  • Crystal structure of bis(1,2-diaminopropane)palladium(II) bis(oxalato)palladate(II) has been determined by X-ray crystallography. Crystal data: $Pd_2C_{10}H_{10}N_{4}O_{8}$, $M_W$ = 573.09, orthorhombic, space group $P_{ccn}$ (No = 56), a = 16.178(5), b = 16.381(6), c = 6.685(2)$\{AA}$, V = 1771.6 $\{AA}^3$, $M_W$W = 573.09, $D_c$ = 2.014 g${\cdot}c\;m^{-3}$, Z = 4, T = 294K, F(000) = 1056.0 and $\mu$ = 20.466 c$m^{-1}$. The intensity data were collected with $Mo-K\alpha$ radiation (${\lambda}$ = 0.7107 $\AA)$ on an automatic four-circle diffractometer with a graphite monochromater. The structure was solved by Patterson method and refined by full matrix least-squares methods using Pivot weights. The final R and S values were R = 0.065, $R_W = 0.059, R_{all}$ = 0.065 and S = 4.315 for 605 observed reflections. Both cation and anion complexes are essentially planar and have dihedral angle of $18(l)^{\circ}$ between thier planes. In the crystal structure, they do not have the Magnus's salt type mixed stacks; instead, the complex anions form regular stacks along the c-axis with the M-M bond length of $3.343(5)\AA$ and their stacks are surrounded by the complex cations through hydrogen bonds with the nitrogen-oxygen distances of 2.94(3) and $3.31(4)\AA.$

Estimation of Attenuation Relationship Compatible with Damping Ratio of Rock Mass from Numerical Simulation (수치해석을 통한 진동감쇠식 맞춤형 암반의 감쇠비 산정)

  • The stability of the adjcent structures or slopes under blasting is typically evaluated using an empirical vibration attenuation curve or dynamic numerical analysis. To perform a dynamic analysis, it is necessary to determine the blast load and the damping ratio of rock mass. Various empirical methods have been proposed for the blast load. However, a study on representative values of damping ratio of a rock mass has not yet been performed. Therefore, the damping ratio was either ignored or selected without a clear basis in performing a blast analysis. Selection of the dampring ratio for the rock mass is very difficult because the vibration propagation is influenced by the layout and properties of the rock joints. Besides, the vibration induced by blasting is propagated spherically, whereas plane waves are generated by an earthquake. Since the geometrical spreading causes additional attenuation, the damping ratio should be adjusted in the case of a 2D plane strain analysis. In this study, we proposed equivalent damping ratios for use in continuum 2D plane strain analyses. To this end, we performed 2D dynamic analyses for a wide range of rock stiffness and investigated the characteristics of blast vibration propagation. Based on numerical simulations, a correlation between the attenuation equation, shear wave velocity, and equivalent damping ratio of rock mass is presented. This novel approach is the first attempt to select the damping ratio from an attenuation relationship. The proposed chart is easy to be used and can be applied in practice.

Induction of Apoptosis by Samgibopae-tang in Human Non-small-cell Lung Cancer Cells (인체폐암세포 NCI-H460 및 A549의 증식에 미치는 삼기보폐탕의 영향 비교)

  • In the present study, we investigated the antiproliferative activity of the water extract of Samgibopae-tang (SGBPT) in NCI-H460 and A549 non-small-cell lung cancer cell lines. We found that exposure of A549 cells to SGBPT resulted in the growth inhibition in a dose-dependent manner as measured by MTT assay, however SGBPT did not affect the growth of NCI-H460 cells. The antiproliferative effect by SGBPT treatment in A549 cells was associated with morphological changes such as membrane shrinking and cell rounding up. SGBPT treatment did not induce the cell cycle arrest in both cell lines, however the frequency of sub-G1 population was concentration-dependently increased by SGBPT treatment in A549 cells. SGBPT treatment partially induced the expression of tumor suppressor p53 in A549 cells and the expression of cyclin-dependent kinase inhibitor p21(WAF1/CIP1) was markedly increased in both transcriptional and translational levels in A549 cells. The up-regulation of p21 by SGBPT occurred in a similar a concentration dependent manner to that observed with the inhibition of cell viability and induction of sub-G1 population of the cell cycle. However SGBPT treatment did not affect other growth regulation-related genes such as early growth response-1 (Egr-1), nonsteroidal anti-inflammatory drug-activated gene-1 (NAG-1), inducible nitric oxide synthease (iNOS), cyclooxygenases (COXs), telomere-regulatory factors in A549 as well as NCI-H460 cells. Taken together, these findings suggested that SGBPT-induced inhibition of human lung carcinoma A549 cell growth was aoosciated with the induction of p21 and the results provided important new insights into the possible molecular mechanisms of the anti-cancer activity of SGBPT.

