Improvement of Ergone Production from Mycelial Culture of Polyporus umbellatus

  • Ergone, a fungal metabolite derived from ergosterol, was previously isolated and identified from Polyporus umbellatus. Ergone is a major component of P. umbellatus known to have anti-aldosteronic diuretic effect and also displays cytotoxic activities. Most of mushroom's fruit bodies used for test contained less than 10 ${\mu}g/g$ of ergone. But P. umbellatus have larger amount of ergone than any other mushrooms. In order to improve the ergone production from the submerged culture of P. umbellatus, several factors including medium composition, culture conditions (temperature and pH) and different combinations of co-cultivation with various mycelia were studied. Among various carbon sources examined, starch proved to be most effective for the production of mycelia. The optimum pH and temperature for a flask culture of P. umbellatus mycelia were found to be 4.5 and $25^{\circ}C$, respectively. Under the optimized culture conditions, both the ergone production (86.9 ${\mu}g/g$) and mycelial growth (3.5 g/l) increased when P. umbellatus was cultured with Armillariella mellea. When the optimized conditions were applied, both mycelium and ergone production were significantly enhanced.

분말 성형체로부터 양축정렬 집합조직을 갖는 니켈 테이프의 제조 (Fabrication of Biaxially Textured Ni Tapes from Ni Powder Compact Rods)

  • Biaxially textured Ni tapes were fabricated by a cold working and recrystallization heat treatment processes from powder compact rods. The processing parameters associated with the cube texture formation in Ni tapes were systematically investigated by using X-ray diffraction and pole-figure analysis. The Ni powder used in this study was 5 $\mu$m in size and 99.99% in purity. To find the optimum sintering temperature, tensile tests were performed for Ni rods sintered at various temperatures. The Ni rods sintered at 100$0^{\circ}C$ showed poor elongation and low fracture strength, while the Wi rods sintered above 100$0^{\circ}C$ revealed good mechanical properties. The higher elongation and fracture strength of the Ni rods sintered at higher temperatures than 100$0^{\circ}C$ are attributed to the full densification of the sintered rods. The sintered Ni rods were cold-rolled with 5% reduction to the final thickness of 100 $\mu$m and then annealed for development of rube texture in rolled Ni tapes. The annealed Ni tapes depicted strong cube texture with FWHM(full-width at half-maximum) of in-plane and out-of-plane in the range of 8$^{\circ}$ to 10$^{\circ}$. The NiO deposited on the Ni tapes by MOCVD process showed good epitaxy with FWHM=10$^{\circ}$, which indicates that the Ni tapes can be used as a substrate for YBCO coated conductors.

대구지역(大邱地域) 지각(地殼) ${\gamma}$-선(線)의 조사선량율(照射線量率) 측정(測定) (A Measurement of the Exposure Rates by Terrestrial y-rays in Taegu Area)

  • 본 연구는 대구지역의 지각 ${\gamma}$-선에 의한 조사선량율을 측정하고 그에 따른 성분별 공간선량율 및 지역적 분포 등의 환경방사선 특성을 조사하였다. ${\gamma}$-선 스펙트럼의 측정에는 $4^{'}{\phi}{\times}4^{'}$ NaI(T1) 계측기와 휴대용 다중파고분석장치를 이용하여 이 지역내의 28개 지점에서 in-situ spectrometry를 수행하였으며, 측정된 스펙트럼으로부터 조사선량율을 환산하여 지각 ${\gamma}$-선의 총선량율과 일반적으로 자연방사능의 주종이 되는 $^{40}K,\;^{238}U$ 계열 및 $^{232}Th$계열의 성분별 조사 선량율을 구하였다. 조사결과 대구지역의 지각 ${\gamma}$-선에 의한 평균조사선량율은 $9.4{\mu}R/h$였으며 지점별 분포는 $7.6{\sim}11.0{\mu}R/h$ 범위로 다소 차이를 보였다. 측정시에 수반되는 기후나 우주선 등에 따른 일 변화폭을 고려한다면 지점간의 차이는 주로 표토층의 자연방사성핵종인 $^{40}K,\;^{238}U$계열 및 $^{232}Th$ 계열의 핵종에 기인함을 알 수 있었으며, 성분별 선량율은 $^{40}K\;2.9{\sim}4.6{\mu}R/h,\;^{238}U$ 계열 $1.2{\sim}3,\;1{\mu}R/h,\;^{232}Th$ 계열 $2.5{\sim}5.0{\mu}R/h$의 분포를 나타냈다.

