Apical meristems of Wasabia japonica were cultured on Murashige and Skoog's medium supplemented with cytokinins alone or together with 1.0 mg/L IAA. Shoot initials could be induced from leaf primordia on apical meristems. Calli and roots were formed on the medium containing cytokinins and 1.0 mg/L IAA in combination after 30 days of culture, but there were no callus proliferation. Shoot organogenesis began after 60 days of culture and these small shoots elongated when transferred to a medium containing 1.0 mg/L BA or kinetin. Shoots were formed directly without callus induction from apical meristems all the explants on the medium containing cytokinins variously, and most of the shoots proliferated multiple shoots which could be divided to obtain plantlets. Shoot multiplication rate in response to cytokinins was best on the medium containing 1.0 mg/L BA or 2.0 mg/L zeatin. Divided plantlets rooted well on MS medium containing 0.01 mg/L IBA after 15~30 days of subculture and the rooted plantlets developed into whole plants with multiple shoots. After rooting, the regenerated plants were washed and transferred to the pots containing sterilized soil.
In sweet potato cultivars 'Mokpo #29' and 'Sanchunza', shoots from extplants were formed 100% on the MS medium with 0.1 ㎎/L NAA and 2.0 ㎎/L BA after 30 days of culture and roots produced from the base of stem at frequencies of 66.7% ('Mokpo #29') and 69.2% ('Sanchunza'), respectively, The media with 0.5∼4.0 ㎎/L BA were produced the greatest frequency of multiple shoot and the most of shoots developed rapidly into normal plantlets with rooting within 60 days of culture. Whereas the cultivar 'Keumsi' failed to produce normal shoot multiplication on the medium with cytokinins alone because of callusing of adventitious shoots. When single shoots with 1 to 2 nodes were excised from the multiple shoot or shoots covered with callus devoid of root and transferred to MS medium with 4.0 ㎎/L BA or kinetin. Host divided shoots showed the callus induction at the stem base and it was enable to obtain regenerated plantlets with shoot and root normally.
In vitro mass multiplication of Vanilla planifolia was investigated using node as explant. Multiple shoots were developed in MS medium supplemented with $2.0mgl^{-1}$ 6-benzylaminopurine and $1.0mgl^{-1}$$\alpha$-naphthalene acetic acid. Multiple shoots were maintained for 6-T weeks with regular subculturing at the end of $3^{rd}$ week onto fresh medium. The maximum number of shoots at the rate of 12.8 per node segment was achieved over a period of four weeks. The elongated shoots were separated from the shoot clusters and were transferred onto half strength MS medium supplemented with indole-3-acetic acid ($1.0mgl^{-1}$) over a period of 28 days for induction of roots. The development of roots was observed on $7^{th}$ day of incubation. The in vitro raised plantlets were transferred to poly-cups, covered with polyethylene sheets and maintained under shade net for 25 days for hardening. Finally these plants were transferred to field and recorded that 85 % of tissue cultured plants were survived. From the present study, a simple and efficient micropropagation protocol was developed for Vanilla planifolia using single node segments as explants.
Multiple shoots of garlic (Allium sativum L.) were propagated in bioreactors containing MS medium supplemented with 2% sucrose for 3 weeks. Microbulbs were induced on MS medium supplemented with 0.1mg/L NAA and 11% sucrose for 9 weeks. For multiple shoot proliferation, leaves in the shoot must be removed before cultures. When the multiple shoots were cultured without removal of leaves, more than 90% of hyperhydricity and no microbulb formation were observed. Histological observation also indicated irregular size and shape of the cells in hyperhydricity of the shoot. Microbulbs were strarted to form from the shoot after 7 weeks of culture by protuberance of adventitious shoot buds followed by inner periclinal divisions and simultaneous anticlinical division in the epidermis of meristematic bulge. Analysis of ploidy level indicated no phenotypic variations in both multiple shoots and microbulbs induced from the mother plant, suggesting genetic homogeneity among the regenerants.

Multiple shoot formation was obtained from excised axillary buds of Achyranthes japonica NAKAI cultured on MS media containing various growth regulators such as auxin and cytokinin. The highest average number of shoots was obtained in 1 mg/L NAA and 2 mg/L BA after 6 weeks (25.8 adventitious shoots per node). Although the regeneration rate was less than the former condition, optimal combination for the production of more shoots with a suitable size was 0.5 mg/L NAA and 1 mg/L BA (19.7 adventitious shoots per node). Roots were induced from regenerated shoots after 3 weeks culture, transferred to 1/2 MS medium supplemented with 0.1 mg/L IBA. Micropropagated plants were successfully transferred to soil.

