CBT-SL5, a Bacteriocin from Enterococcus faecalis, Suppresses the Expression of Interleukin-8 Induced by Propionibacterium acnes in Cultured Human Keratinocytes

  • Propionibacterium acnes is known to playa pivotal role in the pathogenesis of acne vulgaris. CBT-SL5 is one of the antimicrobial peptides from Enterococcus faecalis SL5, and it has shown antimicrobial activity against P. acnes. The aim of this study was to investigate the anti-inflammatory effect of CBT-SL5 on the inflammation induced by P. acnes in cultured human keratinocyes. Cultured human keratinocytes derived from neonatal foreskin were treated with heat-killed P. acnes to induce inflammation, and then various concentrations of CBT-SL5 were added to the P. acnes-treated keratinocytes. The mRNA expression and protein secretion of interleukin (IL)-8, an inflammation marker, was analyzed by real-time reverse transcription polymerase chain reaction and enzyme-linked immunosorbent assay, respectively. We also analyzed the nuclear factor-kappa B (NF-$\kappaB$) p65 translocation by performing immunofluorescent staining. P. acnes treatment up regulated the IL-8 mRNA expression in the keratinocytes, and this was brought about through both toll-like receptor (TLR)2 and TLR4. At the concentrations of 10, 50, and 100 ng/ml, CBT-SL5 significantly down regulated the P. acnes-induced IL-8 mRNA expression and protein production (p<0.05). At 6 hand 12 h of the treatment, CBT-SL5 significantly suppressed the P. acnes-induced IL-8 mRNA expression. Secretion of IL-8 protein was significantly reduced at 24 h. The functional inhibitory activity of CBT-SL5 was shown by CBT-SL5 suppressing the P. acnes-induced NF-$\kappaB$ translocation from the cytoplasm to the nucleus. These results demonstrated that CBT-SL5 suppressed the P. acnes-induced IL-8 expression in keratinocytes. Therefore, CBT-SL5 may be a novel anti-inflammatory treatment for acne.

β-Glucosidase 처리에 의한 치자추출물의 항염증 활성 증진 (Improvement of Anti-Inflammation Activity of Gardeniae fructus Extract by the Treatment of β-Glucosidase)

  • 본 연구에서는, 치자 열매를 항염증 소재로 선택하여, 치자 추출물(GFE)에 ${\beta}$-glucosidase를 처리함으로써 항염증 활성을 증진하고자 하였다. 항염증 활성의 비교를 위해 추출물과 LPS를 함께 처리하여 RAW264.7 세포가 생산하는 NO의 양을 측정하였다. Lactic acid bacteria, 식물 그리고 곰팡이 유래 ${\beta}$-glucosidase를 처리한 결과, 곰팡이(A. niger, A. fumigatus)에서 유래된 효소를 처리하는 것이 GFE의 NO생산 억제활성 증진에 가장 효과적이었다. 그리고 GFE에 대한 A. niger 유래 ${\beta}$-glucosidase의 최적 처리조건은 pH 4.6, $45^{\circ}C$ 그리고 20 U/mL의 효소 농도로 24 h 반응시키는 것이었다. 이와 같은 조건으로 GFE에 효소를 처리한 GFAN은 GFE와 유사한 세포 생존율을 나타냈고, 세포독성은 나타나지 않았다. 또한 GFAN의 처리에 의하여 RAW264.7 세포의염증성 cytokine생산, COX-2생산 그리고 NF-${\kappa}B$활성이 현저히 줄어들었다. 본 연구에서 GFAN의 항염증 활성 증진은 치자의 성분 중 주로 geniposide가 ${\beta}$-glucosidase처리에 의하여 genipin으로 전환되었기 때문이라 생각된다. 이상의 결과는 ${\beta}$-glucosidase를 처리한 치자 추출물(GFE)을 이용하여 안전하고 효과적인 항염증 기능성 식품 소재 개발이 가능함을 제시하였다.

