The Effectiveness of Ulmus Davidiana Planch Herbal Acupuncture to Inhibit $NF-{\kappa}B$ Activation on Type II Collagen-induced Arthritis in Mice (유근피(楡根皮) 약침의 $NF-{\kappa}B$ 활성 억제능이 생쥐의 Type II Collagen 유발 관절염에 미치는 영향)

  • Objectives : The purpose of this study is to investigate the effectiveness of Ulmus davidiana Planch herbal acupuncture(UA) to inhibit nuclear $factor(NF)-{\kappa}B$ activation on type II collagen-induced arthritis (CIA) in mice. Methods : Using an in vitro test, the synoviocytes picked out from the experimental CIA mice were subcultured. The synoviocyte cells were treated with phorbol-12-myristate-13-acetate(PMA) for 1 hour prior to the addition of indicated concentrations($0.4\;-\;1.0mg/m{\ell}$) of UA, and the cells were further incubated for 24 hours. The in vivo test, $NF-{\kappa}B$ p65, inducible nitric oxide synthase(iNOS), cyclooxygenase-2(COX-2), vascular cell adhesion molecule(VCAM)-1 production and apoptosis was observed by immunohistochemical staining. Results : The PMA-induced $I{\kappa}B$ kinase(IKK), iNOS and COX-2 mRNA expression were dose-dependently decreased in UA treated synoviocytes. Using the in vivo test, the number of eosinophils in mice treated with UA noticeably decreased in the the CIA group(P<0.05 using student t test). In mice treated with UA, there was less cartilage erosion. less bone destruction, mild synovial hyperplasia, mild fibrosis, and mild angiogenesis with less MIP-2 production. By immunohistochemical staining, suppression of $NF-{\kappa}B$ p65, iNOS production, inhibition of COX-2 production, inhibition of VCAM-1 production and inducing apoptosis were observed. Conclusions : These results suggest that UA might be applicable to the therapy of RA to suppress $NF-{\kappa}B$ activation.

The Effect of Gamitongkyutang Distillate in Mice with Allergic Rhinitis

  • Objectives : This study aimed to find the curative effect of Gamitongkyutang distillate in mice with allergic rhinitis. Metbods : Forty mice were divided into four groups: the normal group, the control group (allergic rhinitis elicited group), the sample I group (Gamitongkyutang treated group after allergic rhinitis elicitation) and the sample II group (distillate of Gamitongkyutang treated group after allergic rhinitis elicitation). Indexes of AR were investigated such as the histological changes of the nasal mucosa, the changes of eosinophil count, the changes of interleukin-4(IL-4) secretion in the intranasal mucosa, the alteration of inducible nitric oxide synthase(iNOS) mRNA expression and the distribution of the nuclear factor kappa B (NF-kB). ANOVA test was used for statistical analysis (p<0.05). Results : Loss of the cilium and the mucous secretion in the sample I and II groups was rare when compared to the control group. The segment of eosinophil was significantly decreased in the sample I and II groups when compared to the control group (p<0.05). A significant decrease of IL-4 mRNA expression was observed in the sample I and II groups when compared with the control group (p<0.05). Inhibition of iNOS induced by NF-kB p50 in the sample I and II groups was significantly superior to that in the control group (p<0.05). DGT and GT didn't affect AST and ALT. Conclusions : GT was superior to DGT in the IL-4 secretion, eosinophil levels and iNOS production. However, considering the difficulty in taking herbal medicine, the DGT has a meaningful curative effect in mice with allergic rhinitis.

