effective on the cytokine production. Intravenous administration of PLW-0 and PLE-0 significantly augmented natural killer (NK) cell cytotoxicity against Yac-1 tumor cells 3 days after the treatment of polysaccharide fractions. But NK cells obtained from the PLE-treated group showed higher tumoricidal activity even at a low dose of $40{\mu}g$/mouse. In experimental lung metastasis of Colon 26-M3.1 carcinoma cells, prophylactic administration of PLW-0 and PLE-0 significantly inhibited lung metastasis in a dose-dependent manner and PLE-0 was more effective on the inhibition of cancer metasasis. The results lead us to conclude that the pectinase-treated process is indispensable to preparing polysaccharides with higher immune-stimulating activity from persimmon leaves.

Active hexose correlated compound potentiates the antitumor effects of low-dose 5-fluorouracil through modulation of immune function in hepatoma 22 tumor-bearing mice

  • BACKGROUND/OBJECTIVES: A variety of immunomodulators can improve the efficacy of low-dose chemotherapeutics. Active hexose correlated compound (AHCC), a mushroom mycelia extract, has been shown to be a strong immunomodulator. Whether AHCC could enhance the antitumor effect of low-dose 5-fluorouracil (5-FU) via regulation of host immunity is unknown. MATERIALS/METHODS: In the current study Hepatoma 22 (H22) tumor-bearing mice were treated with PBS, 5-FU ($10mg{\cdot}kg^{-1}{\cdot}d^{-1}$, i.p), or AHCC ($360mg{\cdot}kg^{-1}{\cdot}d^{-1}$, i.g) plus 5-FU, respectively, for 5 d. $CD^{3+}$, $CD^{4+}$, $CD^{8+}$, and NK in peripheral blood were detected by flow cytometry. ALT, AST, BUN, and Cr levels were measured by biochemical assay. IL-2 and $TNF{\alpha}$ in serum were measured using the RIA kit and apoptosis of tumor was detected by TUNEL staining. Bax, Bcl-2, and TS protein levels were measured by immunohistochemical staining and mRNA level was evaluated by RT-PCR. RESULTS: Diet consumption and body weight showed that AHCC had no apparent toxicity. AHCC could reverse liver injury and myelosuppression induced by 5-FU (P < 0.05). Compared to mice treated with 5-FU, mice treated with AHCC plus 5-FU had higher thymus index, percentages of $CD^{3+}$, $CD^{4+}$, and NK cells (P < 0.01), and ratio of $CD^{4+}$/$CD^{8+}$ (P < 0.01) in peripheral blood. Radioimmunoassay showed that mice treated with AHCC plus 5-FU had the highest serum levels of IL-2 and $TNF{\alpha}$ compared with the vehicle group and 5-FU group. More importantly, the combination of AHCC and 5-FU produced a more potent antitumor effect (P < 0.05) and caused more severe apoptosis in tumor tissue (P < 0.05) compared with the 5-FU group. In addition, the combination of AHCC and 5-FU further up-regulated the expression of Bcl-2 associated X protein (Bax) (P < 0.01), while it down-regulated the expression of B cell lymphoma 2 (Bcl-2) (P < 0.01). CONCLUSIONS: These results support the claim that AHCC might be beneficial for cancer patients receiving chemotherapy.

Comparison of Immuno-Modulatory Regulatory Activities of Rubus coreanus Miquel by Ultra High Pressure Extracts Process (초고압 공정에 의한 복분자의 면역조절효능 비교)

  • This study was performed to investigate the enhancement of anticancer activities and immuno modulatary activities from R. coreanus. by ultra high pressure extracts process. The cytotoxicity on human kidney cell (HEK293) was showed below 19.5% in adding 1.0 $mg/m{\ell}$ concentration. The anticancer activity was increased over 10% by high pressure processing in AGS and A549 cells. The immune cell growth using human immune B and T cells was improved by the high pressure extracts of Rubus coreanus in adding 1.0 $mg/m{\ell}$ concentration. The secretion of two kinds of cytokine, the IL-6 and $TNF-{\alpha}$ from human immune B and T cells were also enhanced in adding extracts by high pressure process of R. coreanus. The ultra high pressure extraction technique showed high efficiency in extracting of bioactive compound. The ultra high pressure technique could be used combined with other technique to improve the extracting rate and extracting efficiency.

