Influential Characteristics of Decoction Waters on Extraction of Components in Corydalis Tuber Analyzed by Multiple Factor Analysis (다중요인 분석에 의한 전탕용수에 따른 현호색 성분의 추출효과 비교)

  • The characteristics of decoction water are important for the extraction of herbal drugs. In this study, the relationship between the characteristics of water and extraction efficiency of bioactive compounds from Corydalis tuber was investigated. Six different types of decoction water, including three types of mineral water from Jeju Island and three types of bottled water sold in market, were used as decoction water for the extraction of bioactive compounds from Corydalis tuber. The contents of minerals in six types of water were analyzed by ICP-AES and ICP-MS; the contents of extracted bioactive compounds were analyzed by HPLC. Multivariate analysis of variance was used to analyze the differences in total extraction and bioactive compounds extracted with six types of water. Multiple factor analysis was used to analyze the relationship among the pH, content of mineral, anion, total extractions and content of bioactive components as factors. There was a significant difference among the six types of water in the total extracts and the bioactive compounds. The results proved that the content of extracted compounds from Corydalis tuber was influenced by the types of decoction water.

The Relationship Between Odor Unit and Odorous Compounds in Control Areas Using Multiple Regression Analysis (다중회귀분석을 이용한 악취 관리지역에서의 복합취기강도와 개별악취물질들의 관계에 대한 연구)

  • We investigated a trait of odor and the relationship between odor unit and odorous compounds using multiple regression analysis based on data compiled from Sihwa (SIC), Banwol (BIC), Banwol plating (BPIC) and Poseung industrial complex (PIC). These areas are odor control areas in Gyeonggi province. It was revealed that $NH_3$ and styrene concentrations in SIC and BPIC were relatively higher and $H_2S$ concentration especially in mc was more than five times higher than other areas. As a result of regression analysis using SAS, intensity of odor unit was highly related to concentrations of $H_2S$, TMA, styrene and n-valeraldehyde in SIC, $H_2S$, acetaldehyde, and butyraldehyde in BPIC and $NH_3$ in BIC.

Influences of mineral contents in decoction water on the extraction efficiency of bioactive compounds from Eucommiae Cortex (전탕용수의 미네랄 함량이 두충의 유효성분 추출에 미치는 영향 분석)

  • Objective : Eucommiae Cortex is bark of Eucommia ulmoides Oliver, a deciduous tree native to China. Today, teas or herbal drugs made from E. ulmoides leaves are widely consumed as a healthy food especially in Eastern Asia including Japan, China and Korea. This study was aimed to verify the extraction efficiency of bioactive compounds from Eucommiae Cortex depending on characteristics of decoction water. Method : Six types of decoction water with various origin were prepared and the contents of minerals in water were analyzed by ICP-AES and ICP-MS. The contents of extracted bioactive compounds were analyzed by HPLC and the relationship between mineral contents and extracted bioactive compounds in the decoction water were analysed by multiple factor analysis. The decoction waters could be classified in three groups by hierarchical cluster analysis. Result : Extracted amounts of compounds in Eucommiae Cortex showed a positive relationship with Na, Si, K concentration in decoction water, while a negative relationship with Ca, Mg concentration. Conclusion : The study proved that the minerals in water influenced the extraction efficiency of bioactive compounds from Eucommiae Cortex.

Protection by Histidine Dipeptides against Acrolein-induced Neurofilament-L Aggregation

  • The endogenous dipeptides, carnosine and related compounds, are the naturally occurring dipeptides with multiple neuroprotective properties. We have examined the protective effects of carnosine, homocarnosine and anserine on the aggregation of neurofilament-L (NF-L) induced by neurotoxin, acrolein. When NF-L was incubated with acrolein in the presence of carnosine, homocarnosine or anserine, protein aggregation was inhibited in a concentration-dependent manner. These compounds inhibited the formation of protein carbonyl compounds and dityrosine in acrolein-mediated NF-L aggregates. The aggregates of NF-L displayed thioflavin T reactivity, reminiscent of amyloid. This thioflavin T reactivity was inhibited by carnosine and related compounds. This effect was associated with decreased formation of oxidatively modified proteins. Our results suggested that carnosine and related compounds might have protective effects to brain proteins under pathophysiological conditions leading to degenerative damage such as neurodegenerative disorders.

  • Analyzed are recent advances in design of novel boronared conjugates of synthetic and natural porphyrins and chlorins. These compounds showed high efficacy as cytotoxic agents for tumor cells in culture and as phototoxins in photodynamic therapy of tumor xenografts. Thus, boronated porphyrins and chlorins emerge as promising class of anticancer agents with potentially multiple advantages: the chemotherapeutic drugs alone and photo- and radiosensitizers in binary treatments.