Luteolin and luteolin-7-O-glucoside protect against acute liver injury through regulation of inflammatory mediators and antioxidative enzymes in GalN/LPS-induced hepatitic ICR mice

  • BACKGROUND/OBJECTIVES: Anti-inflammatory and antioxidative activities of luteolin and luteolin-7-O-glucoside were compared in galactosamine (GalN)/lipopolysaccharide (LPS)-induced hepatitic ICR mice. MATERIALS/METHODS: Male ICR mice (6 weeks old) were divided into 4 groups: normal control, GalN/LPS, luteolin, and luteolin-7-O-glucoside groups. The latter two groups were administered luteolin or luteolin-7-O-glucoside (50 mg/kg BW) daily by gavage for 3 weeks after which hepatitis was induced by intraperitoneal injection of GalN and LPS (1 g/kg BW and $10{\mu}g/kg\;BW$, respectively). RESULTS: GalN/LPS produced acute hepatic injury by a sharp increase in serum AST, ALT, and $TNF-{\alpha}$ levels, increases that were ameliorated in the experimental groups. In addition, markedly increased expressions of cyclooxygenase (COX)-2 and its transcription factors, nuclear factor $(NF)-{\kappa}B$ and activator protein (AP)-1, were also significantly attenuated in the experimental groups. Compared to luteolin-7-O-glucoside, luteolin more potently ameliorated the levels of inflammatory mediators. Phase II enzymes levels and NF-E2 p45-related factor (Nrf)-2 activation that were decreased by GalN/LPS were increased by luteolin and luteolin-7-O-glucoside administration. In addition, compared to luteolin, luteolin-7-O-glucoside acted as a more potent inducer of changes in phase II enzymes. Liver histopathology results were consistent with the mediator and enzyme results. CONCLUSION: Luteolin and luteolin-7-O-glucoside protect against GalN/LPS-induced hepatotoxicity through the regulation of inflammatory mediators and phase II enzymes.

Functional Analysis of the Heptasequence SPTSPTY in the Transcriptional Activation Domain of Rat Nuclear Factor 1-A

  • Nuclear Factor 1 (NF1) proteins are a family of transcriptional factors consisting of four different types: NF1-A, -B, -C, and -X. Some NF1 transcription factors contain a heptasequence motif, SPTSPSY, which is found as a repeat sequence in the carboxy terminal domain (CTD) of the largest subunit of RNA polymerase II. A similar heptasequence, SPTSPTY, is contained in rat liver NF1-A at a position between residues 469 and 475. In order to investigate the roles of the individual amino acids of the heptasequence of rat liver NF1-A in transcriptional activation, we systematically substituted single and multiple amino acid residues with alanine residue(s) and evaluated the transcriptional activities of the mutated NF1-A. Substitution of a single amino acid reduced transcriptional activity by 10 to 30%, except for the proline residue at position 473, whose substitution with alanine did not affect transcriptional activity. However, changes of all four serine and threonine residues to alanine or of the tyrosine residue along with the serine residue at position 469 to alanine reduced the activity to almost background levels. Our results indicate that multiple serine and threonine residues, rather than a single residue, may be involved in the modulation of the transcriptional activities of the factor. Involvement of the tyrosine residue is also implicated.

The Treatment Effect of Neurofeedback Training on Executive Function in Attention-Deficit Hyperactivity Disorder (뉴로피드백 훈련이 Attention-Deficit Hyperactivity Disorder 아동의 실행기능에 미치는 영향)

  • Objectives : Children with attention-deficit hyperactivity disorder (ADHD) have been shown to display more inhibitory deficits and executive function deficits. This study investigated the treatment effects of neurofeedback (NF) training on executive function by comparing the results of neuropsychological tests of the trained children at pre- and post-training. Methods : Fifteen children with ADHD, aged 6 to 14 years, participated in the study. The NF treatment consisted of slow cortical potential (SCP) training and these sessions took place once a week. The ADHD children performed 20 sessions of NF training within 6 months. Pre-training and post-training assessments encompassed Continuous Performance Test (CPT), Stroop Test, Children's Color Trails Test I&II (CCTT) and Wisconsin Card Sorting Test (WCST). Results : Patients receiving NF training showed significant improvement in visual commission error and standard deviation of auditory response time on CPT ; and total errors on WCST. But there was no significant improvement in the Stroop test and CCTT. Conclusion : SCP training using NF improves the self-regulatory capacities and impulsivity in ADHD patient, especially impulsivity in visual stimulation tasks. This study showed evidence of clinical efficacy of NF on executive function in ADHD.

