This study aimed to monitor the distribution of Cobitidae in Korea by the identification of species using genetic analysis. Based on the genetic analysis, Cobitidae species in four of five domestic fish farms consisted of only Chinese muddy loach Misgurnus mizolepis, but muddy loach Misgurnus anguillicaudatus was also present it in one fish farm. In the case of imported Cobitidae species, in addition to Chinese muddy loach and muddy loach, the harmful species Paramisgurnus dabryanus, was also present. Chinese muddy loach accounted for 20%, 67%, and 60% of the S6, S7, and S8 samples, respectively. An analysis of the total length, body length, and weight showed that domestic Chinese muddy loach showed higher values than imported muddy loach, and imported Chinese muddy loach showed similar values to P. dabryanus. There were no significant differences in the country of origin of the three species. Thus, the mitochondrial cytochrome c oxidase subunit I gene sequence was analyzed and compared the verification of species identification. The three species of Cobitidae were genetically divided into three groups and determined to have genetic differences. These results indicate that it is necessary to reduce the heterogeneous mixing rate through discriminating species by genetic analysis.
Differences in carotenoid composition in the integuments of wild and cultured chinese muddy loach Misgurnus mizolepis and muddy loach Misgurnus anguillicaudatus were compared. Total carotenoid contents in the integuments of the wild and cultured chinese muddy loach were $4.76mg\%\;and\;3.43mg\%$, respectively. The important carotenoids in the integuments of the wild chinese muddy loach were lutein($30.5\%$), ${\beta}$-cryptoxanthin($24.6\%$), ${\beta}$-carotene($20.6\%$) and cynthiaxanthin($11.7\%$). In addition, zeaxanthin($4.7\%$), tunaxanthin ($4.5\%$), and a-cryptoxanthin($1.0\%$) were present in small amounts. In the integuments of the cultured chinese muddy loach, lutein($35.4\%$), ${\beta}$-cryptoxanthin($17.9\%$), cynthiaxanthin($16.0\%$) and ${\beta}$-carotene($12.7\%$) were present as important carotenoids. In addition, zeaxanthin($8.1\%$), tunaxanthin($5.0\%$), a-cryptoxanthin($0.9\%$) were found in small amounts. Total carotenoid contents in the integuments of the wild and cultured muddy loach were $4.00mg\%\;and\;2.99mg\%$, respectively. The important carotenoids in the integuments of the wild muddy loach were lutein($32.9\%$), ${\beta}$-cryptoxanthin($18.8\%$), cynthiaxanthin($17.0\%$) and ${\beta}$-carotene($15.1\%$). In addition, zeaxanthin($6.5\%$), tunaxanthin($6.0\%$) and a-cryptoxanthin($1.5\%$) were found in small amounts. In the integuments of the cultured muddy loach, lutein($51.8\%$), cynthiaxanthin($19.9\%$) and ${\beta}$-cryptoxanthin($10.8\%$) were observed as important carotenoids. In addition, ${\beta}$-carotene($5.0\%$), zeaxanthin($4.8\%$), tunaxanthin($4.5\%$) and a-cryptoxanthin($0.2\%$) were found in small amounts.

A feeding trial was conducted to determine the optimum dietary protein and lipid levels for the growth of juvenile muddy loach Misgurnus anguillicaudatus. Eight experimental diets (designated as P20L7, P20L14, P30L7, P30L14, P40L7, P40L14, P50L7 and P50L14) were formulated to contain 20%, 30%, 40% and 50% protein with 7% and 14% lipid. Three replicate groups of muddy loach (1.5 g/fish) were fed one of the experimental diets ad libitum for 8 weeks. Survival of fish fed the P20L14 diet was lower than in other groups (P<0.05). Specific growth rate (SGR) and feed efficiency (FE) of fish were affected by dietary protein (P<0.05) and lipid (P<0.005) levels. SGR and FE of fish tended to decrease with increasing dietary lipid level at the same protein level. There were no significant differences in SGR and FE among fish fed diets containing different protein levels ranging from 30% to 50% with the same lipid level. Whole body moisture and lipid contents were affected by dietary protein and lipid levels. Crude lipid content of fish fed the 14% lipid diets was higher than that of fish fed 7% lipid diets at each protein level. The results of this study indicate that a diet containing 30% protein with 7% lipid would be suitable for the optimum growth and effective feed utilization of juvenile muddy loach.
