Objectives In this study, the author intended to investigate whether Gami-cheongpetang (GCP), Gagam-jeongkitang (GJG), Gami-samooltang (GSM) and Gami-ijoongtang (GIJ) significantly affect in vivo (animal model) and in vitro (cultured cells) mucin secretion and MUC5AC gene expression in airway epithelial cells. Methods For in vivo experiment, the author induced hypersecretion of airway mucin in rats by introducing SO2 for 3 weeks. Enzyme-linked immunosorbent assay (ELISA) was used to assess the effects of orally-administered GCP, GJG, GSM and GIJ in vivo mucin secretion from tracheal goblet cells of rats after 1 week. Also, the effects of the agents on TNF- or EGF-induced MUC5AC gene expression in human airway epithelial cells (NCI-H292) were investigated. Possible cytotoxicities of the agents were assessed by examining the rate of survival and proliferation of NCI-H292 cells. Results (1) GCP and GJG significantly inhibited hypersecretion of in vivo mucin, although GSM and GIJ did not affect hypersecretion of in vivo mucin; (2) GCP and GJG significantly increased in vitro mucin secretion from cultured HTSE cells. However, GSM and GIJ did not affect in vitro mucin secretion from HTSE cells; (3) GCP and GJG significantly inhibited the expression levels of EGF-induced MUC5AC gene in NCI-H292 cells. However, GSM and GIJ increased the expression levels of EGF-induced MUC 5AC gene in NCI-H292 cells; (4) GCP, GJG, GSM and GIJ did not significantly inhibit the survival and proliferation of NCI-H292 cells. Conclusions These results suggest that GCP, GJG, GSM and GIJ can not only affect the secretion of mucin but also affect the expression of mucin gene. The author suggests that the effects of GCP, GJG, GSM and GIJ with their components should be further investigated by using animal experimental models that simulate the diverse pathophysiology of pulmonary diseases.
In the present study, the author tried to investigate whether four oriental medical prescriptions named, jawan-chihyosan (CHS), gwaru-jisiltang (GJT), and several single compounds, kaempferol, coumarin, betaine and ursolic acid significantly affect mucin release from cultured hamster tracheal surface epithelial (HTSE) cells. Confluent HTSE cells were metabolically radiolabeled with 3H-glucosamine for 24 hrs and chased for 30 min in the presence of CHS, GJT, kaempferol, coumarin, betaine and ursolic acid, respectively, to assess the effect of each agent on 3H-mucin release. Possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase(LDH) release. Additionally, total elution profiles of control spent media and treatment sample (CHS and GJT) through Sepharose CL-4B column were analysed and effect of CHS and GJT on MUC5AC mRNA expression in cultured HTSE cells were investigated. The results were as follows : (1) CHS and GJT significantly stimulated mucin release from cultured HTSE cells, with significant cytotoxicity , (2) CHS and GJT chiefly stimulated the 'mucin' release and did not affect significantly the release of the other releasable glycoproteins with less molecular weight than mucin. This result suggests that the three herbal prescriptions specifically stimulate the release of mucin ; (3) CHS and GJT significantly increased the expression levels of MUC 5AC mRNA. This result suggests that the three herbal prescriptions can affect the synthesis of mucin at gene level in cultured HTSE cells ; (4) Kaempferol and coumarin did not affect mucin release, however, betaine and ursolic acid stimulated mucin release. All the agents did not show significant cytotoxicity. We suggest that the effects of CHS and GJT, betaine and ursolic acid should be further investigated and it is of great value to find, from oriental medical prescriptions, novel agents which have the effective expectorant or mucoregulative effect on mucin secretion from airway goblet cells.

Confluent HTSE cells were metabolically radiolabeled with ³H-glucosamine for 24 hrs and chased for 30 min in the presence of SCRT or KCHS to assess the effect of each agent on ³H-mucin release. Possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase(LDH) release. Also, the effects of SCRT and KCHS on contractility of isolated tracheal smooth muscle were investigated. The results were as follows: (1) SCRT significantly inhibited mucin release from cultured HTSE cells, without cytotoxicity; (2) KCHS significantly increased mucin release without cytotoxicity; (3) SCRT and KCHS did not affect contractility of isolated tracheal smooth muscle. We suggest that the effects of SCRT and its components should be further investigated and it is of great value to find, from oriental medical prescriptions, novel agents which have the possible inhibitory effects on mucin release from the viewpoint of management of hypersecretion of airway mucus.
