The seed-borne mycoflora of sorghum and foxtail millet collected from different growing areas in South Korea were isolated and taxonomically identified using dry inspection, standard blotter and the agar plate method. We investigated the in vitro and in vitro germination rates of disinfected and non-disinfected seeds of sorghum and foxtail millet using sterilized and unsterilized soil. The percent recovery of seed-borne mycoflora from the seed components of sorghum and foxtail millet seeds was determined and an infection experiment using the dominant species was evaluated for seedling emergence and mortality. A higher number of seed-borne fungi was observed in sorghum compared to that of foxtail millet. Eighteen fungal genera with 34 fungal species were identified from the seeds of sorghum and 13 genera with 22 species were identified from the seeds of foxtail millet. Five dominant species such as Alternaria alternata, Aspergillus flavus, Curvularia lunata, Fusarium moniliforme and Phoma sp. were recorded as seed-borne mycoflora in sorghum and 4 dominant species (Alternaria alternata, Aspergillus flavus, Curvularia lunata, Fusarium moniliforme) were observed in foxtail millet. The in vitro and in vitro germination rates were higher using disinfected seeds and sterilized soil. More seed-borne fungi were recovered from the pericarp compared to the endosperm and seed embryo. The percent recovery of seed-borne fungi ranged from 2.22% to 60.0%, and Alternaria alternata, Curvularia lunata and 4 species of Fusarium were isolated from the endosperm and embryo of sorghum and foxtail millet. Inoculation of the dominant seed-borne fungi showed considerable mortality of seedlings. All the transmitted seed-borne fungi might well be a primary source of infection of sorghum and foxtail millet crops.
Mycoflora was assessed in the commercial meju from four well-separated geographic origins. A total of 112 fungal isolates were identified by phenotypic characteristics and molecular taxonomy using sequencing the internal transcribed spacer of the rDNA and revealed 19 species from 13 genera. Enzymatic characteristics of protease and amylase, and mycotoxin production were analyzed.
The mycoflora of Korea was rather poorly studied in the past and the fungi belonging to the genus Septoria are no exception. For this reason, taxonomic studies on Septoria have been initiated, with the eventual aim of producing a monograph of the Septoria species present in Korea. The present study circumscribes 10 species; viz., Septoria artemisiae, S. callistephi, S. chrysanthemella, S. erigerontis, S. lycopersici, S. lysimachiae, S. oenotherae, S. phlogis, S. rohlenae, and S. sonchi. Distinguishing morphological characters are described and illustrated for each species.
This study was carried out to determine the causative agent of dermatophytosis in 7 horses, and to examine the skin mycofloras on 84 healthy and 7 diseased horses which were derived from Jae-ju and Kyonggi, Korea in 1994~1995. Specimens of hair and scale were collected from skin lesions(or normal skins) and inoculated directly on potato dextrose agar and mycobiotic agar. These agar plates were incubated at $25^{\circ}C$ for 2 weeks. Growing fungi were isolated and identified by the morphological and nutritional characteristics. Lesions were found on the hind legs of an infected horses and each lesion was round or oval(1~4 cm) in shape accompanied by severe itching. The causative agent of the 7 equine dermatophytosis was identified as Trichophyton equinum. The skin mycofloras were Penicillium(69.0%), Aspergillus(63.2%), Cladosporium(51.7%), Fusarium(31.0%), Mucor(28.7%), Absidia(18.4%), Alternaria(17.2%), Acremonium(11.5%), Paecilomyces and Phycomyces(6.9%), Rhizopus(5.6%), Trichoderma(4.6%), Scopulariopsis and Trichophyton(3.5%), Beauveria(2.3%), Tritiracheum, Sporothrix, Curvularla, Aureobasidium and Chaetomium(1.2%), and Yeast(27.6%).
Fungicides and antagonists were tested for their efficacy in the management of fungal pathogens of watermelon. The fungal species in different genera were isolated from the seeds of watermelon and their vulnerability was assessed against an array of chemicals and bioagents. Among the fungal pathogens, Fusarium species were effectively controlled by Bavistin. Topsin also showed the promising effects against all the fungal pathogens, and Dithane M-45 effectively controlled Didymella bryoniae. Seed treatment with antagonists like Trichoderma harzianum and T. viride improved the seed germination, seedling vigour and reduced the incidence of seed-borne fungal pathogens. Bavistin and Topsin among chemicals increased significantly the seed germination and vigour index. Trichoderma harzianum showed its efficacy against all Fusarium species and even stood effective than Captan and Blitox. However, Pseudomonas fIuorescens also showed promising effect against Didymella bryoniae over fungicides. Under field condition, Topsin and Dithane M-45 showed better yield than Bioagents.
The colonization pattern and extent of decay produced in paddy stubble by soil inhabiting mycoflora were done by using nylon net bag technique. Among the three methods used for isolation of fungi, dilution plate technique recorded the highest number of fungi followed by damp chamber and direct observation method. Nutrient availability and climatic conditions (temperature, humidity and rainfall) influenced the occurrence and colonization pattern of fungi. Maximum fungal population was recorded in October ($48.99{\times}10^4/g$ dry litter) and minimum in May ($11.41{\times}10^4/g$ dry litter). Distribution of Deuteromycetous fungi was more in comparison to Zygomycetes, oomycetes and ascomycetes. In the early stage of decomposition Mucor racemosus, Rhizopus nigricans, Chaetomium globosum and Gliocladium species were found primarly whereas at later stages of decomposition preponderance of Aspergillus candidus, Torula graminis, Cladosporiun cladosporioides and Aspergillus luchuensis was recorded.