  • In this study, We evaluated the removal efficiency of natural organic matters(NOM) in the Ultrafiltration(UF) and Nanofiltration(NF) membranes with molecular weight cutoff of 2500(GH), 8000(GM) and 250(HL), respectively. Filtration type was cross-flow filtration. The investigation result about raw water structure was hydrophobic 28%, hydrophilic 53% and transphilic 19%, in conjunction with XAD8/4 resin fractionation method. We were compared with UF(GM, GH) and NF(HL), in operation characteristic. In spite of poor MWCO, GM(8000Da) was superior than GH(2500Da), in the efficiency of total operation. It was showed the NF(HL) 80%, UF(GM) 50%, in the removing efficiency of HAAFP.

  • In this study the nanofiltration (NF) membrane treatment of a sulfuric acid waste solutions containing copper ion ($Cu^{+2}$) discharging from the etching processes of the printed circuit board (PCB) manufacturing industry has been studied for the recycling of acid etching solution. SelRO MPS-34 4040 NF membrane from Koch company was tested to obtain the basic NF data for recycling of etching solution and separation efficiency (total rejection) of copper ion. NF experiments were carried out with a dead-end membrane filtration laboratory system. The pure water flux was increased with the increasing storage time in sulfuric acid solution and lowering pH of acid solution because of the enhancement of NF membrane damage by sulfuric acid. The permeate flux of acid solution was decreased with the increasing copper ion concentration. Total rejection of copper ion was decreased with the increasing storage time in sulfuric acid solution and copper ion concentration, and lowering the pH of acid solution. The total rejection of copper ion was decreased from initial 37% to 15% minimum value.

Effect of Orostachys japonicus on Apoptosis and Autophagy in Human monocytic leukemia Cell line THP-1 via Inhibition of NF-κB and Phosphorylation of p38 MAPK (와송이 인간 백혈병 세포주 THP-1에서 NF-κB 활성 억제와 p38 활성을 통해 세포사멸과 자가포식에 미치는 영향)

  • Objectives: Orostachys japonicas (O. japonicus) has been known for its anti-tumor effect. In the present study, it was investigated whether O. japonicus EtOH extracts could induce apoptosis and autophagy which are part of the main mechanism related to anti-tumor effect in THP-1 cells. Methods: Cells were treated with various concentrations of O. japonicus EtOH extracts ($0-300{\mu}g/ml$) for 24, 48, and 72h. Cell viability was evaluated by MTS/PMS assay and apoptosis rate was examined by flow cytometry and ELISA assay. The mRNA expression of apoptosis-related genes (Bcl-2, Mcl-1, Survivin, Bax) and autophagy-related gene (mTOR) was evaluated using real-time PCR. The protein expression of Caspase-3, Akt, LC3 II, Beclin-1, Atg5, $NF-{\kappa}B$, p38, ERK was evaluated using western blot analysis. Results: O. japonicus EtOH extracts inhibited cell proliferation and apoptosis rate was increased in both flow cytometry and ELISA assay. Bcl-2, Mcl-1, Survivin (anti-apoptosis factors) mRNA expressions were decreased and Bax (pro-apoptosis factor) mRNA level was increased. mTOR mRNA expressions was decreased and LC3 II protein expressions was increased. Activation of $NF-{\kappa}B$ was decreased and phosphorylation of p38 was increased. Conclusion: O. japonicus is regarded to inhibit cell proliferation, to induce apoptosis and to regulate autophagy-related genes in THP-1 cells via $NF-{\kappa}B$ and p38 MAPK signaling pathway. This suggests O. japonicus could be an effective herb in treating acute myeloid leukemia.
