Simulations of Thermal Stratification of Daecheong Reservoir using Three-dimensional ELCOM Model (3차원 ELCOM 모형을 이용한 대청호 수온성층 모의)

  • The transport of contaminants and spatial variation in a deep reservoir are certainly governed by the thermal structure of the reservoir. There has been continuous efforts to utilize three-dimensional (3D) hydrodynamic and water quality models for supporting reservoir management, but the efforts to validate the models performance using extensive field data were rare. The study was aimed to evaluate a 3D hydrodynamic model, ELCOM, in Daecheong Reservoir for simulating heat fluxes and stratification processes under hydrological years of 2001, 2006, 2008, and to assess the impact of internal wave on the reservoir mixing. The model showed satisfactory performance in simulating the water temperature profiles: the absolute mean errors at R3 (Hoenam) and R4 (Dam) sites were in the range of $1.38{\sim}1.682^{\circ}C$. The evaporative and sensible heat losses through the reservoir surface were maximum during August and January, respectively. The net heat flux ($H_n$) was positive from February to September, while the stratification formed from May and continued until September. Instant vertical mixing was observed in the reservoir during strong wind events at R4, and the model reasonably reproduced the mixing events. A digital low-pass filter and zero crossing method was used to evaluate the potential impact of wind-driven internal wave on the reservoir mixing. The results indicated that most of the wind events occurred in 2001, 2006, 2008 were not enough to develop persistent internal wave and effective mixing in the reservoir. ELCOM is a suitable 3D model for supporting water quality management of the deep and stratified reservoirs.

Derivation of Embryonic Germ Cells from Post Migratory Primordial Germ Cells, and Methylation Analysis of Their Imprinted Genes by Bisulfite Genomic Sequencing

  • The embryonic germ cell (EGCs) of mice is a kind of pluripotent stem cell that can be generated from pre- and post-migratory primordial germ cells (PGCs). Most previous studies on DNA methylation of EGCs were restricted to 12.5 days post coitum (dpc). This study was designed to establish and characterize murine EGC lines from migrated PGCs as late as 13.5 dpc and to estimate the degrees of methylation of their imprinted genes as well as of the non-imprinted locus, Oct4, using an accurate and quantitative method of measurement. We established five independent EGC lines from post migratory PGCs of 11.5-13.5 dpc from C57BL/6 ${\times}$ DBA/2 F1 hybrid mouse fetuses. All the EGCs exhibited the typical features of pluripotent cells including hypomethylation of the Oct4 regulatory region. We examined the methylation status of three imprinted genes; Igf2, Igf2r and H19 in the five EGC lines using bisulfite genomic sequencing analysis. Igf2r was almost unmethylated in all the EGC lines irrespective of the their sex and stage of isolation; Igf2 and H19 were more methylated than Igf2r, especially in male EGCs. Moreover, EGCs derived at 13.5 dpc exhibited higher levels of DNA methylation than those from earlier stages. These results suggest that in vitro derived EGCs acquire different epigenotypes from their parental in vivo migratory PGCs, and that sex-specific de novo methylation occurs in the Igf2 and H19 genes of EGCs.

Formation of Liquid Crystal Gel with Hydrogenated Lecithin and Its Effectiveness

  • This study described about method that form liquid crystal gel (LCG) by main ingredient with hydrogenated lechin (HL) in OW emulsion system. Result of stability test is as following with most suitable LCG's composition. Composition of LCG is as following. To form liquid crystal, an emulsifier used $4.0\;wt\%$ of cetostearyl alcohol (CA) by $4.0\;wt\%$ of HL as a booster, Moisturizers contained $2\;wt\%$ of glycerin and $3.0\;wt\%$ of 1.3-butylene glycol (1,3-BG). Suitable emollients used $3.0\;wt\%$ of cyclomethicone, $3.0\;wt\%$ of isononyl isononanoate (ININ), $3.0\;wt\%$ of cerpric/carprylic triglycerides (CCTG), $3.0\;wt\%$ of macademia nut oil (MNO) in liquid crystal gel formation. On optimum conditions of LCG formation, the pHs were formed all well under acidity or alkalinity conditions. Considering safety of skin, PH was the most suitable $\pm61.0$ ranges. The stable hardness of LCG formation appeared best in $32\;dyne/cm^2.$ Particle of LCG is forming size of $1{\~}20\;{\mu}m$ um range, and confirmed that the most excellent LCG is formed in $1{\~}6\;{\mu}m$ range. According to result that observe shape of LCG with optical or polarization microscope, LCG could was formed, and confirmed that is forming multi-layer lamellar type structure around the LCG. Moisturizing effect measured clinical test about 20 volunteers. As a result, moisturizing effect of LCG compares to placebo cream was increased $30.6\%$. This could predicted that polyol group is appeared the actual state because is adsorbed much to round liquid crystal droplets to multi-lamellar layer's hydrophilic group. It could predicted that polyol group is vast quantity present phase that appear mixed because is adsorbed to round liquid crystal to multi-lamellar layer's hydrophilic group. This LCG formation theory may contribute greatly in cosmetics and pharmacy industry development.