The present survey was undertaken to determine the mycoflora of broiler houses. Attempts were made to isolate and identify fungi in the dust, feed, litter and water from 21 broiler houses. A total of 166 isolates of fungi was identified as yeast spp. (44%), Aspergillus spp. (30.7%), Verticillium spp. (7.2%), Penicillium spp. (3.6%), Paecilomyces spp. (3.6%), Scopulariopsis spp. (3.0%), Cephalosporium spp. (3.0%), Chrysosporium spp. (2.4%), Cladosporium spp. (1.8%) and Absidia spp. (0.6%). Isolated of Aspergillus(A) spp. and Penicillium(P) spp. were identified as A. fumigatus, A. flavus, A. terreus, A. nidulans, A. niger, P. citrinum and P. palitan. Fungal contamination of the broiler houses predominated in the dust. The important point is that must of the isolates were pathogenic fungi.
Five cereal grains(wheat, barley, rice, maize and sorghum) were collected from three Egyptian provinces known to be grain producers(Daqahlia, Gharbia and Kafer el-Sheikh). Two species of Alternaria(A. raphani and A. tenusinae); two species of Aspergillus(A. flavus and A. niger); one species of Cunninghamella(C. elegans); one Dreschslera species(D. myakt); three Fusarium species(F. graminearum, F. moniliform and F. solani); one Rhizopus species(R. stolonifer) and two species of Penicillium(P. digitatum and P. notatum) were isolated from the grains. The densities of these fungi and their frequencies of occurrence have been investigated. All the fungal isolates were tested for the production of toxic metabolites in culture media and the percentages of toxigenic isolates were calculated. The biological assay of the toxigenic fungal isolates showed significant variations in toxigenic activity. Thin layer chromatography revealed that the most active isolate produces moniliformin in culture media. The effect of culture conditions on the production of moniliformin was studied.

Diverse fungi are present in Korean traditional meju and they are known to play an important role in fermented soybean products. To determine the origin of the fungi in meju, we examined the mycoflora of soybeans from 10 traditional meju factories. The samples were untreated or treated with sodium hypochlorite, and placed on malt extract agar (MEA), dichloran 18% glycerol agar (DG18), and dichloran rose bengal chloramphenicol agar (DRBC) medium. A total of 794 fungal strains were isolated and they were identified as 41 genera and 86 species. From sodium hypochlorite untreated soybeans, the genera, Cladosporium (55%), Eurotium (51%), Fusarium (33%), Penicillium (22%), and Aspergillus (exclusion of Eurotium) (20%), were mainly isolated, and Eurotium herbariorum (22%), Eurotium repens (18%), Cladosporium tenuissimum (18%), F. fujikuroi (18%), Aspergillus oryzae/flavus (7%), and Penicillium steckii (6%) were the predominant species. In case of sodium hypochloritetreated soybeans, Eurotium (31%) and Cladosporium (5%) were frequently isolated, but Aspergillus (excluding Eurotium), Penicillium and Fusarium which were frequently isolated from untreated soybeans, were rarely isolated. Eurotium herbariorum (21%), Eurotium repens (8%), and Cladosporium tenuissimum (3%) were the predominant species. Of the 41 genera and 86 species isolated from soybeans, 13 genera and 33 species were also found in meju. These results suggest that the fungi on soybeans may influence the mycoflora of meju.
A total of twenty-nine species and one species variety belonging to 12 genera was isolated from 30 samples of molasses on 1% glucose(10 genera, 22 species and 1 variety) and 50% sucrose(7, 21 and 1) Czapek's agar at $25^{\circ}C$ media. Aspergillus, Mucor, Mycosphaerella and Penicillium were the most common genera on the two types of media. From the above genera, the most prevalent species were: Aspergillus flavus, A. fumigatus, A. niger, Mycosphaerella tassiana, Penicillium chrysogenum, P. oxalicum and P. purpurogenum. Also, some species were only isolated on 50% sucrose such as Eurotium amstelodami, E. chevalieri, E. repens, Humicola fuscoatra, Penicillium aurantiogriseum and P. puberulum. About 65 fungal isolates isolated from 50% sucrose agar were tested for their ability to produce invertase enzyme in liquid medium and 93.8% of the isolates could produce this enzyme. From the positive isolates, 32 showed high invertase activity, 21 had moderate activity and the remaining 8 isolates were of weak activity. Sixty isolates of Aspergillus, Emericella, Eurotium, Mycosphaerella and Penicillium from the preceding study were screened for the presence of their respective mycotoxins. Larva of brine shrimp(Artema sauna L.) were used for toxicity test of the fungal crude extracts. Three isolates out of 60 tested were toxic. Using thin-layer chromatographic technique, 5 different known mycotoxin were detected aflatoxins : B1, B2, G1, G2 and citrinin.