Bioleaching of Heavy Metals from Shooting Range Soil Using a Sulfur-Oxidizing Bacteria Acidithiobacillus thiooxidans (황산화균 Acidithiobacillus thiooxidans를 이용한 사격장 토양 내 중금속 용출)

  • Applicability of bioleaching techniques using a sulfur-oxidizing bacteria, Acidithiobacillus thiooxidans, for remediation of shooting range soil contaminated with toxic heavy metals was investigated. The effects of sulfur concentration, the amount of bacterial inoculum and operation temperature on the efficiency of heavy metal solubilization were examined as well. As sulfur concentration and the amount of bacterial inoculum increased, the solubilization efficiency slightly increased; however, significant decrease of heavy metal extraction was observed with no addition of sulfur or bacterial inoculum. Bacteria solubilized the higher amount of heavy metals at $26^{\circ}C$ than $4^{\circ}C$. Lead showed the highest removal amount from the contaminated soil but the lowest removal efficiency when compared with Zn, Cu and Cr. It was likely due to formation of insoluble $PbSO_{4(s)}$ as precipitate or colloidal suspension. Sequential extraction of the microbially treated soil revealed that the proportion of readily extractable phases of Zn, Cu and Cr increased by bacterial leaching, and thus additional treatment or optimization of operation conditions such as leaching time were required for safe reuse of the soil. Bioleaching appeared to be a useful strategy for remediation of shooting range soil contaminated with heavy metals, and various operating conditions including concentration of sulfur input, inoculum volume of bacteria, and operation temperature exerted significant influence on bioleaching efficiency.

The Characteristic of Te Recovery in Gold Concentrate Using Electrolysis (전기분해법을 이용한 금정광내 Te 회수 특성)

  • In order to obtain pure metallic Te from gold concentrate, roasting treatment, hypochlorite leaching, Fe removal and electrolysis experiments were carried out. The contents of Au, Ag and Te from the concentrate sample and roasted sample were much more soluble in the hypochlorite solution than in aqua regia digestion, whereas the metals Pb, Zn, Fe and Cu were easier to leach with the aqua regia than the hypochlorite. With the addition of NaOH in the hypochlorite leaching solution prior to electrolysis, the Fe removal rate achieved was only 96% in the concentrate sample, while it reached 98% in the roasted sample. The results of electrolysis for 240 min, 98% of the metallic copper was recovered from the concentrate sample, while 99% was obtained from the roasted sample due to the removal of S by roasting. The amount of anode slime was also greater in the electrolytic solution with the roasted sample than with the concentrate sample. The results on the anode slime after the magnetic separation process showed the amount of metallic pure native tellurium recovered was greater in the roasted sample than in the concentrate sample.