Experiment Report about the Effect on the Anti-cancer of Herbal-acupuncture with Evodiae Fructus Infusion Solution (오수유(吳茱萸)약침의 항암효과에 대한 실험적 연구)

  • This study was done to investigate the effect of anti-cancer, anti-metastasis and immune response improvement of herbal-acupuncture with Evodiae Fructus herbal infusion solution(EF-HAS). To study the effects of anti-cancer, anti-metastasis and immune response improvement of EF-HAS, we injected EF-HAS into Chung-wan(CV12) of C57BL/6 mice implanted intravenously with Bl6-Fl0 melanoma. Then, we have examined the effect on the increasing of spleen cells, the effect on the number of CD25$^+$/CD4$^+$, CD8$^+$/CD3e$^+$, CD69$^+$/B220$^+$, NK1.1$^+$/CD3e$^+$ cells in mice's PBMCs, the effect on the pulmonary colony number, and the effect on median survival time(MST) and increase of life span(ILS) of C57BL/6 mice implanted intravenously with Bl6-Fl0 melanoma. The following results were obtained; in the experiment groups treated with EF-HAS, the spleen cell proliferation in C57BL/6 mice, the percentage of CD25$^+$/CD4$^+$, CD8$^+$/CD3e$^+$, CD69$^+$/B220$^+$, NK1.1$^+$/CD3e$^+$ cells in C57BL/6 mice's PBMCs, and MST and ILS of C57BL/6 mice implanted intravenously with Bl6-Fl0 melanoma were significantly increased compared with that of the control group. And in the experiment groups treated with EF-HAS, the pulmonary colony number of C57BL/6 mice implanted intravenously with Bl6-Fl0 melanoma was decreased significantly compared with that of the control group. These results support a role for EF-HAS might De usefully applied in treatment of cancer.

Comparison of Immune modulatary and Anticancer Activities according to the Parts of the Styrax japonica Sieb. et Zucc. (때죽나무의 부위별 면역 및 항암활성 비교)

  • This study was performed to anticancer activities and immune modulatary activities according to the parts of the S. japonica Sieb. et Zucc. The cytotoxicity on human kidney cell (HEK 293) was showed below 27.4% in adding the methanol extracts. The anticancer activity were increased in over 60% by barks extracts in AGS and MCF-7 cells. The immune cell growth using human immune B and T cells was improved by the barks extracts of S. japonica Sieb. et Zucc. in adding 1.0mg/ml concentration. The secretion of the IL-6 and TNF-${\alpha}$ from human immune B and T cells was showed secretion for the amount of cytokines by bark extracts of S. japonica Sieb. et Zucc. NK cell growth was increased against control all of the extracts of S. japonica Sieb. et Zucc. Densitometric analysis of Bcl-2 revealed that possible to decrease potentialities of taking cancer in adding of extracts from S. japonica Sieb. et Zucc. From the results, the roots and barks extracts of S. japonica Sieb. et Zucc. were showed useful biological activities.

Anticancer and Immune Activities of the Extracts from Amorpha fruticosa L. (족제비싸리 부위별 추출물의 항암 및 면역활성)

  • This study was performed to experiment about useful biological activities of the parts of the extracts from Amorpha fruticosa LINNE. Experimental studies were progressed through the anticancer activities and immune activities such as cell cytotoxicity, inhibition activities of cell growth, cell growth of human B and T cell, productivity of cytokines and natural killer cell activities. The cell cytotoxicity using human Lung normal cell (HEL299) was showed cytotoxicity of below 22% by extracts of Amorpha fruticosa L. in 1.0 g/l concentration. The anticancer activities were increased in over 70% by roots extracts of Amorpha fruticosa L. in A549 and AGS cells. The immune cell growth using human immune B and T cells was increased against control by barks extracts of Amorpha fruticosa L. The secretion of cytokines $(IL-6,\;TNF-{\alpha})$ from human immune B and T cells was showed secretion of the amount of cytokines by roots extracts of Amorpha fruticosa L. Also NK cell growth was increased against control all of the extracts of Amorpha fruticosa L. From the results, the roots and barks extracts of Amorpha fruticosa L. were showed useful biological activities.