열처리 사균체 엔테로코커스 패칼리스 EF-2001의 항염증 효과 (Anti-inflammatory Effect of Heat-Killed Enterococcus faecalis, EF-2001)

  • 염증은 인체에서 가장 흔히 나타나는 증상으로 조직이 손상되면 염증 반응이 발생하고 염증 부위에서 혈관 확장과 혈류가 증가하여 부종이 생긴다. Lipopolysaccharide (LPS)는 Toll-like receptor 4에 의해 인식되고 염증 반응을 일으킨다. 열로 사멸시킨 Enterococcus faecalis 사균체(EF-2001)는 면역 조절 및 예방 활동을 하는 것으로 사전 보고되었고, 항 종양 효과가 있다고 보고되었지만 염증에 미치는 영향에 대해서는 지금까지 연구되지 않았다. 본 연구에서는 LPS에 의한 대식세포 염증 반응에 대한 EF-2001의 효과에 대해 연구하였다. 연구결과에서 EF-2001은 LPS에 의해 유도된 산화 질소의 생성을 감소시켰다. 우리는 EF-2001의 세포 독성이 있는지 확인했으며, 산화 질소의 감소는 세포독성에 의한 것이 아님을 확인하였다. 또한 이러한 EF-2001의 항염증 효과에 대한 분자기전을 연구하였다. LPS에 의한 유도된 iNOS와 COX-2의 발현은 EF-2001에 의해 감소되었다. 더해진 분자기작 분석에서 EF-2001은 LPS로 유도된 ERK, JNK 및 p38 인산화를 농도 의존적으로 억제하였다. 더해진 실험에서 EF-2001은 Akt 인산화를 억제하고 $NF-{\kappa}B$ 억제제인 $I{\kappa}B$ 단백질 발현을 증가시켰다. 또한, EF-2001은 p65의 핵으로의 이동을 억제함을 알수 있었다. 따라서, 이러한 결과는 EF-2001이 항염증 효과를 가지며 염증 질환 치료에 유용 할 수 활용될 수 있음을 시사한다.

항염증 및 항산화 활성 보유 유용 식물 탐색 (Screening of Useful Plants with Anti-inflammatory and Antioxidant Activity)

  • 본 연구는 식물자원을 재료로 하여 in vitro 항염증 및 항산화 활성을 분석함으로써 기능성소재로의 활용 가능성이 있는 유망 후보자원을 발굴하고자 수행하였다. 이를 위해 식물추출물 38종을 대상으로 세포증식에 대한 영향, 염증 관련지표(nitric oxide, TNF-${\alpha}$, IL-6, $I{\kappa}B{\alpha}$, iNOS, COX-2) 및 항산화항목(DPPH 라디칼 소거능, LDL 산화저해능) 및 총페놀 함량에 대한 효과를 탐색하고 결과로부터 8종의 시료를 1차로 선발하였다. 선발된 8종의 시료에 대해서는 다시 농도별로 세포증식에 대한 효과, iNOS, COX-2의 발현 및 NF-${\kappa}B$ 전사에 대한 영향, peroxynitrite, ROS, nitric oxide 생성에 대한 $IC_{50}$ 값을 분석하여 보다 가능성이 있는 식물을 유망후보자원으로 선발하고자 하였다. 그 결과, 소사나무(가지), 등골나물(잎), 으름(꽃) 및 개모시풀(뿌리)은 항염증 혹은 항산화활성이 우수하나 농도에 따라 세포독성을 나타내었으므로 활용에 주의가 필요해보였으며, 어저귀(잎)은 항염증활성과 항산화활성이 비교적 우수하고 실험된 처리농도에서 세포독성이 없어 향후 in vivo 활성 검정 등 심화연구를 통해 그 활용 가능성을 검토할 필요가 있다고 사료되었다.

TI-I-174, a Synthetic Chalcone Derivative, Suppresses Nitric Oxide Production in Murine Macrophages via Heme Oxygenase-1 Induction and Inhibition of AP-1

  • Chalcones (1,3-diaryl-2-propen-1-ones), a flavonoid subfamily, are widely known for their anti-inflammatory properties. Propenone moiety in chalcones is known to play an important role in generating biological responses by chalcones. In the present study, we synthesized chalcone derivatives structurally modified in propenone moiety and examined inhibitory effect on nitric oxide (NO) production and its potential mechanisms. Among the chalcone derivatives used for this study, TI-I-174 (3-(2-Hydroxyphenyl)-1-(thiophen-3-yl)prop-2-en-1-one) most potently inhibited lipopolysaccharide (LPS)-stimulated nitrite production in RAW 264.7 macrophages. TI-I-174 treatment also markedly inhibited inducible nitric oxide synthase (iNOS) expression. However, TI-I-174 did not significantly affect production of IL-6, cyclooxygenase-2 (COX-2) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), implying that TI-I-174 inhibits production of inflammatory mediators in a selective manner. Treatment of macrophages with TI-I-174 significantly inhibited transcriptional activity of activator protein-1 (AP-1) as determined by luciferase reporter gene assay, whereas nuclear factor-${\kappa}B$ (NF-${\kappa}B$) activity was not affected by TI-I-1744. In addition, TI-I-174 significantly inhibited activation of c-Jun-N-Terminal kinase (JNK) without affecting ERK1/2 and p38MAPK, indicating that down-regulation of iNOS gene expression by TI-I-174 is mainly attributed by blockade of JNK/AP-1 activation. We also demonstrated that TI-I-174 treatment led to an increase in heme oxygenase-1 (HO-1) expression both at mRNA and protein level. Transfection of siRNA targeting HO-1 reversed TI-I-174-mediated inhibition of nitrite production. Taken together, these results indicate that TI-I-174 suppresses NO production in LPS-stimulated RAW 264.7 macrophages via induction of HO-1 and blockade of AP-1 activation.