Association of Plasma Osteoprotegerin with Adiponectin and Difference according to Obesity in Men with Metabolic Syndrome (대사증후군 남성에서의 혈중 Osteoprotegerin의 아디포넥틴과의 상관성과 비만도에 따른 차이)

  • Osteoprotegerin (OPG) plays a core role in bone reformation by antagonizing the effect of receptor activator of nuclear factor ${\kappa}$-B ligand (RANKL), and mediates vascular calcification in cardiovascular disease patients. Thus, we aimed to examine the relationship between serum OPG levels and cardiovascular factors and inflammatory markers in metabolic syndrome patients (MS). This cross-sectional study included 96 men who visited the diet clinic between May and July 2011. Patients were classified into 2 groups based on NCEP-ATP guidelines: normal and with MS (n = 50 and 46, respectively). Physical measurements, biochemical assay were measured. Serum OPG and IL-6, diponectin and hs-CRP were assessed. MS were aged $50.02{\pm}10.85$ years, and normal patients $52.07{\pm}9.56$ years, with no significant differences. Significant differences were not observed in BMI between the 2 groups. Moreover, significant differences were not observed in serum OPG, however, the serum OPG level ($4.41{\pm}1.86pmol/L$) differed significantly between an overweight MS (BMI > 25) and normal patients. OPG was correlated to age (r = 0.410, p = 0.000), HDL-cholesterol (r = 0.209, p = 0.015), and log adiponectin (r = 0.175, p = 0.042). Multiple regression analyses using the enter method showed that age (${\beta}$ = 0.412, p = 0.000) and BMI (${\beta}$ = 0.265, p = 0.000) considerably affected OPG. In conclusion, out study showed that serum OPG levels are correlated with cardiovascular risk factors, such as BMI, HDL-cholesterol and adiponectin in MS and adiponectin, suggesting that serum OPG has potential as a cardiovascular disease indicator and predictor.

Bioassay Study on fresh Water Fish with PCP and DDT (PCP와 DDT의 독성이 담수어에 미치는 영향)

  • Four-day bioassay method was used in this study to find out toxicity levels of DDT and PCP. From this study it was found that M. chrysophekadion was the most sensitive to DDT with a 96 h-TL sub(m) value of 0.0044 mg/l followed by P. Sutchi with a 96 h-TL sub(m) value of 0.0056 mg/l, and the most resistant was C. Siamensis with a 96 h-TL sub(m) value of 0.0133 mg/l. In the case of PCP, it was also found that M. Chrysophekadion and P. Sutchi were the first and second most sensitive to PCP with a 96 h-TL sub(m) value of 0.065mg/l PCP and 0.125mg/l PCP respectively, as in the case of DDT study. T. Nilotica was found to be the most resistant to PCP with a 96 h-TL sub(m) value of 0.264mg/l PCP. The resistant power of fish to pollution varies with the species of fish and the types of pollutants. In order of decreasing sensitivity of fish to DDT based on TL sub(m), the following sequence is obtained M. Chrysophekadion, P. Sutchi, T Nilotica, P. Gonionotus, K. Bicirrhis, L. Bicolor, C. Carpio, R. Heteromorpha, C. Siamensis while in the case of PCP, M. Chrysophekadion, P. sutchi, L. Bicolor, K. Bicirrhis, P. Gonionotus (R. Heteromorpha, R. Trilineata), C. Siamensis, C. Carpio, T. Nilotica.

Isolation and Identification of Active Antimicrobial Substance against Listeria monocytogenes from Ruta graveolens Linne (운향으로부터 Listeria monocytogenes에 대한 항균 활성 물질의 분리 및 구조동정)

  • Ethanol extracts from Ruta graveolens Linne exhibited strong antimicrobial activities by disc diffusion method against 5 strains of Listeria monocytogenes (ATCC 19111, ATCC 19112, ATCC 19113, ATCC 19114 and ATCC 15313). Ethanol extract from Ruta graveolens Linne was subsequently fractionated by n-hexane, chloroform, ethyl acetate and water. Chloroform fraction of Ruta graveolens Linne showed strong growth inhibition at concentrations as low as 40 ppm level in broth culture medium against 5 strains of L. monocytogenes for 72 hr at $30^{\circ}C$. Single substance(RTG1-1) was isolated by silica gel column chromatography from chloroform fraction of Ruta graveolens Linne. RTG1-1 showed a strong bactericidal activity against L. monocytogenes at a concentration of 20 ppm level. Purified RTG1-1 was identified as gravacridonechlorine by analyses of EI-Mass, $^1H-NMR$ and $^{13}C-NMR$.