We designed a 16-week feeding experiment to test the effects of dietary supplementation with distillers' dried grain (DDG) on the growth, feed utilization, and body composition of juvenile muddy loach Misgurnus anguillicaudatus. Seven isonitrogenous and isocaloric diets were formulated to contain 0% DDG (Control); 10%, 20%, and 30 % DDG from rice (diets RM10, RM20 and RM30); and 10%, 20%, and 30% DDG from rice and wheat flour (diets RWM10, RWM20 and RWM30), respectively. Three replicate groups of juvenile muddy loach averaging $1.5{\pm}0.10$ g were fed one of the diets to satiation twice daily. Survival of juvenile muddy loach fed the RWM20 and RWM30 diets was higher than that of the control group (P<0.05), while weight gain tended to decrease when fish were fed diets containing DDG. Weight gain of fish fed the RWM10 diet was greater than that of fish fed other diets, but the RM30 diet resulted in less weight gain than did the control (P<0.05). The feed and protein efficiency ratios of fish fed the RM10, RM20, RM30, RWM10, RWM20 and RWM30 diets were all similar to the control (P>0.05). Neither daily feed intake nor proximate composition of the whole body was affected by dietary DDG (P>0.05). The compositions of isoleucine, leucine, lysine, threonine, and valine were all elevated in fish fed the RM30, RWM10, RWM20 and RWM30 diets relative to those of other groups (P<0.05). In contrast, methionine + cystine and phenylalanine + tyrosine were all lower in fish fed the RM30, RWM10, RWM20 and RWM30 diets, as compared to those of other groups (P<0.05). Our results suggest that DDG is suitable as a partial replacement for soybean meal and wheat flour, and could be used at a rate of up to 20% for rice, or 30% for rice and wheat flour, for optimum growth performance of juvenile muddy loach.
On September in 2010, myxosporidian parasites were found on the fins of Chinese muddy loach, Misgurnus mizolepis, Several cysts of approximately 2 mm diameter were observed on the fins. Numerous spores were found in the cyst. The total length of the spore was 13.0 to 15.8 ${\mu}m$, and the width of spore was 6.0 to 7.5 ${\mu}m$. As the results, the myxosporidians were finally identified as Thelohanellus misgurni (Kudo, 1919) on the basis of the presence of characteristic cysts and size of spores.

The objective of the present study was to investigate fish farm and growth performances of Chinese muddy loach (Misgurnus mizolepis) under indoor and outdoor aquaculture conditions for the domestic loach aquaculture industry. The investigation was conducted from August 2017 to October 2019 at one indoor farm (D-site) and two outdoor farms (P1-site and P2-site). Results showed that WG (weight gain), DWG (daily weight gain), and SWG (specific weight gain) based on growth measurements were 175.27±36.2%, 7.98±3.3%, and 0.94±0.2% at D-site, 320±34%, 4.07±0.6%, and 0.55±0.1% at P1-site, and 295.9±53%, 4.15±0.5%, and 0.62±0.1% at P2-site, respectively. WG was higher in outdoor farms (P1-site and P2-site), while DWG and SWG were higher in the indoor farm (D-site). Such differences of the present study might be due to differences in growth, water quality, plant and sale frequency depending on aquaculture types. In particular, outdoor farms takes more than a year to have sales whereas indoor farms can have sales three times in a year.
During the winter, the rice field-dwelling muddy loach Misgurnus mizolepis is buried in burrows constructed of mud and are subjected to exposure to air at times of shortage of water. To investigate the environmental factors that lead to changes of the skin mucus cells of the muddy loach in rice fields, we carried out an experiment where we artificially exposure the fish to air, duplicating as close as possible winter conditions in nature. During the summer, a water tank containing M. mizolepis was filled with mud, and the water was allowed to evaporate. After a month of evaporation, the loach constructed burrows similar to those in a winter rice field. The epidermis in the experimental fish was mostly occupied by large elongated mucus cells, whose numbers drastically increased in all observed regions of the dorsum, lateral region, and the occiput. Such features are typically seen in fishes in wild habitats during the winter season.