섬오갈피나무 부정근(不定根) 배양(培養)에서 부정근(不定根)의 생장(生長)과 Eleutheroside 류(類)의 생산(生産)에 미치는 Jasmonic acid 처리(處理)의 영향 (Effects of Jasmonic Acid on Root Growth and Eleutheroside Accumulation in Adventitious Root Culture of Eleutherococcus koreanum)

  • 생물반응기를 이용한 섬오갈피 부정근 배양시에 jasmonic acid를 농도별 (0, 0.01, 0.05, 0.1, 0.2, 0.4 mg/L)로 처리하여 부정근의 생장과 eleutheroside류 생산과의 관계를 조사하였다. Jasmonic acid 처리농도별 부정근의 생장은 무처리구에서 2.9 g Dry Weight/L로 가장 높았으며 jasmonic acid 농도가 증가할수록 부정근 생장은 감소하였다. 그러나 eleutheroside B와 E의 함량은 jasmonic acid 농도가 높을수록 증가하여 eleutheroside B는 0.4 mg/L처리구에서 $49.0{\mu}g/g\;DW$, eleutheroside E는 0.2 mg/L처리구에서 $64.7{\mu}g/g\;DW$로 가장 생산량이 많았다. 반면에 eleutheroside $E_1$은 무처리구에 $19.0{\mu}g/g\;DW$로 가장 생산량이 많았다. 배지 lL당 eleutheroside류의 총생산량은 0.01 mg/L처리구에서 $259.9{\mu}g/L$를 생산하여 가장 우수하였다. 0.01 mg/L의 jasmonic acid 처리 후 12일간 eleutheroside류의 함량을 조사한 결과 eleutheroside B는 jasmonic acid처리 후 감소하였으나, eleutheroside E는 처리 후 10일째, eleutheroside $E_1$은 처리 후 4일째 가장 많은 생산량을 보여 주었다.

주암댐 호저 퇴적물에서의 수직적 중금속 분산과 $^{210}Pb$를 이용한 퇴적속도산정 (Vertical Distribution of Heavy Metal Concentrations in Sediment Cores and Sedimentation Rate Using $^{210}Pb$ Dating Technique in the Juam Reservoir)

  • 주암댐에 유입된 호저퇴적물의 입도(2mm-200{\mu}m,\;200-100{\mu}m,\;100-50{\mu}m,\;50-20{\mu}m,\;<20{\mu}m)별 중금속(As, Cd, Cr, Cu, Ni, Pb, Zn)의 이동과 퇴적 후 중금속의 수직적 분산 특성을 규명하기 위하여 12개 지점의 호저퇴적물과 3개 지점의 코아시료를 채취하였으며, 코아시료는 2-5 cm 간격으로 절단하여 깊이별 간극수와 퇴적물의 중금속 함량을 측정하였다. 호저퇴적물의 퇴적속도를 측정하기 위하여 코아시료에 대한 $^{210}Pb$ 동위원소 연구를 수행하였다. 주암댐 호저 퇴적물의 중금속 및 미량원소의 함량은 퇴적위치와 상관없이 거의 일정한 함량을 보이며.,평균 함량은 $As\;14.9\;{\mu}g/g,\;Cd\;0.81\;{\mu}g/g,\;Cu\;30.7\;\;{\mu}g/g,\;Ni\;34.7\;{\mu}g/g,\;Pb\;63.3\;{\mu}g/g,\;Zn\;87.9\;{\mu}g/g$이다. Cu, Pb, Zn, Ni 및 Cd의 함량은 $20\;{\mu}m$ 이하 입도의 퇴적물에서 가장 높은 반면, As는 $2mm-200{\mu}m$ 혹은 $200-100\;{\mu}m$ 입도의 퇴적물에서 가장 높다. 간극수에 용해된 원소 중에서 가장 함량이 높은 것은 Fe와 Mn 이었으며, 코아시료의 상부로부터 심부로 갈수록 함량이 증가하는 수직적 분산특성을 나타낸다. 반면에 용존 Zn과 Cu함량은 코아시료의 최상부에서 가장 높게 나타나며, 이는 산화:환원 전위차에 따른 유기물질 및 산화광물의 용해작용과 관련이 있다. 코아시료의 퇴적물 내 Cu, Ni, Pb 및 Zn 함량은 0-4 cm구간에서 가장 높은 표면집적 현상이 관찰되며, Pb와 Cu 함량이 현저하게 높은 데, 이는 인위적인 오염에 기인하는 것으로 해석된다. $'unsupponed\;^{210}Pb'$을 이용하여 계산된 호저퇴적물의 퇴적 속도는 $\omega=0.91\;cm\;year^{-1}$ 이며, 이를 적용하면 주암댐이 완공된 이후 약 10 cm의 퇴적물이 평균적으로 퇴적된 것으로 추정된다. 이러한 자료는 주암댐 호수 바닥에 퇴적된 퇴적물의 관리방안을 마련하는 데 중요한 정보로 사용될 수 있을 것이다.

