Effects of Vitexin from Mung Bean on 3T3-L1 Adipocyte Differentiation and Regulation According to Adipocytokine Secretion (녹두의 Vitexin이 비만전구세포에서 세포분화 및 아디포사이토카인 분비능에 미치는 영향)

  • Obesity is an important issue worldwide as it may associated with increased prevalence of metabolic diseases. Mung bean is known as a functional food for decreasing the glycemic index and lipid profile of plasma. The purpose of this study was to investigate the anti-obesity effects of vitexin from mung bean on the regulation of adipocyte differentiation and adipocytokine secretion. When 3T3-L1 adipocytes were treated with vitexin from days 0 to 14 at various levels of 25, 50, 100, and $200{\mu}M$, there was no change in cell viability. Vitexin treatment at 50, 100, and $200{\mu}M$ decreased triacylglycerol levels in cells, but only $100{\mu}M$ vitexin induced lipolysis. At $200{\mu}M$ of vitexin, phosphorylation of p38 and ERK, which causes secretion of inflammatory adipocytokines, was depressed, whereas there was an increase in expression of $PPAR{\gamma}$, the key regulator of adipocyte differentiation. Phosphorylation of AMPK increased at $100{\mu}M$ vitexin. TNF-${\alpha}$ and aP2 mRNA expression increased at $25{\mu}M$ vitexin, whereas only TNF-${\alpha}$ mRNA expression increased at $200{\mu}M$ vitexin. Further, the mRNA levels of TNF-${\alpha}$ and aP2 decreased at other concentrations in a dose-dependent manner. Since we observed that mRNA expression of C/EBP, SREBP1, and $PPAR{\gamma}$ did not change upon vitexin treatment, our future studies will investigate other genes such as mTOR, which is related with apoptosis signaling, or SIRT1, which is associated with inhibition of adipogenesis. Our results indicate that vitexin at concentrations between 100 and $200{\mu}M$ is suitable in vivo for the development of mung bean as an anti-obesity therapy or functional food.

Contents of Phytic Acid of Various Cereal Crops Produced in Korea

  • The main objective of the study was to investigate the phytic acid contents of various cereals and legumes produced in Korea. The results showed that the phytic acid contents of buckwheat, foxtail millets, sorghum, millet, barley, jobs' tears, yellow corn and wheat flour (Urimil) were in a range of 0.13 to 2.27%. The contents of the phytic acid ranged from 1.03 to 1.16% for legumes including red Indian bean, black Indian bean, mung bean, and black soybean. The phytic acid content of brown rice was five times higher than those of 100% polished rice. The polishing process of the rice decreased significantly the content of the phytic acid. We estimate that the daily phytic acid intake from rice was changed from 653 mg/day in 1995 to 430 mg/day in 2005 based on the results of a national nutrition survey.

Nutrient Intake Status of College Students Based on Their Consumption of Black Beans (검정콩 섭취여부에 따른 일부 대학생의 영양소 섭취상태)

  • This study analyzed the intake of calories and nutrients based on the consumption of black beans. The survey was conducted on 124 college students (male: 42, female: 82), of whom 63.7% (79 students) were consumers of black beans. The calorie intake of the black bean intake group (1599.81±555.48 kcal) was significantly higher than that of the non-black bean intake group (1259.99±507.58 kcal) (P<0.01). The black bean intake group also showed a significantly higher intake of crude protein (P<0.05), plant protein (P<0.01), animal protein (P<0.05), crude fiber (P<0.001), plant calcium (P<0.001), plant iron (P<0.001), zinc (P<0.01), vitamin B2 (P<0.01), vitamin C (P<0.01), vitamin E (P<0.01) and folic acid (P<0.001) compared to the non-black bean intake group. There was a positive correlation between the frequency of black bean intake and crude fiber (P<0.05), plant calcium (P<0.05), plant iron (P<0.05), and folic acid (P<0.05). Overall, the black bean intake group was more likely to eat black sesame 13.82 times (95% CI=5.37, 35.55), white beans 10.79 times (95% CI=3.53, 33.02), mung beans 7.22 times (95% CI=2.58, 20.23), and brown rice 4.72 times (95% CI=1.88, 11.84), than the non-black bean intake group. In conclusion, we believe that black beans constitute a vital food ingredient that is necessary to provide Korean college students with a well-balanced diet.

Isolation and Characterization of ACC Synthase Gene Family in Mung Bean (Vigna radiata L.): Differential Expression of the Three ACC Synthase enes in Response to Auxin and Brassinosteroid

  • By screening a cDNA library of auxin-treated mung bean (Vigna radiata L.) hypocotyls, we have isolated two full-length cDNA clones, pVR-ACS6 and pVR-ACS7, for 1-aminocyclopropane-1-carboxylate (ACC) synthase, the rate-limiting enzyme in the ethylene biosynthetic pathway. While PVR-ACS6 corresponds to the previously identified PCR fragment pMBA1, pVR-ACS7 is a new cDNA clone. A comparison of deduced amino acid sequences among auxin-induced ACC synthases reveal that these enzymes share a high degree of homology (65-75%) to VR-ACS6 and VR-ACS7 polypeptides, but only about 50% to VR-ACS1 polypeptide. ACS6 and ACS7 are specifically induced by auxin, while ACS1 is induced by cycloheximide, and to lesser extent by excision and auxin treatment. Results from nuclear run-on transcription assay and RNA gel blot studies revealed that all three genes were transcriptionally active displaying unique patterns of induction by IAA and various hormones in etiolated hypocotyls. Particularly, 24-epibrassinolide (BR), an active brassinosteroid, specifically enhanced the expression of VR-ACS7 by distinct temporal induction mechanism compared to that of IAA. In addition, BR synergistically increased the IAA-induced VR-ACS6 and VR-ACS7 transcript levels, while it effectively abolished both the IAA- and kinetin-induced accumulation of VR-ACS1 mRNA. In light-grown plants, VR-ACS1 was induced by IAA in roots, whereas W-ACS6 in epicotyls. IAA- and BR-treatments were not able to increase the VR-ACS7 transcript in the light-grown tissues. These results indicate that the expression of ACC synthase multigene family is regulated by complex hormonal and developmental networks in a gene- and tissue-specific manner in mung bean plants. The VR-ACS7 gene was isolated, and chimeric fusion between the 2.4 kb 5'-upstream region and the $\beta$-glucuronidase (GUS) reporter gene was constructed and introduced into Nicotiana tobacum. Analysis of transgenic tobacco plants revealed the VR-ACS7 promoter-driven GUS activity at a highly localized region of the hypocotyl-root junction of control seedlings, while a marked induction of GUS activity was detected only in the hypocotyl region of the IAA-treated transgenic seedlings where rapid cell elongation occurs. Although there was a modest synergistic effect of BR on the IAA-induced GUS activity, BR alone failed to increase the GUS activity, suggesting that induction of VR-ACS7 occurs via separate signaling pathways in response to IAA and BR.