In the present study, we tried to investigate whether protein kinase C (PKC) activator, phorbol 12-Myristate 13-Acetate (PMA), and PKC inhibitors, $G\"{o}-6976$, Ro-31-8220 and rottlerin significantly affect basal mucin relesed from cultured airway goblet cells. Confluent primary hamster tracheal surface epithelial (HTSE) cells were metabolically radiolabeled with $^3H$-glucosamine for 24 hr and chased for 30 min in the presence of each agent to assess the effects on $^3H$-mucin release. The results were as follows: (1) PMA increased mucin release from cultured HTSE cells, during 30 min of treatment period; (2) However, $G\"{o}-6976$, Ro-31-8220 and rottlerin did not significantly affect mucin release, during 30 min of treatment period. This finding suggests, at least in part, that PKC might playa minor role in the signaling pathways involved in basal - physiological or constitutive - mucin release from airway goblet cells, although further studies are needed.
In the present study, we tried to investigate whether polymerized basic amino acid e.g. poly-L-lysine (PLL) which has the degree of polymerization under 50mer significantly affects the physiological and stimulated mucin release from cultured hamster tracheal surface epithelial cells. Confluent primary hamster tracheal surface epithelial (HTSE) cells were metabolically radiolabeled with $^3{H}$-glucosamine for 24 hr and chased for 30 min in the presence of either PLLs or adenosine triphosphate (ATP) and PLL to assess the effects on basic or ATP-stimulated $^3{H}$-mucin release. Possible cytotoxicities of PLLs were assessed by measuring lactate dehydrogenase (LDH) release from HTSE cel1s during treatment. The results were as follows: PLLs significantly inhibited basic mucin release from cultured HTSE cells in a dose-dependent manner from the range of 46mer to 14mer; PLL 46mer significantly inhibited the stimulated mucin release by ATP from cultured HTSE cells; there was no significant release of LDH from cultured HTSE cells during treatment. We conclude that PLLs inhibit both physiological and stimulated mucin release from airway epithelial cells without significant cytotoxicity and PLL lost its activity under the range of 14mer. This finding suggests that polymer of basic amino acid like PLL might function as a regulator for hypersecretion of mucus manifested in various respiratory diseases.
In the present study, we investigated whether quercitrin, quercetin and afzelin derived from Houttuynia cordata affect the production and gene expression of MUC5AC mucin from airway epithelial cells. Confluent NCI-H292 cells were pretreated with quercitrin, quercetin or afzelin for 30 min and then stimulated with epidermal growth factor (EGF) or phorbol 12-myristate 13-acetate (PMA) for 24 h. The MUC5AC mucin gene expression and production were measured by RT-PCR and ELISA, respectively. The results were as follows: (1) Quercitrin, quercetin and afzelin inhibited EGF- and PMA-induced MUC5AC mucin production from NCI-H292 cells; (2) The three natural products also decreased EGF- and PMA-induced MUC5AC mucin gene expression in NCI-H292 cells. These results suggest that quercitrin, quercetin and afzelin showed the regulatory effect on the steps of gene expression and production of mucin, by directly acting on airway epithelial cells.
Microbial dysbiosis in the gut is associated with human diseases, and variations in mucus alter gut microbiota. Therefore, we explored the effects of mucin on the gut microbiota using a community of 19 synthetic gut microbial species. Cultivation of these species in modified Gifu anaerobic medium (GAM) supplemented with mucin before synthetic community assembly facilitated substantial growth of the Bacteroides, Akkermansia, and Clostridium genera. The results of 16S rRNA microbial relative abundance profiling revealed more of the beneficial microbes Collinsella, Bifidobacterium, Ruminococcus, and Lactobacillus. This increased acetate levels in the community cultivated with, rather than without (control), mucin. We identified differences in predicted cell function and metabolism between microbes cultivated in GAM with and without mucin. Mucin not only changed the composition of the gut microbial community, but also modulated metabolic functions, indicating that it could help to modulate microbial changes associated with human diseases.

