Comparison of Genetic Parameter Estimates of Total Sperm Cells of Boars between Random Regression and Multiple Trait Animal Models

  • The objective of this study was to compare random regression model and multiple trait animal model estimates of the (co) variance of total sperm cells over the active lifetime of AI boars. Data were provided by Smithfield Premium Genetics (Rose Hill, NC). Total number of records and animals for the random regression model were 19,629 and 1,736, respectively. Data for multiple trait animal model analyses were edited to include only records produced at 9, 12, 15, 18, 21, 24, and 27 months of age. For the multiple trait method estimates of genetic and residual variance for total sperm cells were heterogeneous among age classifications. When comparing multiple trait method to random regression, heritability estimates were similar except for total sperm cells at 24 months of age. The multiple trait method also resulted in higher estimates of heritability of total sperm cells at every age when compared to random regression results. Random regression analysis provided more detail with regard to changes of variance components with age. Random regression methods are the most appropriate to analyze semen traits as they are longitudinal data measured over the lifetime of boars.

A Case of Disseminated Multiple Glomus Tumors (파종성 다발성 사구종양 1례의 치험례)

  • Purpose: Glomus tumors are neoplasms that are composed of modified smooth muscle cells of the glomus body and multiple glomus tumor comprises 10% of all glomus tumors. We report a case of disseminated multiple glomus tumors. Methods: A 14 - year - old boy presented with multiple subcutaneous purple nodules on the right cheek, back, right arm, right hand dorsum, right fourth finger, and left ankle. Nodules on the back and right fourth finger were completely excised under local anesthesia and histopathologic examination was followed. Results: Histopathologic findings showed numerous dilated, cavernous - like, thin - walled, vascular spaces surrounded by one or a few layers of glomus cells. On immunohistochemical examination, glomus cells stain for smooth muscle actin, and endothelial cells stain for CD31. Those revealed multiple glomangiomas. Conclusion: A review of Korean literature revealed only one reported cases of disseminated multiple glomus tumors, so this is the second case to be reported in the Korean literature. In case of multiple soft tissue tumors, thorough physical examination and preoperative evaluation is needed.

Development of W/O/W Multiple Emulsion Formulation Containing Burkholderia gladioli

  • W/O/W (water-in-oil-in-water) type multiple emulsion was applied to improve the storage stability of an antagonistic microorganism, Burkholderia gladioli. Encapsulation of microorganism into a W/O/W emulsion was conducted by using a two-step emulsification method. W/O/W emulsion was prepared by the incorporation of B. gladioli into rapeseed oil and the addition of polyglycerin polyriconolate (PGPR) and castor oil polyoxyethylene (COG 25) as the primary and secondary emulsifier, respectively. Microcrystalline cellulose was used as an emulsion stabilizer. To evaluate the usefulness of W/O/W emulsion formulation as a microbial pesticide for controlling the bacterial wilt pathogen (Ralstonia solanacearum), the storage stability and antagonistic activity of emulsion formulation were tested in vitro. The storage stability test revealed that the viability of formulated cells in emulsion was higher than that of unformulated cells in culture broth. At $4^{\circ}C$, the viabilities of formulated cells and unformulated cells at the end of 20 weeks decreased to about 2 and 5 log cycles, respectively. At $37^{\circ}C$, the viability of formulated cells decreased to only 2 log cycles at the end of storage. On the other hand, the viable cells in culture broth were not detected after 13 weeks. In activity test, formulated cells in emulsion were more effective in inhibiting the growth of pathogen than unformulated cells in culture broth. Unformulated cells completely lost their antagonistic activity during storage under similar conditions. The W/O/W multiple emulsion formulation was shown to be useful as the novel liquid formulation for biological control.

  • Somatic embryogendesis is one of good examples of the basic research for plant embryo development as well as an important technique for plant biotechnology. This paper describes the direct somatic embryogenesis from zygotic embryos of Panax ginseng is reversely related to normal axis growth of zygotic embryos by the experiment of various chemical treatments. Under the normal growth condition, the apical tips of embryo axis produced an agar-diffusible substance, which suppressed somatic embryo development from cotyledons. Although the cells of zygotic embryos were released from the restraint of embryo axis, various factors were still involved for somatic embryo development. Electron microscopic observation revealed that the ultrastructure of cells of cotyledon epidermis markedly changed before initiation of embryonic cell division, probably indicating reprogramming events into the cells embryogenically determined state. Polar accumulation of endogenous auxin or cell-cell isolation by plasmolysis pre-treatment is the strong inducer for the somatic embryo development. The cells for the process of somatic embryogenesis might be determined by the physiological conditions fo explants and medium compositions. Direct somatic embryos from cotyledons fo ginseng were originated eithrer from single or multiple cells. The different cellular origin of somatic embryos was originated either from single or multiple cell. The different cellular origin of somatic embryos was depended on various developmental stages of cotyledons. Immature meristematic cotyledons produced multiple cell-derived somatic embryos, which developed into multiple embryos. While fully mature cotyledons produced single cell-derived single embryos with independent state. Plasmolysis pretreatment of cotyledons strongly enhanced single cell-derived somatic embryogenesis. Single embryos were converted into normal plantlets with shoot and roots, while multiple embryos were converted into only multiple shoots. GA3 or a chilling treatment was prerequisite for germination and plant conversion. Low concentration of ammonium ion in medium was necessary for balanced growth of root and shoot of plantlets. Therefore, using above procedures, successful plant regeneration of ginseng was accomplished through direct single embryogenesis, which makes it possible to produce genetically transformed ginseng efficently.