Analysis of CTOD Tests on Steels for Liquefied Hydrogen Storage Systems Using Hydrogen Charging Apparatus (수소 장입 장치를 활용한 액체수소 저장시스템 강재의 CTOD 시험 분석)

  • Hydrogen infiltration into metals has been reported to induce alterations in their mechanical properties under load. In this study, we conducted CTOD (Crack Tip Opening Displacement) tests on steel specimens designed for use in liquid hydrogen storage systems. Electrochemical hydrogen charging was performed using both FCC series austenitic stainless steel and BCC series structural steel specimens, while CTOD testing was carried out using a 500kN-class material testing machine. Results indicate a notable divergence in behavior: SS400 test samples exhibited a higher susceptibility to failure compared to austenitic stainless steel counterparts, whereas SUS 316L test samples displayed minimal changes in displacement and maximum load due to hydrogen charging. However, SEM (Scanning Electron Microscopy) analysis results presented challenges in clearly explaining the mechanical degradation phenomenon in the tested materials. This study's resultant database holds significant promise for enhancing the safety design of liquid hydrogen storage systems, providing invaluable insights into the performance of various steel alloys under the influence of hydrogen embrittlement.

  • X-ray Fluorescence Spectrographic method has been applied for the rapid determination of main components, such as $SiO_2$, $Al_2O_3$, $Fe_2O_3$, CaO, MgO and $K_2O$ in Silicate Minerals. In this method, Boric Acid was used as a binder after fusion with Lithium Tetraborate in the briquet-making process. The Lithium Flubride, Ammonium di-Hydrogen Phosphate and Ethylene Diamine d-Tartrate crystals were used with Scintillation counter and Gas Flow counter as the detectors. Several influences on this method were discussed, including the particle size of samples and reducing of the matrix effects by dilution with Boric Acid and addition of Lanthanum Oxide with the diluent. In order to test the reproducibility of this method described above, the determination of the same kind of samples were carried out repeatedly, and the results obtained were presented in the table. Calibration curves for each element were presented, and the application of the method was tested with International Rock Standard T-Ⅰ. All the results obtained by X-Ray Fluorescence Spectrographic method were compared with the results by conventional chemical method.

A Study of the Effect of a Mixture of Hyaluronic Acid and Sodium Carboxymethyl Cellulose ($Guardix-sol^{(R)}$) on the Prevention of Pericardial Adhesion (Hyaluronic Acid와 Sodium Carboxymethyl Cellulose 혼합용액($Guardix-sol^{(R)}$)의 섬유막유착 방지 효과에 관한 연구)

  • Background: This study was designed to evaluate the efficacy of a mixture of hyaluronic acid and sodium carboxymethyl cellulose ($Guardix-sol^{(R)}$) on experimental pericardial adhesion. Material and Method: Thirty rats were divided into 2 groups of 15 rats each and pericardial mesothelial injury was induced during surgery by abrasion. In the control group, blood and normal saline were administered into pericardium; in the test group, blood and HA-CMC solution were administered. Pericardial adhesions were evaluated at 2 weeks (n=5), 4 weeks (n=5), and 6 weeks (n=5) after surgery. The severity of adhesions was graded by macroscopic examination, and the adhesion tissue thickness was analyzed microscopically with Masson trichrome stain and an image processing program. Result: The test group had significantly lower macroscopic adhesion scores ($2.9{\pm}0.6$ : $3.9{\pm}0.4$, p<0.000) compared with the control group. For microscopic adhesion tissue thickness, the test group had lower scores compared with the control group, but this difference was not statistically significant ($91.73{\pm}49.91$ : $117.67{\pm}46.4$, p=0.106). Conclusion: We conclude that an HA-CMC solution ($Guardix-sol^{(R)}$) reduces the formation of pericardial adhesions in this animal model.

Effect of an Individually Tailored Program Based on Self-Measurement of Blood Glucose on Health Behavior and HbA1c in Diabetes and Pre-diabetes Patients (자가혈당측정 기반의 개별 맞춤형 프로그램이 당뇨병 및 당뇨병 전단계 환자의 건강행태와 당화혈색소에 미치는 영향)

  • Objective: This study was to evaluate the effectiveness of an individually tailored program based on self-measurement of blood glucose on health behavior and HbA1c in diabetes and pre-diabetes patients. Methods: The program consisted of seven sessions for 12 weeks which were carried out every two weeks. Almost all sessions were progressed on untact method except for the first and last session. The 71 subjects were assessed for their knowledge of diabetes, health behavior, the experience of self-measurement of blood glucose, body mass Index (BMI) and hemoglobin A1c (HbA1c) at before and after the program. They were also evaluated on their degree of utilization of blood glucose measurements after the program. Results: Each mean score on their knowledge of diabetes, health behavior and the experience of self-measurement of blood glucose was significantly increased from 14.77, 25.50, and 2.70 to 15.41, 28.40, and 4.81, respectively. Each mean score on both BMI and HbA1c (n=53) was significantly decreased from 24.47kg/m2 and 7.27% to 24.01kg/m2 and 6.67%, respectively. The post-HbA1c had a significant negative correlation(r=-0.415) with the degree of utilization of blood glucose measurements. The degree of utilization of blood glucose measurements had a significant positive correlation(r=0.581) with post-health behavior. Conclusions: The program shows effectiveness in improving HbA1c in Type 2 diabetes and pre-diabetes patients. The post-HbA1c might be related to the degree of utilization of blood glucose measurements which might be related to the health behavior.
