Histochemical Properties Study on the Mucosubstances of the Intestinal Mucosa in the Raja kenojei (홍어 장점막 점액의 조직화학적 성상에 관한 연구)

  • This study is performed in order to clarify the histochemical structure, the distribution of mucous cell and goblet cell, and the histochemical properties of the mucosubstances in the middle region of intestinal mucosa and rectum of Raja kenojei. In the H&E stain, distribution of the mucous cells and acidophilic cells were a more compacted middle portion than other regions of intestine, but the former was more than the latter in the number of mucous cells to rectum. The mucosubstances were stained with aldehyde fuchsin pH 1.7-alcian blue(pH 2.5) stain and then compared to distribution of the mucosubstances used in image and microscope technology(IMT-Size5). The middle intestine of Raja kenojei was composed of mucous cells having only large amounts of mucosubstances in the distal region was much more than that of proximal region. It was two types of mucous cells to rectum, one type was the same as proximal intestine while the other had small amounts of weakly sulfated and large amounts of carboxylated mucins.

  • The species of the slug used in this experiment is the Korean terrestrial slug (Incilaria fruhstorferi), which is examined for the cytochemical and ultrastructural research on the mucous granule-producing cells and the epithelial cells. I. Epidermal tissue According to the part of the epidermal tissue of this slug, the epidermal tissue is divided into the mantle, the foot and the dorsal epidermis. These epidennal tissue are composed of the irregular simple columnar epithelium, which are formed into the sensory epithelial cells, the supporting epithelial cells, the mucous granule-producing cells, and the clear epithelial cells are similar to the sensory epithelial cells. Both the sensory epithelial cells and the supporting epithelial cells are observed between the mantle and the foot epidermis, but the clear epithelial cells are only seen in the dorsal epidermis. II. Mucous granule-producing cell The acid mucous granule-producing cells and the neutral mucous granule producing cells are observed between the irregular simple columnar epithelium of the mantle, the foot and the dorsal epidermis. According to the part of the epidermal tissue, the number of these mucous granule-producing epithelial cells are differently distributed between the epidermis respectively.

Morphology and Histochemistry of the Skin of the Mud Loach, Misgurnus mizolepis, in Relation to Cutaeneous Respiration

  • The skin structure of Misgurnus mizolepis was studied based on the microanatomical investigation of skin fragments taken from four regions. The epidermis was distinguished by two types of skin glands, a small mucous cell and a large club cell. The mucous cell was acid sulfomucins (some sialomucins) but the club cell did not give any histochemical tests for mucosubstances. The presence of a well defined lymphatic system with small lymphocytes was established in the stratum germinativum layer of the epidermis. A large number of blood capillaries run very close to each other just below the basement membrane, and a definite area giving AB and PAS positive was present between the basement membrane and scale. These structural features of skin in M. mizolepis seem to be closely related with cutaneous respiration.

  • Skin mucous cells in the three regions of the body in Pseudobagrus fulvidaraco and Leiocassius nitidus were investigated using three histochemical methods (PAS, AB-PAS and HID). In the two species, components of mucous cell were not distinguished in each region, but presented a differences in position and type, size, and count in all regions. Mucous cells of P. fulvidraco were located in two layers, a superficial epithelial cell and the space within alarm substance cells (ASCs). Mucous cells of L. nitidus were situated only in the superficial epithelial cell layer. The size of mucous cells in the ASC layer were larger than those of the superficial epithelial cells for all measured values: diameter, length, width, and area. In the superficial epithelial cell layer, the average number of mucous cells within the three regions did not differ significantly between species by unpaired T-test, but the average number of those within two cell layers of P. fulvidraco had a similarity at each region by paired T-test. The dismilarity in dorsal and ventral regions in average number of ASCs between P. fulvidraco and L. niditus was confirmed by unpaired T-test.

  • This study investigated the relationship between attachment site preference of the skin parasite, Entobdella hippoglossi and mucous cell density, moucus cell size and epidermis thickness on the surface of the Atlantic halibut Hippoglossus hippoglossus. Parasites occupying the ventral surface of their host were significantly longer and wider than those found on the other zones of the fish (P<0.05). The mean size of the mucous cells on the front region was significantly greater than the other regions on the dorsal and ventral surface (P<0.05). The average numbers of mucous cells and the epidermal thickness in the skin of the halibut were shown that the front region had significantly higher numbers of mucous cells and thicker layer than the rear region on the dorsal and ventral surface of the halibut, respectively (P<0.05).