  • Multiple solutions in natural convection of air (Pr=0.7) between two horizontal walls with mean temperature difference and the same periodic nob-uniformities are investigated. An analytical solution is found for small Rayleigh number, and the general solution is investigated by using a numerical method. In the conduction-dominated regime, two upright cells are formed between two walls over one wave length. When the wave number is small, the flow becomes unstable with increase of the Rayleigh number, and multicellular convection occurs above a critical Rayleigh number. The multicellular flows at high Rayleigh numbers consist of approximately square-shape cells. And several kinds of multiple flows classified by the number of cells are found.

Harmonic Suppression and Broadening Bandwidth of Band Pass Filter Using Aperture and Photonic Band Gap Structure

  • In this paper, we introduced a band-pass filter employed the PBG structure and the aperture on the ground together. The harmonics of band pass filter have been suppressed by employing the PBG structure and the bandwidth of it has been broadened by using the aperture on the ground. The designed PBG cells have three different sizes. The largest cells, the middle cells, and the smallest cells have suppressed the multiple of second harmonics, the multiple of third harmonics, and the multiple of fifth harmonics, respectively. The center frequency has been 2.18 GHz. The bandwidth has been increased from 230 MHz up to 310 MHz(80 MHz, about $35\%$) by the aperture and the ripple characteristics in passband have been improved and the harmonic frequencies have been suppressed about 30 dB by the PBG.

Successful engraftment after infusion of multiple low doses of CD34+ cells from a poorly matched sibling donor in a patient with severe aplastic anemia

  • The dose of CD34+ cells is known to influence the outcome of allogeneic peripheral blood stem cell (PBSC) and/or T-cell-depleted transplantation. A previous study proposed that $2{\times}10^6\;CD34+\;cells/kg$ is the ideal minimum dose for allogeneic transplantation, although lower doses did not preclude successful therapy. In the case we present here, CD34+ cells were collected from a matched sibling donor on the day of allogeneic hematopoietic stem cell transplantation; however, the number of cells was not sufficient for transplantation. Consequently, PBSCs were collected three additional times and were infused along with cord blood cells from the donor that were cryopreserved at birth. The cumulative dose of total nuclear cells and CD34+ cells was $15.9{\times}10^8\;cells/kg$ and $0.95{\times}10^6\;cells/kg$, respectively. White blood cells from this patient were engrafted on day 12. In summary, we report successful engraftment after infusion of multiple low doses of CD34+ cells in a patient with severe aplastic anemia.

  • A case of multiple myeloma with massive pleural effusion is reported. A 53 year-old previous known multiple myeloma patient vistited our hospital complaining of cough with sputum. Radiologic study revealed multiple osteolytic bony lesions and left side pleural effusion. The effusion were bloody exudates containing numerous atypical plasma cells. The tumor cells showed pleomorphism, eccentric nuclei, prominent nucleoli, perinuclear halo, multincleation, and chromatin patterns of occasional cart-wheel appearance. The cytological examination of pleural fluid established the malignant nature of the effusion with multiple myeloma.

Stimulatory Effects of Ginsan on the Proliferation and Viability of Mouse Spleen Cells

  • Ginsan is an acidic polysaccharide purified from Panax ginseng, a famous oriental herb. Although a variety of biological activities of ginsan have been studied, the effects of ginsan on spleen cells are not fully elucidated. We investigated the effect of ginsan on the viability and proliferation of spleen cells. Using Cell Counting $Kit-8^{(R)}$ solution and trypan blue solution, we found that ginsan significantly enhanced viability and proliferation. Multiple clusters, indicating proliferation, were observed in ginsan-treated spleen cells and, carboxyfluorescein succinimidyl ester and surface marker staining assay revealed that ginsan promoted proliferation from $CD19^+$ B cells rather than $CD4^+$ or $CD8^+$ T cells. In addition, ginsan decreased the percentage of late apoptotic cells. Ginsan increased the surface expression of CD25 and CD69 as well as production of interleukin-2 from spleen cells, suggesting increased activation. Taken together, these results demonstrate that ginsan increases the viability and proliferation of spleen cells via multiple mechanisms, valuable information for broadening the use of ginsan in clinical and research settings.

A Flexible Conveying System using Hybrid Control under Distributed Network

  • In this research, we propose a flexible conveying system (FCS) which consists of multiple arrays of cells. Each cell is a wheel driven by a two degree-of-freedom mechanism. The direction and velocity of cell are controlled based on the concept of hybrid control under a distributed network. Each cell has its own controller under a subsumption architecture for low-level control. A cell communicates with its four neighboring cells to manipulate n targeted object towards its desired position. The high-level control assigns a desired position and direction of the object to each cell. The path of each object is generated by many supporting cells. Moreover, the FCS can handle multiple objects simultaneously. To study the flexible conveying system, a GUI-based simulator of flexible conveying system is constructed. The simulated results show that the system can handle multiple objects independently and simultaneously under the proposed hybrid control architecture.
