Lipid Peroxidation, $NF-_{\kappa}B$ Activation and Cytokine Production in Neutrophil-Stimulated Pancreatic Acinar Cells

  • Reactive oxygen species (ROS), generated by infiltrating neutrophils, are considered as an important regulator in the pathogenesis and deveolpment of pancreatitis. The present study aims to investigate whether neutrophils primed by $4{\beta}-phorbol\;12{\beta}-myristate\;13{\alpha}-acetate$ (PMA) affect the productions $H_2O_2$ and lipid peroxide (LPO), $NF-_{\kappa}B$ activation and cytokine production in pancreatic acinar cells, and whether these alterations were inhibited by an antioxidant, N-acetylcysteine (NAC) and superoxide dismutase (SOD). $H_2O_2$ (ferrithiocyanate method), LPO (as thiobarbiturate reactive substances), and cytokines $(IL-l{\bata},\; IL-6,\;TNF-{\alpha};\;enzyme-linked\;immunosorbent\;assay)$ and $NF-_{\kappa}B$ activation (electrophoretic mobility shift assay) were analyzed in acinar cells treated with or without PMA-primed neutrophils in the absence or presence of NAC (10 mM) or SOD (300 U/ml). As a result, the productions of H2O2, LPO and $TNF-{\alpha}$ were increased with the ratio of PMA-primed neutrophils to acinar cells while the productions of LPO, $IL-l{\beta},\;IL-6\;and\;TNF-{\alpha}$ were increased with time. PMA-primed neutrophils resulted in the activation of $NF-_{\kappa}B.$ Both NAC and SOD inhibited neutrophil-induced alterations in acinar cells. In conclusion, ROS, generated by neutrophils, activates $NF-_{\kappa}B,$ resulting in upregulation of inflammatary cytokines in acinar cells. Antioxidants might be clinically useful antiinflammatory agents by inhibiting oxidant-mediated activation of $NF-_{\kappa}B$ and decreasing cytokine production.

LPS로 유도한 염증반응에서 해죽순의 항염증 효과 (The Anti-inflammatory Effect of Nypa fruticans Wurmb. Fruit on Lipopolysaccharide-induced Inflammatory response on RAW 264.7 cells)

  • Objective : Nypa fruticans Wurmb. Fruit (NF) has been used as a conventional medicine to treat inflammatory peridontal diseases in Myanmar and Eastern Asia. However, the anti-inflammatory effect of NF aqueous extract on lipopolysaccharide (LPS)-induced inflammatory responses was not well-investigated. Therefore, this study was aimed to investigate the anti-inflammatory effect of NF on LPS-induced inflammatory responses on RAW 264.7 cells.Methods : To induce inflammation on the macrophage cell line, RAW 264.7 cells were treated with 500 ng/mL of LPS. Water extracts of NF was treated 1 h prior to treatment of LPS. Cell viability was measured by MTT assay. Production of nitrite was measured with Griess assay and pro-inflammatory cytokines such as interleukine (IL)-1β and IL-6, and tumor necrosis factor (TNF)-α was measured by enzyme-linked immunosorbent assay (ELISA) and real-time polymerase chain reaction (PCR). In addition, we examined the inhibitory mechanisms of NF by western blot and immunocytochemistry.Result : Water Extract from NF itself did not have any cytotoxic effect at the concentration of 200 ㎍/ml in RAW 264.7 cells. Treatment of NF inhibited the production of nitrite, and pro-inflammatory cytokines inlcuding IL-1β, IL-6 and TNF-α in a dose dependant. In addition, NF treatment inhibited the LPS-induced activation and translocation of nuclear factor (NF)-κB.Conclusion : In summary, our result suggest that treatment of NF could reduce the LPS-induced inflammatory responses via deactivation of NF-κB. This study could suggest that NF could be a beneficial drug or agent to prevent inflammation.