Variation of Nephrotoxicity Biomarkers by Urinary Storage Condition in Rats

  • Recently, there has been an increase in the use of several nephrotoxicity biomarkers in preclinical experiments. In addition, it has been indicated that the result may have been influenced by secondary factors, such as sample storage condition or storage period. In this study, we have assessed the variation in urinary nephrotoxicity biomarkers as a result of urine storage conditions and storage period of the urine. Urine was sampled from specific pathogen-free Sprague-Dawley rats (19 weeks old), which were housed individually in hanged stainless steel wire mesh cages. Urine was stored at $20^{\circ}C$, at $4^{\circ}C$, or at $-70^{\circ}C$ after sampling. The levels of the biomarkers such as beta-2 microglobulin (B2M), cystatin-C (Cys-C), N-acetyl-${\beta}$-D-glucosaminidase (NAG), micro albumin (MA), micro protein (MP) were measured at 6, 24, 48 and 144 hr after sampling. The B2M level was significantly decreased at 6, 24, 48, and 144 hr compared to 0 hr at $-70^{\circ}C$ (p < 0.05, p < 0.01, p < 0.05, and p < 0.05, respectively) and 24 and 144 hr at $20^{\circ}C$ (p < 0.01, p < 0.01, respectively). The Cys-C level was significantly decreased at 144 hr compared to 0 hr at $4^{\circ}C$ (p < 0.01), at $20^{\circ}C$ (p < 0.05) and at $70^{\circ}C$ (p < 0.01). MP and MA levels were not different for 144 hr in all storage conditions. Taken together, B2M and Cys-C levels were modulated by storage temperature and period. For the enhancement of test accuracy, it is suggested that strict protocols be established for samples to minimize the effects of the storage conditions on the detected levels of biomarkers.

A Comparative Study between Microbial Fermentation and Non-Fermentation on Biological Activities of Medicinal Plants, with Emphasis on Enteric Methane Reduction (천연 약용식물의 미생물 발효를 통한 장내 메탄 생성 억제 효과 비교 연구)

  • A study was conducted to improve the biological activity of two medicinal plants, Eucommia ulmoides Oliv. and Glycyrrhiza uralensis, by fermentation. The biological activity was assessed by determining antibacterial, antioxidant and antimethanogenic properties. Fermentation was achieved by adding the plant materials in MRS broth at 10% (w/v) and different starter cultures at 1% (v/v). Condition for fermentation were incubation temperature of $30^{\circ}C$ and agitation at 150 rpm for 48 h. Six starter cultures, Weissella confusa NJ28 (Genbank accession number KJ914897), Weissella cibaria NJ33 (Genbank accession number KJ914898), Lactobacillus curvatus NJ40 (Genbank accession number KJ914899), Lactobacillus brevis NJ42 (Genbank accession number KJ914900), Lactobacillus plantarum NJ45 (Genbank accession number KJ914901) and Lactobacillus sakei NJ48 (Genbank accession number KJ914902) were used. Antibacterial activity was observed in L. curvatus NJ40 and L. plantarum NJ45 only as opposed to other treatments, including the non-fermented groups, which showed no antibacterial activity. Both plants showed antioxidant activity, although E. ulmoides Oliv. had lower activity than G. uralensis. However, fermentation by all strains significantly improved (p<0.05), antioxidant activity in both plants compared to non-fermented treatment. Six treatments were based on antibacterial activity results, selected for in vitro rumen fermentation; 1) non-fermented E. ulmoides, 2) fermented E. ulmoides NJ40, 3) fermented E. ulmoides NJ45, 4) non-fermented G. uralensis, 5) fermented G. uralensis NJ40, 6) fermented G. uralensis NJ45. A negative control was also added, making a total of 7 treatments for the in vitro experiment. Medicinal plant-based treatments significantly improved (p<0.05) total volatile fatty acid (VFA) concentration. Significant methane reduction per mol of VFA were observed in G. uralensis (p<0.05). Based on the present study, fermentation improves the biological activity of E. ulmoides Oliv. and G. uralensis. Fermented G. uralensis could also be applied as an enteric methane mitigating agent in ruminant animals.