  • Two Known compounds, syringaresinol monoglucoside(8), chimaphilin(14), together with three new compounds, (9)[mp. $106{\sim} 111^{\circ}C$, C_{29}H_{44}O_{13}$], (10)[mp. $180{\sim}182^{\circ}C$, $C_{15}H_{20}O_{9}$] and (11)[mp. $100{\sim}105^{\circ}C$, $C_{18}H_{28}O_{8}$] were isolated from the BuOH fractionof Pyrola japonics(Pyrolaceae). The structures of the known compounds were determined by chemical and spectorscopic methods. The assignments of the $^{1}H-$ and $^{13}C-NMP$ spectra of these compounds were carried out by two-dimensional $^1H-^1H-COSY$, NOESY and $^1H-^{13}C$ multiple-bond, multiplequantum spectroscopic correlation techniques. The characterization of the three new compounds is now in progress.

Removal of Edge Artifact due to Pertial Volume Effect in the Adaptive Template Filtering (적응 템플릿 필터링에서 복셀의 부분 볼륨 효과로 인한 헤지 아티팩트의 제거)

  • Adaptive template filtering has been proposed recently for enhancement of signal-to-noise ratio without loss of resolution. In the adaptive template filtering, an optimal template among multiple templates is selected, then linear least square error filtering based on the template is applied in vowel by vowel basis. In some magnetic resonance imaging, where the distribution of gray level has relatively small dynamic range, e.g., $T_1$ imaging, however, artificial stair-like artifact is observed at near edges. This is partially due to the edge enhancement effect in such yokels that contain multiple compounds at the boundaries of tissues. The gray levels of these yokels become similar gray levels of near dominant vowels that contain single compound by the adaptive filtering, which enlarges edge discontinuities. In this paper, we propose a technique to eliminate such artifact by identifying those yokels that contain multiple compounds and assigning the largest template for them. Filtered images with the proposed technique show substantial visual enhancement at the edges without degradation of peak signal-to-noise ratio compared to the original adaptive template filtering for both magnetic resonance images and phantom images.

Fungal Metabolism of Environmentally Persistent Compounds: Substrate Recognition and Metabolic Response

  • Mechanism of lignin biodegradation caused by basidiomycetes and the history of lignin biodegradation studies were briefly reviewed. The important roles of fungal extracellular ligninolytic enzymes such as lignin and manganese peroxidases (LiP and MnP) were also summarized. These enzymes were unique in their catalytic mechanisms and substrate specificities. Either LiP or MnP system is capable of oxidizing a variety of aromatic substrates via a one-electron oxidation. Extracellular fungal system for aromatic degradation is non-specific, which recently attracts many people working a bioremediation field. On the other hand, an intracellular degradation system for aromatic compounds is rather specific in the fungal cell. Structurally similar compounds were prepared and metabolized, indicating that an intracellular degradation strategy consisted of the cellular systems for substrate recognition and metabolic response. It was assumed that lignin-degrading fungi might be needed to develop multiple metabolic pathways for a variety of aromatic compounds caused by the action of non-specific ligninolytic enzymes on lignin. Our recent results on chemical stress responsible factors analyzed using mRNA differential display techniques were also mentioned.

Study on Photodegradable Water-Soluble Compounds of Expanded Polystyrene

  • Many previous studies have focused on revealing the harmfulness of microplastic particles, whereas very few studies have focused on the effects of chemicals, particularly photooxidation product. In this study, products of photodegradation from expanded polystyrene (EPS), compounds produced by photolysis by ultraviolet (UV) light, were investigated. EPS was directly irradiated and photolyzed using a UV lamp, and then the extracted sample was analyzed using high-resolution mass spectrometry (HRMS). Multiple ionization techniques, including electrospray ionization, atmospheric pressure chemical ionization, and atmospheric pressure photoionization, were used. In total, >300 compounds were observed, among which polystyrene monomer, dimer, and oxidized products were observed. In this work, the data presented clearly demonstrate that it is necessary to identify and monitor oxidized plastic compounds and assess their effect on the environment.

Development of kNN QSAR Models for 3-Arylisoquinoline Antitumor Agents

  • Variable selection k nearest neighbor QSAR modeling approach was applied to a data set of 80 3-arylisoquinolines exhibiting cytotoxicity against human lung tumor cell line (A-549). All compounds were characterized with molecular topology descriptors calculated with the MolconnZ program. Seven compounds were randomly selected from the original dataset and used as an external validation set. The remaining subset of 73 compounds was divided into multiple training (56 to 61 compounds) and test (17 to 12 compounds) sets using a chemical diversity sampling method developed in this group. Highly predictive models characterized by the leave-one out cross-validated $R^2$ ($q^2$) values greater than 0.8 for the training sets and $R^2$ values greater than 0.7 for the test sets have been obtained. The robustness of models was confirmed by the Y-randomization test: all models built using training sets with randomly shuffled activities were characterized by low $q^2{\leq}0.26$ and $R^2{\leq}0.22$ for training and test sets, respectively. Twelve best models (with the highest values of both $q^2$ and $R^2$) predicted the activities of the external validation set of seven compounds with $R^2$ ranging from 0.71 to 0.93.