Effect of saccharin on inflammation in 3T3-L1 adipocytes and the related mechanism

  • BACKGROUND/OBJECTIVES: Excessive intake of simple sugars induces obesity and increases the risk of inflammation. Thus, interest in alternative sweeteners as a sugar substitute is increasing. The purpose of this study was to determine the effect of saccharin on inflammation in 3T3-L1 adipocytes. MATERIALS/METHODS: 3T3-L1 preadipocytes were differentiated into adipocytes. The adipocytes were treated with saccharin (0, 50, 100, and 200 ㎍/mL) for 24 h. Inflammation was induced by exposure of treated adipocytes to lipopolysaccharide (LPS) for 18 h and cell proliferation was measured. The concentration of nitric oxide (NO) was measured by using Griess reagent. Protein expressions of nuclear factor kappa B (NF-κB) and inhibitor κB (IκB) were determined by western blot analysis. The mRNA expressions of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), interleukin 1β (IL-1β), interleukin 6 (IL-6), monocyte chemoattractant protein-1 (MCP-1), and tumor necrosis factor-α (TNF-α) were determined by real-time PCR. RESULTS: Compared with the control group, the amount of NO and the mRNA expression of iNOS in the LPS-treated group were increased by about 17.6% and 46.9%, respectively, (P < 0.05), and those parameter levels were significantly decreased by saccharin treatment (P < 0.05). Protein expression of NF-κB was decreased and that of IκB was increased by saccharin treatment (P < 0.05). Saccharin decreased the mRNA expression of COX-2 and the inflammation cytokines (IL-1β, IL-6, MCP-1, and TNF-α) (P < 0.05). CONCLUSIONS: The results of this study suggest that saccharin can inhibit LPS-induced inflammatory responses in 3T3-L1 adipocytes via the NF-κB pathway.

아크 플라즈마를 이용한 과불화합물 처리공정에서 반응가스에 의한 효과 (Effect of Reaction Gases on PFCs Treatment Using Arc Plasma Process)

  • 화학적으로 안정한 과불화합물을 처리하기 위해서는 많은 양의 에너지를 필요로 한다. 이러한 단점을 극복하기 위해서 저전력 아크 플라즈마 시스템을 개발하였다. 분해대상은 $CF_4$, $SF_6$, $NF_3$가 플라즈마 토치로 직접 주입되었으며, 아크 플라즈마 토치의 열효율을 측정하여 실출력을 계산하였다. 실출력과 폐기체 유량 변화 그리고 추가적인 반응가스에 의한 분해효율을 확인하였다. 또한 열역학적 평형조성 분석을 수행하여 실험 결과와 비교하였다. 토치의 열효율은 60~66%의 결과를 보였으며 폐가스 유량이 증가함에 따라 분해효율이 감소하였고 입력전력이 늘어남에 따라 분해효율이 상승되었다. 추가적인 반응 가스가 없이 $CF_4$, $SF_6$, $NF_3$의 분해효율은 입력전력이 3 kW, 폐가스 유량이 70 L/min인 조건에서 각각 4, 15, 90%를 보였다. 반응가스로 산소와 수소를 이용하여 분해효율을 급격하게 증가시킬 수 있었으며, 실험 결과 산소보다 수소를 사용하였을 경우가 분해효율 상승효과와 부산물 제어에 효과적인 것을 알 수 있었다. 수소의 경우, 발생되는 부산물은 불화수소산이었으며 이는 일반적인 습식 스크러버를 이용하여 처리가 용이한 물질이다. 수소를 이용한 화학반응에서 입력전력이 3 kW, 폐가스유량이 100 L/min인 조건에서 $CF_4$가 25%, $SF_6$가 39%, $NF_3$가 99%의 분해효율을 각각 나타냈다.

L-ascorbic acid induces apoptosis in human laryngeal epidermoid Hep-2 cells by modulating the nuclear factor kappa-light-chain-enhancer of activated B cells/mitogen-activated protein kinase/Akt signaling pathway

  • L-ascorbic acid (L-AA; vitamin C) induces apoptosis in cancer cells. This study aimed to elucidate the molecular mechanisms of L-AA-induced apoptosis in human laryngeal epidermoid carcinoma Hep-2 cells. L-AA suppressed the viability of Hep-2 cells and induced apoptosis, as shown by the cleavage and condensation of nuclear chromatin and increased number of Annexin V-positive cells. L-AA decreased Bcl-2 protein expression but upregulated Bax protein levels. In addition, cytochrome c release from the mitochondria into the cytosol and activation of caspase-9, -8, and -3 were enhanced by L-AA treatment. Furthermore, apoptosis-inducing factor (AIF) and endonuclease G (EndoG) were translocated into the nucleus during apoptosis of L-AA-treated Hep-2 cells. L-AA effectively inhibited the constitutive nuclear factor-κB (NF-κB) activation and attenuated the nuclear expression of the p65 subunit of NF-κB. Interestingly, L-AA treatment of Hep-2 cells markedly activated Akt and mitogen-activated protein kinase (MAPK; extracellular signal-regulated kinase 1/2, p38, and c-Jun N-terminal kinase [JNK]) and and LY294002 (Akt inhibitor), SB203580 (p38 inhibitor) or SP600125 (a JNK inhibitor) decreased the levels of Annexin V-positive cells. These results suggested that L-AA induces the apoptosis of Hep-2 cells via the nuclear translocation of AIF and EndoG by modulating the Bcl-2 family and MAPK/Akt signaling pathways.

