Structure and Function of the Genes Involved in the Biosynthesis of Carotenoids in the Mucorales

  • Carotenoids are widely distributed natural pigments which are in an increasing demand by the market, due to their applicatins in the human food, animal feed, cosmetics, and pharmaceutical industries. Although more than 600 carotenoids have been identified in nature, only a few are industrially important (${\beta}$-carotene, astaxanthin, lutein or lycopene). To date chemical processes manufacture most of the carotenoid production, but the interest for carotenoids of biological origin is growing since theire is an increased public concern over the safety of artificial food colorants. Although much interest and effort has been devoted to the use of biological sources for industrially important carotenoids, only the production of biological ${\beta}$-carotene and astaxanthin has been reported. Among fungi, several Mucorales strains, particularly Blakeslea trispora, have been used to develop fermentation processes for the production of ${\beta}$-carotene on almost competitive cost-price levels. Similarly, the basidiomycetous yeast Xanthophyllomyces dendrorhous (the perfect state of Phaffia rhodozyma), has been proposed as a promising source of astaxanthin. This paper focuses on recent findings on the fungal pathways for carotenoid production, especially the structure and function of the genes involved in the biosynthesis of carotenoids in the Mucorales. An outlook of the possibilities of an increased industrial production of carotenoids, based on metabolic engineering of fungi for carotenoid content and composition, is also discussed.

한국전통 식품의 원료인 메주와 누룩에서 분리된 접합균에 대한 연구 (Studies on the Fungal Isolates of Mucorales Collected from Korean Home Made Mejus and Nuluks)

  • The fungal isolates of Mucorales, directly collected from Korean traditional raw materials of Nuruk (raw material for Korean rice wine) and Meju (raw material for Korean soysauces), were compared with those of Rhizopus oryzae purchased. The fungal isolates of Rhizopus, Mucor, and Absidia mostly identified as based on the morphological observations, were evaluated with the PCR-polymorphic bands. The PCR-polymorphic bands of the genomic DNA reacted with the primers of OPD series tenmer were various, but showed averaged 4 to 6 in the agarose-electrophoresis. The dissimilarity coefficient (DC) between two isolates were compared by the cluster analyses, dendrogams and polar ordinations. The isolates of R. oryzae known. showed several groupings within the lower value of DC and were divided to two groups of amylo-process and other fungi with other purposes. The isolates unidentified were identified by the DC made of this results. Taxonomy of these isolates made by the morphological observations were consistent with those resulted above in most case but not in all aspects. More works were needed with the isolates known for detail informations of Mucorales.

Isolation and Characterization of Two Rare Mucoralean Species with Specific Habitats

  • The order Mucorales, the largest in number of species within the Mucoromycotina, comprises typically fast-growing saprotrophic fungi. During a study of the fungal diversity of undiscovered taxa in Korea, two novel mucoralean strains, CNUFC-GWD3-9 and CNUFC-EGF1-4, were isolated from specific habitats including freshwater and fecal samples, respectively. On the basis of their morphological characteristics and sequence analyses of internal transcribed spacer and large subunit ribosomal DNA, the CNUFC-GWD3-9 and CNUFC-EGF1-4 isolates were confirmed to be Gilbertella persicaria and Pilobolus crystallinus, respectively. It is ecologically, pathologically, and mycologically significant to find such rare zygomycetous fungi in such specific habitats.

Characterization of Two New Records of Mucoralean Species Isolated from Gut of Soldier Fly Larva in Korea

  • While surveying the diversity of fungi of the order Mucorales, two isolates, EML-PUKI12-1 and EML-PUKI06-1, were obtained from the gut of soldier fly larvae inhabiting the bulrush at a pond located in the Chonnam National University Arboretum, Gwangju, Korea. The isolates were confirmed as Mucor irregularis and Mucor fragilis species, respectively, based on the morphological characteristics and phylogenetic analysis of rDNA internal transcribed spacer region. Such mucoralean species belonging to undiscovered taxa has not previously been described in Korea.

  • During an investigation of fungi of the order Mucorales from freshwater and sediment samples in Korea, we isolated six strains, NNIBRFG6649, NNIBRFG6255, NNIBRFG1498, CNUFC-YJ13, CNUFC-YR7, and NNIBRFG2739. The morphology and phylogeny of these strains were analyzed. Based on the morphological characteristics and molecular data from internal transcribed spacer (ITS) region, the isolates NNIBRFG6649 and NNIBRFG6255 were identified as Mucor abundans, and M. aligarensis, respectively. The isolates NNIBRFG1498 and CNUFC-YJ13 were identified as M. moelleri, whereas the isolates CNUFC-YR7 and NNIBRFG2739 were identified as M. heterogamus. To the best of our knowledge, M. abundans, M. aligarensis, M. moelleri, and M. heterogamus have not yet been reported in Korea.

