A fungal isolate was newly collected from Zoysiagrass 'Anyang-Joongji' in small circular patches on a fairway ofa golf course in Korea, which seriously occurred during the early summer period of 2005. The isolate presented on PDAmedium, named Scz1, was closely identical to Sclerotinia homoeocarpa, a casual fungus of dollar spot disease, in cool season turf grasses such as creeping bentgrass. Hereby, this study was accomplished to characterize the isolate and compare it with the fungus, named Scb1, isolated from dollar spot-infected creeping bentgrass (Agrostis palustris Huds. cv Penncross). On PDAmedium, individual mycelial appearance of three isolates was very similar except for the pigment. Mycelial pigments of Scz1 and Scz2 (another analogous isolate collected) were light pinkish on the reverse side of PDA medium but that of Scb1 was dark brownish. In a microscopic study, three isolates were barely distinguishable in the appearance of mycelia. As expected, in the temperaturesensitivity assay, all pathogens were very delicate to $32^{circ}C$ above but not to $30^{circ}C$ below, in which was explained to be one of typical characteristics in S. homoeocarpa. In an artificial inoculation assay, disease symptoms including leaf spots in Zoysiagrass were appeared within 6-7 days after inoculation through the hand inoculation method with the isolate-infested soil. Then the fungus was re-identified from the infected leaf tissues. Interestingly, inoculation of isolate Scz1 gave rise to distinct symptoms in only Zoysiagrass but not in creeping bentgrass 'Penncross' and Kentucky bluegrass 'Midnight'. The observation might be involved in host specific pathogenecity of S. homoeocarpa Scz1 to Zoysiagrass. In a chemical sensitivity assay for the isolate, Scz1, showed a high mycelial inhibition against two fungicides, iprodione and propiconazole. All results described above suggest that S. homoeocarpa Scz1 is a primary pathogen of Zoysia dollar spot disease.

The present study was conducted to determine the effect of electron beam irradiation on control of Botrytis cinerea and postharvest quality of cut roses. Electron beam doses of 0.1, 0.2, 0.4, 0.6, 0.8, 1, 2, 10, and 20 kGy were applied with a 10-MeV linear electron beam accelerator (EB Tech, Korea). Electron beams inhibited spore germination and mycelial growth of B. cinerea with increasing irradiation doses. Conidia of B. cinerea were more tolerant to irradiation than were mycelia: the effective irradiation doses for 50% inhibition ($ED_{50}$) of spore germination and mycelial growth were 2.02 kGy and 0.89 kGy, respectively. In addition, electron beam irradiation was more effective in reducing mycelial growth of B. cinerea at $10^{\circ}C$ than at $20^{\circ}C$. Analysis of in vivo antifungal activity revealed that elevated irradiation doses exhibited increased control efficacy for tomato gray mold. Flower longevity and fresh weight of cut roses decreased when the irradiation dose was increased. In addition, flower bud opening tended to be inhibited in a dose-dependent manner. Although 'Decoration', 'Il se Bronze', 'Queen Bee', and 'Revue' roses tolerated and maintained overall postharvest quality up to 0.4 kGy, 'Vivian' did not, demonstrating that the irradiation sensitivity of cut roses varies according to cultivar.
The Rhizoctonia blight causing by Rhizoctonia spp. is an important disease of turfgrass, requiring fungicide application to maintain acceptable conditions for turfgrass good qualities in the golf course. The experiment was conducted to determine the mean 50% effective concentration inhibiting mycelial growth ($EC_{50}$) value of to flutolanil, pyraclostrobine and hexaconazole to Rhizoctonia solani AG-1 IB, Rhizoctonia cerealis and Rhizoctonia solani AG2-2IV isolated from Gyeongbuk province of Korea in vitro. Five discriminatory concentrations of each fungicide were used to detect in vitro sensitivity. The mean of $EC_{50}$ values to three systemic fungicides was the lowest isolate of R. solani AG-1 IB. However, the sensitivity of fungicides to Rhizoctonia solani AG2-2IV were higher mean $EC_{50}$ value of 0.026 ${\mu}g\;a.i.\;ml^{-1}$ of pyraclostrobine and 0.044 ${\mu}g\;a.i.\;ml^{-1}$ of flutolanil. R. cerealis was the lowest sensitivity to hexaconazole which was an average $EC_{50}$ value of 0.022 ${\mu}g\;a.i.\;ml^{-1}$. Inhibition of mycelial growth rate (%) by three combine fungicides using the $EC_{50}$ value of each fungicide was the highest R. solani AG2-2IV. Results of this study were may confirmed in vitro response fungicide of three Rhizoctonia species for control of Rhizoctonia blight in the field.