Isolation and Characterization of Two Isoperoxidases from Mung Bean Seedling (녹두(綠豆)에서 Peroxidase 동위(同位) 효소(酵素)들의 분리(分離)와 효소적(酵素的) 특성(特性))

  • The changes in peroxidase activity and its isozyme pattern in the different parts of mung bean sprout were investigated; The enzyme activity in cotyledon and root showed a tendency to increase at an early stage and then decreased gradually as germination continued. However, the crude homogenate of epicotyl and hypocotyl showed a continuous decline in the enzyme activity. In particular, the enzyme activity of the root was $1.5{\sim}3.5$ times higher than that of other parts. Gel electrophoresis of the crude homogenate revealed that the number of isozyme in every part of the mung bean sprout increase during germination up to 6th days. Two isozymes from root were partially purified by ammonium sulfate fractionation, gel filtration by Sephadex G-75 and DEAE cellulose column chromatography. One of the isozymes (A) was purified 16-fold by the present procedure, but the purity of the other isosyme (B) was not increased , significantly. Isozyme A was the most active at $65^{\circ}C$ and isozyme B at $70^{\circ}C$, while both isozyme (A, B) have a optimal pH of 5.6. The Km values of isozyme A and B for 0-dianisidine as a hydrogen donor determined to be 0.071 mM and 0.052mM, respectively, and those for isozyme A and B using $H_2O_2$ as a hydrogen acceptor were 0.28mM and 0.23mM, respectively.

Study on the Positional Distribution of Fatty Acids, and Triacylglycerol Separation, of Seed Oils (종실유(seeds oil)의 위치별 지방산 및 트리아실글리세롤의 조성 연구)

  • Six types of oil were extracted from pomegranate seed, mung bean, pepper seed, safflower seed, seeds of Cassia tora Linnaeus, and perilla seed. The extracted seed oils were analyzed for total and positional fatty acid composition, triacylglycerol (TAG) level, and tocopherol content. Crude fat levels measured by the Folch method were 21.64% in perilla seed, 13.85% in safflower seed, 9.60% in pepper seed, 8.85% in pomegranate seed, 2.25% in mung bean, and 2.00% in C. tora,respectively (all w/w). Linoleic acid (C18:2) was the most abundant fatty acid at the sn-2 position of triacylglycerols (TAGs), ranging from 15.99-88.3 wt%. The composition of TAGs was analyzed by reverse-phase HPLC, and TAGs of seed oils showed partition numbers of 36-48. The highest content (377.74 mg/100 g) of total tocopherol was found in pomegranate seed whereas the total tocopherol content of mung bean, C. tora, pepper seed, perilla seed, and safflower seed were 141.16, 107.23, 33.88, 30.05, and 29.80 mg/100 g, respectively.

  • This experiment was conducted to verify the responses of major crop species to saline stress.To determine the saline tolerance of crop species, sevral crop species were cultivated under sand-culture system using Hoagland's nutrient solution whit 200mM NaCl. The crop species showing saline tolerance were cotton(Gossypium indicum), maize(Zea mays), barely(Hordeum vulgare), and wheat(Triticum aestivum) but perilla (Perilla frutescens) and leguminous crops, mung bean(Phaseolus radiatus), azuki bean(Phaseolus angularis) and soy bean(Glycin max) showed very por tolerance. One the typical symptom was the darkening of leaf color due to increase of chlorophyll concentration.Among of the plant families, Fabaceae was the most susceptible but crop species belonging to Poaceae were more proper for cultivating on reclaimed tidal land in the course of desalinazation. It was suggrsted that the crop species belonging to Fabaceae, a sensitive family to soil salinity, must be cultivated when the soil salinity decreased below 10ds/m. To know the critical salinity level for crop growth,salinity of saline soil collected from reclaimed tidal land was adjusted from 10.0ds/m tp41.7ds/m with tap water. It was suggested that the ECs of the soil in which the plant height of each crop spicies was reduced to 50% of control plant were 22.6 and 21.7 ds/m in rice, barley, corn, mung bean, and soy bean,respectively.

  • Penicillium sp. PS-113 showed no toxicity against loach(Misgurnus mizolepis) and monnow(Oryzias latipes), a kind of biological indicator, for 96 hours at the concentrations of $1.0\;{\times}\;10^8$ conidia/ml inoculum. Additionally, it showed no toxicity against several plants such as Chinese cabbage(Brassica campestris subsp. napus var. pekinensis), mung bean(Phaseolus radiatus), etc. during their germination at the same concentration of inoculum.