Pharmacological Activities and Applications of Spicatoside A

  • Liriopogons (Liriope and Opiopogon) species are used as a main medicinal ingredient in several Asian countries. The Liriopes Radix (tuber, root of Liriope platyphylla) has to be a promising candidate due to their source of phytochemicals. Steroidal saponins and their glycosides, phenolic compounds, secondary metabolites are considered of active constituents in Liriopes Radix. Spicatoside A, a steroidal saponin, could be more efficacious drug candidate in future. In this review, we summarized the available knowledge on phytochemical and pharmacological activities for spicatoside A. It significantly suppressed the level of NF-${\kappa}B$, NO, iNOS, Cox-2, IL-$1{\beta}$, IL-6 and MAPKs in LPS-stimulated inflammation. The production of MUC5AC mucin was increased. MMP-13 expression was down-regulated in IL-$1{\beta}$-treated cells and reduced glycosaminoglycan release from IL-$1{\alpha}$-treated cells. The neurite outgrowth activity, PI3K, Akt, ERK1/2, TrkA and CREB phosphorylation and neurotropic factors such as NGF and BDNF were upregulated with increased latency time. It also showed cell growth inhibitory activity on various carcinoma cells. From this, spicatoside A exerts anti-inflammation, anti-asthma, anti-osteoclastogenesis, neurite outgrowth, memory consolidation and anticancer activities. Further studies are needed on spicatoside A in order to understand mechanisms of action to treat various human diseases.

화피(樺皮) 에탄올 추출물의 Ultraviolet B로 자극한 피부 각질 세포 보호 작용 (Protective Effect of Betula Platyphylla on Ultraviolet B-irradiated HaCaT Keratinocytes)

  • Objectives: Betula Platyphylla(BP) has been used as a analgesic, anti-microbial, anti-oxidant drug in Eastern Asia. However, it is still unknown whether BP ethanol extract could exhibit the inhibitory activities against ultraviolet B(UVB)-induced skin injury on human keratinocytes, HaCaT cells. This study was aimed to investigate the protective activity of BP ethanol extract on UVB-irradiated skin injury in HaCaT cells. Methods: The skin injury model of HaCaT cells was established under UVB stimulation. HaCaT keratinocyte cells were pre-treated with BP ethanol extract for 1 h, and then stimulated with UVB. Then, the cells were harvested to measure the cell viability, production of reactive oxygen species(ROS), pro-inflammatory cytokines such as interleukin(IL) 1-beta, IL-6, and tumor necrosis factor(TNF)-𝛼, hyaluronidase, type 1 collagen, matrix metalloproteinase(MMP)s. In addition, we examined the mitogen activated protein kinases(MAPKs) and inhibitory kappa B alpha(I𝜅;-B𝛼) as inhibitory mechanisms of BP ethanol extract. Results: The treatment of BP ethanol extract inhibited the UVBinduced cell death and ROS production in HaCaT cells. BP ethanol extract treatment inhibited the UVB-induced increase of IL-1beta, IL-6, and TNF-𝛼. BP ethanol extract treatment inhibited the increase of hyaluronidase, MMP and decrease of collagen. BP ethanol extract treatment inhibited the activation of MAPKs and the degradation of I𝜅-B𝛼. Conclusions: Our result suggest that treatment of BP ethanol extract could inhibit the UVB-induced skin injury via deactivation of MAPKs and nuclear factor kappa B(NF-𝜅B) in HaCaT cells. This study could suggest that BP ethanol extract could be a beneficial agent to prevent skin damage or inflammation.

