Unusual Mitochondrial DNA Polymorphism of the Blue Mussel (Mytilus edulis) Species Complex on the Southern Coast of Korea

  • Mitochondrial DNA (mtDNA) from 54 specimens of the blue mussel (Mytilus edulis) species complex sampled from the southern coast of Korea was assayed for polymorphism with a portion of the COIII gene (336 bp). Fifteen haplotypes were found. PAUP, one-step networks, and PHYLIP analyses revealed the presence of two clearly differentiated mitochondrial clades (termed clades B and E), separated by 3.6% of minimum sequence divergence. The distribution pattern of the species appears to be consistent with category II of the phylogeographic pattern sensu (Avise et al., 1987): the presence of two discontinuous and distinct mtDNA genotypes in the same geographic region. This unusual mitochondrial polymorphism was explained by the presence of the Mediterranean species, M. galloprovincialis, possessing mtDNA of both M. galloprovincialis and M. edulis.

A Taxonomical Study on the Shell Morphology of Blue Mussel, Mytilus edulis galloprovincialis Lamarck in Korea and Japan (한국산 및 일본산 진주담치의 패각형태에서 본 분류학적 고찰)

  • The internal shell morphology of the blue mussel, which were collected from Korea and Japan were studied. The range of the mean ratio between anterior adductor muscle scars to shell height in each locality were between 62.47 in Yongdok to 54.17 in Uichang. These values were very similar to that of M galloprouincialis in Mediterranean than that of M edulis in Europe. And the range of the mean ratio between hinge plate length to shell height were between 61.31 in Jukbyon to 56.15 in Otuschi. Also these values were similar to that of M. galloprovincialis in Mediterranean. Based on the mean ratio between anterior adductor muscle scars and hinge plate length to shell height, it was suggested that the Korean and Japanese blue mussel is certainly identical to the Mediterranean species, Mytilus galloprovincialis.

  • We studied the attachment-repelling activity of flavonoids, stilbenes, and their glycosides against the blue mussel Mytilus edulis galloprouincialis. Molecular mechanics calculations have shown that the most stable conformations of potent repellents 29 and 32 were the same. On the basis of these results, structure-activity relationships of these compounds are discussed.

  • Attempts were made to analyze the toxin composition of the toxic mussel Mytilus edulis galloprovincialis which were collected from aquaculture pond in Apr. 1988 in Hachung, Koje, southern Korea. The toxins were partially purified from the ethanolic extract of the mussel digestive glands by activated charcoal and Bio Gel P-2 column chromatography. HPLC analysis demonstrated that the toxin consisted mainly of gonyautoxin 1-4 (GTX 1-4), along with trace amounts of saxitoxin (STX) and protogonyautoxin 1-2 (PX 1-2).

  • Mitochondrial DNA (mtDNA) polymorphism of the blue mussel (Mytilus edulis) species complex sampled from the east coast of Korean was studied using a partial sequence of COIII gene (336 bp). Samples obtained from three localities on the east coast of Korea revealed four haplotypes with two clearly differentiated mitochondrial clades (termed clades B and E), separated by 4.2% of minimum sequence divergence. This pattern indicates no difference between east and south coasts of Korea. According to population genetic theory on evolutionary characteristics of mtDNA, we concluded that mtDNA introgression from M. edulis to M. gallprovincialis might be a source for mtDNA polymorphism found in mussels on the east coast of Korea.

  • 홍합감시(Mussel Watch) 연구의 일환으로 1988년과 1989년에 남한 연안의 24곳에서 홍합(Mytilus)속에 속하는 종들을 채집하였다. 채집된 표본들의 패각의 형태 특성과 분포를 조사 분석한 결과 3종-홍합, 진주담치(M. edulis galloprovincialis), Mytilus sp. 로 밝혀졌다. 이 3종은 패각의 형태에서 뚜렷한 차이를 보이고 있으며, 지리분포와 서식지 수직분포에서도 나름대로의 특성을 보여주고 있다. 지리분포에서는 수온이 수직분포에서는 종간 경쟁이 분포의 제한 요인인 것 같다.

