  • Objectives Hataedock is the treatment that dispels toxic heat and meconium gathered at the fetus for the new born baby by orally administered herbal extracts. The purpose of this study was to evaluate whether Hataedock alleviate inflammatory skin damages in AD-induced NC/Nga mice through regulating of skin barrier maintain and Th2 differentiation. Methods We established an AD model in the 3-week-old NC/Nga mice through the repeated application of DNFB (dinitrochlorobenzene) on days 28, 35, 42 after Hataedock treatment which was orally administered. We identified the changes of skin barrier and Th2 differentiation through the histological and immunohistochemical changes of protein kinase C (PKC), interleukin (IL)-4, degranulated mast cell, Substance P and MMP-9. Results Our results suggested that Hataedock treatment significantly down-regulated levels of PKC by 82% (p < 0.001), as well as IL-4 by 56% (p < 0.001). Hataedock also suppressed mast cell infiltration, ear edema formation. and Substance P in the tissue of NC/Nga mice were decreased by 57% (p < 0.001), and MMP-9 by 55% (p < 0.001). Conclusions These results suggest that Hataedock alleviates AD through the down-regulation of PKC and Th2 cytokines, which are involved in the initial steps of AD development. Hataedock have potential application for the treatment of AD.

  • In this study, a test for the immunity control effect by ethanolic extract of Eurya emarginata (EE-70E) on NC/Nga mice as the models for atopic dermatitis was conducted with the following results. Atopic dermatitis in NC/Nga mice was induced by repeated application of 1-chloro-2,4-dinitrobenzene (DNCB) for 5 weeks. Mice were orally administered EE-70E or terfenadine, positive control for 3 weeks. Scratching behavior, clinical skin severity, and the levels of IL-4, L-13, IL-17, total serum IgG1, and total serum IgE were measured. The oral administration with EE-70E doses of 200 or 400 mg/kg significantly decreased scratching behavior scores and clinical skin severity score in a dose-dependent manner (p<0.05). The administration of EE-70E at 400 mg/kg significantly decreased cytokines within the blood serum, that is, IL-4, L-13, and IL-17 compared to the control group (p<0.05). The level of blood histamine was statistically significantly decreased. Administration of EE-70E at 400 mg/kg significantly decreased the levels of total serum IgE (p<0.05). The above results indicated that EE-70E was effective in improving the symptoms of atopic dermatitis through various immunity control mechanisms.

  • Objectives : Atopic dermatitis is a chronic inflammatory skin disease which is characterized by severe pruritis, erythema, edema, effusion and scabs. The aim of this study was to understand effects of Arctium lappa Linne, which is well known for its efficacy for various skin diseases, on atopic dermatitis Methods : We conducted this experiment using the DNFB-induced NC/Nga mice. After two weeks of applying DNFB to NC/Nga mice, severe symptoms of atopic dermatitis occurred. We divided the mice into three different groups: a control group which was given no treatment at all, a group treated with dexamethasone (1mg/kg), and another group treated with Arctium lappa Linne (300mg/kg). After one week of treatment, results were recorded according to their improvement on skin, itching behavior, IL-4, and INF-$\gamma$ measurement, which is a significant criterion for diagnosing atopic dermatitis. Results : Itching behavior showed significant improvement in the Arctium lappa Linne group. However Arctium lappa Linne failed to reduce IL-4 and INF-$\gamma$ count. Also there was no satisfying improvement on AD-like skin lesions on the rostral back of the NC/Nga mice treated with Arctium lappa Linne. Conclusions : Arctium lappa Linne showed improvement in itching behavior in NC/Nga mice induced by DNFB. Arctium lappa Linne showed neither significant improvement on skin lesions nor in IL-4 and INF-$\gamma$ measurement.

  • Background: Atopic dermatitis (AD) is a common chronic inflammatory skin disease. To understand AD, there have been many trials establishing AD animal models. Although various trials to establish AD animal models have been existed, even the mechanisms of AD in animal models are not enough clarified. Objectives: This study assessed AD characteristics induced in Nishiki-nezumi Cinnamon/Nagoya (Nc/Nga) mice following trinitrochlorobenzene (TNCB) treatment for different periods and house dust mite (HDM) treatment to compare each model's immunological patterns, especially with cytokine antibody array tool. Methods: In this study, we exposed Nc/Nga mice to TNCB or HDM extract to induce AD. Nc/Nga mice were divided into 4 groups: control, TNCB 2 weeks-treated, TNCB 8 weeks-treated, and HDM-treated groups. After AD induction, all mice were evaluated by serum immunoglobulin E (IgE) concentration and serum cytokine antibody assays, scoring of skin lesions, scoring of scratching frequency, and histological analysis. Results: The results showed significant differences between groups in serum IgE concentration, skin lesion scores, and scratching frequency. The analysis results for serum cytokine antibody arrays showed that in the TNCB 8 weeks- and HDM-treated groups, but not in the TNCB 2 weeks-treated group, expressions of genes related to the immune response were enriched. Among the histological results, the skin lesions in the HDM-treated group were most similar to those of AD. Conclusions: We confirmed that immunological pattern of AD mice was markedly different between HDM and TNCB treated groups. In addition, the immunological pattern was quietly different dependent on TNCB treated duration.