  • The ultrastructures of the mucous glands in the Asiatic land salamander (Hynobius leechi) skin were observed by means of electron microscope. The results were as follows; 1. The mucous gland of the Asiatic land salamander skin consisted of a body of gland and a duct. The body of gland consisted of the glandular epithelial and the myoepithelial cells. 2. The light, the dark, the mitochondria-rich, the endoplasmic reticulum-developed, the secretory granule-containing and the vesiculated cells were observed in the mucous glandular epithelium. 3. It is supposed that the light, the dark and the mitochondria-rich cells are undifferentiated cells, the endoplasmic reticulum-developed cell is pre-secretory cell, the secretory granule-containing cell is mature cell and the vesiculated cell is evacuated cell.

  • The ultrastructure of the gills of Oplegnathus fasciatus was examined by means of light and transmission electron microscopes. The gills have primary and secondary filaments (lamellae). The following cells are identified and described : epithelial cell, pillar cell, chloride cell and mucose cell. The simple epithelial layer consists of squamous epithelium containing a large nucleus and the surface is covered with some of microridges. The lamella pillar structures are characterized by axial microtubules and lateral membrane interdigitations. Chloride cells contain a lot of mitochondria and specifically developed tubular systems. The rough endoplasimic reticulum and golgi complex, and some of mucous granules were observed in immature mucous cells. The mature mucous cells were AB-PAS positive, globular in shape, and had mucous granules of similar size with various electron densities.

Effects of Mokdanpijihwang-tang on gastric mucosal damage in mice (목단피지황탕(牧丹皮地黃湯)이 위점막손상(胃粘膜損傷)에 미치는 영향(影響))

  • 1. Back ground and purpose An experimental study has done to examine the effect of defense and cure gastric mucasal damage of Mokdan-pijihwang-tang. 2. Methods Mice had intragastric injected with MJ extract before indomethacin treatment which induces homorrhage infarct and erosion artificially. Degree of lipid peroxidation, general morphology, change of mucous cell, the distribution of PNA, ICAM and distribution of apoptotic cell were objected. (Abbreviation) MJ : Mokdanpijihwang-tang, PNA : Peanut Agglutinin, ICAM : Intercellular Adhesion Molecule 3 Results 1) The degree of lipid peroxidation in INDO-group had increased conspicuously than control group. But the degree of lipid peroxidation in MJ-group had decreased than INDO-group and these decline had probability. 2) After indomethacin treatment, hemorrhage infarct and erosion had increased in stomach body. But in MJ-group, the configuration is normal, except the group intragastric injected with MJ extract at hour 24 before indomethacin treatment. 3) Surface mucous cell and neck mucous had disappeared in INDO-group. But in MJ-group tormal distribution had shown like control group except the group intragastric injected with MJ extract at hour 24 before indomethacin treatment. 4) PNA positive reaction had not shown in INDO-group. But medium PNA positive reaction had shown In Mj-group. 5) ICAM positive reacted cell had shown in INDO-group. The decrease of ICAM positive cell were shown than INDO-group. 6) A number of apoptotic cell was distributed in hemorrhagic erosion. A few number of apoptotic cell was distributed in MJ-group except some surface mucous. 4. Conclusion These results suggest that MJ has an effect on cure of gastric mucosal damage.

  • We investigated ultrastuctural change of small intestinal mucosal epithelial cell, columnar cell and mucous cell, of hibernating ground squirrel during activating and hibernating stages. In active columnar cells, many mitochondria and rough endoplasmic reticulum were observed. In hibernating columnar cells, more free nhosome than rough endoplasmic reticulum were observed. In active mucous cells, large and many mucosal granules, mitochondria and rough endoplasmic reticulum were observed. Mucosal granules have been secreted excellently. In hibernating mucous cells, small and little mucosal granules and many free ribosome were observed.

  • The histochemical characteristics of the cutaneous mucous glands and its fine structure of the Korean fire bellied toad, Bombina orientolis Boulenger, were studied with light and electron microscopes. The mucous glands of the toad are simple alveolar gland, and are composed of inner glandular epithelial cells and outermost myoepithelial cells. Histochemical studies suggest that main secretory materials released from the mucous glands might be acid mucopolysaccharides which is more abundant in the ventral skin than in the dorsal skin. The scanning electron microscopic observation showed that the morphology of the opening sites of the gland duct is similar to the morphology of stomata on the plant leaves. Glandular epithelial layers of the mucous glands are composed of two cell types, the mucous secreting cells and the mitochondria rich cells. The mucous secreting cells could be classified according to the morphology and inner textures of cytoplasm and those characteristics were seemed to be related to the degree of maturation of the cells. Furthermore the mucous within the mucous secreting cells appears to be synthesized in the rough endoplasmic reticulum.