Leaves of Raphanus sativus L. Shows Anti-Inflammatory Activity in LPS-Stimulated Macrophages via Suppression of COX-2 and iNOS Expression.

  • Raphanus sativus L. (RS) is a cruciferous vegetable that is widely consumed in Korea. The anticancer activity of leaves of RS (RSL) extract has been investigated; however, no studies focused on its anti-inflammatory effects. Therefore, the aim of the current study was to evaluate the anti-inflammatory effects of RSL extract. In brief, RSL powder was fractionated into n-hexane, chloroform, ethyl acetate, n-butanol, and water-soluble fractions. Lipopolysaccharide (LPS)-stimulated RAW264.7 cells were treated with each fraction for initial screening. It was found that the chloroform fraction significantly inhibited nitric oxide release in LPS-stimulated RAW264.7 cells with a half maximal inhibitory concentration value of $196{\mu}g/mL$. In addition, the mRNA and protein expression levels of inducible nitric oxide synthase, measured using reverse transcriptase-polymerase chain reaction and western blotting, respectively, were reduced in a concentration-dependent manner. Moreover, the inflammatory cyclooxygenase-2 enzyme expression decreased. Furthermore, the expression of nuclear factor-kappa B ($NF-{\kappa}B$), the key regulator of the transcriptional activation of the inflammatory cytokine genes, was reduced by the RSL chloroform fraction. Therefore, the results of our study suggest that RSL exhibits anti-inflammatory effects in LPS-stimulated macrophages via $NF-{\kappa}B$ inactivation.

복분자가 U937세포의 apoptosis에 미치는 효과 (Effects of Rubus Coreanus Miquel on the Apoptosis of U937 Cells)

  • Methanol extracts of Rubus Coreanus Miquel (RCM) were found to exhibit apoptosis induction of U937 human histiocytic leukemia cells. Treatment of RCM exerted strong cytotoxicity against U937 human leukemia cells. RCM induced apoptosis of U937 leukemia cells in a dose dependent manner. Nitric oxide (NO) production was increased in RCM-treated RAW264.7 macrophage cell line. RCM increased the p53 and $NF{\kappa}B$ gene and decreased the $I{\kappa}B$ gene expression in U937 cells. RCM also increased the $NF{\kappa}B$ protein expression, but decreased the PCNA and BcL-xL protein expression in cultured U937 cells. These data suggest that RCM are effective on the apoptosis induction of human leukemia cells and anti-cancer properties.

Helicobacter pylori 감염에 의한 Cytokines 유전자 발현에 대한 치양탕(治瘍湯)의 효과 (Effects of Chiyangtang on Helicobacter pylori-induced increase of cytokines gene expression)

  • Effects of Chiyangtang(CYT) on H. pylori-induced increase of interleukin 8 and interleukin 1 gene expression was studied in Kato Ⅲ cell line, a human stomach epithelial cell line. Treatment of H. pylori to the cell culture signifant!y increased IL-8 and IL-1 mRNA synthesis. When CYT was added along with H. pylori, the increase of IL-8 and IL-1 mRNA synthesis was blocked. Activation of transcription factor $NF-{\kappa}B$ and AP-1 which were known to important in IL-8 and IL-1 gene expression was also studied using chloramphenicol acetyltransferase(CAT) assay. Treatment of H. pylori increased activation of $NF-{\kappa}B$ and AP-l and CYT effectively protected the activation. Electrophoretic mobility shift assay suggested that CYT effectively inhibited DNA binding of $NF-{\kappa}B$ and AP-l to their cognate site. These results suggested that CYT could prevent stomach diseases through the down regulation of IL -8 and IL-l gene expression which might be mediated by the inhibition of $NF-{\kappa}B$ and AP-1 activities and their binding to DNA.