  • This paper described design as a purposeful human activity for solving its problems. This means, problem solving is finding a way to get from some initial situation to a desired goal.In order to do this, like a voyage, there should be a compass, and a route map to guide the designer through the design process. Evaluation gives information about the way a design is proceeding and suggests the direction in which change should be made in order that the complex of design concepts should be fulfilled in a satisfactory manner.I think Pugh's total design model discussing in this paper takes a layered approach through a series of sequential actions, part of which is to find the product status. Each layer has the $$\mu$ti-dimensional elements, Product Design Specification (PDS) around the design core. I made a comparative study of several evaluation tools wi th Pugh's model and examined the criteria for evaluation and the rating / weighing methods.It needs, however, more study in order that PDSs may have its values and be fixed from the dynamic design concepts.

Time Resolced Molecular Beam Characteristic in a Pulsed Supersonic Jet

  • A pulsed molecular beam source having short pulse duration (typically 70 ${\mu}s)$ and narrow velocity distribution (${\Delta}$v/v=8% for helium) has been costructed utilizing a commercial fuel injector. Beam characteristics of helium and ammonia seeded in helium expansions are accomplished by the use of an electron impact time-or-flight mass spectrometer. The comparisons between experimental data and theoretical calculations show that the proper beam speed is important to predict the evolution of stream temperature and valve shutter function. The decreasing tendency of pulse duration with increasing cluster size leads to the conclusion that the cluster beam property is described as a function of cluster mass and disinct cluster temperature.

  • Cadmium (Cd) is an inorganic toxicant of great environmental and occupational concern which was classified as a human carcinogen in 1993. Occupational cadmium exposure is associated with lung cancer in human. In the present study, we established the mechanistic basis of apoptotic cell death induced by Cd in WI38 human lung fibroblast. Cd at 20-80 ${\mu}$M decreased viablility of cells in a concentration-dependent manner. PI staining, TUNEL staining and DNA fragmentation analysis demonstrated the apoptotic cell death by Cd. The principle of treatment in the previous medicine is to treat each disease by 'Assisting-Good Qi' and 'Removig-Bad Qi'. but The principle of treatment in Sasang Constitutional medicine is to manage incurable disease by helping 'Essential Qi of each constitution(體質正氣)' 2. Incurable disease is classified into a chronic disease by 'Nature(性氣)' and a acute disease by 'Emotion(情慾)'. Both diseases became serious through 'Noi-Ok(牢獄)' and 'Wi-Gyoung(危傾)'. A chronic disease is much in the middle years of life and become senile disease. A acute disease is much in the young years of life and make patients die young. 3. prognoses of incurable disease are different from degree of Inherent vitality(命脈實數) and term of disease. The case in which Inherent vitality is exhausted is thought that is unable to treat. 4. The prevention of incurable disease Is more important the treatment of one in Sasang Constitutional Medicine. but if incurable disease is caught, Medicine(醫藥) and management(調養) must be used together for treatment of incurable disease. Medicine is more important in the level of 'Noi-Ok(牢獄)' and management is more important in the level of 'Wi-Gyoung(危傾)'. 5. Therefore, incurable disease should be treated by method that 'Essential Qi of each constitution(體質正氣)' is recovered and declination is removed through 'controlling mind(治心) and correcting Qi(正氣), so then the state of 'Golden mean(中庸)' is reached.