Development of an Environmental Friend Additive Using Antibacterial Natural Product for Reducing Enteric Rumen Methane Emission (항균활성 천연물질을 이용한 반추위 메탄저감용 친환경 첨가제 개발)

  • The present study was conducted to investigate effective starter culture to improve biological activity of Asarum sieboldii. Antibacterial activity, antioxidant activity and reduction of enteric rumen methane production were used as criterions for biological activity. Ground A. sieboldii was added in MRS broth at 10% (w/v) and fermented by different starter cultures. Weissella confusa NJ28, Weissella cibaria NJ33, Lactobacillus curvatus NJ40, Lactobacillus brevis NJ42, Lactobacillus plantarum NJ45 and Lactobacillus sakei NJ48 were used for starter culture strains. Each starter culture was inoculated with 1% (v/v) ratio and fermentation was performed at $30^{\circ}C$ with agitation (150 rpm) for 48 h. MRS broth for the control was employed without starter culture. Then the fermentation growth was dried and extracted using ethyl alcohol. The growth of starter culture was detected at NJ40, NJ42, NJ45 and NJ48. And the highest cell growth was found in NJ40. Antibacterial activity against to Staphylococcus aureus, Listeria monocytogens, Mannheimia haemolytica and Salmonella gallinarum were observed in the extract fermented by NJ40 and NJ45. All treatments showed antioxidant activities, however, there were no significant differences (p>0.05). In in vitro rumen fermentation, negative control (NC) and positive control (PC) were assigned to without extract and with non-fermented A. sieboldii extract. Significant suppression of gas productions were detected in positive control and treatments compared to negative control (p<0.05). However, total volatile fatty acid production was not suppressed. Significant methane reduction per total volatile fatty acid productions were found in positive control and NJ45 treatment (p<0.05). The present study suggested a fermentation of A. sieboldii using NJ45 strain could improve its biological activity and make possible for its use in bio additive for enteric rumen methane mitigation without suppression of animal productivity.

Development of Polyclonal Antibodies to Abdominal and Subcutaneous Adipocyte for Reducing Body Fat of Rat (흰쥐 체지방 감소를 위한 지방부위별 특이 다클론 항체 개발 연구)

  • Polyclonal antisera against regional (abdominal and subcutaneous) fats were developed to reduce body fat in rats. Isolation and culture of abdominal and subcutaneous adipocytes of rats were performed for analyzing lactate dehydrogenase (LDH) concentration. At the level of 1:1,000 dilution, little antibody reactivity appeared in non-immunized serum whereas both of antisera against abdominal (AAb) and subcutaneous adipocyte plasma membrane proteins (SAb) had relatively strong reactivity till the level of 1:128,000 dilutions. Compared with regional fats, extremely low reactivities of AAb and SAb were detected with PMP of the organs (p<0.001). Both AAb and SAb were most strongly reacted with each adipocyte plasma memebrane proteins and showed statistically (p<0.01) higher cross-reactivities compared with non-immunized serum based on LDH analysis. In conclusion, these results may indicate that the present polyclonal antibodies against regional inedible adipocyte plasma membrane proteins are well developed and have safety in cross-reactivities with body organs.

A New Rice Variety Developed from an Interspecific Cross, "Hwaweon 2" (종간교잡 유래 중생 다수성 벼 품종 "화원 2호")

  • "Hwaweon 2" was developed from an interspecific cross between Hwaseongbyeo and a wild species, Oryza rufipogon L. (IRGC 105491) based on marker-aided selection. The recurrent parent "Hwaseongbyeo" is a high grain quality cultivar with medium-maturity. Hwaweon 2 is nearly isogenic to Hwaseongbyeo except a small O. rufipogon introgression on chromosome 9. This segment was associated with genes controlling a number of traits including grain weight, heading date, culm length, and spikelets per panicle. The preliminary and replicated yield trial was conducted at Chungnam National University in 2004, 2005 and 2006. The local adaptability test was carried out by the National Seed Management Office (NSMO) in 2007 and 2008. This cultivar was registered to NSMO with a cultivar designated as "Hwaweon 2". This cultivar averaged 98cm in culm length and has a medium to late growth duration. This variety is resistant to stripe virus as the recurrent parent. Milled rice of "Hwaweon 2" is translucent and the grain quality traits are comparable to those of the recurrent parent. The yield potential of "Hwaweon 2" in grain is about 7.68 MT/ha at the ordinary fertilizer level about 14% higher than that of Hwaseongbyeo due to increase in grain weight and spikelets per panicle.