  • Background and Objectives : Atopic dermatitis is a recurrent or chronic eczematous skin disease with severe pruritus, and has increased in Korea. Although the pathogenic mechanisms of atopic dermatitis are yet unknown, recently skin barrier dysfunction and hyperresponsive Th2 cells in the acute phase have been reported as important mechanisms. Cheonggi-san(CGS) is used in oriental clinics for treatingacute skin lesions of eczema or urticaria. There have been no studies on the therapeutic mechanism of CGS for curing atopic dermatitis. We aimed to find out the therapeutic effects of its internaluse on atopic dermatitis-like skin lesions, induced in NC/Nga mice by the mite antigen D. pteronyssinus and disrupting skin barrier. Materials and Methods : The NC/Nga mice were classified into three groups: control group, atopic dermatitis elicitated group(AD), and CGS treated group (CT). Atopic dermatitis-like skin lesions were induced on the back of female NC/Nga mice, 12 weeks of age, by tape stripping, 5% SDS applied to disrupt skin barrier and painting 3 times a week with D. pteronyssinus crude extract solution for 3 weeks. CT was treated with CGS orally after atopic dermatitis was elicitated. We observed changes of skin damage, mast cells, substance P, angiogenesis, skin barrier, Th2 cell differentiation, nuclear factor-${\kappa}B(NF-{\kappa}B)$ p65 activation and COX-2 in NC/Nga mice with atopic dermatitis-like skin lesions. Results : The skin damages as eczema were seenin AD, but mitigated in CT. The degranulated mast cells in dermal papillae increased in AD, but decreased in CT. The substance P positive reacted cells in CT remarkably decreased. The angiogenesis increased in AD, but decreased in CT. The decrease of lipid deposition and ceramide in AD was seen, but anincrease of lipid deposition and ceramide in CT was seen. The distribution of IL-4 positive reacted cells in dermal papillae increased in AD, but decreased in CT. The distribution of NF-${\kappa}B$ p65 positive reacted cells & COX-2 positive reacted cells in CT decreased. Conclusion : The results may suggest that the CGS per os decreases the dysfunction of the skin barrier, inhibits Th2 cell differentiation and inhibits NF-${\kappa}B$ p65 activation in NC/Nga mice with atopic dermatitis-like skin lesions.

  • Gromwell (LE, Lithospermum erythrorhizon), a perennial herbal plant, has been used for the treatment of various problems associated with atopic dermatitis of the skin, such as water loss, epidermal hyperproliferation, and severe inflammation. Previously, it was shown that oral supplementation with a 70% ethanol extract of gromwell prevented the development of atopic dermatitis in NC/Nga mice. In this study, in order to identify the fraction that mediates gromwell's efficacy, the dietary effects of water and ethyl acetate fractions from the ethanol extract of gromwell were assessed in the development of dermatitis using NC/Nga mice. Dietary supplementation of the hot water fraction significantly reduced scores for epidermal hyperproliferation in parallel with a marked increase of ceramides. Supplementation of the gromwell hot water fraction also decreased scratching behavior, which was accompanied by a decrease in plasma levels of IgE. These results showed that the hot water fraction of the gromwell ethanolic extract prevented the development of atopic dermatitis by increasing ceramides in NC/Nga mice.

  • This research was performed to examine an anti-inflammatory effects of Gamisangryosamul-Tang(GSS) and anti-pruritus effects of Ziyang-Go(Salve). This study was processed by three experiments; Experiment 1: Inhibitory activity of GSS extract on the degranulation of mast cell and histamine release in plasma induced by compound 48/80 i.p, injection after the pretreatment of GSS extract i.p, injection in Sprague-Dawley rats, Experiment 2: Anti-inflammatory effect of GSS extract on macropharge raw 264,7 cells treated by LPS 250 ppm (before 2 hours). Experiment 3: Measurement of passive cutaneous anaphylaxis and atopic dermatitis using NC/Nga mice, GSS extract inhibited histamine release by 70% compared to compound 48/80 treated control group and histologically significantly reduced (P<0,01) the degranulation of mast cell in SD rats. In GSS extract treated group, the expression of TNF-α in macropharge cell showed the remarkable inhibitory effect about 62% (P<0,01) compared to LPS treated control group. The expression of IL-6 appeared more effective by 46% than the LPS treated control group and by 6% compared to hydrocortison treated group, Comparing with steroid (0.05% prednisolon) ointment, Ziyan-Go treated group showed the significant(30%) recovery on skin response index in atopic dermatis like anaphylaxis mice(NC/Nga), Finally, in scratching behavioral tests of NC/Nga mice for three weeks, Ziyang-Go treated group significantly (P<0.05) suppressed the pruritus on the face, neck, ears and dorsal skin than inbred NC/Nga mice. However, the change of IgE and IFN-γ from the spleen cell of NC/Nga mice was not significantly different between the oral intake of GSS extract group and of saline intaked control group. Summary and Conclusion: This study demons trates that Ziyang-go have the equal anti-pruritus effect to steroid ointment and GSS extract have the notable immunologic activity on inflammatory in vivo and in vitro model. Advanced experiment of this study will be required for more reliable information about the correlation between the lymphokine (i.e. IgE) and the anti-allergic effects of GSS.