The thirteen commercial insecticides to Lycoriella mali were investigated on their insecticidal activities and mycelial growth of two Letinula edodes strains. For the adults, insecticides showing over 95% insecticidal activity were chlorpyrifos-methyl, fenthion, fenitrothion, benfuracarb, furathiocarb and deltamethrin. For tile larvae, diflubenzuron and cyromazine showed over 90% insecticidal activity. Fenthion, benfuracarb, furathiocarb, deltamethrin, diflubenzuron and cyromazine did not affect the mycelial growth of L. edodes strain, in Imhyup 1 variety. And deltamethrin, diflubenzuron and cyromazine did not affect that in Sanlim-5. Insecticides showing over 80% residual effect for 14 days were benfuracarb to the adults and diflubenzuron and cyromazine to the larvae. Control effect of furathiocarb, fenthion, benfuracarb, deltamethrin, diflubenzuron and cyromazine against larvae of Lycoriella mali showed 90% ten days after application under sawdust cultivation. Among them, benfuracarb, fenthion and furathiocarb revealed excellent control effect against adults. These results indicate that benfuracarb, fenthion, and furathiocarb can be used for the control of Lycoriella mali in the field. However, further studies are needed on the effect of insecticides treatment oil fruit-body yield and chemical residue in the mushroom tissues.
Fifty-three actinomycetes antagonistic to Phytophthora capsici and Magnaporthe grisea were isolated from rhizosphere soils in six pepper-growing areas and ashore soils. Thirty-two antagonistic actinomycetes, showing inhibition zone larger than 5 mm, were classified into 20 groups according to their colony morphology and color. The antagonistic activity against P. capsici greatly varied, which showed inhibition zone sizes in the ranges from 5.7 to 17.5 mm on V-8 juice agar and from 2.5 to 17 mm on tryptic soy agar. The antagonistic activity of some actinomycetes tested was remarkably different between the two test media. The antagonists showed a relatively broad antifungal spectrum, but their antibacterial activity was negligible, except for Pseudomonas solanacearum. Butanol extracts of culture filtrates from antagonistic actinomycetes inhibited mycelial growth of P. capsici and M. grisea, thereby confirming strongly antibiotic production in culture. Culture filtrates of some antagonistic actinomycetes completely inhibited Phytophthora blight in pepper plants.

Since the mass mortality of Quercus mongolica has been first reported in Gyeonggi province at 2004, the disease spread rapidly over Korean peninsula annually. Ambrosia beetle (Platypus koryoensis) was known as the insect vector of oak wilt fungus, Raffaelea quercus-mongolicae, and control methods of the disease had mainly been focused on eradication of insect vector. However, for the efficient management of the disease, combined control methods for both of the pathogenic fungus and insect vector are strongly required. As one of the efforts to suppress the pathogenic fungus, antifungal activities of Streptomyces isolated from oak forest soil were assayed in this study. Optimum culture condition for the selected isolates was also studied, As a result, Streptomyces blastmyceticus cultured in PDB (Potato Dextrose Broth) at $25^{\circ}C$ for 1 week showed the strongest antifungal activity against oak wilt fungus. Mycelial growth inhibition rates (MGIRs) of Streptomyces isolates were compared on culture media supplemented with heated and unheated culture filtrates of S. blastmyceticus. MGIRs on culture media with unheated culture filtrates were generally higher than those on culture media with heated culture filtrates. Antagonistic mechanism to get involved in the inhibition of hyphal growth and spore formation of the pathogen is due to the antifungal metabolites produced by Streptomyces. This study will provide the fundamental information in developing biocontrol agents for the environment-friendly management of oak wilt disease.
Response surface methodology (RSM) was successfully applied to optimize for the production of Ganoderma lucidum in batch fermentations using the whey (40,000 mg latose/L) as substrate. This study was performed according to the central composite design (CCD) with respect to pH and temperature, where the designed intervals were 3.3$22.9^{\circ}C$$37.1^{\circ}C$, respectively. A second-order factorial design of the experiments was used to build empirical models providing a quantitative interpretation of the relationships between the two variables. The optimum conditions to maximize the production of G. lucidum were pH 4.2 and $28.3^{\circ}C$. At optimum conditions, the mycelial dry weight (MDW) and residual soluble COD (SCOD) were simultaneously used to evaluate the biokinetic coefficients assocoated with substrate inhibition model by nonlinear least squares method with 95% confidence interval. The. maximum microbial growth rates (${\mu}m$), half saturation coefficient ($K_s$), and the inhibition substrate concentration ($K_{is}$) were determined to be 0.095 l/hr, 128,000 mg SCOD/L and 49,000 mg SCOD/L, respectively. And the microbial yield coefficient (Y), biomass decay rate coefficient ($K_d$), and the maintenance energy coefficient ($m_s$) were determined to be 0.37 mg MDW/mg SCOD, 0.001 1/hr, and 0.0015 1/hr, respectively.