닭에 집단 발병한 뮤코마이코시스 (Mucormycosis in a group of chickens)

  • Mucormycosis는 자연 환경속에 상채하고 있는 Mucorales 목의 곰팡이류에 의해 발생하는 질병인데, 강원도 속초시의 한 양계장에서 82수(연령 100일~2년)의 닭에 집한 발병하였다. 임상중상은 식욕감퇴, 하리, 영양실조, 호흡곤란, 마비 등으로 발병 후 수 주간에 폐사하였으며, 부검시 호흡기관을 비롯하여 간장, 심장, 비장, 신장, 소화관 등에서 회황색의 다발성 결절(0.14~4.6cm)과 소출혈반점을 발견하였다. 병리 조직학적으로 병소는 화농성 혹은 건락성 괴사소와 이를 둘려싸는 위산성 백혈구, 대식세포, 임파구, 거대핵세포의 침륜 등 급성 육아종성염증 소견을 나타내었으며, 본균 특이성의 균사(두께 $4{\sim}24{\mu}$)를 괴사병소 내에서 관찰하였다. 본 질병의 발생은 사료 첨가제와 치료제로써 항생제의 남용이나 타질병의 일차 감염 등이 전소인으로 작용하는 것으로 사료된다.

국내 기수역 환경의 균류 다양성 (Diversity of Fungi in Brackish Water in Korea)

  • 본 연구에서는 국내 기수 환경의 물과 토양에서 균류의 다양성을 확인하기 위해 을숙도, 금강하구둑, 순천만습지, 대호방조제, 신두리사구 및 부남방조제 총 6지점의 현장조사 및 시료를 채집하였다. 현장에서 여과한 담수시료와 담수퇴적물로부터 곰팡이를 순수분리하여 PDA배지에 접종한 후 25℃에서 배양하였다. 기수역 시료에서 순수분리된 균주는 총 173균주로 ITS 부분을 분석하여 속 수준으로 동정하였다. 분리 동정된 균주를 다양성 지수를 이용하여 분석하였다. 분리된 균류는 Pleosporales, Eurotiales, Capnodiales, Hypocreales, Polyporales, Saccharomycetales, Agaricales, Glomerellales, Mucorales, Dothideales, Russulales, Xylariales, Sordariales, Myrmecridiales, Tubeufiales, Onygenales, Cantharellales, Amphisphaeriales 목에 속하며 Cladosporium속이 20%로 가장 우점하며, 그 다음은 Penicillium (19%), Fusarium (5%)의 순서로 많이 분포하였다. Sarocladium kiliense NNIBRFG3280와 Fusicolla merismoides NNIBRFG23708를 국내 기수환경에서 발굴하였으며, 이를 국내 미기록종으로 보고하고자 한다.

이분지털곰팡이(털곰팡이목): 송이의 괴사성 균기생균 (Syzygites megalocarpus (Mucorales): A necrotrophic mycoparasite of Tricholoma matsutake)

  • 외생균근성 버섯인 송이는 동북아시아지역에서 가장 비싼 버섯이다. 이 버섯은 식용버섯이므로 자실체 표면에 병원균이 발생한다면 매우 심각한 문제가 아닐 수 없다. 송이 자실체의 기생균을 발견하였으므로 이에 대하여 자세히 설명하고 Syzygites megalocarpus Ehrenberg ex Fries로 동정하고 이분지털곰팡이로 명명하였음을 보고한다.

  • Rhinocerebral mucormycosis is a rare but acutely fatal disease caused by fungi belonging to the order Mucorales, and characterized by an aggressive necrotizing infection spreading from the nose to the paranasal sinuses, orbit, and then to the central nervous system. It most frequently develops in individuals with poorly controlled diabetes mellitus. The authors report a 65-year-old woman who developed intracerebral hemorrhage during treatment for rhinocerebral mucormycosis associated with diabetes mellitus. Despite the treatment, she became worse. The pertinent literatures were reviewed.

Phylogenetic Status of an Undiscovered Zygomycete Species, Syncephalastrum monosporum, in Korea

  • During a survey of undiscovered taxa in Korea, two zygomycete fungal isolates, EML-BT5-1 and EML-BT5-2, were isolated from the seed of a pumpkin (Cucurbita pepo) fruit in Korea. Based on their morphological characteristics and a sequence analysis of four genes, ITS1-5.8S-ITS2, 18S, 28S rDNA, and EF-$1{\alpha}$, the isolates were confirmed to be Syncephalastrum monosporum in the family Syncephalastraceae. To our knowledge, the zygomycete fungal species S. monosporum has not been previously described in Korea.