Paralytic Shellfish Poisoning Toxin Accumulation in Four Mussel Species Fed on Toxic Alexandrium tamarense (독성 Alexandrium tamarense를 섭취한 담치류 4종의 마비성패독 축적)

  • Cultured cells of the toxic Alexandrium tamarense were fed to four mussel species, Mytilus coruscus, M. edulis, M. galloprovincialis and Septifer vulgatus, to examine the interspecies and interlocality differences in the ability to accumulate paralytic shellfish poisoning (PSP) toxins. Toxin content of A. tamarense cells varied during culture period. In contrast, toxin composition in the cell (C1,2, GTX1-4 and neoSTX) was constantly stable. In feeding experiment, the four mussel species collected from Geoje intoxicated after uptake of A. tamarense. Toxin content ($average{\pm}SD\;{\mu}g$ STXeq/100 g) of M. coruscus, M. edulis, M. galloprovincialis and Septifer vulgatus were $1,660{\pm}79,\;3,914{\pm}2,242,\;5,626{\pm}1,620\;and\;958{\pm}163$, respectively. Toxin profiles included C1,2, GTX1,4 and neoSTX as the major components, and dcGTX2,3, GTX2,3, neoSTX and STX as the minor ones. Toxin accumulation of three mussel species collected from Pohang, Geoje and Anmyon-do showed interspecies and interlocality differences. Toxin content ($average{\pm}SD\;{\mu}g$ STXeq/100 g) were $91{\pm}4,\;151{\pm}14,\;39{\pm}3$ in M coruscus, $189{\pm}1,\;231{\pm}11,\;206{\pm}15$ in M edu/is and $214{\pm}28,\;326{\pm}30,\;291{\pm}26$ in M. galloprovincialis in order of Anmyon-do, Geoje and Pohang.

  • This study was carried out with blue mussels at Otsuchi Bay in Japan from August 1981 to May 1982, in order to investigate the effect of the parasitic pea crab (Pinnotheres pholadis) on the reproductive capacity of host blue mussels (Mytilus edulis galloprovincialis). The results show that pea crabs delay gonad development of host mussels about one month and lower their fatness rate by intercepting nutrients. The parasitization with pea crabs is thought not to prevent spawning activity itself, but to reduce the number of germ cell resulting in the reduced space of gonads.

  • Sessile marine bivalves including mussels, oysters and clams are often used as a sentinel species in coastal environmental monitoring since changes in the environmental quality are often well preserved in their tissues and shells. In this study, we investigated overall health condition of the Mediterranean Mussel, Mytilus galloprovincialis on the south coast using histology. Reproductive condition as gonad index (GI), condition index (CI) as a ratio of the tissue weight to the shell weight, digestive gland atrophy (DGA), types of parasites, and pathologic conditions including erosion, necrosis, hemocyte infiltration, and neoplasia were examined from each histological preparation. GI decreased from March to July then increased from July to September and spawning mussel could be observed as early as in April and the activity continued until September. CI also followed the monthly changes in GI, indicating that decrease in CI was associated with the weight loss due to spawning. DGA increased from March to June, decreased in July and increased from July to September. High DGA values observed in June and September were coincided with spawning and high water temperature. Histology also showed high prevalence of erosion in the digestive gland in June (36.0%) and September (56.4%), suggesting that high water temperature and spawning acted as environmental stressors. No parasitic organism was identified during the survey, although some symbiotic copepods were observed. Histology was found to be useful and affordable technique in monitoring the overall health of mussel, providing useful pathologic information of the cells and tissues.

  • The toxicity of bivalves in the Korean waters was investigated during 1987-1988. The toxicity of paralytic shellfish poison (PSP) was found in the sea mussels (Mytilus edulis galloprovincialis, M. corsucus), scallops (Patinopecten yessoensis, Chlamys farreri), tellin (Peronidia venulosa) and venus clam (Ruditapes philippinarum) among the 28 species screened. The toxicity in mussels, scallop (C. farreri) and venus clam was mainly confirmed from the samples collected around Jindong Bay in the southern coast of the peninsula, whereas the toxicities of tellin and scallop (P. yessoensis) were found in the vicinity of Pohang in the east coast. Samples from the west coast showed low levels of toxicity than those from the east arid south coasts. Although it varied with the regions, the toxicity was detected mainly during April to June, exceptionally during May to August in Pohang. The levels of toxicity were generally below the criteria of 4 mouse unit (MU)/g edible portion in foreign countries, with few exceptions.