(1) GSGT did not affect mucin release without cytotoxicity ; (2) GCPT significantly stimulated mucin release from cultured HTSE cells, with significant cytotoxicity ; (3) GSGT and GCPT did not affect contractility of isolated tracheal smooth muscle. We suggest that the effects of GCPT and its components should be further investigated and it is of great value to find, from oriental medical prescriptions, novel agents which have potent expectorant effects on mucin secretion from airway goblet cells.
경골어류 4종(조피볼락, 용치놀래기, 송곳니베도라치 및 졸복)의 장관 상피 선조연 및 배상세포의 점액질의 조직화학적 성상을 밝히기 위해 PAS 반응, AB pH 1.0 및 pH 2.5, AB pH 2.5-PAS, AF pH 1.7-AB pH 2.5 및 HID-AB pH 2.5 염색법을 사용하였다. 장관 선조연의 점액질은 조피볼락의 근위장과 직장은 중성점액질만을, 중간장과 원위장은 중성점액질과 산성점액질의 혼합성이었으나 용치놀래기는 전 장관에서 중성점액질과 산성점액질을 함유하고 있었으며, 송곳니베도라치와 졸복의 전 장관은 중성점액질만을 함유하고 있었다. 중성점액질의 양은 조피볼락과 용치놀래기는 중등량 내지 상당량, 송곳니베도라치와 졸복은 미량 내지 소량이었다. 장 배상세포 점액질의 양과 성상은 어종 및 장 부위에 따라 차이가 있어 송곳니베도라치와 졸복은 중성점액질만을 함유하고 조피볼락과 용치놀래기는 중성 점액질, sulfomucin 및 sialomucin의 혼합성이었다. 중성점액질의 양은 졸복의 원위장 및 직장은 상당량 내지 다량이었고 조피볼락의 전장, 용치놀래기의 근위장, 원위장 및 직장, 졸복의 근위장 및 중간장은 중등량 내지 상당량이었으며 용치놀래기의 중간장은 미량 내지 소량이었다. 조피볼락의 장 배상세포는 미량의 강 sulfomucin, 약 sulfomucin 및 미량 내지 소량의 sialomucin을 함유하고 있었으며 직장을 제외한 용치놀래기의 장 배상세포는 미량 내지 소량의 강 sulfomucin과 sialomucin을 함유하고 있었다.
농어목에 속하는 경골어류 4종(노래미, 쑤기미, 구실우럭 및 홍감펭)의 장관 점액질의 조직화학적 성상을 밝히기 위해 peridic acid Schiff (PAS) 반응, alcialn blue (AB) pH 1.0 및 pH 2.5, AB pH 2.5-PAS, aldehyde fuchsin (AF) pH 1.7-AB pH 2.5 및 high iron diamine (HID)-AB pH 2.5 염색을 실시하였다. 장 점막에 있는 주름의 모양은 어종 및 부위에 따라 차이가 있었다. 장의 모든 부위에서 관찰되는 배상세포는 원주세포사이에 위치하고 있으며 그 모양은 구형이거나 타원형이었고 원위장에서 수가 가장 많았다. 구실우럭 중간장 및 원위장과 노래미, 쑤기미 및 홍감펭 장에는 산성점액질과 중성점액질이 나타났으며 산성점액질의 성상은 sialomucin이었다. 구실우럭의 근위장에는 중성점액질만 나타났다. 점액질 양과 성상은 장 부위에 따라 차이가 있었다. 쑤기미의 원위장은 산성점액질과 중성점액질 양이 비슷하였다. 노래미의 장관 모든 부위, 쑤기미의 근위장과 중간장, 구실우럭의 중간장과 원위장, 홍감펭의 원위장에는 산성점액질이 중성점액질보다 더 많았다. 홍감펭의 근위장과 중간장에는 중성점액질이 산성점액질보다 더 많았다.