Recently, the importance of management and cultivation of grasses has been increased in Korea. Among these cultural practices, the appropriate control of diseases is considered more important than other cultivation techniques such as fertilization and irrigation. The damages of brown patch and large patch caused by Rhizoctonia spp. and Pythium blight caused by Pythium spp. are serious in the major cultivation area of turfgrass in Korea. Since these diseases are difficult to control by agrochemicals, the damages are very serious if these are occured. The periodic spray of agrochemicals, to protect and control these diseases could make some problems of toxicity and environmental pollution as well as rising of non-target diseases. Therefore, the biological methods to control diseases have been required to decrease problems resulted from overuse of agrochemicals, to conserve natural ecosystem, and to control effectively diseases of grasses in the long period. The number of studies about biological control using antagonistic microorganisms have been increased for last half century. However, the application of biological control method has been very limited. In this study, thirteen isolates of R. cerealis, 8 isolates of R. solani and 3 isolates of Phthyn spp. have been isolated from diseased turfgrass in golf course and grass-culture area that have patch and wilting symptoms of zoysia grass and creeping bentgrass. Isolation frequency of R. cerealis and R. solani was high in especially zoysiagrass, while Pythym spp. was isolated from bent grass at low frequency but showed high pathogenicity. Totally, 205 isolates of soil microorganisms were isolated in this study as primary antagonistic microorganism by Herr's triple agar layer plate and dual culture method using rhizosphere of grasses, soil of crop field as the source of antagonistic microorganisms. Among the 205 isolates, 23 isolates were actinomycetes and 182 isolates were bacteria. All of the actinomycetes were isolated by Herr's method. Antagonistic effect of primary isolated microorganisms was tested for in vitro mycelial growth inhibition against pathogenic fungi isolated from grasses and for inhibition of disease occurrence in 24 well tissue culture plate and pot experiment. Then, four isolated of bacteria which are BG23, BG74, BG136 and BG171 were selected as antagonistic microorganisms against soil-born pathogenic fungi of bentgrass.
This study was conducted to find out antifungal activities of entomopathogenic fungus, Metarhizium anisopliae, against phytopathogenic fungi, Fusarium oxysporum, Botrytis cinerea and Alternaria solani. M. anisopliae was confirmed its antagonistic effect through mycelial inhibition zone of phytopathogenic fungi by culture filtrate of the antagonist. The filtrate (30%: v/v) inhibited the conidial germination of B. cinerea and F. oxysporum to 21.5% (control: 88.2%) and 53.0% (control: 78.6%), respectively and delayed the start of spore germination about 8hours. Microscopic observations proved that the addition of 10% culture filtrate of M. anisopliae restricted the growth of phytopathogenic fungus, F. oxysporum, to the formation of chlamydospore. From these results, we concluded that an addition effect of the filtrate from M. anisopliae on culturing F. oxysporum was fungistatic.
Total 443 isolates of actinomycetes were isolated from turfgrass rhizosphere as potential biological control agents. The two isolates (S11 and S4) showed highest cellulase activity with compared to the other isolates that exhibited a clear zone of 1.2 mm around the colony on cellulose agar medium. S12 strain appeared the most active chitin degrading, which exhibited a 1.2 mm of clear zone. The highest proteolytic activity on skim milk agar was which exhibited a 7.5 mm of clear zone by S2 strain. S1 strain from the soli showed siderophore production ability, which exhibited a 0.6 mm of large clear zone on chrome azurol S agar. The antifungal activity of the volatile compound producing by 4 selected actinomycetes was investigated that inhibition rate against Rhizoctonia solani AG2-2 and Sclerotinia homoeocarpa. Growth inhibition effect of S8 isolate against S. homoeocarpa was appeared to 94.8%, S2 to 76.9%, S5 to 46.1% and S12 to 43.5%. The significant inhibition effects on mycelial growth of S. homoeocarpa were shown on media with four strains. The inhibition effect was the highest with S8 strain treatment at 94.8%.