A rapid micropropagation protocol was established for Exacum travancoricum Bedd. The effect of two cytokinins viz. BA and kinetin were studied to evaluate the propagation of plants through nodal explants. MS medium supplemented with 13.32 ${\mu}M$ BA induced early bud break and subsequent production of multiple shoots. Rooting of shoots occurred when cultured on 1/2 strength MS medium supplemented with 14.7 ${\mu}M$ IBA. Rooted plants were acclimatized to greenhouse conditions. The propagated plants were transferred successfully to field with 65% success. As the plant was amenable to propagation in vitro, this can be employed as a tool for conservation of this critically endangered and endemic ornamental herb.
An effective rapid propagation method was established through in vitro cultures of the medicinal plant, Cudrania tricuspidata. In vitro plantlets were obtained from in vitro germinated seeds. The various levels of cytokinins (BAP, Kinetin and TDZ) were tested on multiple shoot formation from plantlets. BAP (1.0 mg/l) treatment induced highest number of multiple shoots. Single shoot cultures gave higher initial shoot numbers than 5 shoots per culture. Among the various culture media, the shoot elongation was optimal on 2 MS basal medium without growth regulators. The IAA (2.0 mg/l) treatment induced highest number of roots. IBA (2.0 mg/l) treatment more promoted in vitro root growth than other concentrations. Rooted shoots were transferred directly to small pots with an artificial soil and successfully acclimatized.
Whole plants were regenerated from callus induced from leaf explants in hybrid kiwi (Actinidia deliciosa${\times}$A. arguta). Callus was induced from leaf explants which cultured on MS solid medium supplemented with combination of auxin (2,4-D, NAA: 0.1~0.5 mg/l) and cytokinin (BA: 0.1~0.2 mg/l). them, the highest callus formation (96.2%) was obtained from the treatment of 0.5 mg/1 2,4-D+0.1 mg/l NAA+0.05 mg/l BA. In the experiment of adventitious shoots induction from primary shoots, only a few shoots were produced in the treatment of 1.0 mg/l BA+0.05 mg/l IBA or 2.0 mg/l BA+0.05 mg/l lBA. As the callus were transferred to the secondary shoot-inducing medium, multiple shoots were obtained from the medium supplemented with 1.0, 2.0 or 5.0 mg/l zeatin in addition to the mixed treatments of BA, thidiazuron (TDZ) or zeatin. However, no multiple shoots were induced on the BA-contained medium of concentrations. Therefore it turned out that addition of BA to medium was less effective for induction of multiple shoots from callus in Actinidia deliciosa${\times}$A. arguta. For producing adventitious roots from shoots, the best frequency of rooting (83.3%) were recorded on the treatment of in vitro rooting (Standardi (St)+1.0 mg/l IBA). On the other side, the lowest result (40.0%) were shown in the treatment of 500 mg/l IBA, 1 hr. Whole plants with shoots and roots were recovered and acclimatized successfully.

A successful, efficient system for multiple shoot induction and plant regeneration of soybean (Glycine max) was established. Four soybean genotypes were compared for organogenic responses on various media cultured under light conditions. The adventitious shoots (98%, 2.6 shoots/cotyledon) directly from one-day-old cotyledon after germination induced by the hormone treatment and its efficiency was higher than any other conditions. The optimal medium for the induction of multiple shoots from cotyledon in Pungsannamulkong(shoot formation rate, 98%), Lx 16 (83%) and IIpumgeomjeongkong(63%) was MS medium supplemented with 2 mg/L BAP, but for Alchankong(75%), MS medium supplemented with 1mg/L zeatin and 1mg/L IAA, 3% sucrose, 4% Phytagel. Higher root induction (88%) was observed from the shoots placed on rooting medium (hormone-free MS basal). Plantlets were transferred onto the same medium supplemented with 1% activated charcoal for further development. With this treatment, regenerated plantlets were obtained within 7-8 weeks (shoot induction for 4 weeks, rooting and shoot elongation for 3-4 weeks).

A method for rapid propagation of mature Jack fruit was developed. Four types of explants (mature embryos, apical meristems of young seedlings, apices from mature plants and nodal segments) were used. It has been found 88% of young apical meristems produced shoots in Campbell and Durzan (CD) medium compared to 60% in Murashige and Skoog (MS) medium. Only 1/3 of them produced multiple shoots. Shoot idtiation from nodal segments was very rare. Mature apices produced callus. Although removed of the sheathing cover around mature buds enhanced the shoot initiation but success rate was low in growth regulator free medium. Embryos respond to the CD medium but not to the MS medium. Embryos from seeds soaked in water for 24 hours produced shoots after 8 weeks of incubation and the success rate was 70% while embryos from dry seeds only produced roots. There was no significant effect of cold storage (refrigeration) for 7 days on shoot initiation from mature embryos (65%) but the ability for shoot induction declines with storage time (55% after 21 days of cold storage). Mature axillary buds were established in Modified Campbell and Durzan (CD) medium supplemented with 0.5mg/1 and IBA. There was a significant difference in the growth performance of shoots according to the period of the year in which explants were collected. Highest (60%) was observed in November-January period. It was only 30% when the explants were collected in February-April or May-July and decreased to 20% in August-October. The shoots produced in November-January showed a higher vigor than those produced in other months. Since Jak fruit show seasonal changes in fruit bearing and shedding of leaves, it can be suggested that the difference in growth performances of tissues cultured in artificial culture media would have been affected by endogenous rhythms.