Promoting the organic farming, much of the plant extracts used for controlling pests and fungi have been imported from China, India and Myanmar. But, it is so worrisome that aquatic animals such as muddy loach inhabiting in paddy field and common carps in river exposed to the pests and fungi likely be harmed. This study was conducted in order to evaluate the risks of aquatic animals influenced by the three plant extracts, i.e. Sophora flavescens, Azadirachta indica and Derris elliptica. The toxicities of common carp (Cyprinus Carpio), muddy loach (Misgurnus anguillicaudatus) and PEC (Predicted environmental concentration) exposed to the three plant extracts were estimated by the typical spray volume method. Risks were determined by the toxicity value as 48-hr $LC_{50}$ (Lethal concentration, median) or NOEC (No observed effect concentration) into PEC. 48-hr $LC_{50}$ of Common carp and NOEC by Sophora flavescens extracts was 7.9 and 6.2 mg/L, 26.8 and 21.8 mg/L by Azadirachta indica extracts and 47.0 and < 24.0 mg/L by Derris elliptica extracts, respectively. 48-hr $LC_{50}$ of Muddy loach and NOEC by Sophora flavescens extracts was 16.9 and 10.0 mg/L, 35.6 and 30.0 mg/L by Azadirachta indica extracts, and 73.9 and < 40 mg/L by Derris elliptica extracts, respectively. Therefore, acute toxicities of the three plant extracts for aquatic animals were proved to be very low level. PEC of Sophora flavescens extracts in paddy, drainage and river water was 68.0~3.0, 11.33~0.50 and 3.0~0.0018 mg/L, respectively. TER of Sophora flavescens extracts in the three water was 0.2~5.6, 1.5~33.8 and 2.6~4388.9, respectively. PEC of Azadirachta indica extracts in paddy, drainage and river water was 90.9~1.2, 15.2~0.2 and 4.8~0.00075 mg/L, respectively. TER of Azadirachta indica extracts in the three water was 0.4~29.7, 2.3~178.0 and 4.5~35733.3, respectively. PEC of Derris elliptica extracts in river water was 0.0063 mg/L. TER of Derris elliptica extracts in river water was 5222~15667.
To evaluate the effect of endocrine disruption chemicals (EDCs) to aquatic organisms, muddy loach (Misgurnus anguillicaudatus) was exposed to low concentration methomyl for 21 days in order to identify the effect of biomarkers and endocrine. Vitellogenin (VTG) in blood plasma, which used widely as validated biomarker for endocrine disruption, was significantly greater in male fish exposed to 0.4 mg/L and 2 mg/L methomyl, and in female fish exposed to 0.08 mg/L, 0.4 mg/L, and 2 mg/L methomyl for 21 days (p<0.05). This results suggest that methomyl have probability of endocrine disruption to organism on aquatic system. While inhibition of Acetylcholinesterase (AChE) activity and increase of DNA damage in comet assay were verified by fish exposed to methomyl, change of ethoxyresorufin-O-deethylase (EROD) activity was not occurred, comparing the control group (p<0.05). Indicators at the level of organism such as condition factor (CF), hepato-somatic index (HSI), and gonado-somatic index (GSI) were not influenced by exposure of methomyl. In conclusion, these results showed the possibility of methomyl in regard to not only endocrine disruption but also impacts on biochemical biomarkers to aquatic organisms.
The histological morphology on the skin of Misgurnus anguillicaudatus was described in the three regions such as dorsal, lateral, occiput and subsequently morphological variations of the skin were monthly observed for a year. The skin consisted of epidermis having epithelial cell, club cell and mucus cell, and dermis of mainly connective tissue fiber, embedded scale and blood capillary. Unicellular mucus cells situated at the epidermis underwent seasonal change in its size, as well as number and amounts of mucus-secreting materials, which they greatly increased in winter, but did not in summer. As it is getting cold, the mucus cells' shape changed from initial spherical to oval or elongated form. Such considerable changes in the mucus cell were particularly most evident in the occiput during winter. Moreover, the dermis largely thickened about 2~3 times in winter than in summer. Based on these results, we discussed function for the mucus on what it mainly acts in nature and information on whether mucus cells' seasonal variations affect on hibernation and cutaneous respiration.