미세먼지로 인한 피부 각질 세포 손상에서 몰약 에탄올 추출물의 항염증 효과 (Anti-inflammatory Effects of Myrrh Ethanol Extract on Particulate Matter-induced Skin Injury)

  • Objectives: Myrrh have been used as a traditional remedy to treat infectious and inflammatory diseases. However, it is largely unknown whether myrrh ethanol extract could exhibit the inhibitory activities against particulate matter (PM)-induced skin injury on human keratinocytes, HaCaT cells. Therefore, this study was aimed to investigate the inhibitory activity of myrrh ethanol extract on PM-induced skin injury in HaCaT cells. Methods: To investigate the inhibitory effects of myrrh ethanol extract in HaCaT cells, the skin injury model of HaCaT cells was established under PM treatment. HaCaT keratinocyte cells were pre-treated with myrrh ethanol extract for 1 h, and then stimulated with PM. Inhibitory activity of gintonin on inflammation in human IL-1β-stimulated fibroblast-like synoviocytes and collagen-induced arthritis in mice

  • Background: Gintonin is a newly derived glycolipoprotein from the roots of ginseng. The purpose of this study is to investigate the anti-arthritic efficacy of Gintonin on various proteases and inflammatory mediators that have an important role in arthritis. Methods: Fibroblast-like synoviocytes (FLS) were treated with Gintonin and stimulated with interleukin (IL)-1β 1 hour later. The antioxidant effect of Gintonin was measured using MitoSOX and H2DCFDA experiments. The anti-arthritic efficacy of Gintonin was examined by analyzing the expression levels of inflammatory mediators using RT-PCR, western blot, and ELISA. The phosphorylation of mitogen-activated protein kinase (MAPK) pathways and translocation of nuclear factor kappa B (NF-κB)/p65 into the nucleus were also analyzed using western blot, ELISA, and immunocytochemistry. Collagen-induced arthritis (CIA) mice model was used. Mice were orally administered with Gintonin (25, 50, and 100 mg/kg) every 2 days for 45 days. The body weight, arthritis score, squeaking score, and paw volume were measured as the behavioral parameters. After sacrifice, H&E and safranin-O staining were performed for histological analysis. Results: Gintonin significantly inhibited the expression of inflammatory intermediates. Gintonin prevented NF-κB/p65 from moving into the nucleus through the JNK and ERK MAPK phosphorylation in FLS cells. Moreover, Gintonin suppressed the symptoms of arthritis in the CIA mice model. Conclusion: As a result, the antioxidant and anti-inflammatory effects of Gintonin were demonstrated, and ultimately the anti-arthritic effect was proved. Collectively, Gintonin has a great potential as a therapeutic agent for arthritis treatment.

야관문(夜關門)의 항염증효과(抗炎症效果) (Anti-inflammatory effects of Lespedeza Cuneata in vivo and in vitro)

  • Objectives : Lespedeza Cuneata has been used to treat leukorrhea, asthma, stomach pain, diarrhea, acute mastitis, in Korean traditional medicine. According to recent studies, Lespedeza Cuneata has antioxidation, hypoglycemia, cell protective, insulin secretion, whitening, corpora cavernosa smooth muscle relaxation and antimicrobial activities, but it has been rarely conducted to evaluate the immuno-biological activity. The present study was examined to evaluate the anti-inflammatory effects of the Lespedeza Cuneata MeOH extract (LCE) in vivo and in vitro. Methods : In vitro, inflammatory mediators, such as cytokines, nitric oxide and prostaglandin $E_2$ were detected after the addition of LPS with or without LCE in Raw 264.7 macrophage cell line. In vivo, anti-edema effect of LCE was determined in the carrageenan-induced paw edema model in rats. Results : In vitro assay, LCE decreased release of nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) via suppression of iNOS and COX-2 expression. LCE inhibited the phosphorylation of $I{\kappa}B$ indicating the suppression of NF-${\kappa}B$ pathway. In vivo assay, LCE significantly inhibited the formation of paw edema induced by carrageenan injection in rats. LCE effectively inhibited increases of hind paw skin thickness and inflammatory cell infilterations. Conclusion : These findings demonstrate that LCE has inhibitory effect on inflammatory mediators in LPS-activated Raw 264.7 cells and on paw edema in carrageenan-stimulated rats, showing the possibility of anti-inflammatory use of Lespedeza Cuneata.