렌즈콩(Lens culinaris) 추출물이 HepG2 인간 간암 세포에서 Proteasome 활성과 Nuclear Factor κB 활성화에 미치는 영향 (Effects of Lentils(Lens culinaris) Extract on Proteasome Activity and Nuclear Factor κB Activation in HepG2 Human Liver Cancer Cells)

  • Proteasome inhibitors can improve the efficiency of cancer treatments by inhibiting nuclear factor ${\kappa}B$($NF-{\kappa}B$) activation in cancer cells. Lentils are a type of beans of which consumption of such beans is increasing. The purpose of this study was to investigate the effects of lentils extract (LE) on the proteasomal activities, $NF-{\kappa}B$ activation, and cell cycle in HepG2 human liver cancer cells. LE treatments inhibited proteasomal activities at concentrations of 10, 50, and $100{\mu}g/mL$ respectively, and repressed $NF-{\kappa}B$ activation at concentrations of 1, 10, and $100{\mu}g/mL$ respectively, in HepG2 cells. LE treatments at concentrations of 1, 10, and $100{\mu}g/mL$ respectively, increased sub-G1 cell population in HepG2 cells, which may be the result of apoptosis. The results suggest that LE inhibited $NF-{\kappa}B$ activation partially with its proteasome inhibitory activities, and the increase of sub-G1 cell population was induced partially, by inhibition of $NF-{\kappa}B$ activation in HepG2 cells.

생리적 반응이 다른 비료 종류가 '설향' 딸기의 영양생장에 미치는 영향 (Influence of Fertilizer Type on Physiological Responses during Vegetative Growth in 'Seolhyang' Strawberry)

  • '설향' 딸기를 재배하는 동안 비료의 종류와 농도를 변화시켜 시비하고 작물 생장과 양분 흡수량 변화에 미치는 영향을 구명하여 시비를 위한 기초자료를 확보하고자 본 연구를 수행하였다. 증류수를 원수로 산성, 중성 및 알칼리성 비료를 조제한 후 N 농도를 기준으로 $100mg{\cdot}L^{-1}$$200mg{\cdot}L^{-1}$로 그리고 처리용액의 pH를 약 6.0-6.3으로 조절하여 100일간 시비하였다. 동일한 시비농도에서 시비 100일 후 '설향' 딸기의 생체중 및 건물중은 산성비료를 시비한 처리가 가장 가벼웠고 알칼리비료 처리가 무거웠다. 또한 알칼리비료의 $100mg{\cdot}L^{-1}$보다 $200mg{\cdot}L^{-1}$로 시비한 처리의 생체중과 건물중이 더 무거웠다. 식물체 무기원소 함량을 분석한 결과 N, P 및 Na은 중성비료 $200mg{\cdot}L^{-1}$ 처리에서 각각 3.08, 0.54 및 0.10%로, K은 산성비료 $200mg{\cdot}L^{-1}$ 처리에서 2.83%로, Ca과 Mg은 알칼리비료 $100mg{\cdot}L^{-1}$ 처리에서 0.98 및 0.42%로 다른 처리들보다 유의하게 식물체 내 함량이 높았다. Fe, Mn, Zn 및 Cu 함량은 산성비료를 시비한 처리에서 높았고, 알칼리비료를 시비한 처리에서 뚜렷하게 낮았다. 근권부 pH는 산성비료 $100mg{\cdot}L^{-1}$$200mg{\cdot}L^{-1}$, 그리고 중성비료 $200mg{\cdot}L^{-1}$으로 시비한 처리에서 지속적으로 낮아졌다. 근권부 무기원소 농도는 산성비료 $200mg{\cdot}L^{-1}$, 중성비료 $200mg{\cdot}L^{-1}$, 그리고 산성비료를 $100mg{\cdot}L^{-1}$으로 시비한 처리 순으로 $NH_4$ 농도가 낮아졌다. 근권부 K 농도는 시비 42일 후부터 모든 처리에서 지속적으로 상승하였으며, Ca 및 Mg 농도는 시비 84일 후 까지는 알칼리비료를 시비한 처리의 농도가 가장 낮고 중성 비료 및 산성 비료 순으로 높아지는 경향이었다. $NO_3$는 산성비료에서 낮았고, 중성 및 알칼리비료의 순으로 높아졌다. 세 종류 비료를 동일한 농도로 시비한 경우 $PO_4$ 농도가 유사한 경향을 보이며 변화하였고, $SO_4$은 시비 70일 후까지 산성 비료가 중성이나 알칼리성 비료를 시비한 처리들보다 높았다. 이상의 연구결과는 '설향' 딸기 재배에서 알칼리성 비료를 시비해야 함을 의미하며, 알칼리 원소의 농도가 상승하도록 관비용액의 조성을 변화시켜야 한다고 판단하였다.
